Uranium-stressed soil caused by nuclear industry development and energy acquisition have attracted extensive attentions for a long time. This study investigated the effects of biochar application with different pyrolysis temperatures (300 ℃, 500 ℃ and 700 ℃) on remediation of uranium-stressed soil. The results showed that higher pyrolysis temperature (700 ℃) was benefit for ryegrass growing and caused a lower uranium accumulation in plants. At the same time, uranium immobilization was more effective at higher biochar pyrolysis temperature. Careful investigations indicated that activities of soil urease and sucrase were promoted, and bacterial diversity was strengthened in C700 group, which may contribute to uranium immobilization. The biochar application could activate metabolic of lipids and amino acids , organic acids and derivatives, and organic oxygen compounds. Nicotinate and nicotinamide metabolism, and Benzoxazinoid biosynthesis were unique metabolic pathways in the C700 group, which could enhance the uranium tolerance from different perspectives. Based on these results, we recommend to use biochar with 700 °C pyrolysis temperature when processing remediation of uranium-stressed soil. This study will facilitate the implementation of biochar screening and provide theoretical helps for remediation of uranium-stressed soil.

The simple and efficient detection of nucleic acids is important in the diagnosis of tuberculosis (TB) caused by Mycobacterium tuberculosis (M. tuberculosis). However, base mismatch will lead to false positive and false negative nucleic acid test, which seriously interferes with the accuracy of the final results. Herein, we demonstrated a CRISPR/Cas-9-mediated fluorescent strategy utilizing fluorescence resonance energy transfer (FRET) for the detection of bacteria. High-variable region of M. tuberculosis 16S rDNA fragment was used as the target, and CRISPR/Cas9 was used as the recognition element. The binding of the P1 probe of upconversion nanoparticles (UCNPs) @SiO2-P1 and the P2 probe of Fe3O4@Au-P2 caused the fluorescence quenching of UCNPs. In the presence of the target, the P2 probe hybridized with the target to form double-stranded DNA (dsDNA), which was recognized and cleaved by CRISPR/Cas9, resulting in the breaking of the P1–P2 duplex linkage. UCNPs moved away from Fe3O4@Au under a magnetic field, and the fluorescence signal was restored; bacteria were detected under the excitation of a 980 nm laser source. Using the CRISPR/Cas-9-mediated system, the sensor could distinguish single-base mismatches in 10 bases from the protospacer adjacent motif (PAM) region. The limit of detection (LOD) was 20 CFU mL−1 and the detection time was 2 h. It developed a new way of accurate nucleic acid detection for disease diagnosis.

Objective: This study aimed to investigate the impact of electroacupuncture (EA) on blood glucose levels, gut microbiota, short-chain fatty acids (SCFAs), and glucagon-like peptide-1 (GLP-1) in a rat model of type 2 diabetes mellitus (T2DM).Methods: Forty Sprague–Dawley (SD) rats were randomly assigned to five groups (n = 8/group) using a random number table: normal control, T2DM model, electroacupuncture (EA), EA + antibiotics (EA + A), and antibiotics (A). The normal rats received a standard diet and saline gavage, while the other groups were fed a high-fat diet and emulsion. The EA + A and A groups received additional antibiotic solution gavage. The normal, model, and A groups were immobilized and restrained for 30 min, six times per week, for 4 weeks. The EA and EA + A groups received EA treatment at specific acupoints for 30 min, six times per week, for 4 weeks. EA parameters were continuous waves at 10 Hz and 1–2 mA. During the intervention, water and food consumption, body weight, fasting blood glucose (FBG), and oral glucose tolerance test (OGTT) were monitored. Pancreatic tissue was examined using hematoxylin and eosin (H&E) staining. Fecal microbial communities were analyzed by 16S rDNA sequencing, and short-chain fatty acids (SCFAs) were measured using gas chromatography–mass spectrometry (GC–MS). Serum levels of fasting insulin (FINS), glycated hemoglobin (HbA1c), and glucagon-like peptide-1 (GLP-1) were determined using enzyme-linked immunosorbent assay (ELISA).Results: EA significantly improved daily water intake, food consumption, and body weight in T2DM rats (p < 0.01). EA also reduced FBG, the area under the curve of the OGTT, FINS, and homeostasis model assessment of insulin resistance (HOMA-IR) in T2DM rats (p < 0.05). The ELISA results showed a lower concentration of HbA1c in the EA group (p < 0.05). EA improved the overall morphology and area of pancreatic islets, increased the number of β-cell nuclei, and alleviated β-cell hypertrophy. The abundance of operational taxonomic units (OTUs) in the EA group increased than the model group (p < 0.05), and EA upregulated the Shannon, Chao1, and Ace indices (p < 0.05). EA increased the concentrations of acetic acid, butyric acid, and GLP-1 (p < 0.05). Correlation analysis revealed negative associations between Lactobacillaceae (R = −0.81, p = 0.015) and Lactobacillus (R = −0.759, p = 0.029) with FBG. Peptostreptococcaceae and Romboutsia were negatively correlated with HbA1c (R = −0.81, p = 0.015), while Enterobacteriaceae was positively correlated with OGTT (R = 0.762, p = 0.028). GLP-1 was positively correlated with acetic acid (R = 0.487, p = 0.001), butyric acid (R = 0.586, p = 0.000), isovaleric acid (R = 0.374, p = 0.017), valeric acid (R = 0.535, p = 0.000), and caproic acid (R = 0.371, p = 0.018). Antibiotics disrupted the intestinal microbiota structure and weakened the therapeutic effects of EA.Conclusion: EA effectively improved glucose metabolism in T2DM rats. The hypoglycemic effects of EA were associated with the regulation of gut microbiota, SCFAs, and GLP-1.

Purpose When blended, animal and plant proteins can complement each other in terms of amino acid composition and release time. In this study, we investigated whether the blended protein diet has a better feeding effect than the single protein diet, and to reveal the differences in growth and intestinal microbiota composition caused by the blended protein diet. Methods Forty Sprague Dawley (SD) rats received diets with different protein sources, including casein (C), whey protein (WP), black soybean protein (BSP), and black soybean-whey blended protein (BS-WP), for eight weeks. To investigate the effects of blended protein supplement on gut microbiota and metabolites, we performed a high throughput 16S rDNA sequencing and fecal metabolomics profiling. In addition, we determined growth and serum biochemical indices, and conducted intestinal morphology analyses. Results Compared to those in the BSP and WP groups, the daily body weight gain and feed conversion efficiency increased in the BS-WP group. Serum biochemical indices indicated that the protein utilization efficiency of the WP and BS-WP groups was relatively high, and the BS-WP blended protein diet improved the protein adoption rate. The BS-WP blended protein diet also improved intestinal tissue morphology and promoted intestinal villi development compared to the single protein diets. Furthermore, dietary protein altered the composition of gut microbiota, the gut microbial diversity of rats fed with the BS-WP diet was significantly ( P < 0.05) higher than that of the other groups. The difference in dietary protein corresponded with an alteration of fecal amino acids and their metabolites, and tryptophan and tyrosine metabolism were the key mechanisms leading to the changes in fecal microbial composition. Conclusion Dietary protein sources played an important role in the growth and development of rats by influencing intestinal metabolism and microbial composition. The BS-WP blended protein diet was more conducive to nutrient absorption than the single protein diet. Furthermore, blended protein increased the diversity of intestinal microbes and aided the establishment of intestinal barrier function.

The addition of soil amendments to facilitate plant-based remediation of soil contaminated with radioactive nuclides is considered a promising approach. Here, we tested different levels of biochar to help clean uranium-contaminated soil in the potted plants. Adding 1% biochar had the best results in deactivating uranium, increasing soil enzyme activity, and promoting ryegrass growth. Microbiological and metabolomic analysis further revealed that 1 wt % biochar significantly enhanced the abundance of microorganisms such as Actinobacteriota and Myxococcota and accelerated the production of differential metabolites such as lipids and lipid-like molecules, organic acids and derivatives, and organic oxygen compounds. The analysis of biological and nonbiological interaction networks indicates that the coordinated interaction between bacteria, enzymes, and metabolites significantly improves the expression level of the ABC transporter’s metabolic pathway. This enhances the resistance of living cells to uranium and maintains system homeostasis under uranium stress. This study provides an example of the application of biochar-assisted phytoremediation and offers theoretical guidance for the remediation of soil contaminated with radioactive nuclides.

Lactoferrin is a natural multifunctional glycoprotein with potential antidepressant-like effects. However, the mechanism of its antidepressant effect has not been explored from the perspective of gut flora metabolism. Therefore, we employed both 16S rDNA gene sequencing and LC–MS metabolomics analysis to investigate the regulatory effects and mechanisms of lactoferrin in a rat model of depression. After one week of acclimatization, twenty-four 7-week-old male Sprague–Dawley rats were randomly and equally assigned into three groups: the control group, the model group, and the lactoferrin intervention group. The control group rats were housed under standard conditions, while the rats in the model and lactoferrin intervention groups were individually housed and exposed to chronic unpredictable mild stress for 44 days simultaneously. The lactoferrin intervention group was provided with water containing 2% lactoferrin (2 g/100 ml). Behavioural tests were conducted at week 7. Upon completion of the behavioral tests, the rats were anesthetized with isoflurane, humanely euthanized using a rat guillotine, and tissue samples were collected for further experiments. The results indicated that lactoferrin intervention led to an increase in sucrose solution consumption, horizontal movement distance, number of cross platforms, and residence time in the target quadrant. Additionally, it resulted in an increase in jejunal tight junction protein ZO-1 expression and a suppression of serum expression of inflammatory factors, Lipopolysaccharide and Diamine oxidase. In summary, lactoferrin can regulate the metabolic disorder of intestinal flora, reduce intestinal permeability, and further regulate the metabolic balance of hippocampal tissues through the microbiota-gut-brain axis. This process ultimately alleviates the depression-like behavior in rats.

As a radioactive heavy metal element with a long half-life, uranium causes environmental pollution when it enters the surrounding soil. This study analyzed the changes about soil enzyme activity , non-targeted metabolomics, microbial community structure and function microbial community structure and function to assess the differences in the effects of uranium stress on rhizosphere and non-rhizosphere soil. Results showed that uranium stress significantly inhibited the activities of urease and sucrase in rhizosphere and non-rhizosphere, which had less effect on rhizosphere. Compare to the non-rhizosphere soil, the uranium stress induced the production of gibberellin A1, to promoted several metabolic pathways, such as nitrogen and PTS (Phosphotransferase system) metabolic in rhizosphere soil. The species and abundance of Aspergillus , Acidobacter , and Synechococcus in both rhizosphere and non-rhizosphere soil were decreased by uranium stress. However, the microorganisms in rhizosphere soil were less inhibited according to the soil metabolism and microbial network map analysis. Furthermore, the Chujaibacter in rhizosphere soil under uranium stress was found significantly positively correlated with lipid and organic oxygen compounds. Overall, the results indicated that ryegrass roots significantly alleviated the effects of uranium stress on soil microbial activity and population abundances, thus playing a protective role. The study also provided a theoretical basis for in-depth understanding of the biological effects, prevention and control mechanisms of uranium-contaminated soil.

Objectives To explore the mechanism underlying the effect of Fructus Akebiae (FAE) against hepatic fibrosis in mice through combined network pharmacology, liver metabolomics, and 16S rDNA analyses of the gut microbiota. Methods In this study, we randomly divided mice into the control, model, FAE high-dose, FAE medium-dose, and FAE low-dose groups to analyze the pathological changes in the hepatic fibrosis and levels of the α -SMA, collagen 1, Nuclear Factor Kappa B (NF- κ B), Toll Like Receptor 4 (TLR4). The gut microbiota was analyzed through 16S rDNA sequencing analysis of liver metabolites using liquid chromatography-mass spectrometry. Furthermore, network pharmacology was used to determine the specific molecular regulation mechanism of FAE in hepatic fibrosis treatment. Results FAE treatment markedly improved the pathological changes in the hepatic fibrosis. Analysis revealed that FAE administration reversed the carbon tetrachloride (CCl 4 )-induced dysbiosis by increasing the abundance of Akkermansia and reducing that of Cyanobacteria . Additionally, metabolomic analysis showed that FAE treatment reversed the CCl 4 -induced metabolic disorders by regulating amino and nucleotide sugar metabolism. Furthermore, correlation analysis showed that Akkermansia and Verrucomicobiota were closely related to D-tolasaccharide and maltotetraose saccharide. Moreover, network pharmacology indicated that FAE might regulate the signaling pathway through the JUN/CASP3/NOS3/PTGS2/HSP90AA1 during treatment. Conclusion FAE may be a promising treatment for hepatic fibrosis, and its protective effects are associated with improvements in the microbiome and metabolic disorders.

Xiexin Tang (XXT) is a classic Chinese medicine formula that can be used to treat Atherosclerosis (AS). This study aimed to investigate the mechanism by which XXT regulated AS lipid levels. Firstly, the mixture components of XXT were analyzed by High-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Then, the AS model based on Apolipoprotein E knockout (ApoE -/- ) mice was established. Cytokines related to lipid metabolism and bile acid metabolism were detected by Quantitative Real-time PCR (qRT-PCR). 16S rDNA gene sequencing was performed to analyze differential bacterial populations, and the mechanism of XXT regulation of bile acids affecting lipid metabolism was further explored by targeted metabolomics. Further, antibiotic-treated mice were used to investigate the role of gut microbiota in the anti-AS effect of XXT. The results showed that XXT attenuated the lipid levels and reversed the abnormal elevation of cytokines, such as hepatic lipid metabolism and inflammatory reaction in AS mice. XXT also repaired the gut barrier damage and reversed gut microbiota disorders in AS mice. Furthermore, the metabolic levels of bile acids were reshaped by XXT. Whereas, in the absence of gut microbiota, XXT failed to attenuate lipid levels and inhibit the expression of cytokines related to inflammation and bile acid metabolism in AS mice and failed to play a role in ultimately treating AS. In conclusion, XXT could effectively inhibit the inflammatory reaction and lipid accumulation in AS mice, and this effect was closely related to its remodeling of gut microbiota to regulate bile acid metabolism.

Ulcerative colitis (UC) is a severe inflammatory bowel disease marked by intestinal inflammation, compromised barrier function, and gut microbiota imbalance, with a restricted range of therapeutic options currently available. Quercetin, a flavonoid extracted from fruits and vegetables, have been shown significant anti-inflammatory and microbiota-modulating effects. However, the interactions between gut microbes and quercetin in colitis remain insufficiently elucidated. This research delved into the potential involvement of the gut microbiota-isovanillic acid (IVA)-intestinal barrier axis in the anti-colitis effects of quercetin via 16S rDNA sequencing and metabolomics. Quercetin administration effectively enhanced gut barrier integrity and mitigated colitis by boosting IVA production through modulating gut microbiota composition, particularly increasing Clostridium_XIVa and Clostridium_XI abundances. Notably, gut microbiota depletion with antibiotics (ABX) treatment markedly diminished IVA production and concurrently negated quercetin's positive effects on colitis. More importantly, IVA supplementation was also effective in alleviating colitis via enhancing gut barrier function, suggesting gut microbiota-derived IVA supported beneficial effects of quercetin on colitis. Our research highlighted the crucial involvement of gut microbial-derived IVA in the anti-colitis effects of quercetin and underscored its therapeutic potential, along with IVA, as a treatment for colitis and other intestinal inflammatory conditions.

Introduction Mastitis outbreaks in dairy goats at a farm in western Jiangxi Province, have recently imposed significant economic losses on farmers. This study aimed to identify the causative pathogen and implement targeted control measures for early disease eradication. Methods Pathological autopsy was performed on diseased goats submitted for examination. Pathological mammary gland tissues and milk cistern fluid were collected aseptically for bacterial isolation and purification. The isolated pathogen was characterized through colony morphology observation, Gram staining, biochemical tests, and 16S rDNA gene sequencing. Pathogenicity was assessed using an animal challenge test in mice. In vitro antibacterial susceptibility testing was conducted using 30 Antibiotics (e.g., cefradine, cephalexin, cefuroxime), 18 Chinese herbal medicines (CHMs; e.g., Rhus chinensis (Galla Chinensis), Coptis chinensis (Chinese Goldthread), Punica granatum (Granati Pericarpium)), and 15 phytogenic extracts (e.g., Berberine, Gallic acid, Tannic acid). Results A Gram-positive bacterium was isolated from the diseased tissues. Biochemical profiling and 16S rDNA sequencing identified the isolate as Staphylococcus caprae . The animal challenge test confirmed the isolate’s strong pathogenicity in mice. Susceptibility testing indicated that the isolate was susceptible to Antibiotics such as cefradine, cephalexin, cefuroxime, and ciprofloxacin, while showing the poorest susceptibility to penicillin among the tested antibiotics. Among the CHMs and phytogenic extracts, Rhus chinensis (Galla Chinensis), Coptis chinensis (Chinese Goldthread), Punica granatum (Granati Pericarpium), Berberine, Gallic acid and Tannic acid demonstrated superior antibacterial efficacy. Discussion This study provides multiple therapeutic and preventive options (CHMs, Antibiotics, phytogenic extracts) for mastitis control on this farm. It also offers valuable insights into the identification and treatment of mastitis in dairy goats, contributing to safeguarding goat health and ensuring dairy product safety.

Effects of octenylsuccinate (OS) starch on body composition and intestinal environment in high-fat diet-fed mice were investigated. C57BL/6J mice were treated with a regular-fat (RF) diet, a high-fat (HF) diet, or a high-fat diet supplemented with OS starch (HFOSS). Fecal short-chain fatty acids (SCFAs) were quantified using gas chromatography, and the fecal microbiota profile was analyzed by 16S rDNA sequencing. One-way ANOVA and metastats analysis were performed for statistical analysis. After 22 weeks of feeding, mice in the HFOSS group had significantly lower body weight, body fat, liver weight, and cumulative food intake than those in the HF group but higher than that of the RF group. Fecal total SCFA, acetic, propionic, and butyric acid concentrations were significantly higher in the HFOSS group than that in the HF and RF groups. OS starch intervention increased the relative abundance ofParabacteroides,Alistipes, andRuminiclostridium_5and decreased that ofTyzzerella,Oscillibacter,Desulfovibrio, andAnaerotruncuscompared with the RF and HF groups. The relative abundance ofLachnospiraceae_UCG-006in the HFOSS group was lower than that in the HF group but higher than that in the RF group. In conclusion, OS starch prevents fat accumulation in high-fat diet-fed mice and might provide potential health benefits due to its fermentability in the gut and its ability to regulate gut microbial community structure. Keywords:octenylsuccinate starch;high-fat diet;fecal short-chain fatty acids;gut microbiota;gas chromatography;16S rDNA sequencing
