Establishment of a visual assessment platform for total antioxidant capacity of Ganoderma sichuanense based on Arg-CeO2 hydrogel nanozyme utilizing a self-programmed smartphone app and a self-designed 3D-printed device

Tan Suhui,Tan Mengyuan,Zhang Tao,Gao Lu,Li Yi,Xu Xuechao,Yang Zhen-quan

A multienzyme-like L-argininemodified CeO 2  (Arg-CeO 2 ) nanozyme was prepared for accurate total antioxidant capacity (TAC) determination of  Ganoderma sichuanense  ( G. sichuanense ). It exhibits oxidase-like and peroxidase-like activities catalyzing the generation of superoxide anion free radicals and hydroxyl radicals, accelerating 3, 3′, 5, 5′-tetramethylbenzidine (TMB) chromogenic reaction. Antioxidants can effectively eliminate the generated free radicals, inhibiting the TMB chromogenic reaction. Hence, TAC can be assessed by Arg-CeO 2  catalyzed TMB chromogenic reaction using ascorbic acid (AA) as a reference substance. TAC of  G. sichuanense  extracted using different solvents was determined, indicating the highest TAC observed in the  G. sichuanense  extracted by the mixture solvent (deionized water/ethanol/ethyl acetate = 1 : 1 : 1). A visual assessment platform based on Arg-CeO 2  hydrogel nanozyme utilizing a self-programmed smartphone app and a self-designed 3D printed device is established. TAC using AA as a reference substance (3.33 ~ 83.33 µM) can be detected with a detection limit as low as 0.58 µM. The platform can realize one-step quantitative determination of TAC in  G. sichuanens e within 40 min. The proposed visual assessment platform presented exhibits promising application prospects in the field of TAC assessment in food. A 3D-printed integrated maneuverable device for sensitive colorimetry of Pseudomonas aeruginosa

Jizhou Li,Shiruoyu Zhou,Ruining Bai,Zhifeng Fu

Timely and sensitive monitoring of pathogens in clinical specimen is highly demanded for efficient control and precise treatment of infectious diseases. Herein, a 3D-printed maneuverable device integrating incubation, washing, and detection functions was manufactured for colorimetry of Pseudomonas aeruginosa ( P. aeruginosa ) using transparent resin. The device combined with magnetic beads (MBs) can achieve specific separation and efficient enrichment of P. aeruginosa . Furthermore, it can be directly fixed onto a 96-well plate holder for colorimetry. Specifically, a P. aeruginosa bacteriophage termed as JZ1 acquired from river water was applied as a recognition reagent to functionalize the separation vectors MBs. Then, a nanoconfinement MOFs material termed as PCN-222(Pt) with remarkable peroxidase-like activity was conjugated with polymyxin B to act as a signal tracer. With the formation of target bacterial complexes, the bound PCN-222(Pt) catalyzed the color reaction of 3,3′,5,5′-tetramethylbenzidine to enable quantitative colorimetry of P. aeruginosa by the maneuverable device. With this device, P. aeruginosa can be quantified within 40 min, with a dynamic range of 1.9 × 10 2 ∼ 1.9 × 10 6 cfu mL −1 . The results for colorimetry of P. aeruginosa in diverse sample matrixes demonstrated its satisfactory practicability. This work provides a facile, timely, and cost-effective technique for point-of-care testing of pathogens.

Applying CRISPR/Cas system as a signal enhancer for DNAzyme-based lead ion detection

Yanju Chen,Hui Wu,Siwenjie Qian,Xiaoping Yu,Huan Chen,Jian Wu

Heavy metal lead accumulation in the environment pollutes the ecology systems and further threatens the human health. It is necessary to develop a sensitive method to detect it. Here, we propose a highly sensitive lead detection method by combining DNAzyme and CRISPR system. Once the lead ion is recognized, the substrate chain of DNAzyme is cleaved to produce single strand DNA . The produced single strand DNA can be detected by Cas protein/guide RNA complex and further trigger the collateral cleavage effect of CRISPR system, which can indiscriminately cut short single strand DNA reporters. By this way, the detection signals can be greatly amplified. This method can detect lead ions as low as 0.48 nM. The sensitivity is higher than the DNAzyme method. Furthermore, the portable 3D printing device is designed to observe the fluorescent signals so the end-point detection results can be visualized by the naked eyes. The entire detection process can avoid using bulky and expensive instruments, which can promote on-site lead ion detection.

Copper ion as a peroxidase mimic for fluorescent detection of casein phosphopeptide by a 3D-printed portable device

Zhihong Guan,Ruobo Chen,Kaixin Liu,Li Fu,Ling Li,Li Jia

Developing an on-site sensing platform for casein phosphopeptide (CPP) is essential to ensure the quality of infant formula. This study is the first report on a fluorescence detection method for CPP. The method is based on the readily available copper ions (Cu 2+ ). Cu 2+ has peroxidase-like activity and can catalyze the oxidation of o -phenylenediamine (OPD) to produce 2,3-diaminophenazine with a fluorescence property. CPP can inhibit the peroxidase activity of Cu 2+ through the coordination of the phosphate groups in CPP and suppress the oxidation of OPD, decreasing the fluorescence intensity of the catalytic system. Based on this principle, a fluorescence approach was developed to detect CPP in milk and milk powder samples. The method does not require the preparation of nanomaterials, and the detection process takes 25 min. Furthermore, a self-designed portable detection device was constructed to detect CPP, demonstrating the potential of the fluorescence sensing platform in the on-site detection of CPP.

A ratiometric fluorescent sensor based on S-doped BCNO quantum dots and Au nanoclusters combined with 3D-printing portable device for the detection of malachite green

Li Wenhao,Liu Fang,He Yu,Song Gongwu

Sulfur-doped BCNO quantum dots (S-BCNO QDs) emitting green fluorescence were prepared by elemental doping method. The ratiometric fluorescence probe with dual emissions was simply established by mixed S-BCNO QDs with gold nanoclusters (GSH-Au NCs). Because the emission spectrum of Au NCs (donor) at 615 nm overlapped well with the ultraviolet absorption of malachite green (MG), fluorescence resonance energy transfer (FRET) can be achieved. When the concentration of MG increased, the fluorescence intensity ( F 495 ) of S-BCNO QDs decreased slowly, while the fluorescence intensity ( F 615 ) of Au NCs decreased sharply. The fluorescence intensity ratio of F 615 / F 495 decreased with the increase of MG. By plotting the F 615 / F 495 values against MG concentration, a sensitive and rapid detection of MG was possible with a wide detection range (0.1–50 µM) and a low detection limit of 10 nM. Due to the accompanying fluorescence color change from pink to blue-green, it can be used for visual detection. A three dimensional-printing device utilizing digital image colorimetry to capture color changes through the built-in camera, enables quantitative detection of MG with a good linearity between the values of red/green ratio and MG concentrations at the range 1–50 µM. This sensing platform had a range of advantages, including high cost-effectiveness, portability, ease of operation, and high sensitivity. Furthermore, the sensing platform was successfully applied to the detection of MG in real water sample and fish samples, thereby verifying the reliability and effectiveness of this sensing platform in water quality monitoring and food safety. In situ generation of polydopamine as photothermal reporter for on-site testing of azodicarbonamide in flour using an integrated 3D-printed device

Guo Lan,Zhao Dong-Mei,Hu Bao-Hua,Wu Shuo,Chen Shuai,Yu Yong-Liang

The detection of the illegal additive azodicarbonamide (ADA) in flour and related products is crucially important due to the harmful effects of excessive consumption on human health. However, the requirement for expensive instruments and trained operators poses a major challenge for on-site testing of ADA, particularly in field enforcement and consumer self-monitoring. Herein, an economical and portable 3D-printed photothermal (PT) device constructed by a 3D-printed sample bracket, a separable 96-well plate, an 808 nm laser, and a thermometer was rationally designed for quantitative on-site testing of ADA in flour. Since the target ADA could convert reducing glutathione (GSH) into non-reducing glutathione disulfide (GSSG), manganese dioxide nanosheets (MnO 2 NSs) in this sensing system remain intact to catalyze the conversion of dopamine (DA) into polydopamine nanoparticles (PDA NPs). During the detection process, the PT signal changes induced in the sample driven by the 808 nm laser could be easily recorded with a digital thermometer. The proposed PT method demonstrates an excellent linear response for ADA in the range of 30–1000 μM, with a detection limit as low as 10 μM, which was significantly lower than the maximum allowable ADA level in flour extracts (388 μM). This portable and cost-effective PT sensing system shows great potential for quantitative on-site testing, enabling immediate food safety identification in resource-limited regions. Graphical
