In recent years, a new type of micro-porous material, namely metal organic framework material, has received more and more attention from many basic and industrial fields because these materials possess unique advantages. In this work, through the powerful sonochemical preparation method, a three-dimensional cluster-based Cd II -MOFs, {[Cd(abtz) 2 (H 2 O) 2 ]·(ClO 4 ) 2 ·H 2 O} n ( 1 , abtz = 1-(4-aminobenzyl)-1H-1,2,4-triazole) can be quickly synthesized in the facile ultrasonic method. Powder X-ray diffraction (PXRD) measurement confirms that these bulky samples 1 (synthesized on different ultrasonic powers and ultrasonic time conditions) were pure . In addition, ultrasonic chemical time and irradiation power did not change the structure of composites materials 1 . SEM and morphological changes of 1 in the ultrasonic synthesis are also determined. Moreover, 1 exhibits good stability, the structure of 1 can be maintained not just in various solvents, and in aqueous environments with pH values from 2 to 12. Photo-luminescent experiment also reveals that complex 1 has the excellent application prospect as highly sensitive sensing material for the biomarker DPA (2,6-pyridine dicarboxylic acid) and 5-HT (5-hydroxytryptamine) through the photo-luminescence “turn-on” and “turn-off” effect, respectively. Further photo-luminescent measurements also show that different ultrasonic powers and ultrasonic time can effectively induce fluorescent sensing enhancement for biomarkers DPA and 5-HT based on the water stable clustered-based cadmium(II) coordination framework.

Growing evidence suggested that energy deficiency might be involved in the pathophysiological mechanism of depression. Energy deficiency, mainly results from mitochondrial damage, can lead to the dysfunction of synaptic neurotransmission, and further cause depressive-like behavior. The antidepressant effect of resveratrol had been widely demonstrated in previous studies; however, the underlying mechanism remains poorly understood. The present study aimed to investigate whether the antidepressant effects of resveratrol involved in the energy levels and neurotransmission in the hippocampus. We found that resveratrol and fluoxetine significantly attenuated depressive-like behaviors induced by chronic unpredictable mild stress (CUMS), which evidenced by the increased sucrose preference and the reduced immobility time in a forced swimming test. In addition, resveratrol increased hippocampal ATP levels, decreased Na + -K + -ATPase and pyruvate levels, and upregulated the levels of mitochondrial DNA (mtDNA), mRNA expression of sirtuin (SIRT)1 and peroxisome proliferator-activated receptor γ coactivator (PGC)1α. Furthermore, resveratrol and fluoxetine increased serotonin (5-HT) levels and downregulated the mRNA expression of 5-HT transporter (SERT) in the hippocampus. The decreased protein expression of growth-associated protein (GAP)-43 induced by CUMS was also ameliorated by resveratrol and fluoxetine. These findings demonstrated the antidepressant effects of resveratrol and suggested that resveratrol was able to promote mitochondrial biogenesis, enhance ATP and 5-HT levels, as well as upregulate GAP-43 expression in the hippocampus.

Metabolites of dietary fibers which consist of non-digestible carbohydrates (usually as plant-based polysaccharides) exert many benefits on gastrointestinal health. Previous studies proved that polysaccharides can change intestinal metabolic pathways, increase metabolite levels and alleviate intestinal inflammation. Our previous studies have shown that polysaccharide from Aloe vera gel (AGP) alleviated intestinal barrier dysfunction. However, whether intestinal protective function of AGP was related to metabolites need further exploration. Therefore, the effects of AGP on intestinal metabolites and the relevant mechanisms for subsequent mitigation of intestinal damage was investigated in this work. Metabolomics analysis results showed that AGP promoted tryptophan metabolism to 5-hydroxytryptamine (5-HT). Intriguingly, antibiotics treatment decreased the expression of tryptophan hydroxylase 1 (TPH1) in the intestinal epithelium of mice, and TPH1 inhibitor (PCPA) down-regulated 5-HT level in colonic contents, proving that AGP up-regulated TPH1 expression in the intestinal epithelium by restoring intestinal microbiota homeostasis, thus promoted the intestinal tryptophan metabolism to 5-HT. Combination of bioinformatics analysis of RNA-sequencing (RNA-Seq) in mice colon and the reported relationship among “5-HT, Htr4 (one of the 5-HT receptors), Kit + goblet cells, canonical Wnt pathway and intestinal stem cells (ISCs)” guided the confirmation of related mechanism in intestinal organoids (IOs) that 5-HT regulated ISCs function and intestinal barrier via Htr4-Kit-Wnt3 axis. Overall, AGP promoted 5-HT production to alleviate intestinal injury. This study revealed the effects of polysaccharides on intestinal health, which was critical and may provide new insights for the reasonable selection of dietary fiber to prevent intestinal inflammation and maintain intestinal function.

Background The traditional herbal medicine Atractylodes macrocephala Koidz. ( A. macrocephala ) is commonly utilized for alleviating symptoms associated with spleen deficiency, abdominal distension, diarrhea, and constipation. These pharmacological effects are attributed to a variety of active constituents. However, the specific bioactive compounds responsible for promoting defecation and gastrointestinal transit in A. macrocephala remain unidentified. Methods The primary polysaccharide characteristics of PAMK was elucidated by HPLC, FT-IR, and HGPGC. Efficacy of PAMK (0.07, 0.14, and 0.28 mg/g) on mice was evaluated in a spleen deficiency constipation mouse model by analyzing stool parameters, constipation-related physiological indexes, and SCFAs. The expression levels of 5-HT 3 R, 5-HT 4 R, and related receptor genes were examined by RT-qPCR, and neurotransmitters were examined using ELISA. Finally, the diversity of gut microbiota was analyzed with 16S rDNA sequencing. Key Results The results showed that PAMK significantly reduced the gastrointestinal transport time and increased the number of fecal pellets and fecal water content in spleen deficiency constipation model mice. PAMK kept the balance of 5-HT, SCFAs, TPH-1, SERT, CgA, and neurotransmitter levels (VIP, SP, MTL) in mice colon. In addition, PAMK could regulate the abundance of gut microbiota such as Alistopes , Bacteroides , and Odoribacter in spleen deficiency constipation model mice gut. Conclusions and Inferences It can be concluded that PAMK effectively ameliorated the symptoms of spleen deficiency constipation in mice by modulating the expression of 5-HT and its associated receptors. The underlying mechanism was elucidated, providing a solid theoretical foundation for the therapeutic application of A. macrocephala in treating spleen deficiency constipation and offering potential for developing novel approaches to address this condition.

Depression is a worldwide affliction that seriously compromises social adaptation and physical well-being. Citrus medica L. var. sarcodactylis Swingle (also known as Bergamot) is a citrus plant in the Rutanceae family. Bergamot essential oil, a naturally derived product, has been reported to possess neuroprotective effects. The present study aimed to reveal the neuroprotective mechanisms of bergamot essential oil in PC12 cells, and compare its efficacy with that of its predominant constituent, D-limonene. To confirm the target of action, this study sought to use PI3K inhibitor LY294002 to validate the mechanism in modulating autophagy and apoptosis of bergamot essential oil. By constructing a corticosterone-induced PC12 cell model of depression in vitro , we observed that bergamot essential oil stimulated the PI3K/AKT/mTOR signaling cascade, reducing autophagy markers LC3-II and p62 approximately by 22 % and 37 % respectively, and decreasing apoptosis rates by 37 %. Bergamot essential oil intervention also upregulated 5-HT levels and played a positive role in the BDNF/TrkB pathway which associated with synaptic plasticity. D-limonene (56.6 % of bergamot essential oil) was demonstrated to be a key contributor to these mechanisms. It exhibited protective effects comparable to bergamot essential oil against autophagy and BDNF/5-HT signaling activation, however, bergamot essential oil showed better efficacy in alleviating apoptosis. This study provides in vitro mechanistic evidence supporting the potential application of bergamot essential oil in the treatment of depression.

Environmental endocrine disruptors 4-nonylphenol (NP) and 4-tert-octylphenol (OP) may cast huge harm to human health. We used a rat model to observe the influence of NP or/and OP exposure on anxiety-related behaviors and the underlying mechanisms. Eighty male Sprague-Dawley (SD) rats were randomly divided into 10 groups: control group (corn oil), NP groups [30, 90, 270 mg/kg], OP groups [40, 120, 360 mg/kg] and NO groups [(mixed with the corresponding NP, OP alone exposed low, medium and high dose according to the natural environment exists NP:OP = 4:1]. The rats were orally administered every other day for 30 days. The neurobehaviors of rats were evaluated by open-field test (OFT) and elevated plus-maze test (EPM), and the concentrations of 5-HT, monoamine oxidase (MAOA), serotonin transporter (SERT), vesicular monoamine transporter 2 (VAMT2), 5-hydroxytryptamine 1A (5-HT 1A ), 5-hydroxytryptamine 2A (5-HT 2A ),and 5-hydroxytryptamine 2C (5-HT 2C ) in the rat prefrontal cortex were analyzed by ELISA . OFT and EPM tests showed that NP or/and OP exposure induced anxiety-related behaviors in rats. 5-HT levels were significantly increased compared with the control group. The levels of MAOA, SERT, VAMT2, 5-HT 1A , 5-HT 2A, and 5-HT 2C in the prefrontal cortex reduced in different degrees by high-doses NP or/and OP exposure. In summary, NP or/and OP exposure might cause anxiety-related behaviors in rats through regulating neurotransmitter 5-HT levels by altering the expression of 5-HT decomposition enzyme MAOA, transporters SERT and VMAT2, and 5-HT receptors 5-HT 1A , 5-HT 2A, and 5-HT 2C .

The real-time detection of neurotransmitters had attracted more attention due to the potential to provide vital information for pathology research, disease diagnosis, and clinical efficacy evaluation. Here, a magnetic-mediated nano sensor was developed to improve sensitivity for detection of serotonin (5-HT). The PDMS membrane doped with magnetic particles serves as a conversion element, enhancing the deformation of the thin film caused by surface stress under the action of a magnetic field and amplifying the resistance response signal of the nano sensor. The 5-HT detection associated with neurotransmitter diseases serve as a proof of concept revealing the magnetic sensitization. The detection limit of the 5-HT was decreased to 3.21 nM in the linear range of 5–500 nM, which was lower than that of the nano sensor without magnetic field. The clinical 5-HT samples were also been accurately detected, indicating a positive application potential of the as-prepared magnetic-mediated nano sensor. All these results reveal the high sensitivity and practicality of the as-prepared magnetic-mediated nano sensor and provide a new method for neurotransmitter diseases diagnosis.

Pediococcus pentosaceus Li05, a strain of lactic acid bacteria isolated from the faeces of healthy volunteers, exhibited potential protective effects against various diseases. This study performed third-generation sequencing and detailed characterisation of its genome. The Li05 chromosome harboured conserved genes associated with acid resistance ( atp ), bile salt resistance ( bsh ), oxidative stress resistance ( hsl , dltA , and et al.), and adhesion ( nrd , gap , and et al.), whereas the plasmid did not contain antibiotic resistance or virulence genes. Following intervention with Li05 in loperamide-induced constipated mice, constipation symptoms improved. Meanwhile, alterations in gut microbiota, increased BSH activity in faeces, and modifications to the faecal bile acid profile were observed. Additionally, expression levels of TGR5 and TPH1 in the colon of the mice increased, leading to elevated 5-HT levels. When the TGR5 gene was knocked out or the TPH1 inhibitor LX1606 was administered to suppress 5-HT synthesis in constipated mice, the beneficial effects of Li05 on gastrointestinal motility and mucus secretion were reversed. Culturing intestinal organoids demonstrated that increased bile acids such as DCA, Iso-LCA, and EALCA could enhance 5-HT levels through the TGR5/TPH1 axis. Therefore, we concluded that Li05 regulated bile acid metabolism, subsequently increasing 5-HT levels through the TGR5/TPH1 axis, thus alleviating constipation. Graphical The mechanism of Li05 in alleviating constipation. Li05 intervention regulated gut microbiota and bile acid metabolism, increasing 5-HT levels through the TGR5/TPH1 activation to ameliorate constipation.

Summary The coincidence between conditioned stimulus (CS) and unconditioned stimulus (US) is essential for associative learning; however, the mechanism regulating the duration of this temporal window remains unclear. Here, we found that serotonin (5-HT) bi-directionally regulates the coincidence time window of olfactory learning in Drosophila and affects synaptic plasticity of Kenyon cells (KCs) in the mushroom body (MB). Utilizing GPCR-activation-based (GRAB) neurotransmitter sensors, we found that KC-released acetylcholine (ACh) activates a serotonergic dorsal paired medial (DPM) neuron, which in turn provides inhibitory feedback to KCs. Physiological stimuli induce spatially heterogeneous 5-HT signals, which proportionally gate the intrinsic coincidence time windows of different MB compartments. Artificially reducing or increasing the DPM neuron-released 5-HT shortens or prolongs the coincidence window, respectively. In a sequential trace conditioning paradigm, this serotonergic neuromodulation helps to bridge the CS-US temporal gap. Altogether, we report a model circuitry for perceiving the temporal coincidence and determining the causal relationship between environmental events.

As a widely used anticancer drug in the clinic, cisplatin has obvious side effects, especially nephrotoxicity . Previous studies have suggested that the accumulation of intracellular reactive oxygen species (ROS) is a hallmark of cisplatin-induced acute kidney injury . This study aimed to investigate the relationship between ROS accumulation induced by cisplatin and 5-HT degradation. In vivo, by HE and TUNEL staining, we found that cisplatin-induced renal lesions and apoptotic regions, which were located in proximal tubular epithelial cells, were also the regions in which tryptophan hydroxylase 1 (Tph1), aromatic l -amino acid decarboxylase (AADC), 5-HT 2A receptor (5-HT 2A R) and monoamine oxidase A (MAO-A) were overexpressed, as determined by immunohistochemistry. Notably, the 5-HT 2A R antagonist sarpogrelate hydrochloride (SH) and the AADC inhibitor carbidopa (CDP) significantly attenuated cisplatin-induced increases in serum creatinine and blood urea nitrogen levels, renal ROS levels, oxidative stress (SOD activity and MDA), proinflammatory cytokine levels (NF-κB, TNF-α and IL-1β), proapoptotic factor levels (Bax, Bcl-2, C-caspase 3 and C-caspase 9) and the phosphorylation of p38 and STAT3 , as well as renal lesions and apoptosis . The combination of SH and CDP could almost abolish the effects of cisplatin challenge. In vitro, the effects of cisplatin challenge and the inhibitory effects of SH and CDP were also observed in HK-2 cells. Additionally, similar to the combination of SH and CDP, the MAO-A inhibitor clorgyline could also abolish the effects of cisplatin challenge. More importantly, by western blotting , we detected that the upregulation of Tph1, AADC and MAO-A expression induced by cisplatin both in vivo and in vitro could be obviously suppressed by SH to decrease 5-HT synthesis and mitochondrial 5-HT degradation. Altogether, these findings suggested that cisplatin-induced nephrotoxicity is due to the activation of the 5-HT degradation system in proximal tubular epithelial cells, including 5-HT 2A R and 5-HT synthesis and degradation. 5-HT 2A R plays a role by mediating the expression of MAO-A and the 5-HT synthases Tph1 and AADC.

Constipation is a major health concern worldwide and requires effective and safe treatment options. In this study, we selected ten strains of two species of lactobacilli to identify whether they were effective against constipation induced by loperamide administration in BALB/c mice. Monitoring of constipation-related indicators indicated thatLactobacillus paracasei(L. paracasei) mainly acted on the whole intestinal peristalsis to relieve constipation. Furthermore, through the detection of biological, chemical, mechanical, and immune barriers in mice, it was discovered thatL. paracaseichanged the relative abundance of bacteria related to the levels of acetic acid and 5-hydroxytryptamine (5-HT) (such as by increasing the relative abundance ofOdoribacterandClostridium, and reducing the relative abundance ofMucispirillum,Ruminococcus,Coprobacillus, andDorea), increased the concentration of acetic acid in the intestine, which stimulated enterochromaffin cells, promoted 5-HT synthesis in the colon, enhanced intestinal motility, and relieved constipation. In conclusion, this study provides a theoretical foundation for the development of personalized products for the treatment of constipation. Keywords:lactobacilli;constipation;intestinal microbiota;acetic acid;5-HT;gastrointestinal regulatory peptide;inflammatory factors

Ethnopharmacological relevance Dachengqi decoction (DCQ), a well-known Traditional Chinese Medicine (TCM) formula, has been widely applied for anti-constipation in clinical practice for thousands of years. Nevertheless, its pharmacological mechanisms remain not fully elucidated. Aim of the study This research employed a comprehensive approach integrating network pharmacology and metabolomics, supplemented with molecular docking techniques and both in vitro and in vivo experimental validation, to systematically elucidate the bioactive constituents, critical targets, and underlying mechanisms of DCQ in alleviating constipation. Materials and methods This investigation utilized a comprehensive pharmacology integration strategy to forecast the principal therapeutic pathways through which DCQ alleviates constipation. The establishment of a loperamide-induced constipation model in rats served to evaluate the interventive effects of orally administered DCQ. In addition, ultra-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF-MS/MS) was used to detect the changes of metabolites in plasma and urine of rats, and the metabolic related targets were analyzed. Combined with network pharmacology, the potential mechanism of DCQ in ameliorating constipation was revealed and further verified by molecular docking. According to the results of joint analysis, the intervention of DCQ mainly involved the serotonin (5-HT) pathway, including the inflammatory pathways of arachidonic acid and cAMP downstream of 5-HT. Therefore, the mRNA and protein expression levels of key factors associated with the 5-HT pathway, including phospholipaseA2 (PLA2), cAMP/PKA/AQPs, and PANoptosis, were detected. Furthermore, an inflammatory model of Caco-2 cells was induced to investigate the effect of DCQ on the 5-HT pathway. The expression of related targets (PLA2, PKA and ZBP1) was detected by reverse transcription quantitative PCR and immunofluorescence to further verify the molecular mechanism of DCQ. Results DCQ demonstrated efficacy in ameliorating constipation symptoms induced by loperamide in rats and reducing inflammatory responses in Caco-2 cells, while normalizing aberrant plasma and urinary metabolic profiles and regulating arachidonic acid metabolism in constipated rats. Integrated metabolomics and network pharmacology analyses revealed that DCQ mainly alleviated constipation via the 5-HT pathway, markedly suppressing the PLA2-mediated inflammation and PANoptosis while concurrently activating the cAMP-PKA-AQPs cascade to notably increase the expression of aquaporins (AQPs). Conclusions DCQ could restore the homeostasis of metabolic profiles in plasma and urine in model rats, activate the 5-HT pathway to alleviate inflammation, and may be involved in the regulation of PANoptosis-related mechanisms in intestinal epithelial cells, providing a new perspective for constipation treatment.
