Quercetin boosts gut microbiota-driven production of isovanillic acid to alleviate colitis via enhancing intestinal barrier function

Liang Lei,Jing Wang,Juanjuan Wang,Wenjuan He,Tao Wu,Jing Li,Xiaobin Bi,Mei Mei,Xinlei Guan,Xiaoqiang Zhu

Ulcerative colitis (UC) is a severe inflammatory bowel disease marked by intestinal inflammation, compromised barrier function, and gut microbiota imbalance, with a restricted range of therapeutic options currently available. Quercetin, a flavonoid extracted from fruits and vegetables, have been shown significant anti-inflammatory and microbiota-modulating effects. However, the interactions between gut microbes and quercetin in colitis remain insufficiently elucidated. This research delved into the potential involvement of the gut microbiota-isovanillic acid (IVA)-intestinal barrier axis in the anti-colitis effects of quercetin via 16S rDNA sequencing and metabolomics. Quercetin administration effectively enhanced gut barrier integrity and mitigated colitis by boosting IVA production through modulating gut microbiota composition, particularly increasing Clostridium_XIVa and Clostridium_XI abundances. Notably, gut microbiota depletion with antibiotics (ABX) treatment markedly diminished IVA production and concurrently negated quercetin's positive effects on colitis. More importantly, IVA supplementation was also effective in alleviating colitis via enhancing gut barrier function, suggesting gut microbiota-derived IVA supported beneficial effects of quercetin on colitis. Our research highlighted the crucial involvement of gut microbial-derived IVA in the anti-colitis effects of quercetin and underscored its therapeutic potential, along with IVA, as a treatment for colitis and other intestinal inflammatory conditions.

Isolation, identification, and susceptibility testing of Staphylococcus caprae from dairy goats with mastitis to antibiotics and Chinese herbal medicines

Jie Luo,Yiping Wang,Wenya Zheng,Chaolong Xu,Jianhua Chen,Ben Liu,Xiaoyue Wang,Manxin Fang

Introduction Mastitis outbreaks in dairy goats at a farm in western Jiangxi Province, have recently imposed significant economic losses on farmers. This study aimed to identify the causative pathogen and implement targeted control measures for early disease eradication. Methods Pathological autopsy was performed on diseased goats submitted for examination. Pathological mammary gland tissues and milk cistern fluid were collected aseptically for bacterial isolation and purification. The isolated pathogen was characterized through colony morphology observation, Gram staining, biochemical tests, and 16S rDNA gene sequencing. Pathogenicity was assessed using an animal challenge test in mice. In vitro antibacterial susceptibility testing was conducted using 30 Antibiotics (e.g., cefradine, cephalexin, cefuroxime), 18 Chinese herbal medicines (CHMs; e.g., Rhus chinensis (Galla Chinensis), Coptis chinensis (Chinese Goldthread), Punica granatum (Granati Pericarpium)), and 15 phytogenic extracts (e.g., Berberine, Gallic acid, Tannic acid). Results A Gram-positive bacterium was isolated from the diseased tissues. Biochemical profiling and 16S rDNA sequencing identified the isolate as Staphylococcus caprae . The animal challenge test confirmed the isolate’s strong pathogenicity in mice. Susceptibility testing indicated that the isolate was susceptible to Antibiotics such as cefradine, cephalexin, cefuroxime, and ciprofloxacin, while showing the poorest susceptibility to penicillin among the tested antibiotics. Among the CHMs and phytogenic extracts, Rhus chinensis (Galla Chinensis), Coptis chinensis (Chinese Goldthread), Punica granatum (Granati Pericarpium), Berberine, Gallic acid and Tannic acid demonstrated superior antibacterial efficacy. Discussion This study provides multiple therapeutic and preventive options (CHMs, Antibiotics, phytogenic extracts) for mastitis control on this farm. It also offers valuable insights into the identification and treatment of mastitis in dairy goats, contributing to safeguarding goat health and ensuring dairy product safety.

Octenyl Succinic Anhydride-Modified Starch Attenuates Body Weight Gain and Changes Intestinal Environment of High-Fat Diet-Fed Mice

Jia-Ping Chen,Kuerbanjiang Maierheba,Ying Zhang,Hui Cheng,Binbin Lin,Pan Yue,Le-Hua Wang,Feng-Zhi Liu,Jun-Wen Shi,Zhong-Xiao Wan,Xiao-Ping Wang,Jianteng Xu,Li-Qiang Qin,Yanjie Bai

Effects of octenylsuccinate (OS) starch on body composition and intestinal environment in high-fat diet-fed mice were investigated. C57BL/6J mice were treated with a regular-fat (RF) diet, a high-fat (HF) diet, or a high-fat diet supplemented with OS starch (HFOSS). Fecal short-chain fatty acids (SCFAs) were quantified using gas chromatography, and the fecal microbiota profile was analyzed by 16S rDNA sequencing. One-way ANOVA and metastats analysis were performed for statistical analysis. After 22 weeks of feeding, mice in the HFOSS group had significantly lower body weight, body fat, liver weight, and cumulative food intake than those in the HF group but higher than that of the RF group. Fecal total SCFA, acetic, propionic, and butyric acid concentrations were significantly higher in the HFOSS group than that in the HF and RF groups. OS starch intervention increased the relative abundance ofParabacteroides,Alistipes, andRuminiclostridium_5and decreased that ofTyzzerella,Oscillibacter,Desulfovibrio, andAnaerotruncuscompared with the RF and HF groups. The relative abundance ofLachnospiraceae_UCG-006in the HFOSS group was lower than that in the HF group but higher than that in the RF group. In conclusion, OS starch prevents fat accumulation in high-fat diet-fed mice and might provide potential health benefits due to its fermentability in the gut and its ability to regulate gut microbial community structure. Keywords:octenylsuccinate starch;high-fat diet;fecal short-chain fatty acids;gut microbiota;gas chromatography;16S rDNA sequencing

Titanium dioxide nanoparticles drive the enhanced pro-inflammation response, worsening oxidative injure and gut microbiota dysbiosis in experimental colitis mice

Xiaoyan Feng,Tao You,Jiajun Guo,Hengyi Xu

With the wide use of titanium dioxide nanoparticles (TiO 2 NPs) in food products, inflammatory bowel disease (IBD) was inevitably to ingest it. In this work, we daily administered TiO 2 NPs to mice, followed by dextran sulfate sodium (DSS)-drinking for 7 days to obtain the colitis model. Excitedly, shorten colon length, earlier and increased weigh body loss, higher disease activity index score, as well as disorganized pathological structure were observed in colitis mice with TiO 2 NPs exposure. Moreover, the significant changes in overproduction of inflammatory cytokines and oxidative injure were detected in colon. 16S rDNA sequencing results showed that TiO 2 NPs broke the balance of gut microbiota, including decreased α-diversity index, reduced the total operational taxonomic units (OTUs) number and altered the community distribution and compositions. To find the key bacterial regulator, we further analyzed the abundance change of differential species, found that the relative abundance of short-chain fatty acid (SCFAs)-producing bacteria ( Muribaculaceae , Ruminococcus , Clostridia , etc.) noticeably reduced, while the relative abundance of pathogenic bacteria ( Gastranaerophilaceae , Helicobacter , Escherichia-Shigella , etc.) dramatically augmented. The mutual cooperation of reduced SCFAs and elevated inflammatory factors induced the form of inflammation-oxidative cascade cycle. Our work highlighted the risk assessment of dietary nanoparticles on the IBD population and identified the key microbial regulators, looking forward to provide the target therapeutic strategies for IBD affected by environmental factors.

Taxonomic structure and functional assembly of the broad-spectrum sulfonylurea herbicide-degrading microbial consortium L1 under different herbicide substrates

Xiang Li,Yumeng Dai,Xinyu Guan,Zhiyang Han,Xu Li,Xiujuan Wang,Zhencheng Su,Huiwen Zhang,Mingkai Xu

The overuse of sulfonylurea herbicides (SUHs) in agricultural environments has resulted in severe residues that damage ecosystems and harm human health. In this study, we described a microbial consortium L1, capable of broad-spectrum degradation of SUHs, which degraded 200   mg   L -1 of bensulfuron-methyl, chlorimuron-ethyl, metsulfuron-methyl, tribenuron-methyl and thifensulfuron-methyl by 81.27%, 97.59%, 62.34%, 72.06% and 100%, respectively, within 7 days. The 16S rDNA and metagenome sequencing results showed that the dominant genera in consortium L1 under different SUHs were concentrated in Methyloversatilis , Pseudoxanthomonas and Chitinophaga . However, the diversity and structure of consortium L1 varied significantly under different SUHs, reflecting differences in both abundant and rare taxa, which could also interconvert. KEGG annotations showed that consortium L1 had the same functional composition when degrading different SUHs, but the number of gene annotations was significantly different, with benzoate degradation (ko00362) being the most enriched of all treatments. Furthermore, the results showed that functional redundancy existed in degradation high contribution genera, with abundant taxa having a greater impact on the degradation of different SUHs. Additionally, network analysis revealed that both rare and abundant genera had more positive edges, suggesting that the strains engaged in more mutualistic or symbiotic cooperation during the degradation process. More importantly, rare taxa play an important role in connectivity between modules. Overall, the assembly of abundant and rare taxa communities and functional composition were closely related to broad-spectrum consortium degrading. These findings deepen the understanding of bacterial consortia and provide valuable insights to enhance their potential in environmental remediation.

Sinomenine hydrochloride ameliorates dextran sulfate sodium‑induced colitis in mice by modulating the gut microbiota composition whilst suppressing the activation of the NLRP3 inflammasome

Yan Zhou,Shuai Chen,Wenxian Gu,Xiao Sun,Linxiao Wang,Liming Tang

Sinomenine is a pure alkaloid that can be isolated from the root of Sinomenium acutum and has been found to exert anti‑inflammatory and immunosuppressive effects. The present study investigated the effects of sinomenine hydrochloride (SIN) on inflammation and the gut microbiota composition in the colon of mouse models of dextran sulfate sodium (DSS)‑induced colitis. DSS‑induced mice colitis was established by treating the mice with drinking water containing 3% (w/v) DSS for 7 days. The disease activity index of each mouse was calculated on a daily basis. All mice were sacrificed on day 11, then the weight of their spleen and length of their colons were measured. The histological analysis was measured by hematoxylin‑eosin staining. Oral administration of SIN (100 mg/kg/day) attenuated the DSS‑induced increases in the disease activity indices and spleen indices, DSS‑induced shortening of the colon length and histological damage. In addition, reverse transcription‑quantitative PCR data showed that SIN treatment effectively regulated the expression of inflammatory mediators, specifically by suppressing the expression of proinflammatory gene (TNF‑α, IL‑6 and inducible nitric oxide synthase) whilst increasing those associated with inhibiting inflammation (IL‑10 and arginine 1). Gut microbiota analysis was conducted using 16S ribosomal DNA sequencing. The results revealed that SIN improved bacterial community homeostasis and diversity, which were damaged by DSS. Furthermore, western blotting showed that the activation of the NOD‑, LRR‑ and pyrin domain‑containing protein 3 (NLRP3) inflammasome was markedly suppressed by SIN treatment. In conclusion, these results indicated that SIN may ameliorate experimental colitis by modulating the gut microbiota composition and suppressing the activation of the NLRP3 inflammasome in mice. Overall, these findings suggested a broad protective effect of SIN in treating inflammatory gut diseases, including ulcerative colitis.

Fecal metabonomics combined with 16S rDNA sequencing to analyze the changes of gut microbiota in rats fed with different protein source diets

Zhuang Kejin,Meng Weihong,Shu Xin,Liang Defu,Wang Lidong,Zhang Dongjie

Purpose When blended, animal and plant proteins can complement each other in terms of amino acid composition and release time. In this study, we investigated whether the blended protein diet has a better feeding effect than the single protein diet, and to reveal the differences in growth and intestinal microbiota composition caused by the blended protein diet. Methods Forty Sprague Dawley (SD) rats received diets with different protein sources, including casein (C), whey protein (WP), black soybean protein (BSP), and black soybean-whey blended protein (BS-WP), for eight weeks. To investigate the effects of blended protein supplement on gut microbiota and metabolites, we performed a high throughput 16S rDNA sequencing and fecal metabolomics profiling. In addition, we determined growth and serum biochemical indices, and conducted intestinal morphology analyses. Results Compared to those in the BSP and WP groups, the daily body weight gain and feed conversion efficiency increased in the BS-WP group. Serum biochemical indices indicated that the protein utilization efficiency of the WP and BS-WP groups was relatively high, and the BS-WP blended protein diet improved the protein adoption rate. The BS-WP blended protein diet also improved intestinal tissue morphology and promoted intestinal villi development compared to the single protein diets. Furthermore, dietary protein altered the composition of gut microbiota, the gut microbial diversity of rats fed with the BS-WP diet was significantly ( P  < 0.05) higher than that of the other groups. The difference in dietary protein corresponded with an alteration of fecal amino acids and their metabolites, and tryptophan and tyrosine metabolism were the key mechanisms leading to the changes in fecal microbial composition. Conclusion Dietary protein sources played an important role in the growth and development of rats by influencing intestinal metabolism and microbial composition. The BS-WP blended protein diet was more conducive to nutrient absorption than the single protein diet. Furthermore, blended protein increased the diversity of intestinal microbes and aided the establishment of intestinal barrier function.

Modulation of cellular metabolism and alleviation of bacterial dysbiosis by Aconiti Lateralis Radix Praeparata in non-small cell lung cancer treatment

Wen Zhang,Menglei Ding,Yaru Feng,Shuhui Cai,Zichen Luo,Jinjun Shan,Liuqing Di

ABSTRACT Background Non-small cell lung cancer (NSCLC) is a highly prevalent and fatal form of lung cancer. In China, Aconiti Lateralis Radix Praeparata ( Fuzi in Chinese), derived from the lateral root of Aconitum carmichaeli Debx. (Ranunculaceae, Aconitum ), is extensively prescribed to treat cancer in traditional medicine and clinical practice. However, the precise mechanism by which Fuzi treats NSCLC remains unknown. Purpose This article aims to assess the efficacy of Fuzi against NSCLC and elucidate its underlying mechanism. Methods Marker ingredients of Fuzi decoction were quantified using UPLC-TSQ-MS. The effectiveness of Fuzi on NSCLC was evaluated using a xenograft mouse model. Subsequently, a comprehensive approach involving network pharmacology, serum metabolomics, and 16S rDNA sequencing was employed to investigate the anti-NSCLC mechanism of Fuzi . Results Pharmacological evaluation revealed significant tumour growth inhibition by Fuzi , accompanied by minimal toxicity. Network pharmacology identified 29 active Fuzi compounds influencing HIF-1, PI3K/Akt signaling, and central carbon metabolism in NSCLC. Integrating untargeted serum metabolomics highlighted 30 differential metabolites enriched in aminoacyl-tRNA biosynthesis, alanine, aspartate, and glutamate metabolism, and the tricarboxylic acid (TCA) cycle. Targeted serum metabolomics confirmed elevated glucose content and reduced levels of pyruvate, lactate, citrate, α-ketoglutarate, succinate, fumarate, and malate following Fuzi administration. Furthermore, 16S rDNA sequencing assay showed that Fuzi ameliorated the dysbiosis after tumorigenesis, decreased the abundance of Proteobacteria, and increased that of Firmicutes and Bacteriodetes. PICRUSt analysis revealed that Fuzi modulated the pentose phosphate pathway of the gut microbiota. Spearman correlation showed that Proteobacteria and Escherichia_Shigella accelerated the TCA cycle, whereas Bacteroidota , Bacteroides, and Lachnospiraceae_NK4A136_group suppressed the TCA cycle. Conclusions This study firstly introduces a novel NSCLC mechanism involving Fuzi , encompassing energy metabolism and intestinal flora. It clarifies the pivotal role of the gut microbiota in treating NSCLC and modulating the TCA cycle. Moreover, these findings offer valuable insights for clinical practices and future research of Fuzi against NSCLC.

Integrating 16S rDNA and metabolomics to uncover the therapeutic mechanism of electroacupuncture in type 2 diabetic rats

Zhang Yue,Wang Xiang,Deng Duping,Gong Yuanyuan,Chen Xuanyi,Li Juan,Hong Xiaojuan

Objective: This study aimed to investigate the impact of electroacupuncture (EA) on blood glucose levels, gut microbiota, short-chain fatty acids (SCFAs), and glucagon-like peptide-1 (GLP-1) in a rat model of type 2 diabetes mellitus (T2DM).Methods: Forty Sprague–Dawley (SD) rats were randomly assigned to five groups (n = 8/group) using a random number table: normal control, T2DM model, electroacupuncture (EA), EA + antibiotics (EA + A), and antibiotics (A). The normal rats received a standard diet and saline gavage, while the other groups were fed a high-fat diet and emulsion. The EA + A and A groups received additional antibiotic solution gavage. The normal, model, and A groups were immobilized and restrained for 30 min, six times per week, for 4 weeks. The EA and EA + A groups received EA treatment at specific acupoints for 30 min, six times per week, for 4 weeks. EA parameters were continuous waves at 10 Hz and 1–2 mA. During the intervention, water and food consumption, body weight, fasting blood glucose (FBG), and oral glucose tolerance test (OGTT) were monitored. Pancreatic tissue was examined using hematoxylin and eosin (H&E) staining. Fecal microbial communities were analyzed by 16S rDNA sequencing, and short-chain fatty acids (SCFAs) were measured using gas chromatography–mass spectrometry (GC–MS). Serum levels of fasting insulin (FINS), glycated hemoglobin (HbA1c), and glucagon-like peptide-1 (GLP-1) were determined using enzyme-linked immunosorbent assay (ELISA).Results: EA significantly improved daily water intake, food consumption, and body weight in T2DM rats (p < 0.01). EA also reduced FBG, the area under the curve of the OGTT, FINS, and homeostasis model assessment of insulin resistance (HOMA-IR) in T2DM rats (p < 0.05). The ELISA results showed a lower concentration of HbA1c in the EA group (p < 0.05). EA improved the overall morphology and area of pancreatic islets, increased the number of β-cell nuclei, and alleviated β-cell hypertrophy. The abundance of operational taxonomic units (OTUs) in the EA group increased than the model group (p < 0.05), and EA upregulated the Shannon, Chao1, and Ace indices (p < 0.05). EA increased the concentrations of acetic acid, butyric acid, and GLP-1 (p < 0.05). Correlation analysis revealed negative associations between Lactobacillaceae (R = −0.81, p = 0.015) and Lactobacillus (R = −0.759, p = 0.029) with FBG. Peptostreptococcaceae and Romboutsia were negatively correlated with HbA1c (R = −0.81, p = 0.015), while Enterobacteriaceae was positively correlated with OGTT (R = 0.762, p = 0.028). GLP-1 was positively correlated with acetic acid (R = 0.487, p = 0.001), butyric acid (R = 0.586, p = 0.000), isovaleric acid (R = 0.374, p = 0.017), valeric acid (R = 0.535, p = 0.000), and caproic acid (R = 0.371, p = 0.018). Antibiotics disrupted the intestinal microbiota structure and weakened the therapeutic effects of EA.Conclusion: EA effectively improved glucose metabolism in T2DM rats. The hypoglycemic effects of EA were associated with the regulation of gut microbiota, SCFAs, and GLP-1.

Elucidation of the Reinforcing Spleen Effect of Jujube Fruits Based on Metabolomics and Intestinal Flora Analysis

Yi Yan-ling,Li Yao,Guo Sheng,Yan Hui,Ma Xin-fei,Tao Wei-wei,Shang Er-xin,Niu Yang,Qian Da-wei,Duan Jin-ao

Jujube (Ziziphus jujuba Mill.) fruit (JF) is widely consumed as food in Asian countries due to its potential effects for human health. As a traditional Chinese medicine, JF is often used to treat anorexia, fatigue and loose stools caused by spleen deficiency syndromes in China, but the mechanism underlying this effect has not been thoroughly elucidated. In this study, a rat model of spleen deficiency syndromes was adopted to investigate the therapeutic effect of JF extract and its possible mechanism by metabolomics analyses of plasma and urine as well as the intestinal flora analysis. The results showed that the changes in plasma and urine metabolites caused by spleen deficiency were reversed after administration of JF, and these changed endogenous metabolites were mainly involved in retinol metabolism, pentose and glucuronate interconversions, nicotinate and niacinamide metabolism pathways. The 16S rDNA sequencing results showed that JF could regulate intestinal flora imbalance caused by spleen deficiency. The covariance analysis of intestinal flora structure and metabolome indicated that Aerococcus may be a candidate strain for predicting and treating the metabolic pathways of spleen deficiency and related disorders. In summary, it can be revealed that spleen deficiency, which alters metabolic profiles and the intestinal flora, could be alleviated effectively by JF extract.

Sodium Butyrate-Loaded Microspheres With Enhanced Bioavailability for Targeted Treatment of Intestinal Barrier Injury

Yanwei Lv,Mengni Jiang,Yongliang Ouyang,Xiaoyi Zheng,Liang Zhang,Jinpeng Yao,Lianghao Hu,Jiulong Zhao,Zhaoshen Li,Shige Wang

Intestinal barrier dysfunction is related to diseases such as inflammatory bowel disease (IBD) and severe acute pancreatitis (SAP). Butyrate and its derivatives (e.g., sodium butyrate (SB)) can alleviate gut inflammatory responses. Nevertheless, these substances usually cannot fully exert protective effects due to low bioavailability. This research aimed to offer microspheres for treating intestinal barrier injury. Sodium alginate solution is prepared to dissolve SB, followed by mixing with chitosan (CS)-protocatechuic aldehyde (PA)/calcium chloride solution. The required CS-PA/calcium alginate/SB (CPC/SB) microspheres are formed in this manner. Subsequently, the therapeutic effects of CPC/SB microspheres on intestinal barrier injury through in vivo dextran sulfate sodium salt (DSS)-induced IBD and sodium taurocholate (STC)-induced SAP models is explored. Results: The CPC/SB microspheres exhibited excellent antioxidant properties. In vivo bioluminescence imaging experiment confirmed the microspheres effectively targeted the inflammatory gut in IBD. Further in vivo experimental results indicated the microspheres significantly repaired intestinal barrier damage, exerting protective effects in IBD and SAP. Additionally, 16S rDNA sequencing explained the microspheres can regulate the balance between harmful and beneficial bacteria (such as Alistipes, Odoribacter, and Rikenellaceae RC9). This study provides a possible synthetic strategy of microsphere carriers to serve as a potential therapeutic tool for intestinal barrier injury.

Deep-fried Atractylodes lancea rhizome alleviates spleen deficiency diarrhea–induced short-chain fatty acid metabolic disorder in mice by remodeling the intestinal flora

Chunlian Liu,Chengcheng Song,Yan Wang,Yangxin Xiao,Zhongshi Zhou,Guosheng Cao,Xiongjie Sun,Yanju Liu

Ethnopharmacological relevance Atractylodes lancea (Thunb.) DC. is a Chinese herb that has been commonly used to treat spleen-deficiency diarrhea (SDD) in China for over a thousand years. However, the underlying mechanism of its antidiarrheal activity is not fully understood. Aim of the study The antidiarrheal effects of the ethanol extract of deep-fried A. lancea rhizome (EEDAR) due to spleen deficiency induced by folium sennae (SE) were determined on the regulation of the short-chain fatty acid (SCFA) metabonomics induced by the intestinal flora . Materials and methods The effects of EEDAR on a SE-induced mouse model of SDD were evaluated by monitoring the animal weight, fecal water content, diarrhea-grade rating, goblet cell loss, and pathological changes in the colon. The expression of inflammatory factors (tumor necrosis factor [TNF]-α, interleukin [IL]-1β, IL-6, IL-10), aquaporins (AQP3, AQP4, and AQP8), and tight junction markers (ZO-1, occludin , claudin-1) in colon tissues were determined using quantitative polymerase chain reaction and western blotting . SCFA metabonomics in the feces of mice treated with EEDAR was evaluated using gas chromatography–mass spectrometry. Furthermore, 16S rDNA sequencing was used to determine the effect of EEDAR on the intestinal flora of SDD mice, and fecal microbiota transplantation (FMT) was used to confirm whether the intestinal flora was essential for the anti-SDD effect of EEDAR. Results Treatment with EEDAR significantly improved the symptoms of mice with SDD by inhibiting the loss of colonic cup cells, alleviating colitis , and promoting the expression of AQPs and tight junction markers. More importantly, the effect of EEDAR on the increase of SCFA content in mice with SDD was closely related to the gut microbiota composition. EEDAR intervention did not significantly improve intestinal inflammation or the barrier of germ-free SDD mice, but FMT was effective. Conclusion EEDAR alleviated SE-induced SDD in mice, as well as the induced SCFA disorder by regulating the imbalance of the intestinal microbiota.
