Integrated Metabolomic and Transcriptomic Analysis Reveals Differential Flavonoid Accumulation and Its Underlying Mechanism in Fruits of Distinct Canarium album Cultivars

Ruilian Lai,Chaogui Shen,Xin Feng,Minxia Gao,Yongyan Zhang,Xiaoxia Wei,Yiting Chen,Chunzhen Cheng,Rujian Wu

Canarium albumfruit has great potential to be consumed as a raw material not only for food but also medicine. The diverse active metabolites composition and content ofC. albumfruits greatly affect their pharmacological effects. However, up to now, there has been no report on the global metabolome differences among fruits from distinctC. albumcultivars. In our present study, by using non-targeted metabolomics techniques, we identified 87 DAMs (differentially accumulated metabolites) including 17 types of flavonoids from fruits of four differentC. albumcultivars. KEGG (Kyoto Encyclopedia of Genes and Genomes) pathway enrichment analysis revealed that the flavone and flavonol biosynthesis- and flavonoid biosynthesis-related DAMs were major factors determining their metabolome differences. Comparative transcriptomic analysis revealed that 15 KEGG pathways were significantly enriched by genes of the identified 3655 DEGs (differentially expressed genes) among differentC. albumcultivars. Consistent with the metabolome data, flavonoid biosynthesis-related DEGs, including eight key structural genes (such asFLS,CCoAOMT,CHI,C4H,DFR,LAR, andC3′H, etc.) and several regulatory transcription factor (TF) genes (including 32MYBsand 34bHLHs, etc.), were found to be significantly enriched (p< 0.01). Our study indicated that the differential expression of flavonoid biosynthesis-related genes and accumulation of flavonoids played dominant roles in the various metabolome compositions of fruits from differentC. albumcultivars. Keywords:Canarium album;flavonoid;metabolomics;transcriptomics;gene expression

Xanthones from securidaca inappendiculata antagonizes the anti-rheumatic effect of methotrexate by inhibiting reduced folate carrier 1

Sa-sa Zhang,Yi-jin Wu,Shu Pan,Qi-hai Wang,Wen-gang Chen,Zhong-fang Wang,Jin-ying Xu,Jian Zuo,Qin Yin

Background The first-line anti-rheumatic drug methotrexate (MTX) is used in the combination. Because of the unpredictable adverse reactions, optimization of relevant regimens is necessary and meaningful. This study aimed to study the possible interaction between Securidaca inappendiculate Hassk. Derived xanthones and MTX.Methods We established adjuvant-induced arthritis (AIA) model, which was treated with MTX and MTX + xanthone-rich fraction (XRF). The clinical efficacy was evaluated by histopathological examination, and LC-MS was used to monitor the blood concentration of MTX. Western blotting and immunohistochemistry were used to detect protein expression. In vitro, we assessed the activity of related transporters by cellular uptake assay based on HEK-293T cells. Results Compared with MTX-treated rats, inflammation in the immunized rats in the MTX + XRF group was obvious, indicating that XRF antagonized the anti-rheumatic effect of MTX. Meanwhile, XRF reduced liver and kidney injuries caused by MTX in addition to MTX. Results from immunohistochemical and nappendiculat assays suggested that XRF may reduce uptake of MTX by down-regulating reduced folate carrier 1 (RFC1).Conclusion This study indicated that XRF could reduce the plasma concentration of MTX by inhibiting the expression of RFC1, antagonize the therapeutic effect of MTX on AIA rats, and reduce its oral bioavailability. The combination of S. inappendiculate and MTX should be further optimized to achieve the goal of increasing efficiency and reducing toxicity.

Combined application of bud and leaf growth fertilizer improves leaf flavonoids yield of Ginkgo biloba

Daoming Wu,Jiayi Feng,Mingli Lai,Jianhui Ouyang,Dandan Liao,Wanwen Yu,Guibin Wang,Fuliang Cao,Douglass F. Jacobs,Shucai Zeng

Higher flavonoids yield can increase economic and industrial efficiency of Ginkgo biloba Linn. leaf-harvesting plantation. However, high-temperature limits ginkgo leaf flavonoids production in warm climates regions. Few studies have reported on methods that may improve ginkgo leaf flavonoids yield by increasing leaf productivity and flavonoids content simultaneously. This study investigated the effects of combined application of fertilizer for bud growth (FBG) and fertilizer for leaf growth (FLG) on the growth, productivity and flavonoids accumulation of ginkgo leaves, aiming to optimize fertilization schemes for improving leaf flavonoids yield of ginkgo in high-temperature regions. Results revealed that FBG significantly improved twig bud break, twig number, and twig elongation, and increased leaf number (up to 446±27.38 per four-year-old seedling) in spring. Topdressing FLG in summer moderated the decline of leaf SPAD (soil and plant analyzer development) values, modulated leaf nutrient content, and significantly increased single leaf dry weight, antioxidative enzyme activities, and leaf flavonoids content. Dry leaf productivity, flavonoids content, and flavonoids yield of four-year-old ginkgo with combined application of FBG and FLG was 103±16.05 g/plant, 6.54±0.13 mg/g, and 615.71±58.22 mg/plant, respectively, which were comparably high in reference to those in the central production region. These results suggest that the integrated application of FBG and FLG is an effective approach for improving ginkgo leaf flavonoids yield in high-temperature regions.

Salicylic acid-induced flavonoid accumulation in Ginkgo biloba leaves is dependent on red and far-red light

Jun Ni,Lixiang Dong,Zhifang Jiang,Xiuli Yang,Zhehang Sun,Jiaxun Li,Yuhuan Wu,Maojun Xu

Flavonoids play important roles in plant immune responses, but the molecular mechanism and influencing factors on flavonoid accumulation during Ginkgo immune responses are largely unknown. In this research, post-harvest Ginkgo leaves cultured in the dark were treated with salicylic acid (SA). A transcriptome analysis and total flavonoid measurement showed decreased flavonoid biosynthesis and accumulation in leaves. Experiments carried out in seedlings, young and adult trees, showed increased flavonoid content under light conditions but decreased content under dark conditions after treatment with SA. A detailed analysis of individual flavonoids showed similar changes for most of their contents, although some showed different change trends. Single-colored light experiments revealed that red and far-red light, but not blue light, were required in SA-induced flavonoid accumulation. The results clarified the importance of light in SA-induced flavonoid accumulation and provided valuable information to the Ginkgo related pharmaceutical industry.

Eriodictyol From Scutellariae Barbata Alleviates Inflammation and Necrosis via ZBP1-dependent Signaling in Acute Pancreatitis

Simin Tian,Guang Xin,Kun Zhang,Ao Wen,Xiuxiang Yu,Chengyang Ni,Yijiang Liu,Xiaojuan Li,Yu Cao,Wen Huang

Background Acute pancreatitis (AP) is an inflammatory disease of the pancreas marked by inflammation and acinar cell necrosis. Scutellariae Barbata (S. Barbata), a traditional Chinese herb for inflammation, has unclear efficacy for AP. Purpose This study examined the mechanism of eriodictyol, an active component of S. Barbata decoction, targeting pancreatic necrosis and inflammation for the first time. Study Design This study firstly evaluated the pharmacological effects of S. Barbata on AP. Transcriptomic analysis and pharmacological screening were performed to identify essential genes linked to pancreatic necrosis and specific bioactive compounds in S. Barbata. These candidates underwent thorough validation with multiple assays. Our findings strongly demonstrate effectiveness of S. Barbata in combating pancreatic necrosis and reveal the underlying mechanisms involved. Methods Animal experiments were conducted to assess the effects of S. Barbata on AP. Analysis of bulk and single-cell RNA-seq data identified PANoptosis. The protein-protein interaction network and Cytoscape were used to identify AP targets based on PANoptosis. The ConnectivityMap database was used to filter potential compounds from S. Barbatae based on specific targets. After quantifying eriodictyol, its binding to ZBP1 was confirmed through molecular docking, dynamic simulation, cellular thermal shift assay, mtDNA analysis, and ZBP1 overexpression in 266-6 cells. Experiments in vivo and in vitro assessed the effects of eriodictyol on AP. Results S. Barbata decoction reduced pancreatic inflammation and necrosis. Gene Set Enrichment Analysis of PANoptosis revealed that Zbp1, Nlpr3, Ripk1, Casp8, Gsdmd , and Mlkl are ZBP1-dependent targets for AP. Eriodictyol, the active compound in S. Barbata showed a strong affinity for ZBP1. Eriodictyol may enhance pancreatic conditions and reduce ZBP1-dependent protein expression, with minimal impact on mtDNA levels. Conclusion This study broadened the traditional use of S. Barbata suggested eriodictyol as a potential AP treatment candidate via ZBP1-dependent signaling.

Effects of three flavonoids on the metabolism of lenvatinib

Yang Jinzhao,Chen Jie,Li Qingqing,Xu Ren-ai,Chen Xiaohai

Lenvatinib is a first-line therapy for the treatment of hepatocellular carcinoma (HCC), an active multi-target tyrosine kinase inhibitor (TKI). The interaction between Traditional Chinese Medicine (TCM) and chemicals has increasingly become a research hotspot. The objective of this study was to pinpoint the effects of three flavonoids on the metabolism of lenvatinib. Enzyme reaction system was established and optimized in vitro, and in vivo experiments were conducted in Sprague-Dawley (SD) rats, where the analytes were detected by ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). We found that among three flavonoids, luteolin and myricetin had strong inhibitory effects on lenvatinib metabolism, with half-maximal inhibitory concentration (IC50) values of 11.36 ± 0.46 µM and 11.21 ± 0.81 µM in rat liver microsomes (RLM), respectively, and 6.89 ± 0.43 µM and 12.32 ± 1.21 µM in human liver microsomes (HLM), respectively. In Sprague-Dawley rats, the combined administration of lenvatinib and luteolin obviously expanded the exposure to lenvatinib; however, co-administered with myricetin did not have any changes, which may be due to the poor bioavailability of myricetin in vivo. Furthermore, the inhibitory type of luteolin on lenvatinib showed an un-competitive in RLM and a mixed in HLM. Collectively, flavonoids with liver protection, especially luteolin, may inhibit lenvatinib metabolism in vitro and in vivo.

1H NMR-based metabolomic study of the effects of flavonoids on citrinin production by Monascus

Shanshan He,Yanling Wang,Jianhua Xie,Heng Gao,Xiujiang Li,Zhibing Huang

Monascus comprises purple-red molds. Various compounds can be obtained from these species, including statins and food-safe yellow, red, and orange pigments. However, the secondary metabolite citrinin, a mycotoxin, is produced during the late stages of growth. Citrinin biosynthesis should be reduced to apply Monascus pigments safely. Fortunately, this can be achieved by the addition of flavonoids (genistein, daidzein, apigenin, and kaempferol). However, the effects of these flavonoids on other metabolites remain unknown. Here, we report a 1 H NMR-based multivariate metabolomic analysis of the effects of flavonoids on mycotoxin citrinin production by Monascus . Fifteen metabolites involved in lysine and arginine biosynthesis and alanine, aspartate, glutamate, biotin, arginine, proline, and glutathione metabolism were detected. The reduction in glutamate, aspartate, biotin, and 2-phosphoglycerate content suggested their association with the citrinin reduction mechanism. This study identifies the citrinin production pathway in Monascus and will aid in the development of citrinin-control methods.

Antioxidant and Anti-Inflammatory Activities of the Crude Extracts of Moringa oleifera from Kenya and Their Correlations with Flavonoids

Ming-Quan Guo

Moringa oleiferaLam. (M. oleifera) is commonly distributed and utilized in tropical and sub-tropical areas. There has been a large number of reports on the antioxidant and anti-inflammatory activity of its leaves, but only a few about its seeds and roots. Hence, in this work we aimed to systematically compare the antioxidant and anti-inflammatory activities of the ethanol crude extracts of leaves, seeds, and roots ofM. oleiferafrom Kenya, and further correlate the differential activities with the chemical constituents from these three parts. The antioxidant activities were measured by using three different assays (DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2′-azinobis-(3-ethylbenzthiazoline-6-sulfonic acid) and FRAP (Ferric-Reducing Antioxidant Power), respectively). Results showed that the leaf extracts displayed the highest DPPH radical scavenging and FRAP total reducing power activities with IC50values of 1.02 ± 0.13 mg/mL and 0.99 ± 0.06 mM Fe2+/g, respectively; the leaf and root extracts exhibited potential ABTS radical scavenging activities with the IC50values of 1.36 ± 0.02 and 1.24 ± 0.03 mg/mL. Meanwhile, the leaf and seed extracts (11.1–100 µg/mL) also exerted obvious anti-inflammatory activities, as indicated by the inhibition of NO production. To further reveal correlations between these differential activities with the chemical constituents in the three organs, the total flavonoids content (TFC) of the three different extracts were evaluated, and the TFC of leaves, seeds and roots were found to be 192.36 ± 2.96, 5.89 ± 0.65 and 106.79 ± 2.12 mg rutin equivalent (RE)/g, respectively. These findings indicated the important impacts of the total flavonoid contents on antioxidant and anti-inflammatory activities. Additionally, we further determined the phytochemical profiles ofM. oleiferaby HPLC-UV/ESI-MS/MS, and identified most of the chemical constituents of leaves as flavonoids. In summary, the leaves ofM. oleiferaare a better potential natural source of antioxidants and anti-inflammatory agents, and very promising for development into the health promoting dietary supplements. Keywords:Moringa oleifera;antioxidant;anti-inflammatory;flavonoids;phytochemical profile

Identification and quantification of free, conjugate and total phenolic compounds in leaves of 20 sweetpotato cultivars by HPLC–DAD and HPLC–ESI–MS/MS

Chunying Luo,Xixi Wang,Ge Gao,Lian Wang,Yongxin Li,Chengjun Sun

The study systematically investigated free, conjugate and total phenolics (phenolic acids and flavonoids) in leaves of 19 Chinese and one American sweetpotato cultivars grown in China. Three extraction/hydrolytic methods (direct extraction and acidic and basic hydrolysis) for sample preparation were employed to obtain different forms of phenolics. Twenty-nine phenolics were separated and identified using HPLC–DAD and HPLC–ESI–MS/MS. Three quercetin glycosides were characterised for the first time from this plant. Contents of the principal phenolics identified were determined by the HPLC–DAD procedure, which was validated in terms of linearity, precision, accuracy and limit of detection and quantification. Moreover, to the best of our knowledge, it is the first to reveal and demonstrate artifacts of esterification during acidic methanolic and ethanolic hydrolysis, and chromatographic behaviours, UV spectra and MS data of 20 hydroxycinnamic acid methyl and ethyl esters were obtained using acidic methanolic and ethanolic hydrolysis.

Biosynthesis of Gold Nanoparticles by Flavonoids from Lilium casa blanca

Du Juan,Zhou Zhie,Zhang Xueqing,Wu Shuang,Xiong Jianhua,Wang Wenjun,Luo Qiushui

Biosynthesis of gold nanoparticles (GNPs) by flavonoids from Lilium casa blanca has been developed. Several parameters such as pH, reaction temperature, reaction time and concentration of flavonoids were explored to control the formation of the GNPs. The synthesized GNPs were characterized by UV–Vis spectroscopy, transmission electron microscopy and X-ray diffraction. Stability and catalytic activity of the synthesized GNPs were also discussed. The results showed that the synthesized GNPs were in spherical, about 2.6 nm, with a face centered cubic structure. Synthesized GNPs showed good catalytic activity in the reduction of p -nitrophenol ( p -NP) to p -aminphenol ( p -AP). This method for synthesis of GNPs is simple, economic, nontoxic and efficient.

AIEE-Active Flavones as a Promising Tool for the Real-Time Tracking of Uptake and Distribution in Live Zebrafish

Yi Wu,Ying He,Huiqing Luo,Tingting Jin,Feng He

In recent years, aggregation-induced emission enhancement (AIEE) molecules have shown great potential for applications in the fields of bio-detection, imaging, optoelectronic devices, and chemical sensing. Based on our previous studies, we investigated the fluorescence properties of six flavonoids and confirmed that compounds1–3have good aggregation-induced emission enhancement (AIEE) properties through a series of spectroscopic experiments. Compounds with AIEE properties have addressed the limitation imposed by the aggregation-caused quenching (ACQ) of classic organic dyes owing to their strong fluorescence emission and high quantum yield. Based on their excellent fluorescence properties, we evaluated their performance in the cell and we found that they could label mitochondria specifically by comparing their Pearson correlation coefficients (R) with Mito Tracker Red and Lyso-Tracker Red. This suggests their future application in mitochondrial imaging. Furthermore, studies of uptake and distribution characterization in 48 hpf zebrafish larvae revealed their potential for monitoring real-time drug behavior. The uptake of compounds by larvae varies significantly across different time cycles (between uptake and utilization in the tissue). This observation has important implications for the development of visualization techniques for pharmacokinetic processes and can enable real-time feedback. More interestingly, according to the data presented, tested compounds aggregated in the liver and intestine of 168 hpf larvae. This finding suggests that they could potentially be used for monitoring and diagnosing liver and intestinal diseases. Keywords:AIEE;flavones;mitochondria;zebrafish;uptake;distribution
