Toxin Production by Alternaria alternata in Black Spot Disease of Chrysanthemum morifolium ‘Fubai’: Accumulation of Altenuene and Tenuazonic Acid in Flowers

Qingling Zhan,Lina Liu,Wenjie Li,Jingshan Lu,Jiafu Jiang,Fadi Chen,Ye Liu,Zhiyong Guan

Alternariaspecies produce diverse secondary metabolites that act as critical virulence factors during plant pathogenesis. In cultivation areas ofChrysanthemum morifolium‘Fubai’—a key cultivar for herbal tea—black spot disease caused byA. alternatamanifests as necrotic leaf lesions progressing to wilting. Despite this disease’s economic impact, information on its associated toxins is limited, and the types of toxins produced by the black spot pathogen ofChrysanthemum morifolium‘Fubai’ in particular remain unclear. Furthermore, whether toxins are present in the flowers when the leaves show symptoms is uncertain, but their inflorescence is not visibly infected. Using two previously characterizedA. alternatastrains (F16/F20) isolated from ‘Fubai’ in earlier studies, we demonstrated the concomitant production of altenuene (ALT) and tenuazonic acid (TeA) in both strains, with strain-specific yield variations (F20 TeA: 342.16 µg/mL vs. F16: 21.84 µg/mL; ALT: 0.28 µg/mL vs. 0.90 µg/mL). Time-course monitoring revealed coordinated accumulation of both toxins in inoculated petals, reaching 18.07 μg/g ALT and 2.59 µg/g TeA by day 9. Notably, these two toxins were detected only in flower samples from black spot-infected plants, and their concentrations correlated closely with disease severity in the leaves. Moreover, although the inflorescences did not display symptoms, both fresh and dried flowers retained detectable toxin levels. We established a technical system for the extraction and quantitative detection of the toxins ALT and TeA produced by the black spot pathogen in tea chrysanthemum. This work provides the first confirmation of ALT/TeA co-contamination inChrysanthemum morifolium‘Fubai’, revealing substantial dietary exposure risks through tea consumption. Our findings suggest that, from a food safety risk reduction perspective, integrated management strategies should be developed to minimize toxin contamination in tea chrysanthemum, including improved disease prevention measures and potential regulatory considerations.

TBBPA and its alternative TCBPA induced ROS-dependent mitochondria-mediated apoptosis in the liver of Rana nigromaculata

Xiuying Jia,Ruopeng Yan,Huikang Lin,Zhiquan Liu,Lilai Shen,Hongmei Yang,Haoying Wu,Xiaodong Shan,Hangjun Zhang

Tetrabromobisphenol A (TBBPA), which is the most widely employed brominated flame retardant , and its alternative tetrachlorobisphenol A (TCBPA) are widely distributed in aquatic environments. In the present study, the hepatotoxicity induced by TBBPA and TCBPA was investigated in Rana nigromaculata , and the potential mechanisms were investigated with a particular focus on ROS (reactive oxygen species) -dependent mitochondria-mediated apoptosis. Healthy adult frogs were exposed to 0, 0.001, 0.01, 0.1, and 1 mg/L waterborne TBBPA and TCBPA for 14 days. The results showed that liver weight was significantly increased by 51.52%–98.99% in the 0.01, 0.1, and 1 mg/L TBBPA and TCBPA groups relative to the control. Histological examination revealed that the structure of the liver, to some extent, was influenced by TBBPA and TCBPA with nuclear shrinkage and mitochondrial swelling. Meanwhile, TBBPA and TCBPA have significantly increased the alanine transaminase level in serum and the content of ROS, while inhibiting the activity of superoxide dismutase in the liver. In addition, DNA fragments were observed in the TBBPA and TCBPA groups relative to the control. Expression of Cytochrome C was significantly increased by 1.13-, 1.38-, 1.60-, and 2.46-fold in 0.001, 0.01, 0.1, and 1 mg/L TBBPA, and by 1.26-, 1.51-, 2.14-, and 2.98- fold in 0.001, 0.01, 0.1, and 1 mg/L TCBPA, respectively, which indicated that TCBPA may be more toxic than TBBPA. Similarly, the ratio of Bax/Bcl-2 was increased in a dose-dependent manner. These results indicated that apoptosis in the ROS-dependent mitochondrial pathway mediates hepatotoxicity caused by TBBPA and TCBPA. The present study will facilitate an understanding of the toxicity mechanism of flame retardants.

Metformin exposure altered intestinal microbiota composition and metabolites in amphibian larvae

Guang-Li Fu,Qin-Yuan Meng,Yu Chen,Jin-Zhao Xin,Jia-Hui Liu,Wei Dang,Hong-Liang Lu

The antidiabetic pharmaceutical metformin (MET) is largely unmetabolized by the human body. Its residues are readily detectable in various aquatic environments and may have adverse impacts on the growth and survival of aquatic species. To date, its toxicological effects have scarcely been explored in non-fish species. Here, we exposed the tadpoles of black-spotted pond frog ( Pelophylax nigromaculatus ) to different concentrations (0, 1, 10 and 100 μg/L) of MET for 30 days and measured the body size, intestinal microbiota and metabolites to evaluate potential effects of MET exposure in amphibian larvae. MET exposure did not affect the growth and intestinal microbial diversity of tadpoles. However, intestinal microbial composition changed significantly, with some pathogenic bacteria (e.g., bacterial genera Salmonella , Comamonas , Stenotrophomonas , Trichococcus ) increasing and some beneficial bacteria (e.g., Blautia , Prevotella ) decreasing in MET-exposed tadpoles. The levels of some intestinal metabolites associated with growth and immune performance also changed significantly following MET exposure. Overall, our results indicated that exposure to MET, even at environmentally relevant concentrations, would cause intestinal microbiota dysbiosis and metabolite alteration, thereby influencing the health status of non-target aquatic organisms, such as amphibians.

Inhibition of hexose oxidase on the dark spots in fresh wet noodle sheets: A feasible prevention of dark spots

Yang Zhao,Ze-Hua Huang,Hui-Ming Zhou,Ke-Xue Zhu,Xiao-Na Guo,Wei Peng

Hexose oxidase was a feasible prevention for the dark spots in the fresh wet noodle sheets (FWNS). The chemical mechanism that hexose oxidase recucing the melanins of dark spots was discussed basis on the UPLC-TOF-MS analysis of the polyphenol oxidase (PPO)-catechol system. In the process of PPO browning, hexose oxidase catalyzed the oxidation of o -benzoquinone derivatives and their oligomers, hindering the formation of melanins. Hexose oxidase was efficient in FWNS with low ash content when water addition was 24%~44% or pH range was 4 ~ 7.5. Hexose oxidase could inhubit dark spots in the presence of 10 metal ions. The recommended addition amount was 40 ~ 60 ppm, by which the dark spots could be compolitely inhibited. Hexose oxidase was also suitable for wholewheat and oat FWNS, Δ L 6d of wholewheat and oat FWNS were reduced by 4 and 7.98, respectively.

Behavioral effects of polylactic acid microplastics on the tadpoles of Pelophylax nigromaculatus

Zihan Li,Youhua Chen

Polylactic acid microplastics (PLA-MPs), biobased plastics made from renewable resources, are considered to be a potential solution for alleviating the pollution pressure of plastics; however, the potential environmental risks of PLA-MPs must be further evaluated. In this study, the effects of PLA-MPs on the tadpoles of Pelophylax nigromaculatus were investigated by designing different PLA-MP exposure experiments. We found that PLA-MPs negatively affected the survival, growth and development of tadpoles. In addition, in open field tests, PLA-MPs also reduced tadpole locomotion while resulting in more repetitive searching behavior within a restricted area. This effect was more pronounced at higher concentrations of PLA-MPs (20 mg/mL) and in combination with the heavy metal Cd 2+ . In the tank tests, PLA-MPs increased tadpole aggregation, and their combined effect with Cd 2+ resulted in a tendency for tadpole aggregation to increase and then decrease, with the distribution tending to favor aggregation in edge regions. PLA-MPs also strongly inhibited the spatiotemporal exploratory activities of tadpoles in the tanks. This study provides a more detailed investigation of the behavioral effects of PLA-MPs on tadpoles and provides a theoretical basis for subsequent ecotoxicological studies of PLA-MPs.

Antibiotic Effects of Volatiles Produced by Bacillus tequilensis XK29 against the Black Spot Disease Caused by Ceratocystis fimbriata in Postharvest Sweet Potato

Mingjie Xu,Jianheng Guo,Tengjie Li,Chunmei Zhang,Xue Peng,Ke Xing,Sheng Qin

Black spot disease caused by Ceratocystis fimbriata is destructive to the production, transportation, and storage of sweet potato. The antifungal effects of Bacillus tequilensis XK29 against C. fimbriata through volatile organic compounds (VOCs) were evaluated in this study. The activated carbon assay proved that XK29 could exert antibiotic effects through volatiles. By optimizing the wheat seed weight, inoculation method, concentration, volume, and time, the antifungal activity of XK29 was significantly improved. XK29 fumigation inhibited spore formation and germination and changed the cell morphology of C. fimbriata. During the storage of sweet potato tuber roots, XK29 effectively controlled black spot disease and reduced the weight loss and malondialdehyde content. Metabolomic analysis revealed that 21 volatile compounds were released from XK29. Isovaleric acid, isobutyric acid, and 2-methylbutanoic acid effectively inhibited the growth of C. fimbriata. These results indicate that B. tequilensis XK29 has a good potential to be developed as a microbial fumigation agent.

Rapid and Specific Detection of the Poplar Black Spot Disease Caused by Marssonina brunnea Using Loop-Mediated Isothermal Amplification Assay

Qin Xiong,Linlin Zhang,Xinyue Zheng,Yulin Qian,Yaxin Zhang,Lijuan Zhao,Qiang Cheng

Marssonina brunneais the main pathogen that causes poplar black spot disease, which leads to the decrease of the photosynthetic efficiency and significantly affects the production and quality of timber. Currently, no in-field diagnostic exists forM. brunnea. Here, we described a loop-mediated isothermal amplification (LAMP) assay for the rapid and sensitive detection ofM. brunnea. A set of six oligonucleotide primers was designed to recognize eight distinct sequences of the internal transcribed spacer (ITS) region ofM. brunnea. The LAMP assay was optimized by the combination of high specificity, sensitivity, and rapidity for the detection of less than 10 pg/μL of target genomic DNA in 60 min per reaction at 65 °C, whereas with PCR, there was no amplification of DNA with concentration less than 1 ng/μL. Among the genomic DNA of 20 fungalisolates, only the samples containing the genomic DNA ofM. brunneachanged from violet to sky blue (visible to the naked eye) by using hydroxynaphthol blue (HNB) dye. No DNA was amplified from the eight other fungus species, including two otherMarssoninapathogens, three other foliar fungi pathogens of poplar, and three common foliar fungal endophytes of poplar. Moreover, the detection rates ofM. brunneafrom artificially and naturally infected poplar leaves were 10/16 (62.5%) and 6/16 (37.5%) using PCR, respectively, while the positive-sample ratios were both 16/16 (100%) using the LAMP assay. Overall, theITSLAMP assay established here can be a better alternative to PCR-based techniques for the specific and sensitive detection ofM. brunneain poplar endemic areas with resource-limited settings. Keywords:Marssonina brunnea;rapid detection;loop-mediated isothermal amplification;poplar black spot

Characterization of volatile compounds from healthy and citrus black spot-infected Valencia orange juice and essential oil by using gas chromatography–mass spectrometry

Leng Han,Guijie Li,Xuting Wang,Bo Yu,Tenghui Zhang,Yujiao Cheng

Citrus black spot ( Phyllosticta citricarpa, CBS) is an important fungal disease that causes rind blemishes and affects quality of citrus fruits. The response of citrus to CBS in terms of volatiles was evaluated using molecular sensory science approaches. Fifty and twenty-one volatiles were identified in the orange juice and essential oil samples, respectively, via gas chromatography–mass spectrometry (GC–MS). The total volatile content in the samples increased after CBS infection, especially in the severe-infection group (SEG) juice and moderate-infection group (MOG) essential oil, which reached the highest levels. CBS enhanced floral, fruity, and off-flavor aromas and decreased the green aroma in citrus juice. Citrusy, floral, and green aromas increased in the CBS-infected essential oil. Six/five potential markers were screened in citrus juice/essential oil, respectively using the orthogonal partial least-square discriminant analysis (OPLS-DA) model. The changes in aroma profile and the difference in infection levels in citrus were attributed to these odorants.

Rapid detection of linalool by QCM gas sensor based on HKUST-1/MWCNT-gel@MIP in early sweetpotato black spot disease

Suqi Liu,Xiaowei Wu,Shihao Chen,Cong Liu,Wanqi Zhang,Mingyi Yang,Jiyu Cheng,Guoquan Lu,Zhenhe Wang,Wei Chen,Yuge Guan,Linjiang Pang,Xinghua Lu

Linalool, a terpenoid alcohol with bacteriostatic properties, was detected in sweetpotato suffering from black spot disease. In this research, a composite material based on HKUST-1/MWCNT-gel@MIP was employed to detect the linalool content in these infected sweetpotatoes. The micro-morphology and crystal structure of the composite were characterized using scanning electron microscopy (SEM), fourier transform infrared (FTIR), and x-ray diffraction (XRD). Experimental results demonstrated that the quartz crystal microbalance (QCM) gas sensor of HKUST-1/MWCNT-gel@MIP composite exhibited high sensitivity (0.873 Hz/ppm), a low detection limit (0.237 ppm), a small standard deviation (0.83 %), good reproducibility, and outstanding selectivity. The QCM gas sensor data from actual samples showed a strong correlation with gas chromatography-mass spectrometry (GC–MS) results (r = −0.976, p < 0.01), demonstrating its exceptional ability to identify linalool. Thus, it can be used for early detection of sweetpotato black spot disease.

Tetrabromobisphenol a and its alternative tetrachlorobisphenol a induce oxidative stress, lipometabolism disturbance, and autophagy in the liver of male Pelophylax nigromaculatus

Yu Han,Hongmei Yang,Zhiqun Liu,Chao Hu,Imane Lamine,Zhiquan Liu,Panpan Gao,Yanming Sui,Pei Zheng,Hangjun Zhang,Xiuying Jia

Tetrabromobisphenol A (TBBPA) and tetrachlorobisphenol A (TCBPA) have been widely used as flame retardants. However, their potential health risks to organisms have raised concerns, particularly for liver toxicity. Present study aimed to explore the toxic effects of TCBPA and TBBPA on black-spotted frogs ( Pelophylax nigromaculatus ) liver oxidative stress, autophagy, and lipid accumulation. After exposure to 0.001, 0.01, 0.1, and 1 mg/L TBBPA and TCBPA for 14 days, the content of cholesterol and triglyceride were significantly elevated. In addition, the malondialdehyde level rose greatly in dose dependent. However, the glutathione level declined in high TBBPA groups (0.01 and 0.1 mg/L). Furthermore, expressions of Beclin1 , Atg5 , and Atg7 were significantly increased, while p62 was markedly declined, respectively. Results obstained suggested that TBBPA and TCBPA exposure induced liver toxicity in black-spotted frog. This study provided insights into the toxicity mechanism of bisphenol flame retardants in amphibians and will aid in the ecological risk assessment of flame retardants.

Synthesis of a novel β-cyclodextrin chiral stationary phase and its application to the evaluation of the enantioselective bioaccumulation and elimination behavior of tebuconazole in Rana nigromaculata tadpoles

Linbin Zheng,Meng Li,Zhouyu Jiang,Junyan Fan,Zengbo Fang,Jihuai Zheng,Yan Cui

Background The increased production and use of chiral pesticides will enhance their exposure in the environment. Chiral pesticides typically exhibit varied biological effects among these enantiomers. Therefore, it is very essential to develop and validate chiral analytical methods to investigate their potential ecological risks from a stereoselective perspective. Current separation of pesticides enantiomers relies extensively on chiral stationary phases (CSPs), while the development of β-Cyclodextrin derivatives CSPs become the research focus due to their great modifiability and excellent chiral recognition capabilities. Results A novel chiral stationary phase, 3,5-dichlorophenylaminomethyl-6-phenylenediamine-β-cyclodextrin chemically bonded silica gel (MPDCDA), was successfully prepared. Based on that, a stereoselective HPLC-MS/MS method was developed and validated for the determination of tebuconazole enantiomers in Rana nigromaculata tadpoles. After extraction by QuEChERS, the tebuconazole enantiomers were completely separated with the resolutions of 1.63 using the mobile phase of methanol-water (70/30, v/v). Good linearity (r > 0.9990) for both enantiomers over a concentration range of 0.20–500.0 ng/mL was obtained with the accuracy ranged from 6.7 % to 9.3 % and the intra-day and inter-day precisions below 6.2 % at three quality control levels. The proposed method was successfully applied in evaluating the enantioselective bioaccumulation and elimination profiles of tebuconazole in tadpoles. At the tested conditions, there was no significantly enantioselective difference in the bioaccumulation process for S - tebuconazole and R -tebuconazole. However, the elimination process of tebuconazole enantiomers was enantioselective with R -tebuconazole preferentially degraded. Significance This work provided an accurate risk assessment of chiral pesticides to non-target aquatic organisms from a stereoselective perspective. These findings would deepen our understanding of the potential ecological risks of chiral pesticides on aquatic organisms and provide scientific support for the protection of aquatic organisms and their ecological environments, as well as sustainable development.

Non-destructive detection of trans-caryophyllene in early sweetpotato black spot disease using a QCM gas sensor based on modified CAU-1@ZIF-8 composite

Suqi Liu,Linjiang Pang,Xiaowei Wu,Shihao Chen,Mingyi Yang,Jiyu Cheng,Guoquan Lu,Zhenhe Wang,Wei Chen,Yuge Guan,Xinghua Lu

Changes in the content of trans -caryophyllene could identify whether sweetpotatoes are infected with black spot disease. Therefore, in this study, a quartz crystal microbalance (QCM) sensor based on modified CAU-1@ZIF-8 was used for non-destructive detection of sweetpotato black spot. The experimental results showed that the constructed sensor performed best when both modified CAU-1 and ZIF-8 were present. The sensor showed a linear trend (R 2  = 0.996, limit of detection (LOD) = 0.55 ppm) in response to trans -caryophyllene in the concentration range of 20–340 ppm with a sensitivity of −0.489 Hz (ppm) −1 . The developed gas sensor was successfully used for the detection of trans -caryophyllene in real samples of sweetpotato. The experimental data showed a significant correlation with the results of gas chromatography-mass spectrometry (GC–MS) with a correlation coefficient of 0.970 ( p  < 0.01). Therefore, it could be used for early warning of black spot disease in sweetpotato.
