Seasonal dynamics of total flavonoid contents and antioxidant activity of Dryopteris erythrosora

Yinghua Xie,Yunxia Zheng,Xiling Dai,Quanxi Wang,Jianguo Cao,Jianbo Xiao

The seasonal dynamics of the total flavonoid contents in various parts of Dryopteris erythrosora , a traditional Chinese medicinal fern, and their antioxidant activity were investigated. The total flavonoids content in various parts of D. erythrosora showed an obvious seasonal dynamic change. The total flavonoid contents in stems (from 4.3% to 12.5%) were much higher than that in leaves with an average content of 2.01%. In spring, the total flavonoid contents in stems were relatively low, but increased rapidly from summer to winter. However, the seasonal dynamics of total flavonoid contents in leaves showed different model. The total flavonoid contents in the stems showed a negative correlation with that in the leaves from January to July. The correlation coefficient of about −0.7 was obtained. The antioxidant activity of the extracts also altered in proportion to the change of total flavonoid contents. In general, the extracts from stems always showed highest antioxidant potentials and it was suggested that the stems can be used as crude medicine.

Physicochemical and Antioxidant Properties of Four Bee Honey Types and Their Anticancer Effects on Hep-2 Human Laryngeal Cancer Cells

Walaa Kobacy,Ahmed K. Rashwan,Ahmed I. Osman,Ashraf K. Abu-Zaid,Ahmed R. Mazeed,Reda M. Mohamed

This study investigates the physicochemical, antioxidant, and anticancer properties of four Egyptian honey types: clover, fennel, sidr, and multiflower. These honeys exhibited significant variability in color, pH, acidity, sugars, and enzymatic activity. The fennel honey demonstrated the highest content of total phenolic (TPC) of 238.76 mg GAE/100 g, and the total flavonoid (TFC) was 94.06 mg QE/100 g. In addition, the antioxidant activity as measured by DPPH radical scavenging correlated strongly with bioactive compound concentrations, with fennel honey showing the highest inhibition (60.14%). In vitro cytotoxicity assays against Hep-2 human laryngeal cancer cells revealed fennel honey to have the lowest IC50 value (70 μg/mL), followed by multiflower honey (IC50 = 87 μg/mL), confirming a significant relationship between the TPC, TFC, and the anticancer effects. This highlights the role of bioactive compounds in honey as potential natural anticancer agents. Clover honey exhibited the weakest cytotoxicity (IC50 = 130 μg/mL) with the lowest TPC and TFC. These findings underscore the potential of Egyptian honey varieties, particularly fennel and multiflower honeys, as promising natural sources of antioxidants and anticancer agents, suggesting a beneficial therapeutic role in cancer prevention. Further studies are needed to elucidate the detailed mechanisms by which honey compounds exert these anticancer effects.

Quality differences in sea buckthorn (Hippophaë rhamnoides L.) berries of major varieties in China based on key components and antioxidant activity

Yaping Zheng,Kangmeng Sun,Xinyuan Sun,Yue Li,Peigen Xiao,Chunnian He

Sea buckthorn is rich in active ingredients, widespread, and has both medicinal and nutritional value. The present comparative study of wild and cultivated species remains insufficient, which is not conducive to their quality control. Therefore, this study aimed to compare the differences of 21 sea buckthorn samples in total phenolic content (TPC), total flavonoid content (TFC), phenolic components content, secondary metabolites, and antioxidant capacity and the fatty acid, to investigate the quality differences of different varieties. The TPC, TFC and antioxidant activity of wild varieties were higher than those of the cultivated. Multivariate statistical analysis revealed large differences in phenolic content, with higher levels of gallic acid and isorhamnetin-3- O -neohesperidin in the wild, whereas the cultivated were characterized by narcissin and kaempferol. These findings provided the scientific basis for the improvement of quality evaluation standards for different varieties and offered new insights for the further development of sea buckthorn resources.

Antioxidant activity of the different polar solvent extracts of Magnolia officinalis leaves and purification of main active compounds

Tan Li-Hong,Zhang Dan,Yu Bao,Zhao Sheng-Ping,Cao Wei-Guo

The leaf of Magnolia officinalis is abundant in natural resources as a novel potential antioxidant and has great commercial value. In this study, the contents of total phenolics, total flavonoids, magnolol and honokiol in different solvent extracts (acetone, methanol and water) were measured. Meanwhile, the antioxidant activity of extracts from M. officinalis leaves were evaluated using 1,1-diphenyl-2-picrylhydrazyl free radical scavenging assay, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt free radical scavenging assay, FRAP assays and β-carotene/linoleic acid system. In addition, the correlation between the antioxidant activity and the contents of the active compounds was analyzed. Experimental results indicated that three extracts exhibited antioxidant effects in a concentration-dependent manner, and acetone extracts had the highest antioxidant activity, next methanol and water. There was an intimate relativity between the contents of magnolol and honokiol in different solvent extracts and the antioxidant capability of them. So the magnolol and honokiol of M. officinalis leaves as a representative of the active compounds were extracted and purified by using the macroporous resin column. The purity of depurated sample could reach 82.6 %, which indicates the M. officinalis leaves have the potential to substitute the production of magnolol and honokiol in M. officinalis . The results of this study can provide theoretical and technical references for the reasonable use of the M. officinalis leaves and alleviate the shortage of M. officinalis resources.

Antioxidant potential, total phenolic and total flavonoid contents of Rhododendron anthopogonoides and its protective effect on hypoxia-induced injury in PC12 cells

Jing Linlin,Ma Huiping,Fan Pengcheng,Gao Rongmin,Jia Zhengping

Background Rhododendron anthopogonoides Maxim, a kind of traditional Tibetan medicine, has been used to remove body heat, body detoxification, cough, asthma, stomachic and swelling, eliminate abundant phlegm and inflammatory for a long time. In the present study, the total phenols and total flavonoid contents as well as antioxidative properties of the crude extract and solvent fractions of R. anthopogonoides were determined using seven antioxidant assays. Additionally, the protective effect of the extracts on hypoxia-induced injury in PC12 cells was also investigated. Methods The content of total flavonoid and total phenolic was determined by the aluminum colorimetric method and Folin-Ciocalteu assay, respectively. In vitro antioxidant study, the effect of the crude extract and solvent fractions on total antioxidant activity, reducing power, DPPH radical scavenging, ABTS radical scavenging, superoxide radical scavenging, hydroxyl radical scavenging and nitric oxide radical scavenging were examined. The correlation between the phenolic and flavonoid content of the extracts and their antioxidant properties also analyzed. Furthermore, the protective effect of extracts on hypoxia-induced damage on PC12 cells was investigated by cell viability, lactate dehydrogenase (LDH) release, malondialdehyde (MDA) production and the activities of antioxidant enzymes. Results Our results showed that ethyl acetate and n-butanol fractions had higher content of phenolics and flavonoid compounds than other fractions. Except ABTS radical assay, n-butanol fraction exhibited the strongest antioxidant activity. While the hexane fraction showed the lowest antioxidant activity. Ethyl acetate also presented excellent antioxidant activity, which was just lower than n-butanol fraction. Significant correlation between the phenolic, flavonoid content of the extract and fractions with antioxidant assay excluding ABTS, OH scavenging assay was observed. Moreover, ethyl acetate and n-butanol fractions showed protective effect in PC12 cell under hypoxia condition, while crude extract and water fraction had no protective effect. In contrast, hexane fraction exhibited strong cytoprotective effect. Further study indicated that pretreatment of PC12 cells with ethyl acetate and n-butanol fractions, prior to hypoxia exposure, significantly increased the survival of cells and the activities of SOD, CAT, GSH-Px and T-AOC, as well as reduced the level of LDH and MDA. The gathered data demonstrated that ethyl acetate and n-butanol fractions were able to protect PC12 cells against hypoxia induced injury through direct free radical scavenging and modulation of endogenous antioxidant enzymes. Conclusion These findings suggested that ethyl acetate and n-butanol fractions of R. anthopogonoides had significant antioxidant activity and could prevent PC12 cells against hypoxia-induced injury. So it might be regarded as an excellent source of antioxidants and had great potential to explore as therapeutic agent for preventing hypoxia related sickness in future.

Ngoc-Van Thi Nguyen,Cuong Van Nguyen,Ngan Tuyet Duong,Xuan-Trang Thi Dai,Kien Trung Nguyen,Cam-Thuy Thi Le

Response surface methodology was used to estimate the optimum extraction parameters, for which the total phenolic content and total flavonoid content of Avicennia officinalis L. extract were the highest. Based on the 3D surface plots and regression analysis of the independent variables, the optimal conditions for the ultrasound-assisted extraction were as follows: methanol content, 55.27%; liquid-to-solid ratio, 14:1 (mL/g, v/w); temperature, 48.8 °C, and time, 9.66 min. Optimal extraction conditions were applied to validate the quantification. The bioactive compounds from the Avicennia officinalis L. extract were identified through ultra-fast liquid chromatography coupled to diode array detector for the first time. These methods were applied to quantify phenolic acids and flavonoids from A. officinalis extracts collected at six different locations in Vietnam. The results showed that the concentrations of these compounds varied from 0.319 ± 0.022 to 3.524 ± 0.125 mg/g in the methanol extract. The result showed that cinnamic acid is the major compound in A. officinalis collected in Vietnam. In the present study, an optimized ultrasound-assisted extraction was first designed to improve the content of phenolic acids and flavonoid compounds from A. officinalis . Additionally, the bioactive compounds from the A. officinalis extract were identified through UFLC-DAD for the first time.

Comparative Evaluation of Soluble and Insoluble-Bound Phenolics and Antioxidant Activity of Two Chinese Mistletoes

Qing Li,Shihua Yang,Yongqiang Li,Xiaofeng Xue,Yonghua Huang,Hengguo Luo,Yiming Zhang,Zhichao Lu

Mistletoes are used medicinally in order to treat various human illnesses. Few studies have reported on the phenolic content and antioxidant properties of Chinese mistletoes (CMs). In this work, the total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activities of soluble and insoluble-bound phenolic extracts from CMs hosted byCamellia assamica(Mast.) Chang (CMC) andPyrus,i,f.(CMP) were compared. Phenolic compounds in CMC and CMP were identified and quantified using high-performance liquid chromatography (HPLC). The results indicated that the TPC of soluble phenolic extracts was higher than insoluble-bound phenolic counterparts in both CMC and CMP. In addition, the TPC of soluble, insoluble-bound and total phenolic fractions (9.91 ± 0.23, 4.59 ± 0.27 and 14.50 ± 0.35 μmol ferulic acid equivalents per gram (FAE/g) dry sample) extracted from CMP were higher than those extracted from CMC. The soluble phenolic extracts in CMP showed higher antioxidant activities than those in CMC. Eighteen phenolic compounds from soluble and insoluble-bound phenolic extracts from the CMs were identified and quantified by HPLC. This study indicates that CMC and CMP, especially the latter, could be sources of antioxidants in human health care. Keywords:Chinese mistletoes;phenolics;phenolic contents;antioxidant activity

Rapid determination of total flavonoid content, xanthine oxidase inhibitory activities, and antioxidant activity in Prunus mume by near-infrared spectroscopy

Jing-Wen Hao,Nai-Dong Chen,Xuan-Xuan Fan,Wei-Ting Wang,Huan-Huan Jiang,Zi-Yi Zhang,Rui-Ze Gong,Xiao-Li Ruan,Xue Chen

Evaluating the quality of herbal medicine based on the content and activity of its main components is highly beneficial. Developing an eco-friendly determination method has significant application potential. In this study, we propose a new method to simultaneously predict the total flavonoid content (TFC), xanthine oxidase inhibitory (XO) activity, and antioxidant activity (AA) of Prunus mume using near-infrared spectroscopy (NIR). Using the sodium nitrite-aluminum nitrate-sodium hydroxide colorimetric method , uric acid colorimetric method , and 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) free radical scavenging activity as reference methods, we analyzed TFC, XO, and AA in 90  P. mume samples collected from different locations in China. The solid samples were subjected to NIR. By employing spectral preprocessing and optimizing spectral bands , we established a rapid prediction model for TFC, XO, and AA using partial least squares regression (PLS). To improve the model's performance and eliminate irrelevant variables, competitive adaptive reweighted sampling (CARS) was used to calculate the pretreated full spectrum. Evaluation model indicators included the root mean square error of cross-validation (RMSECV) and determination coefficient ( R 2 ) values. The TFC, XO, and AA model, combining optimal spectral preprocessing and spectral bands, had RMSECV values of 0.139, 0.117, and 0.121, with R CV 2 values exceeding 0.92. The root mean square error of prediction (RMSEP) for the TFC, XO, and AA model on the prediction set was 0.301, 0.213, and 0.149, with determination coefficient ( R P 2 ) values of 0.915, 0.933, and 0.926. The results showed a strong correlation between NIR with TFC, XO, and AA in P. mume . Therefore, the established model was effective, suitable for the rapid quantification of TFC, XO, and AA. The prediction method is simple and rapid, and can be extended to the study of medicinal plant content and activity.

In order to screen the Catalpa plant with high antioxidant activity and confirm the corresponding active fractions from Catalpa ovata G. Don, C. fargesii Bur., and C. bungei C. A. Mey., total flavonoid contents and antioxidant activities of the extracts/fractions of Catalpa plant leaves were determined. The determined total flavonoid content and antioxidant activity were used as assessment criteria. Those compounds with antioxidant activity were isolated with silica gel column chromatography and ODS column chromatography. Our results showed that the total flavonoid content in C. bungei C. A. Mey. (30.07 mg/g·DW) was the highest, followed by those in C. fargesii Bur. (25.55 mg/g·DW) and C. ovata G. Don (24.96 mg/g·DW). According to the determination results of total flavonoid content and antioxidant activity in 3 clones of leaves of C. bungei C. A. Mey., the total flavonoid content and antioxidant activity in crude extracts from C. bungei C. A. Mey. 6 (CA6) leaves were the highest. Moreover, the results showed that the total flavonoid content and antioxidant activities of ethyl acetate (EA) fraction in ethanol crude extracts in CA6 leaves were the highest, followed by n -butanol, petroleum ether (PE), and water fractions. Two flavonoid compounds with antioxidant activity were firstly isolated based on EA fraction. The two compounds were luteolin ( 1 ) and apigenin ( 2 ), respectively.

Optimisation and Evaluation of the Correlation of Extraction Conditions for Total Flavonoid Content and Antixanthine Oxidase Activity in Psidium guajava

T.M. Le,C.D.P. Nguyen,A.C. Ha

ABSTRACT Introduction Psidium guajava L. ( P guajava ) and its leaf, one of the most popular plants in traditional medicine and cuisine, were demonstrated to have high flavonoid content with many bioactivities, including antigout activity. However, the correlation between total flavonoid content ( TFC ) and antigout activities and the optimal conditions for extracting P guajava have not been reported. Methods This study established the optimisation of ethanol extracts of P guajava leaves for TFC and antigout activities using the single-factor method and response surface methodology. The antigout activities were determined through the xanthine oxidase inhibition ( XOI ) method. Results The four independent variables (ethanol concentration, plant-solvent ratio, extraction time, and temperature) significantly impacted two response variables (TFC and XOI activity). The optimal extracts for XOI activity reach the maximum TFC of 323.86 ± 2.26 mg quercetin/g and XOI activity of 26.37 ± 0.5 μg/ml. The changes in the chemical structure of extracts were analysed by Fourier transform infrared, which indicated that several flavonoids in P guajava do not have XOI activity. The correlation of TFC and XOI is robust with the Pearson correlation of −0.882 and Spearman Rho test of −0.884. Additionally, the biological activities, such as antioxidant and anti-inflammatory, of optimal extracts for XOI and TFC were determined to extend the application capacity to support the treatment of gout diseases. Conclusions The optimised extraction conditions are critical to isolating flavonoids, which can serve as functional food or medicine for managing gout and its complications.

Cholinesterase, β-amyloid aggregation inhibitory and antioxidant capacities of Chinese medicinal plants

Qin Li,Yanbei Tu,Chao Zhu,Wei Luo,Wencai Huang,Wenbing Liu,Yanfang Li

Traditional Chinese medicinal (TCM) plants have a long history of usage in the treatment of different diseases. Neurodegenerative disorder prevention and treatment using TCM have attracted increasing interest in recent year. In current study, the ethanolic extracts of 26 Chinese medicinal plants were prepared, two cholinesterases (ChEs), β -amyloid aggregation inhibitory, and antioxidant capacities were further investigated for evaluation their anti-Alzheimer’s disease (AD) potential as well as total phenolics and flavonoids contents (TPC, TFC). Of these, Polygonum multiflorum and Spatholobus suberectus were the most active ones against acetylcholinesterase (AChE, IC 50   =   9.11, 9.27   μg/mL), butyrylcholinesterase (BChE, IC 50   =   4.83, 5.37   μg/mL), and 2, 2-diphenyl-1-picrilhydrazyl (DPPH, IC 50   =   5.69, 3.60   μg/mL) with highest TPC (267, 377   mg GAE/g dw) and relative higher TFC (80, 594   mg RE/g dw), respectively. The highest activity was observed with the extracts of Rheum officinale in 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt assay (ABTS, IC 50   =   1.60   μg/mL) and ferric reducing antioxidant power method (FRAP, 5495.43   μmol Fe 2+ /g). Meanwhile, both R. officinale and S. suberectus exhibited the strongest inhibition (>90%) in β -amyloid peptide (A β 1-42 ) aggregation assay. The present findings may reveal the interesting potential of S. suberectus , P. multiflorum, and R. officinale as a valuable plant source of natural bioactive molecules against AD.

RSM-based optimization of microwave- and ultrasound-assisted green extraction of iron-chelating and antioxidant natural products from Azadirachta indica leaves

Mahnoor Anis,Nida Anis,Dildar Ahmed

Azadirachta indica is an abundantly available plant and known for its medicinal properties. Some studies showed its iron chelating activity (ICA) with possible implications for iron overload disorders. The present study aimed to discover efficient and green methods for obtaining ICA-rich extract from A. indica leaves. Microwave-assisted extraction (MAE) and ultrasound-assisted extraction (UAE) methods were optimized using response surface methodology (RSM). Four-factor Box-Behnken designs were used (with 29 runs) with five response factors for each technique. Aqueous choline chloride-glycerol (CCG) DES was employed as an extractant. ICA, total phenolic content (TPC), total flavonoid content (TFC), radical scavenging activity (RSA), and ferric-reducing antioxidant power (FRAP) were the responses. The optimal conditions discovered for MAE were 30 mL/g solvent-to-solid ratio (SSR), 38 % DES concentration, 330 W power and 15 s time with predicted ICA, TPC, TFC, FRAP, and RSA as 127.64 mg EDTAE/g d.w., 23.55 mg GAE/g d.w., 19.94 RE/g d.w., 26.77 mg AAE/g d.w. and 5.37 mg AAE/g d.w., respectively. For UAE, the optimal parameters were 30 mL/g SSR, 39 % DES concentration, 54.8 ͦ C temperature and 17.48 min time with predicted ICA, TPC, TFC, FRAP, and RSA as 127.80 mg EDTAE/g d.w., 18.58 mg GAE/g d.w., 23.36 RE/g d.w., 23.33 mg AAE/g d.w., and 5.79 mg AAE/g d.w., respectively. The models were efficient and supported by statistical and validation studies. MAE, however, used far less time than UAE. The outcomes of the study have important implications for iron overload syndrome and oxidative stress. The developed methods can, thus, be proposed as strong candidates for industrial applications obtaining bioactive compounds from abundantly available leaves of A. indica.
