212 Pb serves as one of the most important nuclides for targeted alpha therapy (TAT), with good potential in the treatment of malignant tumors. However, medical and clinical applications are seriously limited by its extremely short supply, which strongly depends on the acquisition of its parent nuclides such as 228 Ra or 224 Ra. To address this issue, we conducted a systematic investigation on the separation of trace radium from natural thorium using the ion exchange chromatography method, and a novel porous silica-based composite resin functionalized with sulfonic acid groups was prepared, characterized, and employed. Experimental results showed that the silica-supported resin was successfully prepared with the sulfonic acid groups, and possessed small particle size, mesoporous pore structure and large specific surface area. The resin exhibited high selectivity toward thorium compared with barium, and the separation factor reached 123 at low acidity and at low temperature. The resin demonstrated a fast adsorption speed and an excellent separation performance, and the adsorption mechanism was confirmed based on the ion exchange between the sodium carried and the target cations. On the basis, column experiments were further conducted and finally the separation of trace radium from thorium was realized using a small column filled with about 1.0 g resin. The chemical recovery of radium reached 97 %, and the decontamination factor for thorium exceeded 40,000. This work verifies the feasibility of directly extracting 228 Ra and 224 Ra from natural thorium using the sulfonic acid group functionalized materials in the dilute nitric acid solution, and presents an excellent material and systematic experimental data on the separation activities, rendering this study particularly significant.

Targeted alpha therapy (TAT) based on 212Bi has demonstrated potent antitumor activity in preclinical studies. Its short half-life (∼60.5 min) facilitates patient management, yet presents challenges in radiopharmaceutical preparation. 212Pb, the parent nuclide of 212Bi (∼60.5 min), possesses a more practical half-life (∼10.6 h) but is limited by off-target toxicity associated with the redistribution of secondary radioactivity. To address this, we developed an in vivo 212Pb/212Bi generator based on nanohydroxyapatite (nHAP) for the treatment of pulmonary metastases. By leveraging the dense crystal lattice of nHAP to anchor 212Pb/212Bi, we aimed to enhance the radioactivity retention in the targeted organ and reduce systemic toxicity associated with endoradiotherapy. The synthesized 212Pb-nHAP@DOPA exhibited a satisfactory radiochemical yield (68.02 ± 0.25%), excellent in vitro stability, and potent cytotoxicity against cancer cells. Biodistribution studies confirmed significant lung accumulation of 212Pb-nHAP@DOPA with reduced renal uptake. Therapeutic evaluation showed that repeated high-dose treatment (740 kBq × 3) effectively suppressed tumor growth but caused severe systemic toxicity, while lowering the dose to 555 kBq with 3-day intervals could alleviate renal damage of tumor-bearing mice and maintain antitumor efficacy. Nevertheless, persistent immune suppression underscores the need for further carrier optimization to better control hematologic and skeletal toxicity.

212Pb has become a critical parent isotope for 212Bi, which is an alpha-emitting radionuclide utilized in targeted alpha therapy (TAT) for oncology; however, its limited availability impedes broader clinical use. To tackle this significant challenge, this study focuses on a strategic approach to separate 228Th (a precursor to 212Pb) from its parent isotope 228Ra within the decay chain of naturally occurring 232Th. A novel porous silica composite, functionalized with quaternary ammonium groups through in situ polymerization (referred to as SiPVP-N4), was developed to efficiently achieve Th/Ba separation under highly acidic conditions. This material displays a unique spherical morphology with high porosity (25.4 nm pore size), rapid kinetics (achieving equilibrium within 30 min, which is six times faster than commercial resins), and exceptional selectivity for Th over Ba (with a separation factor SFTh/Ba of 1.6 × 103). The adsorption behavior aligns well with that of the Langmuir model, indicating a maximum adsorption capacity of 89.77 mg/g. Column experiments demonstrated 100% thorium recovery with high radiochemical purity under 9 M HNO3 adsorption conditions. Notably, SiPVP-N4 sustains over 95% capacity retention through five regeneration cycles, underscoring its potential for industrial application. This study establishes a robust platform for the production of medical isotopes, presenting a cost-effective alternative to traditional high-acid separation systems and contributing to the advancement of radiopharmaceutical development.
