A novel approach of periodate oxidation coupled with high-performance liquid chromatography (HPLC)–fluorescence detection (FLD) for the quantitative determination of 3-chloro-1,2-propanediol (3-MCPD) has been established. The essence of this approach lies in the production of chloroacetaldehyde by the oxidization cleavage of 3-MCPD with sodium periodate and the HPLC analysis of chloroacetaldehyde monitored by an FLD detector after fluorescence derivatization with adenine. The experimental parameters relating to the efficiency of the derivative reaction such as concentration of adenine, chloroacetaldehyde reaction temperature, and time were studied. Under the optimized conditions, the proposed method can provide high sensitivity, good linearity (r2 = 0.999), and repeatability (percent relative standard deviations between 2.57% and 3.44%), the limits of detection and quantification were 0.36 and 1.20 ng/mL, respectively, and the recoveries obtained for water samples were in the range 93.39–97.39%. This method has been successfully applied to the analysis of real water samples. Also this method has been successfully used for the analysis of vegetable oil samples after pretreatment with liquid–liquid extraction; the recoveries obtained by a spiking experiment with soybean oil ranged from 96.27% to 102.42%. In comparison with gas chromatography or gas chromatography–mass spectrometry, the proposed method can provide the advantages of simple instrumental requirement, easy operation, low cost, and high efficiency, thus making this approach another good choice for the sensitive determination of 3-MCPD.

In this paper, a sensitive method for the determination of 3-chloro-1,2-propanediol (3-MCPD) by high performance liquid chromatography-fluorescence detection (HPLC-FLD) was established, in which 9-(2-Hydroxypropyl) adenine (HP-Ade) was selected as the fluorescent derivatization reagent because of its good water solubility and hydrophobicity. The principle of the method lies in the origination of chloroacetaldehyde with the periodate oxidation of 3-MCPD and its' fluorescence derivatization by HP-Ade to produce a strong fluorescent substance, name as 1-N6-vinylidene-9-(2-hydroxypropyl)adenine (ε-HP-Ade), used for quantitative determination of 3-MCPD by HPLC-FLD. Some mobile phase conditions and incubation parameters related with the method features were studied and optimized. Under the optimal conditions, the method shows high sensitivity, good linearity (r2 = 0.999) and good repeatability (RSD% between 1.04% and 2.55%), the detection limit (LOD) and quantification limit (LOQ) were 0.02 ng/mL and 0.06 ng/mL respectively. It has been successfully applied for the determination of various edible vegetable oil samples, the chromatogram shows symmetrical peak shape, perfect separation and very “clean” profiles. In comparison with gas chromatography-mass spectrum (GC-MS), the pretreatment process of the proposed method was totally operated in aqueous phase, the instrument requirements and cost are relatively lower, thus it will provide another choice for the routine analysis of 3-MCPD.

In this paper, a composite composed of carboxylated multi-wall carbon nanotubes (cMWCNT) incorporated in a metal–organic framework (MOF-199) has been synthesized using 1,3,5-benzoic acid as a ligand through a simple solvothermal method. The synthesized cMWCNT/MOF-199 composite was characterized by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR) and X-ray diffractometry (XRD). The cMWCNT/MOF-199 hybrids were modified on the surface of glassy carbon electrodes (GCE) to prepare a molecularly imprinted electrochemical sensor (MIECS) for specific recognition of 3-chloro-1,2-propanediol (3-MCPD). The electrodes were characterized by differential pulse voltammetry (DPV), electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). Under optimal conditions, the electrochemical sensor exhibited an excellent sensitivity and high selectivity with a good linear response range from 1.0 × 10−9 to 1.0 × 10−5 mol L−1 and an estimated detection limit of 4.3 × 10−10 mol L−1. Furthermore, this method has been successfully applied to the detection of 3-MCPD in soy sauce, and the recovery ranged from 96% to 108%, with RSD lower than 5.5% (n = 3), showing great potential for the selective analysis of 3-MCPD in foodstuffs.

A biosensor for determination of 3-chloro-1, 2-propandiol (3-MCPD) was developed based on the electrochemically catalytic reduction by hemoglobin (Hb). Hb was concentrated and immobilized with magnetic molecularly imprinted nanoparticles (MMIPs NPs). The MMIP NPs had a core-shell structure, with magnetic Fe 3 O 4 NPs as the core and SiO 2 as the shell. MMIPs NPs were modified on the surface of a magnetic electrode to constitute the Hb enzyme-like sensor. The biosensor could catalyze the electrochemical reduction of 3-MCPD, and a direct electrochemical determination of 3-MCPD could be achieved for the first time. The biosensor, where Hb immobilized with MMIPs NPs had high electrochemical response for 3-MCPD, increased by 37.5% compared with Hb immobilized with magnetic non-imprinted nanoparticle (MNIPs NPs). The biosensor increased the response by 19.56% again under an external magnetic field . The reduction peak current had a linear relationship with 3-MCPD concentration ranging from 1.0 mg·L −1 to 160 mg·L −1 with a detection limit (S/N = 3) of 0.25 mg·L −1 . The biosensor was applied to the determination of 3-MCPD in soy sauce samples successfully with recoveries between 97.4% and 99.6%.

(S)-3-chloro-1-phenyl-1-propanol ((S)-CPPO) is an important chiral intermediate predominantly used in the synthesis of the chiral side chain of (S)-fluoxetine. In this study, carbonyl reductase (CBR) from Novosphingobium aromaticivorans was successfully expressed in recombinant E. coli . The enzymatic activity of the recombinant CBR was significantly increased to 1875 U/mL in the fed-batch fermentation in a 10 L fermenter and recombinant CBR was then purified and characterized. By regenerating NADH with glucose dehydrogenase, 100 g/L 3-chloro-1-phenyl-1-propanone (3-CPP) was successfully converted to (S)-CPPO with a conversion of 100% and ee value of 99.6% after 12 h at 30 °C in PBS buffer (pH 7.0), which are the highest reported to date for the bio-production of (S)-CPPO and presented great potential for green production of (S)-CPPO at industrial scale.
