As the first step of resource utilization, the extraction and separation of active ingredients play an important role in the evaluation and application of their activity. In this study, a macroporous resin purification process was optimized to obtain high-purity sea buckthorn flavonoids (SBF). The AB-8 resin exhibited optimal adsorption and desorption properties, with adsorption and desorption rates of 75.25% and 84.78%, respectively. Under optimal purification conditions, the purity of the flavonoid extract was enhanced from 17.17 ± 0.19% to 63.2 ± 0.16%. Kinetic and thermodynamic analyses demonstrated that the purification of SBF on AB-8 resin conformed to the pseudo-secondary kinetic model and the Langmuir adsorption isotherm model. The major components of the SBF were taxifolin, isoquercitrin, narcissoside, and quercetin. The SBF not only inhibited lipopolysaccharide-induced nitric oxide production but also exerted significant anti-inflammatory effects, suppressing the expression of proteins such as nitric oxide synthase, tumor necrosis factor, interleukin 6, and interleukin 1β and inhibiting apoptosis. Therefore, the purified SBF demonstrated improved anti-inflammatory activity, which can be applied in the development of drugs and functional foods.
In this paper, porous Brønsted acidic ionic liquid modified catalyst ILs-AB-8 was successfully synthesized by using 3 steps of Friedel–Crafts hyper-crosslinking synchronous chloromethylation, imidazlole grafting and sulfonic functionalization using macroporous polystyrene AB-8 resin as a raw material. The porous ionic liquid immobilized catalyst ILs-AB-8 exhibits a hierarchical pore structure by the coexistence of abundant macropore, mesopore, and micropore. The catalyst IL 2 -AB-8 with dual Brønsted acid sites(-SO 3 H acid site and p-methylbenzenesulfonic acid site), large surface area (327.19 m 2 /g), high ionic liquid loading amount (IEC 2.03 mmol/g) displayed excellent catalytic activity, with methyl oleate yield up to 95.65% at 338 K in 100 min. The enhanced catalytic performance of IL 2 -AB-8 can be ascribed to the novel Friedel–Crafts hyper-crosslinking process in conjunction with chloromethylation, coupled with th in situ generated activating chloromethyl group incorporated onto the surface of the AB-8 carrier. The hyper-crosslinking approach also effectively transforms the macroporous AB-8 carrier into a multi-level pore structure. This work highlights that IL 2 -AB-8 with high specific surface area, ionic liquid loading content, and strong covalent connection with carrier was coordinated to provide an efficient metal-free heterogeneous catalyst for biodiesel production. Resin AB-8 was screened from seven resins with different polarity and particle size because of its better decoloration and deproteinization efficiency to polysaccharide from the root of Actinidia valvata (AvPs), and then the optimized parameter conditions were applied by the single-factor experiments and were shown as follows: a concentration of crude polysaccharide of 3 mg/mL, an adsorption time of 40 min, an initial pH of 7.0, a flow rate of 1.5 BV/h. In addition, the results of HPGPC, FT-IR, PMP-HPLC-DAD, 1H NMR and Congo red test showed that the sugar chain, structural group, monosaccharide composition and triple-helix conformation structure of AvPs were not broken during resin AB-8 treatment. Moreover, AvPs could keep the stability of biological activity after the resin AB-8 treatment, and revealed an obviously antioxidant and anticancer activities in vitro as well. This research suggests that this method is promising for the deproteinization and decolorization of AvPs which could be explored as a potential antioxidant or anticancer agent for use in functional food or medicine.
