7-Hydroxyflavone improves nonalcoholic fatty liver disease by acting on STK24

Xinyi Qi,Yurou Zhang,Qichao Liao,Yang Xiao,Tianyu Jiang,Siqi Liu,Lei Zhou,Yixing Li

The escalating incidence of nonalcoholic fatty liver disease (NAFLD) is closely associated with a high-fat diet, leading to a decline in quality of life and significant health impairment. 7-Hydroxyflavone (7-HY) is a flavonoid known for its anti-inflammatory, anticarcinogenic, and antioxidant effects. This study aims to assess the ameliorative effects of 7-HY on NAFLD induced by a high-fat diet and elucidate underlying mechanisms. Oleic acid/palmitic acid-induced HepG2 cells and C57BL/6 mice on a high-fat diet were utilized as in vitro and in vivo models. In animal experiments, 7-HY was utilized as a dietary supplement. The 15-week in vivo experiment monitored body weight, body fat percentage, glucose tolerance, insulin tolerance, and metabolic indexes. Commercial kits assessed triglyceride (TG) and total cholesterol levels in cells, liver tissue, and blood. Discovery Studio identified potential targets of 7-HY, compared with NAFLD-associated targets in the GeneCards database. Results indicated 7-HY mitigated fat accumulation, hepatic steatosis, and oxidative stress induced by a high-fat diet. Furthermore, 7-HY showed potential efficacy in ameliorating abnormal glucose metabolism and promoting energy metabolism. Reverse target finding and molecular docking demonstrated a robust interaction between 7-HY and serine/threonine kinase 24 (STK24). Subsequent experimental results confirmed 7-HY's ability to inhibit TG deposition in HepG2 cells through interaction with STK24. In conclusion, 7-HY demonstrated the capacity to alleviate high-fat diet-induced NAFLD, presenting a novel strategy for the prevention and treatment of NAFLD.

Construction of rGO and GSH Electrochemical Sensor by Electrodeposition for Naringenin Sensing

Hui-Ting Hu,Jiang-Tao Xu,Bing-Lun Li,Guo-Cheng Han,Xiao-Zhen Feng,Heinz-Bernhard Kraatz

Naringenin (NRG), a flavanone compound present in citrus fruits, has a variety of beneficial physiological active functions such as antioxidant, anti-inflammatory, and hypoglycaemic. In this study, an sensor was constructed by electrodeposition and used for the electrochemical study of NRG. Reduced graphene oxide (rGO) and glutathione (GSH) showed the ability to synergistically amplify NRG signals and possessed good linearity in the concentration range of 10.00–1200.00 μmol l−1. The linear equation is Ip = 0.0776logc + 0.9353 (R2 = 0.9901), and the limit of detection is 3.33 μmol l−1. The sensor performed well in terms of reproducibility, stability, and selectivity, which in turn enabled the detection of NRG in tomatoes. The average recovery of the sensor is 95.68% to 111.92%, with RSD less than 11.89%. The results were also verified by Ultraviolet–visible spectroscopy(UV-vis). Furthermore, density-functional theory was employed to analyse the front track of the NRG, speculating that the NRG underwent a transfer of two electrons and two protons.

Design and Synthesis of Flavonoid-Based Mono-, Bis-, and Tri-Benzoxazines: Toward Elucidating Roles of Oxazine Ring Number and Hydrogen Bonding on Their Polymerization Mechanisms and Thermal Properties

Rui Yang,Kan Zhang

Designing smart chemical structures and gaining insight into the structure–property relationship can effectively guide the development of advanced green thermosetting resins. Herein, four flavonoid-based biophenols were used as sustainable raw materials for achieving novel mono-, bis-, and trifunctional benzoxazine monomers (HYD-a, CHR-a, API-a, and LUT-a). These monomers were developed to discover the roles of the oxazine ring number and hydrogen bonding on their polymerization mechanisms and thermal properties. Fourier transform infrared spectroscopy (FT-IR), nuclear magnetic resonance, and high-resolution mass spectrometry were employed to confirm the successful synthesis of benzoxazine monomers. Polymerization behavior of each benzoxazine was investigated using differential scanning calorimetry, thermogravimetric analysis (TGA), and in situ FT-IR. The thermal properties of the resulting thermosets were evaluated by TGA and microscale combustion calorimetry. With the designed varieties of flavonoid-based benzoxazine structures, it has been found that the intramolecular hydrogen bonding can act as a latent curing agent for reducing the curing temperature while maintaining excellent shelf life. In addition, the onset curing temperature decreases progressively with increasing the number of oxazine rings in the benzoxazine monomer. Interestingly, the polybenzoxazine derived from bis-benzoxazine (API-a) rather than trifunctional monomer shows the best thermal stability (Td10: 438 °C, Yc: 62%) and flame retardancy (HRC: 11.4 J g–1 K–1, THR: 2.22 kJ g–1), indicating the advantage of designing high-performance thermosets based on flavonoid-based bis-benzoxazines.

Influence of hydroxyl substitution on the inhibition of flavonoids in advanced glycation end-products formation in glucose-lysine-arginine Maillard reaction models

Li-Feng Li,Meng-Di Wang,Chen-Yang Zhang,Meng-Yao Jin,Hua-Lei Chen,Huan Luo,Tian-Yu Hou,Zhi-Jun Zhang,He Li

Advanced glycation end products (AGEs) generated from the Maillard reaction (MR) during food processing in the human physiological environment, have been proven to be significantly associated with various chronic metabolic diseases. In this study, 18 flavonoids were investigated to study their effects on AGEs formation during MR within a glucose-lysine-arginine model system. Five AGEs— N ε -carboxymethyl-lysine (CML), N ε -carboxyethyl-lysine (CEL), pyrraline, pentosidine, and argpyrimidine—were determined by high-performance liquid chromatography-mass spectrometry, with inhibitory rates ranging between 0 % and 71.35 %. Isorhamnetin and naringenin exhibited the strongest inhibitory effect on the formation of CML and CEL, respectively, whereas myricetin exhibited the strongest inhibitory effect on pyrraline, pentosidine, and argpyrimidine formation. In addition, each flavonoid was reacted with glyoxal (GO) and methylglyoxal (MGO) to investigate their trapping activities and adducts. The results showed that the 18 flavonoids could effectively clear GO and MGO, with clearance rates of 1.03 %–71.42 % and 0.93 %–69.37 % for GO and MGO, respectively. Six flavonoids—chrysin, naringenin, apigenin, luteolin, diosmetin, and kaempferol—could form adduct products with mono-/di-MGO, with flavonoid to mono-/di-MGO adduct ratios of approximately 6:1, 2:1, 8:1, 23:1, 10:1, and 3:1, respectively. The number and site of phenolic hydroxyl groups, as well as methoxy substitution on the B-ring in flavonoids, had little effect, but phenolic hydroxyl groups at the C-3 position in the C-ring impeded adduct formation. The flavonoids lowered the degradation of glucose itself, and decreased ammonia-induced degradation and Amadori rearrangement product oxidation due to their antioxidant activities, and they trapped the reactive 1,2-di‑carbonyl species via nucleophilic addition reaction to form AGEs.

Study on the Correlation Between Anthoxanthins Compounds' Reverse-Phase Chromatographic Behavior and Their Solubility

Changhai Sun,Mengxin Xiao,Biyue Cui,Shiyuan Sun,Shushuang Shen,Shuting Zhang,Xinran Zhang,Liting Mu

Based on the current state of research, the identification of flavonoid isomers still faces certain difficulties and challenges, and developing liquid chromatography separation conditions requires substantial resources. Therefore, this study constructed a quantitative structure-retention relationship (QSRR) model for anthoxanthins in reverse-phase chromatography by exploring quantitative relationships between chromatographic retention time (t R ) and implicit solvent model solvation free energy and hydrogen bonding energies in the mobile phase. A database was compiled by calculating the molecular descriptors of 1023 hydroxy-substituted flavonoids. When using UHPLC-HRMS technology combined with a database to identify unknown anthoxanthins, this model provides auxiliary identification to enhance the accuracy of the identification results. The model was constructed using 11 modeling compounds and two molecular descriptors to achieve the goal of predict compound retention times. The chromatographic retention time of anthoxanthins exhibited a linear correlation with their solvation free energy in the mobile phase. Furthermore, the feasibility of calculating the hydrogen bonding energies between anthoxanthins and the mobile phase using quantum chemical methods based on electron density was demonstrated. This study provides novel insights into the reverse-phase chromatographic separation of structurally similar compounds, while the constructed database offers extensive data support for isomer research.
