Mesoporous polydopamine delivering 8-gingerol for the target and synergistic treatment to the spinal cord injury

Yang Jinpei,Wang Meng,Zheng Shuai,Huang Ruodong,Wen Ganjun,Zhou Pan,Wang Wenbo,Zhou Shihao,Jiang Xinlin,Liu Shuangjiang,Li Zhizhong,Ma Dong,Jiao Genlong

In the treatment of spinal cord injury (SCI), the complex process of secondary injury is mainly responsible for preventing SCI repair or even exacerbating the injury. In this experiment, we constructed the 8-gingerol (8G)-loaded mesoporous polydopamine (M-PDA), M@8G, as the in vivo targeting nano-delivery platform, and investigated the therapeutic effects of M@8G in secondary SCI and its related mechanisms. The results indicated that M@8G could penetrate the blood-spinal cord barrier to enrich the spinal cord injury site. Mechanism research has shown that all of the M-PDA,8G and M@8G displayed the anti-lipid peroxidation effect, and then M@8G can inhibit the secondary SCI by suppressing the ferroptosis and inflammation. In vivo assays showed that M@8G significantly diminished the local injury area, reduced axonal and myelin loss, thus improving the neurological and motor recovery in rats. Based on the analysis of cerebrospinal fluid samples from patients, ferroptosis occurred locally in SCI and continued to progress in patients during the acute phase of SCI as well as the stage after their clinical surgery. This study showcases effective treatment of SCI through the aggregation and synergistic effect of M@8G in focal areas, providing a safe and promising strategy for the clinical treatment of SCI.

Online monitoring of Chinese herbal medicine production process toward lean six sigma: multimodal data fusion based on transformer architecture

Mintong Zhao,Zhilong Tang,Mingyang Zhou,Xiaohan Zhang,Xinyu Wang,Xingchu Gong

The manufacturing of Chinese medicines often faces challenges such as poor product consistency, high solvent consumption, and long processing times. The percolation process is a commonly used technique for extracting medicinal herbs. Significant variation in percolate concentration and low concentration near the endpoint make it difficult for existing online detection technologies to accurately determine target component concentrations. To address this, the study developed an online monitoring system integrating multi-modal sensors for physical quantity, image, and spectral data. Using Xiaochaihu capsules, real-time multimodal data were collected, including over 20,000 physical quantity points, 14,000 spectra, and 14,000 images. A Transformer-based framework, PMFormer, was proposed, with interpolation-based data augmentation to alleviate the “data-rich but label-scarce” problem. PMFormer achieved R 2 values of 0.96, 0.94, and 0.91 for 6-gingerol, 8-gingerol, and adenine, with RMSEs below 2.4, 0.4, and 1.8 μg/mL, respectively. A quantitative extraction control strategy was developed, determining the percolation endpoint when the accumulated total mass of collection (ATMC) met quality control limits. Validation showed improved consistency, reduced solvent use, and enhanced efficiency, aligning with Lean Six Sigma concepts. This study provides a reference for online monitoring of TCM percolation processes and demonstrates the potential of multimodal data fusion in pharmaceutical manufacturing.

Effects of lactic acid bacteria on sensory evaluation and flavor compounds of fermented baby ginger jam

Yanyan Nong,Hui Li,Yueen Ou,Luanyu Xie,Caimin Fen,Hong Gao,Guidong Huang,Zhiming Zhang,Senwei Hu

The study optimized the processing technology of baby ginger jam and determined the optimal ratio of thickeners: carboxymethyl cellulose sodium at 0.4%, guar gum at 0.6%, and xanthan gum at 0.2%. Furthermore, the flavor profiles of unfermented and fermented ginger jam were identified using electronic nose and headspace-gas chromatography-ion mobility spectroscopy, identifying 68 volatile compounds including aldehydes, alcohols, ketones, esters, terpenes, acids, and other compounds.Through the combined analysis of relative odor activity value and variable importance in projection, five compounds were identified as characteristic flavor markers of ginger jam: 2-methylpropanal, methyl benzoate, methyl heptenone, linalool, and acetic acid (monomer). The experimental data demonstrated that lactic acid bacteria remarkably improved the flavor profile of ginger jam. Notably, a 1:3 ratio of Lactobacillus plantarum to Lactococcus lactis achieved the most optimal flavor enhancement, and the fermentation did not reduce the content of gingerol. This research provided valuable insights for the industrial production and flavor optimization of baby ginger jam.

Optimized High-Pressure Ultrasonic-Microwave-Assisted Extraction of Gingerol from Ginger: Process Design and Performance Evaluation

Yang Zhang,Siyi Yang,Wensi Li,Xiaoyan Li,Xiangqin Lai,Xiang Li,Wuwan Xiong,Bo Zhang

This study employed high-pressure ultrasonic-microwave-assisted extraction (HP-UMAE) to extract gingerols from ginger. The extraction yield and total polyphenol content of the extracts were determined. Their antioxidant activity was assessed by DPPH and ABTS radical scavenging assays, and compared with extracts obtained by leaching extraction, reflux extraction, ultrasonic-assisted extraction (UAE), microwave-assisted extraction (MAE), and ultrasonic-microwave-assisted extraction (UMAE). The results demonstrated that HP-UMAE achieved the highest extraction yield and the strongest ABTS radical scavenging activity among the evaluated methods. Furthermore, HP-UMAE extracts exhibited the highest concentrations of key gingerol constituents: 6-gingerol (14.29 mg/L), 8-gingerol (0.38 mg/L), 10-gingerol (1.95 mg/L), and 6-shogaol (4.32 mg/L). This enhanced efficacy is attributed to the synergistic combination of ultrasonic cavitation and microwave-induced thermal effects under elevated pressure. This synergy creates conditions promoting cellular wall disruption, facilitating the release of intracellular components, while concurrently enhancing solvent penetration and gingerol solubility. Scanning electron microscopy (SEM) analysis confirmed the significant structural damage inflicted on ginger cell walls following HP-UMAE treatment. The process parameters for HP-UMAE were optimized using single-factor experiments. The optimal extraction conditions were determined as follows: microwave power 800 W, ultrasonic power 1000 W, liquid-to-solid ratio 55:1, and temperature 100 °C (corresponding pressure 2 MPa). Under these optimized parameters, the extraction yield and ABTS radical scavenging rate reached their peak performance, yielding values of 4.52% and 43.23%, respectively.

Analytical quality by design based on knowledge organization: A case study of developing an ultrahigh-performance liquid chromatography method for the detection of phenolic compounds

Yanni Tai,Mintong Zhao,Feng Ding,Gelin Wu,Haibin Qu,Ping Gong,Yongjian Xie,Peng Zhou,Xingchu Gong

Introduction Despite numerous successful cases, there are still some challenges in using analytical quality by design (AQbD) for the development of analytical methods. Knowledge organization helps to enhance the objectivity of risk assessment, reduce the number of preliminary exploratory experiments, identify potential critical method parameters (CMPs) and their scope. Objective In the present study, we aimed to develop a simple, rapid, and robust analytical method for detecting phenolic compounds in Xiaochaihu capsule intermediates utilizing knowledge organization. Methods Knowledge organization and AQbD were combined to obtain the initial analytical conditions through knowledge collection, extraction, reorganization, and analysis. The quantitative relationship between critical method attributes (CMAs) and CMPs was then established by a definitive screening design. The method operable design region was calculated using an exhaustive Monte Carlo approach based on the probability of reaching the standard. Robustness investigation and methodological validation were finally performed. Results Analytical target profiles, CMAs, potential CMPs, and initial analytical conditions were initially identified, and the optimized ranges of operating parameters were obtained. A UHPLC method was successfully established for the analysis of phenolic compounds in ginger–ginger pinellia percolate, and the method validation outcomes were also satisfactory. Conclusion The developed method can be a reliable means to detect the phenolic compounds of Xiaochaihu capsule intermediates. Knowledge organization provides a new approach for making better use of prior knowledge, significantly enhancing the efficiency of analytical method development. The approach is versatile and can be similarly applied to the development of other methods.

Inhibitory mechanism of xanthine oxidase by 6-, 8- and 10-gingerol: Enzyme kinetics, multi-spectroscopy and molecular simulations

Rui Wang,Pei-Feng Guo,Jing Nie,Xing Hu,Yu-Wei Wu,Shu Zhu,Bo-Ru Chen,Jian Li,Xin-An Zeng,Fei-Yue Xu

In this study, the inhibitory activity and mechanism of three gingerols (6-, 8- and 10-gingerol) on xanthine oxidase (XOD), a key enzyme responsible for the formation of uric acid, were evaluated. The three gingerols reversibly inhibited XOD activity in a mixed manner with IC 50 values of (13.16 ± 0.41), (10.18 ± 0.22) and (6.88 ± 0.11) μM, respectively. 10-gingerol exhibited stronger inhibitory ability, which might contribute to its superior antioxidant properties, higher binding affinity and more significant conformational changes for XOD compared to other gingerols. Fluorescence quenching and molecular docking results indicated that gingerols acted in the vicinity of the flavin adenine dinucleotide in XOD, interacting with the surrounding key amino acid residues through hydrogen bonding and van der Waals forces, thereby preventing substrate entry and the release of catalytic products. Molecular dynamics simulations showed that the addition of gingerol decreased the stability and increased the structural density of XOD. These findings could provide valuable data for the application of gingerols in new XOD inhibitors and hypouricemic functional foods.
