Efficient production of 6-hydroxynicotinic acid by newly isolated Pseudomonas poae

Yi Li,Jiacheng Tang,Kaixiang Xin,Zongda Chen,Lele Zhao,Yifan Zhao,Yinbiao Xu,Pei Zhou,Yang Sun,Yupeng Liu,Hua Li

Background Nicotinic acid dehydrogenase possesses the capability to convert nicotinic acid into 6-hydroxynicotinic acid, a compound of significant research value as a pharmaceutical intermediate. The extraction of nicotinic acid dehydrogenase is primarily performed by strains. However, the enzyme activity of the strains reported currently is relatively low, and their potential to catalyze the production of 6-hydroxynicotinic acid is insufficient to meet industrial requirements. Results Due to the revealing properties of 6-hydroxynicotinic acid, this study proposes a technique for calculating the luminescence intensity of colonies, which is based on a fluorescence spectrometer. The developed method establishes a reliable linear relationship (88.2%) between the luminescence intensity and enzyme activity. Consequently, it has been employed to screen strains that produce nicotinate dehydrogenase. This screening approach allows for the evaluation of about 500 enzyme-producing strains daily, presenting an efficient strategy for screening. Conclusions Through this approach, a novel high enzyme activity strain producing nicotinic acid dehydrogenase, Pseudomonas poae have been obtained, which designated as HD530. After process optimization, it was utilized to produce 6-hydroxynicotinic acid, achieving a high yield of 155.45 g/L within 72 h, meeting the requirements for industrial production. The effectiveness and potential of this technique lie in its application for strain screening and improvement.

High-throughput screening of a nicotinate dehydrogenase producing Pseudomonas putida mutant for efficient biosynthesis of 6-hydroxynicotinic acid

Yu-Ting Shang,Jiufu Qin,Jin-Song Gong,Zi-Kai Wang,Heng Li,Hui Li,Jin-Song Shi,Zheng-Hong Xu

A nicotinate dehydrogenase producing strain was isolated and identified to be Pseudomonas putida , which could bio-transform nicotinic acid into 6-hydroxynicotinic acid. The activity of nicotinate dehydrogenase towards nicotinic acid could reach up to 1.11 U/mL and the yield of 6-hydroxynicotinic acid accumulated to 176 g/L within 30 h with a space time yield of 1.36 g/(g•h). In order to further improve its catalytic potential, a mutant library was constructed through atmospheric and room temperature plasma (ARTP) and rapidly screened by a high-throughput screening strategy, which obtained a mutant strain mut-5 with activity of 2.68 U/mL. Via developing a nitrilase-nicotinate dehydrogenase enzymatic cascade system, 54.5 g/L 6-hydroxynicotinic acid could be obtained within 585 min with 3-cyanopyridine as the substrate, which would further reduce the production costs. The results indicated that the nicotinate dehydrogenase (separately or combining with nitrilase) could serve as a superior catalyst for 6-hydroxynicotinic acid production. To our knowledge, the nicotinate dehydrogenase activity reported in this study was the highest level in literature to date.

Three-birds-with-one-stone: MIP-decorated MOF/COF hybrid nanoplatform for multifunctional PEC sensing, adsorption, and photocatalytic degradation of patulin

Wenyan Yan,Shuyue Li,Baoqing Bai,Ying Zhang,Jinhua Zhang,Tao Bo,Qinghua Wang,Ying Gu,Yukun Yang,Shuo Wang

A MIP-modified MOF/COF hybrid nanoplatform for PEC sensing, adsorption, and photocatalytic degradation was developed. • The MOF/COF hybrid enhanced photocurrent through improved light absorption and charge separation. • The MIP layer provided high selectivity. • The platform enabled ultrasensitive PAT detection and efficient adsorption-photocatalytic degradation. Rational design of photoactive materials is essential for the sensitive detection and efficient removal of hazardous contaminants. Herein, a visible-light-responsive covalent organic framework/metal-organic framework (COF/MOF) hybrid with uniform COF layers grown in-situ on the MOF surface, followed by the introduction of a molecularly imprinted polymer (MIP) layer via a sol-gel process, was constructed for the PEC sensing, adsorption, and photocatalytic degradation of patulin. The hybrid architecture enhanced photoelectrochemical performance by facilitating charge separation and improving light harvesting. The resulting PEC sensor exhibited a wide linear range (0.1 nM-1 mM) and a low detection limit (0.055 nM), along with good reproducibility and stability in real samples. Moreover, the MIP/MOF/COF hybrid enabled efficient adsorption and photocatalytic degradation of patulin, driven by photogenerated holes and superoxide radicals. This work provides a promising strategy for the efficient and highly sensitive detection and removal of contaminants. Download: Download high-res image (236KB) Download: Download full-size image

Record-Large Indium-Oxo Clusters: Synthesis, Hierarchical Assembly, and Efficient Optical Limiting

Xiuzhen Wang,Yian Chen,Xiaofeng Yi,Shumei Chen,Jian Zhang

High-nuclearity indium-oxo clusters (InOCs) represent critical molecular models for understanding indium oxide (In2O3) nanoparticles, yet their rational synthesis remains a formidable challenge. Herein, we report a dual-ligand strategy to access bixbyite-type In15-oxo clusters—the largest discrete indium-oxo cores reported to date. Their strategically labile carboxylate sites enable facile functionalization to generate InOC-38, InOC-39, and InOC-40. Notably, the In15 core serves as the highest-nuclearity secondary building unit (SBU) within the InOCs family, which can be extended into an In30 dimer (InOC-41) via a cluster-docking strategy or hierarchically assembled into one-dimensional chains (InOC-42) using multidentate 6-hydroxynicotinate linkers. These architectures, featuring π-conjugated ligands, heavy metals, and dense intermolecular interactions, exhibit exceptional optical limiting (OL) performance. InOC-38 and InOC-41 demonstrate record metrics (Tmin = 0.11 and 0.17; FOL = 0.275 and 0.408 J/cm2), surpassing state-of-the-art cluster-based materials. Furthermore, their processability into flexible transparent films underscores significant practical potential for optical applications.

Determination of patulin using dual-dummy templates imprinted electrochemical sensor with PtPd decorated N-doped porous carbon for amplification

Hu Xiaopeng,Xia Yide,Liu Yiwei,Zhao Faqiong,Zeng Baizhao

A novel dual-dummy templates imprinted electrochemical sensor has been fabricated for the detection of patulin. Herein, 2-oxindole (2-oxin) and 6-hydroxynicotinic acid (6-HNA) as the dummy templates, 4-aminothiophenol as functional monomer, and ionic liquid (IL) as electropolymerization electrolyte are employed to prepare molecularly imprinted polymer (MIP) film. 2-Oxin and 6-HNA have multiple groups and the obtained MIP possesses different types of imprinted sites, thereby achieving a better recognition capacity than that of single-dummy imprinted film. ILs can regulate the density of molecularly imprinted film and facilitate effective molecular recognition. The composite of PtPd decorated N-doped porous carbon has good conductivity and large surface area, and can amplify the signal. With the aid of electrochemical probe [Fe(CN) 6 ] 3-/4- (0.16 V vs. SCE) patulin can be detected. Under the optimal conditions, this sensor shows a detection range from 0.01 to 10 μg L −1 , with a detection limit of 7.5 × 10 −3  μg L −1 ( S / N  = 3). Two spiked juice samples were analyzed by this method, and the recovery ranges from 94 to 99.8% with RSD values of 2.4–4.6% ( n  = 3), indicating that this method can be applied for the detection of patulin in real samples. Graphical abstract A novel dual-dummy templates imprinted electrochemical sensor is firstly fabricated for the detection of patulin. This sensor exhibits high recognition capacity and sensitivity.

Microbial Degradation of Nicotinamide by a Strain Alcaligenes sp. P156

Hu Chunhui,Zhao Shuxue,Li Kuiran,Yu Hao

A novel Alcaligenes sp. strain P156, which can utilize nicotinamide as its sole source of carbon, nitrogen and energy, was enriched and isolated from soil in a solid waste treatment plant. Aerobic growth and degradation with nicotinamide were characterized. Seven nicotinamide degradation-related genes were obtained by sequence alignment from the genome sequence of strain P156. Four genes, designated naaA , naaD , naaE and naaF , were cloned and heterologously expressed. Nicotinamide degradation is initiated by deamination to form nicotinic acid catalyzed by the nicotinamidase NaaA, which shares highest amino acid sequence identity (27.2%) with nicotinamidase from Arabidopsis thaliana . Nicotinic acid is converted to 6-hydroxynicotinic acid, which is further oxidized to 2,5-dihydroxypyridine (2,5-DHP). 2,5-DHP is then transformed to a ring-cleavage product, N -formylmaleamic acid, by an Fe 2+ dependent dioxygenase NaaD. N -formylmaleamic acid is transformed to fumaric acid through maleamic acid and maleic acid by NaaE and NaaF, respectively. To our knowledge, this is the first report of the complete microbial degradation of nicotinamide in bacteria. Nicotinamide is considered as a model compound for the study of microbial degradation of pyridinic compounds, and the nicotinamide degrading related genes in strain P156 were distributed differently from the reported similar gene clusters. Therefore, this study contribute to the knowledge on the degradation of pyridinic compounds.

Surface molecularly imprinted polymer capped Mn-doped ZnS quantum dots as a phosphorescent nanosensor for detecting patulin in apple juice

Wengang Zhang,Yong Han,Xiumei Chen,Xueli Luo,Jianlong Wang,Tianli Yue,Zhonghong Li

A Mn-doped ZnS quantum dots (QDs) based nanosensor for selective phosphorescent determination of patulin (PAT) was synthesized with 6-hydroxynicotinic acid (6-HNA) as dummy template via a surface molecular imprinting sol–gel process. FTIR and XRD indicated the successful graft of molecularly imprinted polymer (MIP) onto crystal QDs. Binding tests revealed that the MIP-QDs presented higher selectivity, adsorption capacity and mass transfer rate than non-imprinted polymers, demonstrating a specific recognition for PAT among competitive mycotoxins and its analogues with the imprinting factor of 2.02. The MIP-QDs could recognize PAT in a linear range of 0.43–6.50   μmol   L −1 with a detection limit of 0.32   μmol   L −1 and a correlation coefficient (R 2 ) of 0.9945. Recoveries of 102.9–127.2% with relative standard deviations <4.95% were achieved in apple juice samples which were in good agreement with high-performance liquid chromatography (HPLC) (P   >   0.05). The results indicated a simple phosphorescent nanosensor for PAT detection in complex matrix.

Joint Synovial Fluid Metabolomics Method to Decipher the Metabolic Mechanisms of Adjuvant Arthritis and Geniposide Intervention

Xiang Zhan,Huan Wu,Hong Wu

Rheumatoid arthritis (RA), a chronic systemic autoimmune disease, is mainly characterized by joint lesions and permanent loss of joint function. To discover the metabolic characteristics of RA and the underlying mechanisms in treatment with geniposide (GE), untargeted metabolomic analysis based on hydrophilic interaction liquid chromatography coupled to high-resolution mass spectrometry (HILIC–HRMS) was performed using the joint synovial fluid samples from adjuvant arthritis (AA) rats. Microdialysis (MD) was utilized to collect the dialysate samples precisely from the articular cavity of AA rats. Multivariate statistical analysis was then conducted to discover the metabolite changes induced by AA and to differentiate GE-related biomarkers. The mass spectrometry data are available on the Chorus website (https://chorusproject.org/pages/index.html) with the data set identifier 1680. The results showed that 20 metabolites differed significantly between AA rats and normal rats. GE treatment recovered the altered levels of the 13 metabolites mentioned above, such as palmitoylethanolamide (PEA), Cer (d18:0/22:0), and PC (18:1(11Z)/16:1(9Z)), and normalized glycerophospholipid metabolism. As evidenced by western blotting, the changes in PEA levels adjusted by GE were associated with the down-regulated expression of N-acylethanolamine-hydrolyzing acid amidase (NAAA) in synovial tissues. Taken together, the elucidation of metabolic changes of joint synovial fluid and how this is influenced by GE will promote future therapeutic interventions of RA.

Screening of a novel and thermostable nicotinate dehydrogenase-producing strain Bacillus paramycoides for efficient synthesis of 6-hydroxynicotinic acid

Zhi Chen,Zhaohe Dong,Xin Ju,Lishi Yan,Liangzhi Li,Donzhi Wei

6-Hydroxynicotinic acid emerged as a pivotal intermediate for fine chemicals. The current study aims to screen novel nicotinate dehydrogenase-producing strains. Based on the high-throughput UV coloration method, a novel nicotinate dehydrogenase-producing strain named Bacillus paramycoides was first and successfully screened from soil and gram staining as 16S rRNA gene sequencing was conducted to further confirm its accuracy. The fermentation medium formula is determined as seignette salt (5 g/L), tryptone (10 g/L), K2HPO4 (2 g/L), and nicotinic acid (4 g/L) after a single factor and orthogonal test. The activity of nicotinate dehydrogenase produced by B. paramycoides after optimization reached 1.23 U/mL, which was the highest ever reported. In addition, the enzyme characterization results revealed that the optimum reaction temperature and pH were 40 °C and 7.0, respectively. Furthermore, B. paramycoides nicotinate dehydrogenase exhibited good thermostability with more than 50% of the highest enzyme activity retained at 50 °C after 8 h incubation, which was better than ever reported. The pH stability results suggested that nicotinate dehydrogenase possessed a narrow pH ranging from 6.0 to 8.0. In addition, shaker speed demonstrated a positive relation with enzyme activity since shaker speed could affect mass transfer and dissolved oxygen concentration. Finally, it could completely transform nicotinic acid to 6-HNA within 3 h when the substrate loading is 15 g/L. In conclusion, these results indicated that B. paramycoides nicotinate dehydrogenase could serve as a potential candidate for bioproduction of 6-HNA.
