Dental caries, particularly secondary caries, which is the main contributor to dental repair failure, has been the subject of extensive research due to its biofilm-mediated, sugar-driven, multifactorial, and dynamic characteristics. The clinical utility of restorations is improved by cleaning bacteria nearby and remineralizing marginal crevices. In this study, a novel multifunctional dental resin composite (DRC) composed of Sr-N-co-doped titanium dioxide (Sr-N-TiO2) nanoparticles and nano-hydroxyapatite (n-HA) reinforcing fillers with improved antibacterial and mineralization properties is proposed. The experimental results showed that the anatase-phase Sr-N-TiO2nanoparticles were synthesized successfully. After this, the curing depth (CD) of the DRC was measured from 4.36 ± 0.18 mm to 5.10 ± 0.19 mm, which met the clinical treatment needs. The maximum antibacterial rate againstStreptococcus mutans(S. mutans) was 98.96%, showing significant inhibition effects (p< 0.0001), which was experimentally verified to be derived from reactive oxygen species (ROS). Meanwhile, the resin exhibited excellent self-remineralization behavior in an SBF solution, and the molar ratio of Ca/P was close to that of HA. Moreover, the relative growth rate (RGR) of mouse fibroblast L929 indicated a high biocompatibility, with the cytotoxicity level being 0 or I. Therefore, our research provides a suitable approach for improving the antibacterial and mineralization properties of DRCs.Keywords:dental resin composite;dental caries;secondary caries;titanium dioxide;antibacterial property;remineralization

Objective The purpose of this work was to develop an anti-CAT-SYI immunoglobulin Y (IgY) antibody that targeted both GtfB (glucosyltransferase B) and GbpB (glucan-binding protein B) and test its anticaries properties in rats. Methods A new CAT-SYI fusion gene was created utilising functional DNA fragments from the GtfB and GbpB genes. The recombinant antigens, comprising the fused CAT-SYI antigen, GtfB, and GbpB, were expressed and purified using a prokaryotic expression and purification system. The purified recombinant antigens were utilised to immunise laying hens against particular IgY production. The biological activities of these particular IgY antibodies were then assessed both in vitro and in vivo, including their capacity to suppress biofilm formation and tooth caries. Results Results indicated that these produced IgY antibodies demonstrated a high antibody titer (>0.1 μg/mL) and could precisely recognise and bind to their respective antigens. Furthermore, it was discovered that these specific IgY antibodies successfully bind to Streptococcus mutans and significantly reduce biofilm development. After 8 weeks of ingesting antigen-specific IgY meals, comprising anti-GtfB IgY and anti-GbpB IgY, the severity of dental caries was dramatically reduced in S mutans– infected Sprague-Dawley rats ( P < .01). Anti-CAT-SYI IgY therapy significantly reduced tooth cavities by 89.0% in vivo ( P < .05) compared to other treatment groups. Conclusions The anti-CAT-SYI IgY, a multitarget antibody that targets both GtfB and GbpB, displayed excellent inhibitory effects against S mutans , making it a promising targeted method with improved anticaries efficacy and significant application opportunities.

Traditional antibacterial treatments, such as chlorhexidine (CHX), destroy cariogenic biofilms. However, they exert negative effects in clinical applications, for example, teeth staining, taste disturbance and harm to oral tissue after a long-term exposure. Therefore, biocompatible strategies for efficient antibacterial drug delivery are in high demand. In this study, aimed at dental caries therapy enhancement, we designed a pH-responsive nanocarrier system, capable of releasing CHX in an acidic environment within cariogenic biofilms. Cationic poly(ethylene glycol)-block-poly(2-(((2-aminoethyl)carbamoyl)oxy)ethyl methacrylate) (PEG-b-PAECOEMA) was synthesized first. Modification of PAECOEMA by citraconic anhydride (CA) forms negatively charged PEG-b-PAECOEMA/CA, which could assemble into core–shell polyionic complex micelles (PICMs) when mixed with cationic CHX via electrostatic interactions. PICMs are stable in healthy neutral oral microenvironments with CHX encapsulated in the core and PEG shell exposed. Once in acidic milieu within caries-producing biofilms, they rapidly disassemble and release CHX cargo owing to degradation of citraconic amide groups. Molecular structures of the above copolymers were confirmed using 1H NMR and gel permeation chromatography (GPC) analysis. The pH-dependent degradation rates of citraconic amide in PEG-b-PAECOEMA/CA copolymer were measured by fluorescamine method. Atomic force microscopy (AFM) studies confirmed successful assembly of well-defined spherical PICMs in aqueous solution. The disassembly of PICMs in acidic microenvironment was observed using dynamic light scattering (DLS). PICMs showed an obvious pH-dependent drug release profile when the pH changed from 7.4 to 5.5. More importantly, the micellar system could reduce drug toxicity of CHX and exhibited outstanding antibacterial capability in the biofilm of Streptococcus mutans. Micelles constructed from pH-sensitive PEG-b-PAECOEMA/CA are highly promising for dental caries therapy and provide guidelines for drug-delivery system design in other acidic pathologic systems.

Biofilm formation is considered as a crucial factor in various oral diseases, such as dental caries. The quorum sensing (QS) signaling system was proved to have a crucial role in the microbial dental plaque biofilm formation of Streptococcus mutans (S. mutans). LuxS was critical to regulating the QS system and survival of the bacterium, and, therefore, compounds which target LuxS may be a potential therapy for dental caries. The binding activities of 37,170 natural compounds to LuxS were virtually screened in this study. Baicalein and paeonol were chosen for further research of the binding mode and ΔG values with LuxS. Both baicalein and paeonol inhibited the biofilm formation without influence on the growth of S. mutans. Baicalein also distinctly reduced the production of both rhamnolipids and acids. The results provide us with a new approach to combat dental caries instead of the traditional use of antibacterial chemicals.

AimsTo investigate the effects of the ginsenoside Rh2 on monospecies and multispecies cariogenic biofilms and explore the mechanism of the antibiofilm effect of Rh2 in vitro. Methods and ResultsStreptococcus mutans, Streptococcus sobrinus and Streptococcus sanguinis were chosen to form the monospecies or multispecies biofilms. Crystal violet staining and laser scanning confocal microscopy were used to observe the effect of Rh2 on biofilms in vitro. Cytotoxicity was examined by the Cell Counting Kit‐8. The effects of Rh2 on bacterial membranes were observed via transmission electron microscopy (TEM). The isobaric tags for relative and absolute quantification (iTRAQ) method were used to profile the common differentially expressed proteins. Gene expression was analysed by reverse transcription quantitative polymerase chain reaction. In general, the treatment of cariogenic biofilms with Rh2 significantly decreased biomass accumulation by inhibiting bacterial growth and extracellular polysaccharide synthesis without any cytotoxic effects. TEM imaging showed that Rh2 could disrupt the cell membranes of these bacteria. The iTRAQ results indicated that the levels of mannose‐specific IIC/D and acetaldehyde/alcohol dehydrogenase were substantially down‐regulated, while the mRNA expression of the corresponding genes were significantly changed.ConclusionsOur data revealed a potential application for Rh2 in the protection against dental caries via the inhibition of cariogenic biofilms.Significance and Impact of the StudyThis study describes the first application of a ginsenoside against multispecies cariogenic biofilms. Rh2 may serve as an alternative agent to prevent dental caries by effectively modulating the pathogenic potentials of oral biofilms.

Dental caries represents one of the most widespread oral health challenges worldwide, impacting people of every age demographic. Traditional anticaries strategies primarily rely on fluoride, yet its chronic overuse may lead to health issues such as dental fluorosis. Lanthanum (La), a rare earth element, has emerged as a promising fluoride alternative due to its low toxicity and strong anticaries activity. However, studies have shown that the direct use of free lanthanum under physiological conditions may cause cellular structural damage, dose-dependent hemolytic reactions, and even liver and kidney fibrosis, highlighting the urgent need to optimize its delivery method. In this study, zeolitic imidazolate framework nanoparticles (La@ZIF) were used to encapsulate lanthanum, enabling its controlled and sustained release, thereby proposing a novel fluoride-free anticaries strategy. Three La@ZIF nanoparticles with varying La incorporation levels (20%, 40%, and 60%) were synthesized via a one-pot method and systematically characterized. Results demonstrated their excellent degradation performance and acid-neutralizing capacity under acidic conditions. In vitro experiments confirmed the favorable biocompatibility of La@ZIF and its ability to effectively promote enamel remineralization and restore surface hardness, achieving comparable efficacy to traditional fluoride treatments. In vivo studies further validated the remineralization potential of La@ZIF in a rat model, with no adverse effects observed on major organs. La@ZIF nanoparticles exhibit remarkable anticaries performance and biosafety, offering a new direction for developing fluoride-free anticaries materials.

Dental caries, a prevalent oral disease, is caused by acid- producing plaque microorganisms-notably Streptococcus mutans ( S. mutans )-which drive enamel demineralization. Current prevention strategies, such as fluoride and chlorhexidine, primarily target planktonic bacteria but exhibit limited efficacy against biofilms and neglect enamel remineralization, thereby reducing their therapeutic efficacy. To address these limitations, we developed dual-functional nanoparticles (AXCP NPs) that can eradicate S. mutans biofilm and promote enamel remineralization. The amphiphilic polymer Arg-XOS-CPP (AXC) was synthesized by covalently conjugating oligosaccharide carbonyl groups to the amino residues of L-arginine and casein. Self-assembly of AXC with calcium and phosphate ions yielded hybrid nanoparticles (AXCP NPs). Experimental results indicated that AXCP NPs had no effect on the viability of HaCaT cells. Under acidic conditions, protonation of arginine triggered a surface charge reversal from negative to positive, enabling targeted binding to negatively charged biofilms. As expected, AXCP NPs exhibited potent anti- S. mutans activity (MIC: 640 μg/mL) and remarkable biofilm eradication efficacy (>80% clearance). In vitro release and remineralization assays demonstrated that AXCP NPs continuously released calcium and phosphorus ions, regulating the mineralization process and effectively restoring enamel functionality. Furthermore, in mice caries model, localized application of AXCP NPs significantly suppressed biofilm accumulation on tooth surface and restored mineral density in demineralized enamel, inhibiting caries progression.

Dental caries is a biofilm-mediated dynamic disease with imbalance in teeth mineralization. In this study, a facile and dual-functional hybrid polydopamine@proanthocyanidin (PDA@PC) coating was designed to inhibit cariogenic biofilm and remineralize demineralized dental hard tissues. PDA@PC showed excellent antibiofilm ability with a lower cariogenic bacterial biofilm biomass (0.15 ± 0.03 OD595nm) than the control group (1.45 ± 0.16 OD595nm). In addition, a dense mineral layer was observed on the surface of demineralized enamel. The microhardness of etched enamel with PDA@PC coating reached 329.28 ± 10.16 HV after incubation with artificial saliva, which was similar with the healthy enamel (368.5 ± 17.18 HV). Furthermore, the in vivo rat dental caries model showed that the PDA@PC coating effectively slowed down the progression of caries, with only superficial demineralization and low dental caries scores. The total score was 3-4 times lower than that of the control group. The successful construction of the dual-functional hybrid PDA@PC coating provides a novel approach for dental caries treatment.

Oral biofilm is the leading cause of dental caries, which is difficult to completely eradicate because of the complicated biofilm structure. What's more, the hypoxia environment of biofilm and low water-solubility of conventional photosensitizers severely restrict the therapeutic effect of photodynamic therapy (PDT) for biofilm. Although conventional photosensitizers could be loaded in nanocarriers, it has reduced PDT effect because of aggregation-caused quenching (ACQ) phenomenon. In this study, we fabricated an oxygen self-sufficient nanodroplet (PFC/TPA@FNDs), which was composed of fluorinated-polymer (FP), perfluorocarbons (PFC) and an aggregation-induced emission (AIE) photosensitizer (Triphenylamine, TPA), to eradicate oral bacterial biofilm and whiten tooth. Fluorinated-polymer was synthesized by polymerizing (Dimethylamino)ethyl methacrylate, fluorinated monomer and 1-nonanol monomer. The nanodroplets could be protonated and behave strong positive charge under bacterial biofilm acid environment promoting nanodroplets deeply penetrating biofilm. More importantly, the nanodroplets had extremely high PFC and oxygen loading efficacy because of the hydrophobic affinity between fluorinated-polymer and PFC to relieve the hypoxia environment and enhance PDT effect. Additionally, compared with conventional ACQ photosensitizers loaded system, PFC/TPA@FNDs could behave superior PDT effect to ablate oral bacterial biofilm under light irradiation due to the unique AIE effect. In vivo caries animal model proved the nanodroplets could reduce dental caries area without damaging tooth structure. Ex vivo tooth whitening assay also confirmed the nanodroplets had similar tooth whitening ability compared with commercial tooth whitener H 2 O 2 , while did not disrupt the surface microstructure of tooth. This oxygen self-sufficient nanodroplet provides an alternative visual angle for oral biofilm eradication in biomedicine.

Polymerization shrinkage of composite resin during curing can form microcracks between the restoration and tooth tissue, facilitating bacterial colonization at the bonding interface and increasing the risk of secondary caries. Thus, developing antibacterial adhesives is crucial. This study synthesized a fluoride-containing dimethylaminohexadecyl methacrylate salt (DMAHDM-F) as a co-monomer to create a fluoride-rechargeable resin adhesive (FD-A) with long-term antibacterial properties. FD-A exhibits stable shear bonding strength, curing kinetics, high mechanical properties, and excellent biocompatibility. Its fluoride release can be sustained for over 1 year, achieving efficient antibacterial effect associated with the quaternary ammonium groups, being superior to those adhesives containing only NaF nanoparticles or quaternary ammonium groups. Of note, the FD-A adhesive is recharged with 10 ppm NaF solution after the one-year release test, and the re-fluorinated sample continues to release more fluoride ion, maintaining its antibacterial efficacy for extended period, which is a welcomed feature for dental applications. In summary, the photopolymerizable fluorinated quaternary ammonium salt provides a potential solution for making dental resin adhesives and composites with stable antibacterial effect to enhance the longevity of dental restorations.

Biomineralization technology has become a trend for the arrest and prevention of dental caries. In particular, the bioactivity and ability to release large amounts of Ca2+ and PO43– ions make amorphous calcium phosphate (ACP) for hard tissue remineralization are highly desired. However, the instability of ACP limits its clinical application. Under continuous bacterial challenge in the oral cavity, the currently developed ACP-based remineralization system lacks the ability to inhibit bacterial adhesion and biofilm formation. Here, a dual-functional nanocomposite with antibiofilm and remineralization properties was designed by combining zwitterionic poly(carboxybetaine acrylamide) (PCBAA) and ACP. The resulting nanocomposite was stable in solution for at least 3 days without any aggregation. The PCBAA/ACP nanocomposite exerted a significant inhibitory effect on the adhesion and biofilm formation of Streptococcus mutans and exhibited bactericidal activities under acidic conditions resulting from bacteria. Moreover, compared with fluoride, this nanocomposite demonstrated superior effects in promoting the remineralization of demineralized enamel and the occlusion of exposed dentinal tubules in vivo and in vitro. The present work provides a theoretical and experimental basis for the use of the PCBAA/ACP nanocomposite as a potential dual-functional agent for arresting and preventing caries.

Secondary caries is the most common reason for restoration failures in general dental practices, resulting from tooth-adherent cariogenic bacteria in the oral environment. The application of polymerizable quaternary ammonium monomers (QAMs), which are capable of copolymerizing with methacrylate monomers and immobilizing antimicrobials in dental materials simultaneously, is expected to be an attractive anticaries strategy. In this work, three types of bis-quaternary ammonium betulin-based dimethacrylate derivatives (Bis-QADM-Bet) were synthesized via a Menschutkin reaction. Their chemical structures mainly differ for alkyl chain lengths of 4, 8, and 12 carbons (namely C4, C8, and C12). We focused on the fabrication of dental resins with 10 wt% Bis-QADM-Bet and a conventional dimethacrylate-based resin (bisphenol A glycerolate dimethacrylate/tri(ethyleneglycol) dimethacrylate, Bis-GMA/TEGDMA). The relationship between the Bis-QADM-Bet structure and the antibacterial activity, physicochemical properties, and cytotoxicity of dental resins were systematically investigated. The results indicated that regulating the alkyl chain length of Bis-QADM-Bet to C8 led to the formulated resin with superior effectiveness in terms of S. mutans inhibition (p < 0.05). Interestingly, this optimal resin exhibited better flowability and lower cytotoxicity in comparison with the Bis-GMA/TEGDMA based resin without compromising the mechanical properties (p > 0.05). Additionally, the potential antibacterial mechanism of contact-killing was proposed to illustrate the structural–morphological changes of S. mutans. In summary, this work shows that dental restorative resins containing Bis-QADM-Bet with the appropriate alkyl chain length are beneficial toward inhibiting the growth of S. mutans without sacrificing physicochemical performance.
