Lilies ( Lilium spp.) are in demand worldwide because of their superior commercial and ornamental value. To develop new and practical preservation technologies, effects of pre-harvest treatment of Promalin (a.i. 1.8% each of 6-benzyladenine (6-BA) and gibberellic acid (GA 4 + 7 )) and 1-Methylcyclopropene (1-MCP) on cut lily under cold storage were investigated. Oriental lily ‘Sorbonne’ was selected for this study. The field plants at coloring stage were sprayed with 25 mg L −1 Promalin, 30 μL L −1 1-MCP additionally, their combination (1-MCP + Promalin) was also tested. Deionized water was used as control. The flower stems were harvested 24 h later and stored at 3 ± 1 °C. Preservation effect and physiological changes, e.g. inflorescence life, flower life, opening rate, dry weight loss rate, respiration rate, ethylene production rate, malondialdehyde (MDA) content, lipoxygenase (LOX) activity, energy metabolism related indexes including adenosine triphosphate (ATP) content, energy charge, activities of ATP synthase (ATPase), succinate dehydrogenase (SDH), cytochrome oxidase (CCO) and Ca 2+ -ATPase were determined. The results showed that 1-MCP or Promalin treatment extended the vase life of stored cut lilies and inhibited the decline of the dry weight of petals, reduced respiration rate, ethylene production rate, LOX activity and the accumulation of MDA. They also maintained the activity of ATPase, Ca 2+ -ATPase, SDH, CCO, ATP content and energy charge of cut lilies at a higher level than control. 1-MCP + Promalin exhibited an additive effect. Combination treatment maintained the flower opening ability as high as 89.3% after five-week storage, control merely maintained it till 3 weeks. These data indicate growth regulators (e.g. 1-MCP or Promalin) can extend the storage life of cut lily ‘Sorbonne’ through cutting down respiration consumption and manipulation of energy balance.

Anthocyanins are bioactive substances that are not only responsible for the pigmentation of various fruits, but also hold potential benefits to human health. In this study, postharvest peaches were treated with 10 ppm 1-MCP, and changes of anthocyanin biosynthesis were then revealed at transcriptional level to gain insight into the underlying molecular mechanisms. The functions of key transcription factors (TFs) were systematically analyzed. Results demonstrated that 1-MCP effectively up-regulated expression of structural genes and stimulated anthocyanin accumulation. A novel transcription factor PpbHLH135 can respond positively to 1-MCP treatment and has been noted for its remarkable gene expression and protein abundance. The transcription factor bHLH3 was also investigated in this study due to its widespread interest and attention, serving to compare the regulatory function of bHLH135 in anthocyanin synthesis, although it didn’t show a significant response to 1-MCP treatment in this study. PpbHLH3 TF showed a stronger regulatory effect on PpF3H , while PpbHLH135 was more dominant over the regulation of PpUFGT . Future genetic and molecular experimentation will elucidate whether PpbHLH135 TF, in combination with other TFs, can play a stronger regulating role in fruit pigmentation, or in other metabolic pathways.

Root mustard is plentiful in vitamins and minerals but shrivels and molds easily. In this study, freshly harvested root mustard was fumigated with various concentrations of 1-Methycyclopropene(1-MCP) (1 µL L−1, 1.5 µL L−1, and 2.0 µL L−1) for 24 h and stored at ambient temperature (17 ± 1 °C) for 35 d. Our data showed that 1-MCP fumigation had a striking preservation effect on maintaining weight loss, fruit firmness, lignin, Vc content, and moisture content, inhibiting respiratory intensity and ethylene release rate, as well as decreasing cell permeability and malondialdehyde (MDA) accumulation and maintaining cell membrane integrity of root mustard. In addition, lipoxygenase (LOX), pyruvate dehydrogenase (PDH), and polyphenol oxidase (PPO) activities were significantly reduced throughout the storage period. In contrast, the activities of succinate dehydrogenase (SDH), superoxide dismutase (SOD), ascorbate peroxidase (APX), phenylalanine deaminase (PAL), and peroxidase (POD) remained at high levels. Results showed that 1-MCP treatments were effective in maintaining the quality of root mustard, and the preservation effect of 1.0 μL·L−11-MCP was better than other concentrations of 1-MCP. This study could serve as a theoretical reference for root mustard preservation.Keywords:1-MCP;root mustard;physiology;storage quality

Chinese bayberry is a highly fragile and perishable fruit that requires protective and functional packaging to maintain its postharvest quality. In this study, a novel bio-based aerogel was developed using a gelatin/dialdehyde starch matrix reinforced with cellulose nanofibrils (CNF) via Schiff-base crosslinking and freeze-drying. The as-prepared aerogel exhibited superior compressibility, elasticity, and biodegradability. This aerogel was further functionalized by loading trans-2-hexenal (E2H) and combined with 1-methylcyclopropene (1-MCP) treatment, thereby constructing a postharvest preservation system for Chinese bayberry with the triple functionality of ethylene inhibition, antifungal activity, and cushioning performance. The optimal aerogel formulation (30% CNF content) demonstrated robust mechanical performance and sustained E2H release behavior, which followed first-order kinetics. In vitro and in vivo assays confirmed its significant inhibitory effect against Penicillium citrinum , the primary postharvest pathogen of bayberry. Storage experiments at 4 °C revealed that the synergistic effect of E2H-loaded aerogel pads and 1-MCP treatment effectively reduced decay, preserved firmness and nutritional quality, and extended the shelf life of bayberries to 18 d, significantly surpassing the control group which lost commercial value after 8 d. Furthermore, the aerogel completely degraded in soil within two weeks, confirming its potential as an eco-friendly packaging material. This work, integrating both protective and active preservation functionalities, provides a sustainable and efficient packaging approach for fragile fruit.

Summary The effect of carnauba wax-based nanoemulsion coating-1 (NC-1) combined with 1-methylcyclopropene (1-MCP) treatment on the storage of honey peach fruit ‘Yulupantao’ for 28 days under 0 ± 0.5 °C, and 90% RH conditions was studied. The results showed that NC-1 or 1-MCP treatment alone and in combination were proved to be more effective in maintaining fruit qualities compared to the control group with no treatment. However, the application with the combination of NC-1 and 1-MCP treatment had the most significant effect on reducing mass loss and decay rate, maintaining firmness, total soluble solids (TSS), ascorbic acid (AsA) and titratable acid (TA) contents, suppressing malondialdehyde (MDA) accumulation and conductivity ascend. Moreover, the activities of peroxidase (POD) were maintained, and polyphenol oxidase (PPO) and phenylalanine ammonialyase (PAL) were effectively suppressed in NC-1, 1-MCP and combined treatment than that of control. Therefore, the combination of NC-1 and 1-MCP treatment could be an efficient way to maintain the qualities of peach fruit during low-temperature storage after harvest.

The effects of 1-methylcyclopropene (1-MCP) treatment, storage time, and shelf life and temperature on phenolic concentrations and antioxidant activity in apple peel and flesh of ‘Jonagold’ apple ( Malus domestica Borkh) have been investigated. Hyperoside and chlorogenic acid were the most abundant phenolic compounds in the peel and flesh, respectively. The flesh had lower concentrations of individual phenolics, total phenolics, flavonoids and lower antioxidant activity (determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay) than the peel. In addition, the profiles of phenolics as well as the changing patterns of individual phenolics also differed between the peel and the flesh. After long-time storage (180 d), the greatest decrease of concentrations was the concentrations of hyperoside (27 %) in the peel and total phenolics (35 %) in the flesh. Also, during the 90–180 d storage period, hyperoside and rutin in the peel, chlorogenic acid in the flesh, and flavonoids and antioxidant activity decreased in both peel and flesh tissues. Individual phenolics exhibited different stabilities during the early stages after harvest. 1-MCP reduced the reduction in the concentrations of hyperoside in the peel, chlorogenic acid in the flesh, and total phenolics, flavonoids and antioxidant activity in both the peel and flesh tissues during storage and shelf life. However, for quercetin and rutin, the protective effect of 1-MCP was only observed during the shelf life period. Overall, phenolic compounds responded variously to storage, shelf life, shelf temperature and 1-MCP treatment.

Nanoemulsion has been shown to extend the postharvest life of horticultural produce. In this study, “Fuji” apple were treated with the combination of nanoemulsion coating-1 (NC-1) and 1-methylcyclopropene (1-MCP). Biochemical and nutritional values were assessed every 35 days during 280 days storage at 4°C and 90% RH. Results show that apples treated with the combination of NC-1 and 1-MCP has significantly reduced mass loss and decay, suppressed malondialdehyde (MDA) content. The combination treatment of NC-1 and 1-MCP also retained significantly higher ascorbic acid (ASC), titratable acid (TA), and total soluble solid (TSS) content. NC-1 and 1-MCP treatments alone or in combination produced higher peroxidase (POD) and catalase (CAT) activities but lower phenylalanine ammonia-lyase (PAL) and polyphenol oxidase (PPO) activities than control. Therefore, the combination of NC-1 and 1-MCP preserved better “Fuji” apple quality, which could be used to extend the postharvest life of “Fuji” apple during long-term storage. Practical applications This research work shows that the combined treatment of NC-1 and 1-MCP on “Fuji” apple reduced mass loss, decay and MDA level, retained higher ASC, TA, and TSS content, promoted POD and CAT activities but suppressed PAL and PPO activities, by which preserved better “Fuji” apple qualities during long-term storage. Moreover, the combination of NC-1 and 1-MCP could provide a potential alternative to extend the storage life, which helps in reducing postharvest losses in the supply chain.

The effects of storage conditions and cooking methods on chlorophyll, glucosinolate (GSL), and sulforaphane content in broccoli florets were investigated in this study. For the storage experiment, fresh broccoli florets were stored for 2, 4, and 6 days at 10, 4, and 0 °C with or without 1-methylcyclopropene (1-MCP) treatment. For the cooking experiment, fresh broccoli florets were cooked for 1, 3, 5, 7, and 9 min under three cooking methods, namely, steaming, microwaving, and boiling. Results showed that the contents of chlorophyll, aliphatic GSL, indole GSL, and sulforaphane in broccoli florets of two cultivars decreased with prolonged storage time. The retained contents of chlorophyll, GSLs, and sulforaphane under 0 °C storage condition were significantly higher than those under 10 °C storage condition after 6 days of storage. The sulforaphane content was increased by 1-MCP treatment but differed among varieties. The cooking experiment showed that aliphatic GSL content decreased with increased cooking time under three cooking methods, and indole GSL and sulforaphane contents had a fluctuating trend with increasing cooking time after steaming and microwaving. Sulforaphane content increased by 17.15–50.16% relative to that in fresh broccoli florets and was considerably affected by cooking time. The highest level of sulforaphane content was retained for 7 min during steaming or 5 min during microwaving. Therefore, the combination of 1-MCP treatment and 0 °C storage condition had the best performance in preserving chlorophyll, GSLs, and sulforaphane. Moreover, steaming for 7 min or microwaving for 5 min is a more effective method for preserving the quality and increasing the sulforaphane content of broccoli florets than boiling. Keywords:broccoli;storage;cooking;glucosinolate;sulforaphane

This study examined the effects of postharvest storage conditions on the fruit quality of a new red-fleshed apple cultivar (‘Meihong’). Mature ‘Meihong’ and ‘Golden delicious’ apples were exposed to room temperature, low temperature, and low temperature and 1-MCP, after which several fruit characteristics were evaluated (i.e., firmness, ethylene release rate, relative content of aroma components, phenolic compounds and antioxidant capacity, fruit softening-related enzyme activities, and related gene expression). Both ‘Meihong’ and ‘Golden delicious’ were ACS1-1/-2 heterozygotes, but the ethylene release rate in ‘Meihong’ fruits was lower than that in ‘Golden delicious’ fruits during storage. Therefore, ‘Meihong’ fruits are more conducive to storage. The low temperature storage with and without 1-MCP delayed fruit softening, decreased the ethylene release rate and ester aroma component content, and maintained total flavonoid and anthocyanin contents. Therefore, storage at low temperatures with 1-MCP or other preservatives may be useful for maintaining the ‘Meihong’ fruit quality.

Food packaging systems with a single function of freshness keeping or monitoring may not be able to meet all practical needs. Herein, cellulose nanofibers (CNF)-based labels with dual functions of fruit freshness keeping and visual monitoring were prepared by coaxial 3D printing . CNF-based ink with blueberry anthocyanin was used to create the shell of fibers, exhibiting high formability and print fidelity as well as sensitive visual pH-responsiveness for freshness monitoring. Chitosan containing 1-methylcyclopropene (1-MCP) was loaded into the hollow microchannels of fibers, in which 1-MCP was trapped by the electrostatic effect of chitosan and CNF and exhibited a sustained release behavior. The 3D printed labels prolonged the shelf life of litchis for 6 days, meanwhile, they sensitively indicated the changes in freshness and the accuracy was confirmed by Headspace-Gas Chromatography-Ion Mobility Spectrometry. The CNF-based integrated labels developed in this work provided a new idea for the development of food intelligent packaging.

Curcumin is a molecule found in turmeric root that has anti-inflammatory, antioxidant, and anti-tumor properties and has been widely used as both an herbal drug and a food additive to treat or prevent neurodegenerative diseases. To explore whether curcumin is able to ameliorate HIV-1-associated neurotoxicity, we treated a murine microglial cell line (N9) and primary rat cortical neurons with curcumin in the presence or absence of neurotoxic HIV-1 gp120 (V3 loop) protein. We found that HIV-1 gp120 profoundly induced N9 cells to produce reactive oxygen species (ROS), tumor necrosis factor-α (TNF-α) and monocyte chemoattractant protein-1 (MCP-1). HIV-1 gp120 also induced apoptosis of primary rat cortical neurons. Curcumin exerted a powerful inhibitory effect against HIV-1 gp120-induced neuronal damage, reducing the production of ROS, TNF-α and MCP-1 by N9 cells and inhibiting apoptosis of primary rat cortical neurons. Curcumin may exert its biological activities through inhibition of the delayed rectification and transient outward potassium (K + ) current, as curcumin effectively reduced HIV-1 gp120-mediated elevation of the delayed rectification and transient outward K + channel current in neurons. We conclude that HIV-1 gp120 increases ROS, TNF-α and MCP-1 production in microglia, and induces cortical neuron apoptosis by affecting the delayed rectification and transient outward K + channel current. Curcumin reduces production of ROS and inflammatory mediators in HIV-1-gp120-stimulated microglia, and protects cortical neurons against HIV-1-mediated apoptosis, most likely through inhibition of HIV-1 gp120-induced elevation of the delayed rectification and transient outward K + current.

Impairment of the blood – brain barrier (BBB) and subsequent inflammatory responses contribute to the development of human immunodeficiency virus (HIV)-1-associated neurocognitive disorders (HAND). Apelin-13, the most abundant member of the apelin family, acts as the ligand of the angiotensin receptor-like 1 (APJ). However, its pharmacological function in HAND and its underlying mechanism are unknown. In the current study, we report that the presence of HIV-1 Tat reduced the levels of Apelin-13 and APJ in the cortex tissue of mice. Importantly, Apelin-13 preserved BBB integrity against HIV-1 Tat in mice by increasing the expression of the tight junction protein zonula occludens-1 (ZO-1) and occludin. Interestingly, increased macrophage infiltration, indicated by elevated CD68-positive staining was observed in the cortex after stimulation with HIV-1, which was mitigated by the administration of Apelin-13. Correspondingly, Apelin-13 reduced the expression of monocyte chemoattractant protein-1; (MCP-1). An in vitro two-chamber and two-cell trans-well assay demonstrated that HIV-1 Tat challenge significantly promoted macrophage migration, which was notably attenuated by the introduction of Apelin-13. Accordingly, treatment with Apelin-13 restored the HIV-1 Tat-induced reduction of occludin and ZO-1, while preventing the upregulation of MCP-1 in human brain microvascular endothelial cells (HBMVECs). Our results suggest that Apelin-13 may reduce macrophage infiltration into brain tissues and mitigate BBB dysfunction in patients with HAND.
