Antioxidant Activity and Total Polyphenols Content of Camellia Oil Extracted by Optimized Supercritical Carbon Dioxide

Weifang Wang,Sai Han,Zhen Jiao,Jiangrui Cheng,Junying Song

In this study, Camellia oil is co-extracted from Camellia oleifera seeds and green tea scraps by supercritical carbon dioxide (SC-CO 2 ), which is optimized on the extraction yield, ABTS-scavenging activity, and total polyphenols content (TPC) of oil by single-factor experiments combined with response surface methodology (RSM). The extraction temperature, pressure, dynamic time, carbon dioxide (CO 2 ) flow rate, and seed mass ratio were investigated with single-factor experiments. The results indicated the optimum CO 2 flow rate and dynamic extraction time were 15 L hour −1 and 60 min (i.e., 2.382 kg CO 2 /100 g sample). Furthermore, the complicated effects of extraction temperature (40–50 °C), pressure (20–30 MPa), and seed mass ratio (0.25–0.75) were optimized by RSM based on the Box–Behnken design (BBD). The models with high R -squared values were obtained and used to predict the optimum operating conditions of the process. Under the optimum operating conditions (i.e., temperature of 46 °C, pressure of 30 MPa, and seed mass ratio of 0.35), the extraction yield, ABTS-scavenging activity, and TPC of oil were 14.43 ± 0.17 g/100 g sample, 73.70 ± 0.34%, and 2.18 ± 0.05 mg GAE/g oil, which were in good agreement with the predicted values. In addition, the experiments indicated that the Camellia oil obtained was rich in polyphenols, resulting in better oxidation stability and antioxidant activity than the original oil.

Isolation, Identification and Activities of Natural Antioxidants from Callicarpa kwangtungensis Chun

Hao Cai,Zhiyong Xie,Guanghui Liu,Xiuman Sun,Guangtian Peng,Baoqin Lin,Qiongfeng Liao

Reactive oxygen species leads to some diseases associated with oxidative stress. Callicarpa kwangtungensis Chun (CK) is a common remedy in traditional Chinese medicine and possesses diverse biological activities involving antioxidant properties; its main compounds phenylethanoid glycosides (PG) and flavonoids are always reported as antioxidants. In order to develop CK as a safe and activated antioxidant, our investigation was performed to validate antioxidant properties and assess which types of compounds (similar polarity or similar structure), even which compounds, played the role of antioxidants. The extracted compounds of CK were analyzed qualitatively and quantitatively by HPLC-DAD-ESI-Trap MS and UV for their contents and antioxidant activities. The correlations between antioxidant activities and known contents were respectively counted and a semi-quantitative experiment was designed to screen antioxidant compounds of CK with HPLC-UV. The n-butanol fraction (BF) showed the highest total phenolic and flavonoid contents (TPC, TFC), and three PG (forsythiaside B, poliumoside and acteoside) contents. BF showed the significantly best ( P <0.05) activities in most assays. There were significant correlations ( P <0.05) between DPPH•, ABTS + •, •O 2 − scavenging, Cu 2+ -chelating, anti-lipidperoxidation activities and TPC. BF also has significant antioxidant activities on CCl 4 -induced acute liver injury Mice and TBHP-reduced HepG2 cells. Nine PG (forsythiaside B, poliumoside, acteoside, alyssonoside, brandioside and their derivatives) and one flavone (rhamnazin) were screened out as antioxidants. BF in CK contained abundant polyphenolic, which reflected some definite antioxidant properties. The antioxidant compounds consisted at the least of nine PG and one flavone.

Unexpected variations in the effects of ultrasound-assisted myofibrillar protein processing under varying viscosity conditions

Jiahui Chen,Jiale Chai,Xiaomei Sun,Ye Tao,Xing Chen,Guanghong Zhou,Xinglian Xu

The efficient synthesis of myofibrillar protein(MRN)-gallic acid (GAD) complex in ultrasound (UID)-assisted processing is a challenging problem in food manufacturing. In this investigation, the effect of viscosity characteristics on the efficiency of UID processing in MRN-based beverages was analyzed. Both viscosity and surface tension can increase sono-physico-chemical effects on the degradation of terephthalic acid and crystal violet, with surface tension having a more significant effect (negative correlation, R 2  = 0.99) than viscosity (positive correlation, R 2  = 0.79). The structural indicators and microstructure demonstrated that the reaggregation and refolding of the MRN structure during the modification procedure occurred with relatively small three-dimensional dimensions. Compared to the MRN/GAD4 group, the water contact angle of the MRN/GAD7 system enhanced by 129.44%, leading to greater system stability. The ABTS-scavenging capacity of the system increased by approximately 19.45% due to the increase in viscosity of these two categories.

Physicochemical properties, antioxidant and anti-inflammatory activities of polysaccharides from lotus leaves by pectinase-assisted extraction

Hu Xu,Gen Li,Cheng Wan,Xiao-Yi Wei,Tao Zhou

In order to improve the extraction yield of lotus leaf polysaccharide (LLP) and offer scientific foundation for utilization of LLPs in the food and pharmaceutical industries, pectinase-assisted extraction method was employed for the preparation of LLP. The chemical composition, physicochemical properties (e.g. rheological and thermal properties etc.), antioxidant and anti-inflammatory effects of LLP and its two purified fractions (LLP1 and LLP3) obtained using DEAE-52 and subsequent Sephadex G-100 column chromatography were also investigated. Antioxidant assays indicated that the polysaccharides had remarkable DPPH• and ABTS +• scavenging activities. In H 2 O 2 -induced RAW264.7 cells, LLPs showed marked protective effect on oxidative stress, reducing the MDA and ROS contents and increasing the CAT and SOD activities. LLPs were also found to effectively modulate the expressions of CAT, SOD1, Nrf-2, HO-1, NQO1 and Keap1 mRNA. Thus, the potent antioxidant effects of LLPs were achieved possibly by regulating the Keap1/Nrf-2/HO-1 signaling pathways. In lipopolysaccharide-treated RAW264.7 macrophages, LLPs reduced phagocytic ability, decreased release of NO, TNF-α, IL-1β and IL-6, and increased release of IL-10 and TGF-β1, indicating their potent anti-inflammatory effect. LLPs downregulated expressions of iNOS, IL-1β, IL-6, TNF-α, COX-2, and TLR-4 mRNA. Therefore, modulation of NF-κB signaling pathway is possibly the important anti-inflammatory mechanism of LLPs.

Optimization of ultrasound-assisted deep eutectic solvent extraction, characterization, and biological studies of polysaccharide from seeds of Toona sinensis

Wen Yao,Chen Geping,Zhang Xiaobing

The present study aimed to evaluate the polysaccharides extracted from Toona sinensis seeds using ultrasound-assisted deep eutectic solvent (DES). The extracted polysaccharides (TSRP) were purified and studied for their hypoglycemic and antioxidant activities. The purified polysaccharides (TSRPs-1) were hydrolyzed and derivatized to study their monosaccharide composition. The optimum extraction conditions were DES (choline chloride to oxalic acid) at a molar ratio of 1.473:1, ultrasound time of 50.643 min, and a material-to-liquid ratio of 1:31.838 g/mL. The ultrasound-assisted DES extraction was more effective (56.1%) as compared to the water extraction (35.89%). The inhibition of α-glucosidase by TSRPs-1 was 42.77% at a concentration of 0.8 mg/mL. At 3.2 mg/mL, TSRPs-1 inhibited α-amylase by 34.38%. The clearance of DPPH by TSRPs-1 was 24.36% at 3.2 mg/mL. The IC 50 for ABTS· + clearance by TSRPs-1 was 0.233 mg/mL. It was shown that the monosaccharides of TSRPs-1 were composed of Man, Rha, GlcA, GalA, Glc, Gal, Xyl, and Ara in a molar ratio of 1:563.13:41.81:51.20:21.81:2244.58:3.73:1130.48. In this paper, the process of extracting TSRP with ultrasound-assisted DES was investigated and optimized. TSRPs-1 possesses certain hypoglycemic and antioxidant activities. Therefore, this paper can provide an experimental basis for the establishment of an industrialized production process of polysaccharides and the study of their biological activities.

Comparative Study on the Antioxidative Effects and α-Glucosidase Inhibitory Potential In Vitro among Ellagic Acid and Its Metabolites Urolithins

Zhao-Rong Li,Rui-Bo Jia,Yurong Mo,Haozheng Wang,Donghui Luo,Chunxia Zhou,Mouming Zhao

The current study compared the radical scavenging and α-glucosidase inhibition potentials of ellagic acid (EA) and its metabolites, urolithins (Uros), and further explored the structure–activity relationship. The outcomes indicated that urolithin M5 (Uro-M5), EA, and urolithin M6 (Uro-M6) exhibited superior 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity; EA and urolithin D (Uro-D) expressed better ABTS scavenging ability, and EA and Uro-M5 showed preferable α-glucosidase inhibition activity. The results of CD spectra and fluorescence spectral analysis explained the interaction between Uros and α-glucosidase. Correlation analysis indicated that hydroxyl groups were crucial for the antioxidative effect, while C-8 OH contributed greatly to the α-glucosidase inhibition activity. Quantum mechanical analysis showed that both EA and Uros exhibited strong electrophilic properties. These comparative results showed a biological discrepancy between Uros and provided essential information for exploring the bioactive application of EA as a functional ingredient or dietary supplement.

Preparation of antibacterial composite film based on arginine-modified chitosan and its application in the preservation of ready-to-eat sea cucumber

Jinghe Sun,Yimeng Li,Tingting Yan,Jingfeng Yang

An edible composite film was developed and applied for ready-to-eat sea cucumber storage to improve the product quality. The PAC film base is first prepared by mixing 0.5 % glycerin (GL) with 4 % polyvinyl alcohol (PVA) and 1 % arginine-modified chitosan (Arg-CTS) in the same volume. After the addition of nano-ZnO (ZnO) and thymol (Thy) to the PAC film base, the mechanical properties and functions were tested. Compared to the PAC film, the PAC-ZnO-ThyH composite film showed a 1.34-fold increase in the DPPH scavenging rate and a 2.19-fold increase in the ABTS scavenging rate. Contrary to the PAC film, the inhibition zone diameter of Escherichia coli and Staphylococcus aureus significantly increased by 2.35 and 4.08 folds in the PAC-Zno-ThyH film, respectively. After applying the PAC-ZnO-ThyH film to store ready-to-eat sea cucumber for 10 days, there was a significant reduction in weight loss, total volatile basic nitrogen (TVB-N), and lipid oxidation levels to 1.47 and 1.26 folds to the Ctrl group. After preservation, the hardness and chewiness of ready-to-eat sea cucumber were maintained at 1079.62 ± 138.86 N and 913.73 ± 175.79 N, respectively. The novel PAC-ZnO-ThyH composite film can be used as an active food packaging for promising seafood applications.

Bianling Jiang,Fuyun Geng,Ran Chang,Mengting Ruan,Ying Bian,Lisheng Xu,Fan Feng,Qiong Chen

The present study was executed to analyze the functional phytochemicals of hulless barley grass grown under different intensities of ultraviolet stress. The wheat seedlings were imposed to 0.5, 1.0, 1.5, 2.0, and 2.5 h ultraviolet radiation and harvested in different times at vegetative stage. Specifically, the contents of total polyphenols, total flavonoids, total triterpenes, total polysaccharides, proanthocyanidins, and chlorophyll were determined and antioxidants capacity was evaluated by OH• and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) scavenging ability. A mathematical model (Technique for Order Preference by Similarity to Ideal Solution, TOPSIS) was also employed for the comprehensive evaluation of functional components of hulless barley grass at different growth stages. The results showed that the UV stress could efficiently improve/preserve the contents of total polyphenols, total flavonoids, total triterpenes, total polysaccharides, proanthocyanidins, chlorophyll a, chlorophyll b, and total chlorophyll, as well as the OH• and ABTS scavenging capacity. TOPSIS evaluation revealed that the highest phytochemical contents were yield on the 15th day under 1.0 h ultraviolet treatment.

Insight into Antioxidant Activity and Peptide Profile of Jinhua Ham Broth Peptides at Different Cooking Times

Ziyi Yang,Jiaming Cai,Evans Frimpong Boateng,Lujuan Xing,Wangang Zhang

This present study aimed to investigate the effects of various cooking times (1 h, 1.5 h, 2 h, 2.5 h, named as JHBP-1, JHBP-1.5, JHBP-2, JHBP-2.5) on the antioxidant activity and peptide profile of Jinhua ham broth peptides (JHBP). The peptides extracted from uncooked ham were used as an uncooked group with the name of JHBP-0. The results revealed that the antioxidant efficacy in the four cooked groups changed dramatically compared to JHBP-0. After cooking, the DPPH radical scavenging activity, hydroxyl radical scavenging activity and superoxide anion radical scavenging activity decreased, except for the Fe2+chelation and ABTS+scavenging capacity which increased significantly. However, the cooked groups still showed a strong antioxidant capacity. In particular, the superoxide anion radical scavenging ability and the Fe2+chelation action were significantly stronger compared to glutathione (GSH) and butylated hydroxytoluene (BHT) (p< 0.05). JHBP-1.5 also displayed stronger antioxidant capacity than the other three cooked groups, and its secondary structure and mass distribution changed significantly after cooking, specifically with an increased proportion of helix and <1 kDa peptides. Moreover, the constitution of free amino acids (FAAs) and the types of peptides released in the broth increased significantly with a longer cooking time. In total, 1306 (JHBP-0), 1352 (JHBP-1), 1431 (JHBP-1.5), 1500 (JHBP-2), and 1556 (JHBP-2.5) peptide sequences were detected using LC-MC/MC. The proportion of <1 kDa peptides also gradually increased as the cooking time extended, which is consistent with the molecular weight distribution measurements. Keywords:Jinhua ham;broth;bioactive peptides;antioxidant activity;peptide profile

UPLC/Q-TOF-MS profiling of phenolics from Canarium pimela leaves and its vasorelaxant and antioxidant activities

Juan Wu,Xiao’ai Fang,Yan Yuan,Yanfen Dong,Yanling Liang,Qingchun Xie,Junfeng Ban,Yanzhong Chen,Zhufen Lv

Canarium pimela K.D. Koenig, Burseraceae, have a long history of use in the Chinese traditional medicine treatment of various ailments including hypertension, and our research team has reported the anti-hypertensive activity and delineated the mechanism involved in the action. The following research aims to evaluate the vasorelaxant and antioxidant activities of ethanol extract from C. pimela leaves and to analyze its chemical composition by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF-MS) that may correlate with their pharmacological activities. The results showed that pre-incubation of aortic rings with the extract (0.3, 1, 3, 10, 30 and 100   mg/l) significantly inhibited the contractile response of the rings to norepinephrine-induced contraction ( p   <   0.01or p   <   0.05). Crude ethanol extract and refined ethanol extract showed a highest inhibitory effect against 2,2dipheyl-2-picrylhydrazyl hydrate scavenging activity (IC 50 of crude ethanol extract   =   15.42   ±   0.14   μg/ml and IC 50 of refined ethanol extract   =   5.72   ±   0.31   μg/ml) and 2,2′-azinobis (3-ethyl-benzothiazoline-6-sulphonic acid ammonium salt) (ABTS (IC 50 of crude ethanol extract   =   3.24   ±   0.18   μg/ml and IC 50 of refined ethanol extract   =   1.88   ±   0.07   μg/ml) scavenging activity, which was considerably higher than that reported for butylated hydroxytoluene and lower of that measured for ascorbic acid. Moreover, its chemical composition was analyzed by UPLC/Q-TOF-MS. Sixteen compounds including nine flavonoids, four tannins, two phenolic acids and one dianthrone were identified for the first time as constituents of this species. And of this, six major phenolic components were simultaneous quantitative analysis by HPLC-UV, chlorogenic acid is the major compounds in C. pimela leaves. These results indicate that the phenolic-rich extract of C. pimela leaves is a promising natural pharmaceutical for combating hypertension and oxidative stress.

Ultrasound-assisted extraction of polyphenols from Phyllanthi Fructus: Comprehensive insights from extraction optimization and antioxidant activity

Haixia Che,Rui Zhang,Xianyao Wang,Hong Yu,Xuepeng Shi,Jie Yi,Jiawen Li,Qi Qi,Ruizhi Dong,Qian Li

Phyllanthi Fructus (PF) is a valuable botanical resource with a long history of traditional use, known for potent antioxidant and anti-inflammatory effects attributed to its rich contents of bioactive compounds, particularly polyphenols. However, current extraction techniques limit the utilization of polyphenols from PF. This study aimed to achieve the maximum polyphenol yield and improve the antioxidant activity of PF extracts to promise PF’s prospects for modern healthcare. Firstly, ultrasonic-assisted extraction (UAE) was employed to extract the polyphenols in PF and a combination of Plackett-Burman designs (PBD) and response surface methodology (RSM) was applied to optimize UAE’s conditions. Next, cellular superoxide dismutase (SOD) and malondialdehyde (MDA) were used to assess the antioxidant activity of extracted polyphenols. Ultra-Performance Liquid Chromatography coupled with Quadrupole Time-of-Flight Mass Spectrometry (UPLC-Q-TOF MS) was utilized to characterize polyphenol components in the PF extracts. Finally, network pharmacology and molecular docking analysis were performed to screen the potential target proteins of polyphenols from PF. As a result, the optimized polyphenol yield was 213.49 mg/g, and the antioxidant activities, measured by ability of DPPH scavenging, ABTS+ scavenging, and FRAP were 76.95 %, 2.24 mmol/g, 2.34 mmol/g, respectively. PF extracts also showed good antioxidant capabilities at cellular level. 26 polyphenol components were identified in the PF extracts. Among these, ellagic acid, myricetin, and eriodictyol may exert antioxidant effects by interacting with AKT serine/threonine kinase 1 (AKT1). In conclusion, our study provides valuable insights into the optimizing PF extraction and underscores its potential applications in enhancing natural polyphenols extraction using UAE with a combination of PBD and RSM. These findings offer a promising avenue for the development and utilization of PF, and could serve as a reference for similar extraction processes in the future.
