The photolysis of colchicine under ultraviolet and visible light irradiation was studied by ultraviolet (UV) scanning and HPLC-MS. The photoproduct was proposed and the cytotoxicity change before and after irradiation was investigated. Results showed that both ultraviolet and visible light irradiation could effectively degrade colchicine into deacetamido-lumicolchicine. The process conformed to first-order kinetics, in which a high degradation rate (K = 0.5862 h − 1 ) was observed when colchicine was dissolved in ethanol and irradiated by UV light. Cell viability and cell cycle studies proved that a photolysis treatment of colchicine could weaken the cytotoxicity effectively. Colchicine inhibited the division of BRL 3 A cells in G2/M phase with an IC 50 value of 0.48 µg/mL, while the toxic effect could be reduced significantly with IC 50 2.1 µg/mL when colchicine was exposed to UV irradiation. Results are beneficial to the toxicity elimination of colchicine in the processing of daylily flower in food industry, and can also provide photochemistry reference for colchicine-related studies.

Hemerocallis citrina Baroni ( H. citrina Baroni) is widely used in the food and medical industries. However, a comprehensive evaluation of the physicochemical, rheological properties, and hypoglycemic activities of its various parts has not yet been reported. In this study, we extracted polysaccharides from the leaves (HCLP) and stems (HCSP) of H. citrina Baroni. The physicochemical properties were investigated using high-performance gel permeation chromatography (HPGPC), high performance liquid chromatography (HPLC), Congo red experiment, Fourier transform infrared spectroscopy (FTIR), ultraviolet-visible spectroscopy (UV-Vis), X-ray diffraction (XRD), thermogravimetric (TG), and scanning electron microscopy (SEM). HCSP exhibited higher total sugar contents, uronic acids, protein, total phenol, and flavonoid contents than HCLP. However, the HCSP extraction yield (2.54 ± 0.68%) was lower than that of HCLP (6.34 ± 0.75%). Both polysaccharides were mainly composed of Mannose (Man) and galactose (Gal), but their contents varied significantly; HCSP contained more glucosamine (GlcN), rhamnose (Rha), arabinose (Ara), gluconic acid (GlcA), galacturonic acid (GalA) and glucose (Glc) than HCLP. The results showed that both HCSP and HCLP displayed macromolecular characteristics with a triple-helix structure. The thermal stability was stronger in HCLP than in HCSP. The rheological results showed that HCLP and HCSP exhibited a pseudoplastic fluid behavior. Compared to HCSP, HCLP had a stronger ability to inhibit α -glucosidase and enhance glucose consumption in HepG2 cells, attributed to its superior thermal stability and higher molecular weight ( M w ). These results indicated that polysaccharides from the leaves and stems of H. citrina Baroni have potential as functional foods and hypoglycemic drugs.

Introduction As a famous traditional Chinese medicine, roots of Platycodon grandiflorus (Jacq.) A.DC. have shown multiple effects against neurodegenerative diseases. To investigate the components against Parkinson's disease (PD), the roots of P. grandiflora were selected as the research subject. Objective Screening and identifying of monoamine oxidase B (MAO-B) inhibitors from the roots of P. grandiflorum via enzyme functionalised magnetic nanoparticles (MNPs)-based ligand fishing combined with high-performance liquid chromatography–mass spectrometry (HPLC–MS) analysis. Method MAO-B functionalised MNPs have been synthesised for screening MAO-B inhibitors from the roots of P. grandiflorum . The ligands were identified by HPLC–MS and nuclear magnetic resonance (NMR) analysis, and their anti-PD activity was evaluated via MAO-B inhibition assay and cell viability assay in vitro . Results Two MAO-B inhibitors were fished out and identified by HPLC–MS as protocatechuic aldehyde ( 1 ) and coumarin ( 2 ), with the half maximal inhibitory concentrations of 28.54 ± 0.39 and 25.39 ± 0.29 μM, respectively. Among them, 1 could also significantly increase the viability of 6-hydroxydopamine-damaged PC12 cells. Conclusion The results are helpful to elucidate the anti-PD activity of the plant, and the ligand fishing method has shown good potential in discovery of MAO-B inhibitors.

Hemerocallis citrina Baroni is a healthy vegetable, and consumption of fresh H. citrina can lead to defecation. However, the laxative ingredients are still not known. In the present study, the laxative effect of fresh H. citrina extracts was primarily evaluated by loperamide-induced constipated mice, and the aqueous extracts (1500 and 3000 mg/kg) showed a significant laxative effect. The aqueous extracts were divided into three parts (0%, 30%, and 50% HCW) by macroreticular resin, and all of them were further evaluated. Based on comprehensive analysis of multiple indices, the constipation-relieving effect of 30% HCW was found to be superior to that of 0% and 50% HCW. A total of 16 compounds were identified from 30% HCW by the ultra-performance liquid chromatography-quadrupole-time of flight-mass spectrometry (UPLC-Q-TOF-MS) method. Three high-level ingredients including chlorogenic acid (3), 4- O -p-coumaroylquinic acid (6) and rutin (13) were selected to further evaluate their laxative effect. The experimental results demonstrate that 4- O -p-coumaroylquinic acid increases defecation frequency in constipated mice, enhances fecal moisture content, and promotes small intestinal motility in constipated mice. The high-dose 4- O -p-coumaroylquinic acid treatment group exhibited higher mean small intestinal transit rates compared to the high-dose chlorogenic acid and rutin groups. Additionally, this compound effectively alleviated loperamide-induced pathological damage, including thinning of the muscle layer and reduced goblet cell numbers, thereby enhancing small intestinal motility, increasing fecal moisture content, and facilitating defecation. This is the first study to identify 4- O -p-coumaroylquinic acid as a key laxative component in fresh H. citrina Baroni, providing novel insights into its application in functional food or pharmaceutical development.

Background Depression is closely associated with dysfunction of the microbiota-gut-brain axis (MGBA). Total phenol from Hemerocallis citrina leaves (HLTP) exhibits antidepressant potential, but its role in relieving depression by regulating MGBA remains uninvestigated. Methods In this study, the chronic unpredictable mild stress (CUMS) method was used to establish a rat model of depression to investigate the ameliorative effects of HLTP on depression. Results Behavioral and pathological results showed HLTP improved weight loss, increased sucrose preference, reduced forced swimming immobility time, novel-suppressed feeding latency, open field movement trajectory, and alleviated hippocampal CA1 neuronal damage in CUMS-depressed rats. At 10, 20, and 40 mg/kg, HLTP increased dopamine (DA), noradrenaline (NE), 5-hydroxytryptamine (5-HT), interleukin-10 (IL-10), brain-derived neurotrophic factor (BDNF), and cAMP response element-binding protein (CREB) in the brain. It also decreased the inflammatory factor (TNF-α and IL-1β) in the brain, as well as corticotropin-releasing hormone (CRH), adrenocorticotropic hormone (ACTH), and corticosterone (CORT) in serum and brain. HLTP modulated gut microbiota diversity and reversed CUMS-induced alterations in 15 bacterial taxa, including Alloprevotella and Monoglobus . Additionally, HLTP reversed 63 CUMS-induced abnormal serum metabolites, primarily affecting tryptophan metabolism, niacin and nicotinamide metabolism, and porphyrin metabolism. Significant correlations were observed among gut microbial composition, serum metabolite levels, biochemical indices, and behavioral outcomes. Conclusion HLTP exerts antidepressant effects in CUMS-induced rats by modulating the MGBA. It enhances metabolic function, reduces neuroinflammatory damage, restores neurotrophic factor levels, suppresses hyperactivity of the hypothalamic-pituitary-adrenal axis, and corrects monoamine neurotransmitter deficiencies, collectively contributing to the attenuation of depression-related symptoms.

Hemerocallis citrina Baroni (HC) is usually soaked with sodium metabisulfite solution after harvest to keep its freshness. However, this will introduce bound sulfites with potential safety risks. In this study, formation pathways of bound sulfite in HC were explored using ultra-high performance liquid chromatography-Q-Exactive HF mass spectrometry (UHPLC-QE-MS) and statistical analysis. The results indicated that the chemical composition of HC changed the most at soaking times of 6–8 h and concentrations of 1.00–2.50 mg/mL. Primary metabolites and phenols in secondary metabolites were greatly affected during soaking. 71 compounds were identified in HC and 8 characteristic markers including 3 bound sulfites were screened which can be used to distinguish between non-soaked HC (NSHC) and sodium metabisulfite-soaked HC (SSHC). Among the 19 commercial HC samples analyzed, 79 % had been treated with sulfites. Mass spectrometry data further revealed two pathways of formation of bound sulfite in HC: one is the sulfonation of amino acids at the amino, the content initially increased with sulfite soaking time and concentration and then decreased until it disappeared; the other is that the addition of aldehyde at the carbonyl, the content increased continuously with sulfite soaking concentration. This study provides insight into bound sulfite formation in HC post‑sodium metabisulfite treatment.

The crude polysaccharides CAPS and CAP of Cynanchum Auriculatum , which were prepared by degrading starch by single-enzymatic method (α-amylase) and double-enzymatic method (α-amylase and glucoamylase) respectively, were compared. CAP had good water solubility and higher non-starch polysaccharide content. A homogeneous neutral polysaccharide CAP-W, with the degree of acetylation about 17 %, was obtained from CAP by anion exchange column chromatography. Its detailed structure was identified by various methods. CAP-W, with the weight average molecular weight of 8.4 kDa, was composed of mannose, glucose, galactose, xylose, and arabinose in a molar ratio of 1.27:1.00:0.25:0.10:1.16. The backbone included β -1,4-Man p , β -1,4,6-Man p , β -1,4-Glc p and β -1,4,6-Glc p residues, with branches at the O-6 position of β -1,4,6-Man p and β -1,4,6-Glc p residues, consisting of α -T-Ara f , α -1,5-Ara f , α -1,2,5-Ara f , α -1,3,5-Ara f , T-Xyl p ,1,4-Xyl p , β -T-Man p and β -T-Gal p residues. In vitro immunological experiments suggested that CAP-W improved the phagocytic ability of macrophages, stimulated the release of NO, TNF-α and IL-6 from RAW264.7 cells, promoted the expression of NF-κB and caused nuclear translocation of NF-κB p65.

Neurological injury, as a major pathogenic mechanism in depression, holds significant importance in the research and development of antidepressant drugs. Hemerocallis citrina Baroni (H. citrina), referred to as “Forgetting Sadness Grass,” has been confirmed to possess remarkable neuroprotective effects. Studies have identified that the total phenolics in H. citrina Baroni leaves (HLTP) consist of flavonoids and phenolic acids and numerous studies have substantiated the neuroprotective effects of them. Based on this, we propose that HLTP may possess neuroprotective properties. To confirm this hypothesis, we initially employed network pharmacology techniques to predict potential targets for the neuroprotective effects of HLTP based on the Swiss Target Prediction database. GO and KEGG analyses were conducted to predict potential pathways, and a component-target-pathway network was constructed. Molecular docking experiments were then performed to analyze the binding abilities of the selected active components with the main targets. Furthermore, we validated the neuroprotective effects of HLTP and key targets selected through network pharmacology using a corticosterone-induced PC12 neuronal cell damage model. Network pharmacology research has identified that in the HLTP, Quercetin, Rutin, Apigenin, and Isoquercitrin are potential active components that may exert neuroprotective effects by modulating key targets such as AKT1, TNF, TP53, and CASP3 through crucial pathways including PI3K/AKT and apoptosis. Molecular docking revealed that 4-O-Caffeoylquinic acid, 5-O-Caffeoylshikimic acid, 4-p-Coumaroylquinic acid, and 5-O-Feruloylquinic acid exhibit low binding energies with key targets. Particularly, 4-O-Caffeoylquinic acid forms stable binding through hydrogen bonding with residues such as LYS389, GLU49, GLN47, LYS30, ASP44, and GLU40 in AKT1. PC12 cells were stimulated with 200 μmol/L Corticosterone (Cort) for 24 h, and then treated with 50, 100 and 200 μg/mL of HLTP for 24 h. The cell viability of damaged cells were significantly increased in a dose-dependent manner by 9.50%, 10.42% and 21.25%, respectively (P < 0.01). Western blot analysis confirmed that HLTP significantly (P < 0.01) increased the protein expression of PI3K and AKT by 15.24%, 30.44%, 41.03%, and 21.78%, 43.63%, 12.86%, respectively. In addition, through biochemical method, flow cytometry and WB analysis, we found that different concentrations of HLTP can all improve cell damage by reducing ROS, MDA, Ca2+, Cyt-C, Caspase-3, TNF-α and IL-1β, and increasing SOD, CAT, MMP, Bcl-2/Bax and IL-10. In particular, the HLTP at 200 μg/mL, compared with the Model group, decreased by 140.2%, 54.66%, 51.34%, 65.26%, 40.32%, 63.87%, and 55.38%, and increased by 39.65%, 35.45%, 38.38%, 28.54%, and 39.98%, respectively. Through the above experiments, we verified that HLTP may exert neuroprotective effects by mediating the PI3K/AKT signaling pathway to counteract oxidative stress damage, improve mitochondrial dysfunction, and alleviate inflammatory injury.
