Single chain morphology and nanofiber-like aggregates of branched β-(1→3)-d-glucan in water/dimethylsulfoxide solution

Cong Chen,Yan Meng,Sheng Li,Wenhua Wu,Chuanjun Liu,Xiaojuan Xu,Lina Zhang

A polysaccharide coded as PR-CA was extracted from Polyporus rhinoceros and determined to be a β-(1→3)- d -glucan with multiple branches. The weight-average molecular weights ( M w ) of PR-CA in dimethylsulfoxide (DMSO) and in water were determined with static light scattering (SLS) to be 3.57   ×   10 5 and 1.79   ×   10 7 , indicating existence of the single chains in DMSO and co-existence of single chains and aggregates in water. Moreover, the stiffness of single chains of PR-CA in water was directly visualized by atomic force microscopy (AFM) and transmission electron microscopy (TEM). The hollow structure of PR-CA nanofibers with width of 30–40   nm and length of ∼350   nm formed in the water/DMSO (9:1, v:v) was demonstrated by a fluorescent probe tetraphenylethylene (TPE) via aggregation-induced emission (AIE). The formation of PR-CA nanofibers was ascribed to the parallel aggregation of the extended PR-CA chains due to the hydrogen bonding and hydrophobic interaction. This work offered valuable results for promising applications of natural branched β-glucans in the biological fields of drug inclusion, delivery and disease diagnosis.

Novel β-1,3-D-glucan porous microcapsule enveloped folate-functionalized liposomes as a Trojan horse for facilitated oral tumor-targeted co-delivery of chemotherapeutic drugs and quantum dots

Xiaonan Li,Ziming Zhao,Yihua Yang,Zhaorong Liu,Jinglei Wang,Yalu Xu,Yanzhuo Zhang

In this study, a new type of β-1,3-D-glucan porous microcapsule (GPM)-enveloped and folate conjugated chitosan-functional liposome (FCL), FCL@GPM, was developed for the potential oral co-delivery of chemotherapeutic drugs and quantum dots (QDs) with facilitated drug absorption and antitumor efficacy. In this dual-particulate system, multiple FCLs serve as the cores for effective loading, folate-mediated tumor-targeting, facilitated intracellular accumulation, and pH-responsive controlled release of chemotherapeutic agents, while a GPM acts as the shell for affording macrophage-mediated tumor selectivity. Gefitinib (GEF) was selected as a chemotherapeutic agent, while acid degradable ZnO QDs were selected due to their dual role as an anticancer agent for synergistic chemotherapy and as a fluorescent probe for potential cancer cellular imaging. The GEF and ZnO QD co-loaded FCL@GPMs (GEF/ZnO-FCL@GPMs) exhibited a prolonged release manner with limited release before uptake by intestinal cells. Furthermore, Peyer's patch uptake, macrophage uptake, cytotoxicity, and biodistribution of FCL@GPMs were tested. In addition, GEF and ZnO QD co-loaded FCLs (GEF/ZnO-FCLs) not only have a tumor acidity responsive release property, but also induce a superior cytotoxicity on cancer cells as compared to GEF. Moreover, a 1.75-fold increase in the bioavailability of GEF delivered from GEF/ZnO-FCL@GPMs as compared to its trademarked drug (Iressa®). As a result, GEF/ZnO-FCL@GPMs exerted a superior antitumor efficacy (1.47-fold) as compared to the trademarked drug in mice. Considered together, the developed FCL@GPMs, combining the unique physicochemical and biological benefits of FCLs and GPMs, possess great potential as an efficient delivery system for the co-delivery of chemotherapeutic agents and quantum dots.

Extraction, purification, structural characterization, and anti-skin aging activity of a homogeneous polysaccharide from Durvillaea antarctica

Wu Jiahui,Yuan Yonglei,Qu Liping

As an edible algae with rich nutrients and high medicinal value, Durvillaea antarctica has certain potential in anti-aging, but there is currently limited research on the separation, purification and activity of its anti-aging active ingredients. We successfully extracted and purified a new β-(1,3)-D-glucan with a special branched structure from Durvillaea antarctica using subcritical water extraction combined with ultrafiltration membrane (SWE-UFM). The polysaccharide, named DAG-1, consists of glucose (92.70%) and has a weight-average molecular weight of 10149 Da. The GC–MS and 1D/2D NMR confirmed that DAG-1 was composed of 3)-β-D-Glcp-(1 → , → 3,6)-β-D-Glcp-(1 → , β-D-Glcp-(1 → and → 6)-α-D-Glcp-(1 → , and β-glycosidic linkages between these sugar units. Animal experiments showed that DAG-1 has significant anti-skin aging effects by inhibiting the reduction of superoxide dismutase (SOD) activity, accumulation of malonic dialdehyde (MDA), and decline in hydroxyproline (HYP) content in collagen fibers of D-gal-induced mouse skin. To further elucidate the mechanism of DAG-1, we found that DAG-1 can alleviate senescence in human dermal fibroblasts (HDFa), inhibit degranulation of human cutaneous mast cells (HCMC), and modulate inflammatory factors (TNF-α, IL-6) production. Additionally, DAG-1 may bind to c-KIT receptors and downregulate the expression of KITLG/KIT, regulating the growth and activation of mast cells, and thus alleviate skin aging. These findings suggest that DAG-1 is a potential anti-aging ingredient for functional food, cosmetic, and medicine development.

Preparation of a water soluble aminated β-1,3-D-glucan for gene carrier: The in vitro study of the anti-inflammatory activity and transfection efficiency

Jing Wei Jin,Min Zhi Rong,Ming Qiu Zhang,Wing-Leung Wong

β-1,3-D-glucan has been reported to have a series of bioactivities including antitumor, antimicrobial, anti-inflammatory and antioxidative effects; however, its insolubility in neutral aqueous solution significantly restricts the potential application in biological and medicine fields. Herein, a water-soluble aminated β-1,3-D-glucan (AG) was synthesized and the anti-inflammatory effect, cytotoxicity and plasmid DNA (pDNA) binding capacity of AG, serum stability, the particle sizes and zeta potentials of AG/pDNA nanocomposites, and the transfection efficiency and mechanism of action were studied. AG shows no obvious cytotoxicity within the range of working concentration (1–64 μg/ml) and it exerts potent anti-inflammatory effect independent on Dectin-1 and TLR2. AG/pDNA nanocomposites prepared by electrostatic interaction possess an appropriate particle size ranged from 192.8 to 118.4 nm and zeta potentials ranged from 20.880 to 27.16 mV with the N/P ratios from 5 to 100. AG/pDNA nanocomposites at the N/P ratios of 10 and 20 were able to show superior transfection efficiencies in RAW 264.7 cells as a result of their suitable particle size, zeta potential, anti-inflammatory effect, and the specific interaction with pattern recognition receptors (Dectin-1 and TLR2). These results indicate that AG is a potential candidate for DNA delivery system due to its potent anti-inflammatory effect and high transfection efficiency.

β-1,3-d-glucan particles-based “nest” protected co-loaded Rhein and Emodin regulates microbiota and intestinal immunity for ulcerative colitis treatment

Yanli Wang,Siwei Zhong,Ke Yang,Ruifeng Luo,Linxin Dai,Wenzhen Zhong,Yan Ye,Chaomei Fu,Dasheng Lin,Nan Li,Jianping Chen,Chuan Zheng,Shu Fu,Fei Gao

Herein, a β-1,3- D -glucan based yeast cell wall loaded with co-loaded nanoparticles of Rhein (RH) and Emodin (EMO), was developed for the combined treatment of ulcerative colitis (UC) by modulating gut microbiota and the Th17/Treg cell balance. This was achieved through an oral “nano-in-micro” advanced drug delivery system. Specifically, RH was grafted onto the HA chain via disulfide bonds to synthesize a reduction-sensitive carrier material and then used to encapsulate EMO to form nanoparticles with a specific drug ratio (denoted as HA-RH/EMO NPs). As anticipated, HA-RH/EMO NPs were encased within the “nests”-yeast cell wall microparticles (YPs), efficiently reach the colon and then released gradually, this occurs mainly due to the degradation of β-1,3- D -glucan by β-glucanase. Additionally, HA-RH/EMO NPs demonstrated a significant reduction-sensitive effect in GSH stimulation evaluations and a remarkable ability to target macrophages in in vitro cell uptake studies. Notably, HA-RH/EMO NYPs reduced inflammatory responses by inhibiting the PI3K/Akt signaling pathway. Even more crucially, the oral delivery and drug combination methods significantly enhanced the regulatory effects of HA-RH/EMO NYPs on gut microbiota and the Th17/Treg balance. Overall, this research marks the first use of YPs to encapsulate two components, RH and EMO, presenting a promising therapeutic strategy for UC.
