Apple necrotic mosaic virus (ApNMV) is associated with apple mosaic disease in China. However, the mechanisms of ApNMV infection, as well as host defence against the virus, are still poorly understood. Mitochondrial ATP synthase plays a fundamental role in the regulation of plant growth and development. However, mitochondrial ATP synthase function in response to virus infection remains to be defined. In the present study, a yeast two-hybrid (Y2H) screening revealed that the apple mitochondrial ATP synthase oligomycin sensitivity-conferring protein (OSCP) subunit (MdATPO) interacts with ApNMV coat protein (CP). It was further verified that overexpression of MdATPO in Nicotiana benthamiana inhibited viral accumulation. In contrast, silencing of NbATPO facilitated viral accumulation, indicating that ATPO plays a defensive role during ApNMV infection. Further investigation demonstrated that ApNMV infection accelerated abscisic acid (ABA) accumulation, and ABA negatively regulated ATPO transcription, which was related to the ability of ABA insensitive 5 (ABI5) to bind to the ABA-responsive elements (ABREs) of the ATPO promoter. Taken together, our results indicated that transcription factor ABI5 negatively regulated ATPO transcription by directly binding to its promoter, leading to the susceptibility of apple and N. benthamiana to ApNMV infection. The current study facilitates a comprehensive understanding of the intricate responses of the host to ApNMV infection.

Volatile compounds were analyzed using gas chromatography-mass spectrometry (GC-MS) and gas chromatography-olfactometry (GC-O). Results showed that nonanal had the highest odor activity value in the filling grains. Nonanal was enhanced by application of JA and inhibited by ABA in the seedlings of Kam sweet rice. C9-aldehydes are significant odorants in many rice types and known products of the 9-lipoxygenase and 9-hydroperoxide lyases encoded by the OsLOX3 and OsHPL1 genes, respectively. OsLOX3 mRNA expression was higher in aromatic rice compared to a nonaromatic cultivar. mRNA expression of ABI5 and MYC2 negatively correlated with OsLOX3 expression ( r = −0.841, P < 0.01; and r = −0.512, P < 0.05). In conclusion, the transcription factors ABI5 and MYC2 might be involved in ABA and JA modulation of nonanal biosynthesis, which could be used to improve rice quality. Practical Applications Nonanal and 2-acetyl-1-pyrroline are important and potent flavor compounds in aromatic Kam sweet rice. The OsLOX3 gene was potentially identified as a critical gene in aromatic Kam sweet rice and the enhancing effect of JA on nonanal biosynthesis was confirmed. The transcription factors ABI5 and MYC2 seem to regulate the expression of the fragrance gene OsLOX3 while MYC2 act as a positive regulator involved in JA-mediated nonanal biosynthesis. ABI5 participate in the ABA signaling pathway to down-regulate the expression of OsLOX3. These results will help understand the pathways involved in the biosynthesis of rice fragrance, which could be used to improve the quality of rice.

Seed dormancy and germination are dynamically regulated by abscisic acid (ABA), with transcription factor ABA INSENSITIVE 5 (ABI5) serving as a key repressor of germination under stress. Here, we identify QUIRKY (QKY), a MULTIPLE C2 DOMAIN AND TRANSMEMBRANE REGION PROTEIN (MCTP) in Arabidopsis , as a critical regulator of ABA signaling through posttranslational control of ABI5. QKY interacts with ABI5 and facilitates its cytoplasmic degradation via the E3 ubiquitin ligase KEEP ON GOING (KEG), promoting the recruitment of ABI5 into phase-separated condensates. Loss of QKY function enhances ABA hypersensitivity, reducing seed germination rates and impairing seedling establishment. These findings highlight QKY as a scaffold that integrates phase separation and targeted ubiquitination to fine-tune ABA responses, balancing dormancy and germination. Our study expands the understanding of posttranslational ABI5 regulation and reveals MCTPs as central players in coordinating growth-stress trade-offs during early plant development.

SUMMARY Abscisic acid (ABA) plays a crucial role in enforcing seed dormancy and suppressing germination under stress conditions by stabilizing key transcriptional repressors. However, the mechanisms by which plants actively attenuate this repression to resume developmental growth remain largely unclear. In this study, we identify chloroplast vesiculation (CV) as a negative regulator of ABA signaling in Arabidopsis. Silencing of CV resulted in hypersensitivity to ABA during seed germination and stomatal closure, whereas its overexpression conferred ABA insensitivity. We demonstrate that CV physically interacts with and directly promotes the vacuolar degradation of two key positive regulators of ABA signaling—the transcription factor ABI5 and the WD40-repeat protein XIW1. Genetic epistasis analyses confirmed that ABI5 and XIW1 function downstream of CV, as loss-of-function mutations in either ABI5 or XIW1 suppressed the ABA-hypersensitive phenotype of CV-silenced plants. Furthermore, we found that ABA itself negatively regulates CV at both the transcriptional and post-translational levels, forming a feedback loop that fine-tunes ABA sensitivity. Our study thus uncovers a previously unknown regulatory module in which CV attenuates ABA responses by targeting ABI5 and XIW1 for vacuolar degradation, thereby facilitating the transition from seed dormancy to germination.
