Here we report on the successful preparation of open-cellular macroporous 3D scaffolds templated from gelatin nanoparticle-stabilized HIPEs with acrylamide (AM) as the monomer in the continuous phase. Tuning the gelatin nanoparticle concentration or AM content led to different porous structures with void diameters varying between 30 and 78 μm. More importantly, keeping HIPEs at room temperature to undergo a limited kinetic coarsening before polymerization could greatly improve the interconnectivity and pore size of the scaffolds, with the average diameters (approx. 118 μm) being enlarged 1.5-fold. Additionally, the scaffolds had a character of soft tissue with compressive modulus more than 150 kPa. The cell culture assay confirmed that HepG2 cells not only could adsorb on but also were grown inside the scaffolds, representing a characteristic of the good biocompatibility of the scaffolds. Our work suggests that the 3D scaffolds fabricated from gelatin nanoparticle-stabilized HIPE templates are promising culture substrates for a wide range of applications in the biomedical field.

Nowadays, the organic pollutants and oil contaminations caused by petroleum exploitation, industrial discharge , and sanitary wastewater are aggravating the water shortage, becoming a global problem. Even though a great number of advanced materials have been developed and devoted to solving these problems, the materials reported recently are usually at cost of sophisticated craft and expensive raw materials, which are unaffordable for underdeveloped countries and regions. Therefore, a facile, scalable as well as costly-available production approach is vital for oil-water separation materials. Using the natural skeleton structure of sugarcane residue, the bio-based 3D scaffold (F-DA-cane) with hydrophobicity and photo-thermal conversion capability is successfully prepared by a scalable and extremely simple craft, which uses polydopamine and fluorine-containing compound as functionalized coating. After two-step surface treatment process, the bio-based 3D scaffold can effectively absorb variety of oils and organic solvents. The absorption capability of F-DA-cane is up to 24.8 g/g (the maximum), which is almost 3 times higher than that of pure sugarcane (8.21 g/g). In order to achieve continuous oil-water separation, pumping-driven oil collection experiments is performed. The oil transport speed of F-DA-cane was significantly improved with the aid of its excellent photo-thermal conversion capability (254.5 L/m 2 h, more than twice of transport speed that obtained without light). These results indicated that these bio-based 3D scaffold could be used as an effective primary treatment method for organic solvents and oily wastewater. In conclusion, by taking advantage of common biowaste and simple coating technology, a facile, scalable as well as affordable production approach for preparing high-efficiency oil-water separation materials was successfully designed.

This paper describes the use of silk protein, including fibroin and sericin, from an alkaline solution of Ca(OH)2for the clean degumming of silk, which is neutralized by sulfuric acid to create calcium salt precipitation. The whole sericin (WS) can not only be recycled, but completely degummed silk fibroin (SF) is also obtained in this process. The inner layers of sericin (ILS) were also prepared from the degummed silk in boiling water by 120 °C water treatment. When the three silk proteins (SPs) were individually grafted with glycidyl methacrylate (GMA), three grafted silk proteins (G-SF, G-WS, G-ILS) were obtained. After adding I2959 (a photoinitiator), the SP bioinks were prepared with phosphate buffer (PBS) and subsequently bioprinted into various SP scaffolds with a 3D network structure. The compressive strength of the SF/ILS (20%) scaffold added to G-ILS was 45% higher than that of the SF scaffold alone. The thermal decomposition temperatures of the SF/WS (10%) and SF/ILS (20%) scaffolds, mainly composed of a β-sheet structures, were 3 °C and 2 °C higher than that of the SF scaffold alone, respectively. The swelling properties and resistance to protease hydrolysis of the SP scaffolds containing sericin were improved. The bovine insulin release rates reached 61% and 56% after 5 days. The L929 cells adhered, stretched, and proliferated well on the SP composite scaffold. Thus, the SP bioinks obtained could be used to print different types of SP composite scaffolds adapted to a variety of applications, including cells, drugs, tissues, etc. The techniques described here provide potential new applications for the recycling and utilization of sericin, which is a waste product of silk processing. Keywords:silk protein;fibroin;sericin;grafting;composite bioink;bioprinting;3D scaffolds;mechanical properties

The healing of critical-sized bone defects (CSD) remains a challenge in orthopedic medicine. In recent years, scaffolds with sophisticated microstructures fabricated by the emerging three-dimensional (3D) printing technology have lighted up the treatment of the CSD due to the elaborate microenvironments and support they may build. Here, we established a magnesium oxide-reinforced 3D-printed biocomposite scaffold to investigate the effect of magnesium-enriched 3D microenvironment on CSD repairing. The composite was prepared using a biodegradable polymer matrix, polycaprolactone (PCL), and the dispersion phase, magnesium oxide (MgO). With the appropriate surface treatment by saline coupling agent, the MgO dispersed homogeneously in the polymer matrix, leading to enhanced mechanical performance and steady release of magnesium ion (Mg 2+ ) for superior cytocompatibility, higher cell viability, advanced osteogenic differentiation, and cell mineralization capabilities in comparison with the pure PCL. The in-vivo femoral implantation and critical-sized cranial bone defect studies demonstrated the importance of the 3D magnesium microenvironment, as a scaffold that released appropriate Mg 2+ exhibited remarkably increased bone volume, enhanced angiogenesis, and almost recovered CSD after 8-week implantation. Overall, this study suggests that the magnesium-enriched 3D scaffold is a potential candidate for the treatment of CSD in a cell-free therapeutic approach.

Gelatin-based inks have a broad range of applications in bioprinting for tissue engineering and regenerative medicine due to their biocompatibility, ease of modification, degradability, and rapid gelation induced by low temperature. However, gelatin-derived inks prepared through low-temperature treatment have poor mechanical properties that limit their applications. To solve this problem, we designed polyacrylamide/gelatin/silver nanoparticle (PAAm-GelatinAgNPs) ink to improve gelatin-based hydrogels. The ink is based on double networks, in which the physically cross-linked gelatin as the first network and covalently cross-linked PAAm as the second network. It was found that the presence of PAAm increased the tensile and compression strength of the gelatin-based ink. Moreover, silver nanoparticles endowed the antibacterial properties to the gelatin-based ink and were able to shield the UV irradiation and damages to rat skin. In addition, this ink showed the shear thinning property; Consequently it succeeded in printing complex 3D scaffolds such as the cube, five-pointed star, flower, and university logo of “SEU”. In summary, this ink presents a new strategy for the modification of gelatin and offers new potential applications for customized therapy of antimicrobial and anti-UV damage to tissues.

Introduction Scaffold structure plays a vital role in cell behaviors. Compared with two-dimensional structure, 3D scaffolds can mimic natural extracellular matrix (ECM) and promote cell–cell and cell–matrix interactions. The combination of osteoconductive scaffolds and osteoinductive growth factors is considered to have synergistic effects on bone regeneration. Materials and methods In this study, core–shell poly(lactide-co-glycolide) (PLGA)/polycaprolactone (PCL)–BMP-2 (PP–B) fibrous scaffolds were prepared through coaxial electrospinning. Next, we fabricated 3D scaffolds based on PP–B fibers with thermally induced self-agglomeration (TISA) method and compared with conventional PLGA/PCL scaffolds in terms of scaffold morphology and BMP-2 release behaviors. Then, rat adipose-derived stem cells (rADSCs) were seeded on the scaffolds, and the effects on cell proliferation, cell morphology, and osteogenic differentiation of rADSCs were detected. Results The results demonstrated that 3D scaffold incorporated with BMP-2 significantly increased proliferation and osteogenic differentiation of rADSCs, followed by PP–B group. Conclusion Our findings indicate that scaffolds with 3D structure and osteoinductive growth factors have great potential in bone tissue engineering.

Although cellular therapy holds enormous promise in treating intractable diseases, its application potential has been significantly hampered due to the scarcity of reliable and consistent cell sources. Therefore, a high-efficiency strategy that improves cell production and storage is desperately needed. Herein, we develop a versatile 3D bioinspired scaffold (Cryosilk) for improving scalable cell manufacture and cryopreservation. A bottom-up fabrication technique integrating electrospinning, in situ surface functionalization and freeze-shaping was explored to construct Cryosilk with biomimetic features and functions of silkworm cocoons. Cryosilk is composed of a core-shell heterostructure with silk fibroin/poly alanine fiber core and silk sericin shell, generating a 3D cocoon-mimicking fibrous structure. Importantly, Cryosilk possesses improved thermal conductivity and ice crystal resistance capability, thus enabling to cryopreserve biological samples with minimal cryodamage. Furthermore, Cryosilk not only promotes cell adhesion and growth, but achieves rapid and uniform rewarming process, which provides high cryopreservation efficacy for immune cells and stem cells. Particularly, Cryosilk can maintain cell viability and biofunctions of stem cell-scaffold constructs after freeze-thawing, which can be directly implanted to promote wound healing. Thus, Cryosilk offers unprecedented efficacy in cell manufacture and cryopreservation, which provides sufficient and high-quality precious cells and tissue engineered scaffolds for cellular therapy.

Assembling natural proteins into large, strong, bone-mimetic scaffolds for repairing bone defects in large-animal load-bearing sites remain elusive. Here this challenge is tackled by assembling pure silk fibroin (SF) into 3D scaffolds with cortical-bone-like lamellae, superior strength, and biodegradability through freeze-casting. The unique lamellae promote the attachment, migration, and proliferation of tissue-regenerative cells (e.g., mesenchymal stem cells [MSCs] and human umbilical vein endothelial cells) around them, and are capable of developing in vitro into cortical-bone organoids with a high number of MSC-derived osteoblasts. High-SF-content lamellar scaffolds, regardless of MSC inoculation, regenerated more bone than non-lamellar or low-SF-content lamellar scaffolds. They accelerated neovascularization by transforming macrophages from M1 to M2 phenotype, promoting bone regeneration to repair large segmental bone defects (LSBD) in minipigs within three months, even without growth factor supplements. The bone regeneration can be further enhanced by controlling the orientation of the lamella to be parallel to the long axis of bone during implantation. This work demonstrates the power of oriented lamellar bone-like protein scaffolds in repairing LSBD in large animal models.

Hydrogen sulfide (H 2 S) serves as the third gaseous signaling molecule in biological systems and is crucial for the regulation of biological cells. However, due to the lack of real time analysis techniques, studies on cellular release of H 2 S may yield inaccurate information and differ significantly from the three-dimensional (3D) cellular in vivo conditions. In this paper, a stretchable conductive 3D scaffold integrated with 3D cell culture, strain loading as well as electrochemical detection was proposed for real time profiling of H 2 S secreted from cells. In our design, single-walled carbon nanotube (CNTs)-impregnated 3D PDMS was used as the conductive substrate scaffold, which was subsequently functionalized with ZIF-8 and gold nanostars (AuNSt). The obtained stretchable AuNSt/ZIF-8/CNTs/PDMS scaffold achieved sensitive electrocatalytic performance toward H 2 S while maintaining excellent mechanical stability. The favorable biocompatibility of the 3D scaffold facilitated the attachment and growth of endothelial cells in culture, enabling in situ inducing and real time tracking of H 2 S secretion during mechanotransduction and pharmacological interventions. This study provided a multifunctional platform for 3D cell culture and to investigate the molecular mechanism related to endothelial dysfunction and vascular disorder.

In this work, novel antibacterial cellulose diacetate/poly(ethylenimine)/hyaluronic acid (CDA/PEI/HA) composite three-dimension (3D) scaffolds were prepared through electrospinning and post-processing. Firstly, CDA/PEI 2D composite nanofiber membranes were prepared by electrospinning, and then the CDA/PEI/HA 3D composite scaffolds were fabricated by post-processing and freeze-drying of CDA/PEI membranes. In particular, HA was added to improve the biocompatibility of composite scaffolds. Compared with CDA/PEI scaffolds, CDA/PEI/HA composite scaffolds showed higher water absorbing rate, higher water retention rate and higher mechanical strength . Moreover, CDA/PEI/HA composite scaffolds with abundant positive charge were benefit for improving antibacterial activity. CDA/PEI/HA scaffolds with biology function of HA were benefit for improving hemocompatibility and cell proliferation of CDA/PEI scaffolds. In summary, antibacterial CDA/PEI/HA scaffolds could protect wound from bacterial infection, improve cellular behavior and accelerate wound healing. Thus, CDA/PEI/HA scaffolds could be potential application for wound dressing.

Large-scale bone defects caused by traumatic injury or tumor resection, are accompanied by relatively high bacterial infection rates. Relevant clinical therapy encounters challenges of infection recurrence, poor vascularization, and bone destruction. Engineered substitutes endowed with multiple functions of bacterial eradication, osteoconductivity, and angiogenesis are urgently needed to address these challenges. Herein, graphene oxide/copper (GO/Cu) nanocomposite-decorated polylactide- co -glycolide (PLGA)/β-tricalcium phosphate (TCP) (P/T/GO/Cu) scaffolds are fabricated using 3D printing technology. A comprehensive evaluation of material characteristics and biological activities of these scaffolds is conducted and compared. This novel bone substitute has unique surface characteristics, and the incorporation of GO/Cu nanocomposites into the scaffolds exhibits improved antimicrobial and osteogenic potential and displays acceptable cytocompatibility. A subcutaneous implantation model in rats confirms the effective inhibition of abscess formation and inflammatory reactions, satisfactory neovascularization and tissue integration, and good biosafety of the implanted P/T/GO/Cu scaffolds. Eventually, a bacteria-contaminated bone defect model is established in rabbits and significantly better new cancellous bone formation featuring collagen or osteoid deposition is clearly found in the defect regions implanted with P/T/GO/Cu scaffolds. Thus, this novel bone substitute with excellent bacterial inhibition, osteogenesis, and angiogenesis capabilities represents a promising strategy to satisfy the multiple demands for the repair of bacteria-contaminated bone defects.

High-molecular biodegradable and bioresorbable polymers are widely used in tissue engineering applications. In this work, a high-resolution 3D printing technique, thermally assisted electrohydrodynamic jet (TAEJ) printing, is developed for patterning high-molecular weight biopolymer 3D structures. Boiling point graded polyvinylpyrrolidone (PVP) and PVP/polycaprolactone biopolymer inks are prepared. The resultant effects of electrohydrodynamic force and thermal field are applied on these polymer ink, to form a stable and controllable sub-micrometer scale jet at the needle tip. Fine jet 2D period patterns and 3D high aspect ratio structures of the biopolymer inks are directly printed using fine jet at a typical feature size of sub-micrometer. Furthermore, TAEJ printing of 3D heterogeneous high-molecular weight biopolymer scaffold is demonstrated. Cell culture shows high biocompatibility and cartilage regeneration in vitro, which provides a great potential for tissue engineering applications.
