Norway spruce is an important coniferous species in boreal forests. Root and stem rot diseases caused by the necrotrophic pathogen Heterobasidion parviporum threaten the wood production of Norway spruce which necessitates the search for durable control and management strategies. Breeding for resistant traits is considered a viable long-term strategy. However, identification of potential resistant traits and markers remains a major challenge. In this study, short-term disease resistance screening was conducted using 218 Norway spruce clones from 17 families. Disease resistance was evaluated based on the size of necrosis lesion length following infection with the pathogen. A subset of needles/branches from clones with small (partial resistant) or large (susceptible) lesions were used for terpene analysis and transcriptomic profiling. The results revealed that the content of monoterpene linalool and 1,8-cineole and their respective encoded genes were significantly more abundant and highly expressed in the partial resistant group. Furthermore, linalool and 1,8-cineole were demonstrated to have inhibitory effect on the growth of the pathogen H. parviporum , with morphological distortion of the hyphae. RNAseq analysis revealed that transcript of pathogen genes involved in the regulation of carbohydrate metabolism and stress responses were significantly decreased in presence of the terpenes. The results suggest the relevance of monoterpenes together with jasmonic acid precursor and some genes involved in phenylpropanoid biosynthesis, as constitutive tolerance factors for Norway spruce tolerance against necrotrophic pathogen. The high level of necrosis related cell death gene expression might be factors critical for host susceptibility and disease development.

Vascular endothelial senescence is an important pathophysiological factor in the development and exacerbation of cardiovascular health problems linked to diabetes mellitusn (DM). Accumulating evidence confirms that 1,8-cineole has multiple pharmacological properties, including anti-inflammatory, anti-microbial, and antioxidant activities. We investigated whether 1,8-cineole could ameliorate cardiovascular diseases and endothelial dysfunction, as the pharmacological properties and mechanism of diabetic vascular ageing remain unknown. Our results revealed notable senescence biomarkers in both in vivo and in vitro models. Treatment with 1,8-cineole alleviated lipid profiles and vascular senescence in mice with DM. Additionally, bioinformatics analysis suggested that peroxisome proliferator-activated receptor-γ (PPAR-γ) plays a crucial role in DM and ageing. We confirmed the binding capacity PPAR-γ with 1,8-cineole. Accordingly, experiments with the PPAR-γ agonist rosiglitazone, the PPAR-γ inhibitor GW9662, and PPAR-γ siRNA were performed to validate the pharmacological characteristics of 1,8-cineole. Finally, we clarified that 1,8-cineole can directly target PPAR-γ protein, as verified by cellular thermal shift assay, drug affinity responsive target stability, and surface plasmon resonance analyses. Taken together, these results provide the first evidence that 1,8-cineole ameliorates DM-induced vascular endothelial ageing via stabilising PPAR-γ protein by promoting deubiquitination at the Lys-466 site.

As an eco-friendly compound from essential oils, 1,8-cineole (cineole, eucalyptol) has the potential to replace the ozone depleting industrial solvents. This paper presents experimental (liquid + liquid) equilibrium (LLE) data for four alcohol–water systems containing 1,8-cineole. To evaluate the phase equilibrium properties of 1,8-cineole in aqueous alcohol mixtures, LLE values for the ternary systems (water + methanol or ethanol or 1-propanol or 1-butanol + 1,8-cineole) were determined with a tie-line method at T = 298.15 K under atmospheric pressure. The well-known Hand, Bachman and Othmer–Tobias equations were used to test the reliability of the experimental results. The binodal curves and distribution ratios of alcohol in the mixtures are shown and discussed. The experimental LLE values were satisfactorily correlated by the NRTL and UNIQUAC models.

The Eucalyptus plant releases allelopathic chemicals into the environment mostly through the essential oils volatilized from the leaves. This study discussed the composition of the leaf oils of few seven-year-old varieties like Eucalyptus pellita (E. pel), Eucalyptus camaldulensis (E. cama), Eucalyptus grandis (E. gra), Eucalyptus dunni (E. dun), Eucalyptus saligna (E. sal), and E. grandis × E. urophylla (E. gra×E. uro) and three-year-old E. grandis × E. urophylla (E. gra × E. uro (three)). It determined the allelopathic mechanism and the types of chemical components playing the leading role. Essential oil was obtained by hydrodistillation and analyzed by the Gas Chromatography-Mass Spectrometry (GC-MS) method. In order to determine the effect of allelopathy, seed germination experiments were carried out at different concentrations (10-100 mL/L) of the E. Gra × E. uro leaf oil (EO) and the major components. The wheat seeds germinated by adding 1,8-cineole were used to determine the activity of α-amylase. Moreover, the mRNA expression of α-amylase in seeds was studied. The major chemical class in the essential oil was oxygenated monoterpene; 1,8-cineole (20.2-67.5%) displayed the highest content. Other substances that were high in content and ubiquitous included α-pinene (0.3-21.8%), α-terpineol (0.44-19.24%), and borneol (0.81-3.05%). The four chemical constituents and EO influenced the germination and growth of the three plants. Among them, 1,8-cineole exhibited the strongest inhibitory effect. The α-amylase activity of the 1,8-cineole-treated wheat seeds had decreased significantly. Molecular evidence suggested that 1,8-cineole decreased the α-amylase gene (AMY) expression.

1,8-Cineole has anti-inflammatory, antibacterial and physiological properties and its high purity is important to application. However, 1,8-cineole usually mixes with monoterpene hydrocarbons in plant essential oils and they are difficult to separate via distillation due to their close boiling points. In this study, a series of lactic acid (LA) based deep eutectic solvents (LaDESs) were formed and used in liquid–liquid separation of 1,8-cineole and monoterpene hydrocarbons. LaDES formed by LA and tyrosine (Tyr) showed 24.8 selectivity of 1,8-cineole to γ-terpinene at 25 ℃. Synergists or auxiliary extractant such as tert -butanol can improve the distribution coefficient but reduce the selectivity. Some effect factors to the extraction were investigated and 99% purity of 1,8-cineole was achieved in the final product. The extraction thermodynamic parameters and kinetic process were studied. The re-extraction and the LaDES recovery were carried out satisfactorily. It further proved that the separation of 1,8-cineole from α-pinene, α-terpinene and ρ-cymene could be performed well with the LaDES respectively. Liquid-liquid extraction separation of 1,8-cineole and γ-terpinene has the advantages of low energy consumption, high selectivity and safety. It would provide an alternative strategy for separating 1,8-cineole and monoterpene hydrocarbons whose boiling points are close.

1, 8-Cineole (Cin), a cyclic monoterpenoid derived from tea trees and eucalyptus species, exhibits diverse pharmacological properties. Yet, its therapeutic impact and underlying mechanism against Staphylococcus aureus (S. aureus) pneumonia remain to be elucidated. In this study, metabolomics based on UPLC-MS/MS was integrated with network pharmacology, molecular biology, and molecular docking to investigate the effects of Cin. The findings demonstrated that Cin markedly reduced mortality and lung bacterial load, lessened pulmonary damage while suppressing the levels of proinflammatory factors, including tumor necrosis factor α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6) in the bronchoalveolar lavage fluid (BALF) of infected mice. Additionally, 19 metabolites, primarily involved in tryptophan metabolism and arginine biosynthesis, were notably modified by Cin via suppressing the enzymatic activity of indoleamine 2, 3-dioxygenase 1 (IDO1) and inducible nitric oxide synthase (iNOS), thereby attenuating the inflammatory response. Notably, knockdown of IDO1 or iNOS significantly diminished the anti-inflammation effect of Cin. In conclusion, our study validates the therapeutic potential of Cin against S. aureus pneumonia via anti-inflammation by downregulating IDO1 and iNOS. Our results provide a theoretical basis of natural substances applied in bacterial pneumonia treatment.

Pork is one of the meats widely consumed all over the world, while stewing is preferred because it can retain the nutritional content of the meat. After stewing, pork still retains some odors, such as bloody aroma, which affects the flavor quality. Therefore, people often add spices during the stewing process to reduce the off-flavor, but the mechanism of the effect of spices on the off-flavor has not been clarified. This study examined the pork broth before and after the addition of spices and determined the off-flavor compounds ( (E) -2-nonenal, (E) -2-heptenal, acetic acid, and valeric acid) in pork broth and key spice compounds (linalool, 1,8-cineole, and (E) -anethole) in pork broth with spices. The mechanism of the influence of spices on the off-flavor compounds of pork broth was clarified through the sensory perceptual interaction between off-flavor compounds and key spice compounds. The S-curve method was used to analyze the perceptual interactions between off-flavor compounds and key spice compounds. The results showed that linalool, 1,8-cineole, and (E) -anethole all had a masking effect on the aroma of off-flavor compounds (S-curve ratios = 1.58, 2.21, and 5.84, respectively), and the inhibitory effect was strongest when the three compounds acted together (S-curve ratio = 42.49).
