The activation of matrix metalloproteinases (MMPs) is responsible for degradation of the hybrid layer . The effect of 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) on biodegradation of the hybrid layer remains unknown. This study aimed to evaluate the effects of 10-MDP and its Ca salt on MMP-9 activity and dentin-bonding durability. 10-MDP-Ca salt was synthesized and characterized by X-ray diffraction (XRD) and nuclear magnetic resonance (NMR). The effects of chlorhexidine (CHX), 10-MDP, and 10-MDP-Ca salt on MMP activity were evaluated by in-situ zymography, MMP-9 inhibition assay, and hydroxyproline (HYP) assay. Dentin-resin bonded specimens were fabricated with using CHX or 10-MDP, including Single Bond 2 (Ctr), CHX + Single Bond 2 (CHX), 10-MDP + Single Bond 2 (10-MDP), and SBU (SBU). Micro-tensile bond strength (μTBS) and nanoleakage tests were performed after 24-h or two-years water storage. XRD and NMR confirmed the presence of 10-MDP-Ca salt. The relative inhibition percentages of recombinant MMP-9 for 10-MDP and 10-MDP-Ca salt were 8.04 ± 0.83% and 55.96 ± 1.82% respectively. HYP assay showed that 10-MDP, CHX, and 10-MDP-Ca salt conditioning decreased collagen degradation. Water storage weakened μTBS of all the groups, with the Ctr and CHX groups showing more significant decreases ( P < 0.05). After aging, the Ctr and CHX groups also presented more nanoleakage, and the CHX group showed more in-situ zymography fluorescence than the 10-MDP and SBU groups. The capacity to inhibit MMP-9 activity, decrease HYP release, and reduce nanoleakage of 10-MDP and its salt contribute to improvement of dentin-bonding durability.

Objectives To examine whether solvents and changing the molecular structure of 10-Methacryloyloxydecyl dihydrogen phosphate (10-MDP) affect its chemical affinity to Yttria-stabilized tetragonal zirconia polycrystals (Y-TZP). Methods The present work investigated the chemical affinity between Y-TZP and 10-MDP dissolved in different solvents (acetone/ethanol/water or mixture) using X-ray photoelectron spectroscopy, Fourier-transform infrared spectroscopy, and thermodynamic calculations. Shear bond strength (SBS) tests were used to evaluate the influence of different solvents on 10-MDP bonding. In addition, several phosphate ester monomer variants were created by changing the 10-MDP molecular structure. Changes included extending/shortening the spacer chain-length, and installing hydroxyl or carboxyl groups as side chains at different positions along the spacer chain. The thermodynamic parameters of the complexes formed between the 10-MDP variants and tetragonal zirconia were evaluated. Results The acquired data indicated that solvent is necessary for the formation of Zr–O–P bonds between 10-MDP and Y-TZP. Solvents affected the chemical affinity of 10-MDP to Y-TZP; acetone facilitated the best bonding, followed by ethanol. Changing the molecular structure of 10-MDP affected its chemical affinity to Y-TZP. The variants 15-MPDP, 12-MDDP, 6-hydroxyl-10-MDP and 6-carboxy-10-MDP all exhibited higher thermodynamic stability than 10-MDP when coordinated with tetragonal zirconia. In contrast, 2-MEP, 5-MPP, 10-hydroxyl-MDP, 10-carboxy-MDP, 5,6-dihydroxyl-10-MDP and 5,6-dicarboxy-10-MDP exhibited lower thermodynamic stability. Significance 10-MDP coordinates with zirconia through dissociating in solvents. Changing the molecular structure of 10-MDP theoretically affects its chemical affinity to Y-TZP.

Introduction and aims This study investigated the synergistic effect of combining 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) and γ-methacryloxypropyl trimethoxysilane (γ-MPTS) in one primer on bonding to lithium disilicate ceramics. Methods A 5% ethanol solution with 10-MDP and γ-MPTS (1:1 by volume) was prepared. The mixed solutions after 24 hours, 72 hours, 7 days, and 6 months storage were characterized by Fourier transform infrared spectroscopy (FTIR). Ultraviolet-visible (UV-Vis) spectrophotometric titration monitored the intermolecular interactions and their dynamics within 30 minutes of mixing. 84 IPS e.max CAD specimens were etched with 5% HF acid and assigned to four surface pretreatment groups: control, 10-MDP, γ-MPTS, and 10-MDP+γ-MPTS. After applying RelyX Ultimate resin cement, 1-mm composite resin cylinders were bonded and light-polymerized. The bonded assemblies were then stored in water for 24 hours or aged via 6 months storage plus 5000 thermocycles before micro-shear bond strength testing. The data were analysed using multivariate analysis of variance ( P < .05). Six machine learning models including Logistic Regression, Decision Tree, Random Forest, Extra Trees, Gradient Boosting, and Extreme Gradient Boosting were employed to evaluate the feature importance influencing bond strength. Results FTIR and UV-Vis analyses indicated that 10-MDP promoted the hydrolysis and siloxane condensation of γ-MPTS, forming a stable organic-inorganic hybrid network. The combined group showed the highest initial (24.22 MPa) and aged (16.07 MPa) bond strength, with more cohesive failures. Machine learning confirmed the combined primer as the most critical factor. Conclusions The coexistence of 10-MDP and γ-MPTS in a single solvent environment provides a synergistic effect, significantly enhancing both the bonding strength and durability of glass-based ceramics. Clinical Relevance A combined 10-MDP/γ-MPTS primer stabilizes the adhesive interface and increases the durability of ceramic restorations for better long-term success.

Intrafibrillar mineralization plays an important role in dentin repair. Current research on intrafibrillar mineralization primarily focuses on the precise positioning of mineral precursors within collagen and the reduction of mineralization time. Inspired by the multifunctionality of noncollagenous proteins (NCPs), we developed a dual-analogue system, 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) followed by 3-(3,4-dihydroxyphenyl)-l-alanine (l-DOPA)-stabilized amorphous calcium phosphate (M + LA), which integrated a nucleation inhibitor l-DOPA and inducer 10-MDP. In this study, we investigated the impact of a preinfiltration strategy using the M + LA system on intrafibrillar mineralization. l-DOPA with a concentration of 250 μg mL–1, acting as a mussel-inspired nucleation inhibitor, was demonstrated to stabilize amorphous calcium phosphate (ACP) precursors within 2 h at 37 °C, with the formed precursors termed “LACP” (l-DOPA-stabilized amorphous calcium phosphate). Numerous LACP precursors were immobilized within collagen via the M + LA system, referred to as “preinfiltration.” The M + LA system exhibited no significant cytotoxicity, as evidenced by the biocompatibility evaluation through a modified trans-dentin disk model. Notably, the M + LA preinfiltration strategy significantly reduced intrafibrillar mineralization time to 3 h, superior to that of bare collagen and MDP-modified collagen. Furthermore, the resulting mineralized M + LA-modified collagen achieved functional mechanical properties. Moreover, 10-MDP-Ca nanolayering was generated via a bottom-up approach during the preinfiltration process. This study introduces a preinfiltration strategy for promoting intrafibrillar mineralization, which provides a reference for understanding the mechanism of intrafibrillar mineralization and shows promising clinical application prospects for dentin repair.

Objectives To evaluate the interactions of two phosphate ester monomers [10-methacryloyloxydecyl dihydrogen phosphate (10-MDP) and dipentaerythritol penta-acrylate phosphate (PENTA)] with hydroxyapatite and collagen and understand their influence on dentine bonding. Methods Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction, nuclear magnetic resonance, ultraviolet-visible, and molecular docking were applied for separately evaluating the interactions of two monomers with hydroxyapatite and collagen. Hydrophilicity tests and morphological observation were employed to characterize pretreated dentine. Microtensile bond strength (μTBS) and nanoleakage were investigated to evaluate the bonding performance. Hydroxyproline assay, in situ zymography, and matrix metalloproteinase-9 (MMP-9) activity assay were used to confirm the MMP inhibition. Results Chemoanalytic characterization confirmed the interactions of 10-MDP and PENTA with hydroxyapatite and collagen. The interactions of PENTA were weaker than 10-MDP. PENTA possessed better dentine tubule sealing after etching than 10-MDP. Dentine treated with PENTA was more hydrophilic than 10-MDP. 10-MDP and PENTA treating significantly increased the initial μTBS than the control group without primer conditioning. μTBS decreased significantly during aging, and the decrease was more severe in the PENTA group than 10-MDP. The 10-MDP and PENTA groups exhibited relatively less fluorescence than the control. The relative inhibition percentages of MMP-9 decreased in the order of 10-MDP–Ca salt, 10-MDP and PENTA. The 10-MDP, PENTA, and 10-MDP–Ca salt groups showed significantly lower hydroxyproline contents than the control. Conclusions Although PENTA adsorbed on hydroxyapatite, it did not form a stable calcium salt. The interactions of 10-MDP with hydroxyapatite and collagen are different than those of PENTA. Clinical significance The sealing of dentinal tubules by PENTA and the inhibition of MMP by 10-MDP and its calcium salts contribute to improving the dentine bonding durability.
