Barnyardgrass [Echinochloa crus-galli (L.) P. Beauv.] is a dominant weed species occurring in rice fields across China. Metamifop, a common herbicide, is frequently applied to control E. crus-galli and other grassy weeds in rice fields. Herein, HS01, an E. crus-galli population suspected to be resistant (R) to metamifop, was collected from Hanshan County in Anhui Province. Whole-plant dose-response testing revealed that, compared with the susceptible (S) population FD03, HS01 had developed high-level resistance to metamifop with a resistance index (RI) of 11.76 and showed cross-resistance to cyhalofop-butyl (RI=9.33), fenoxaprop-P-ethyl (RI=5.80) and clethodim (RI=3.24). Gene sequencing revealed a Cys-2088-Arg mutation in ACCase 1,5 allele of all the R plants, while ACCase gene overexpression was not involved in the resistance. Molecular docking indicated that the less negative binding energies might be the main reason for the resistance of HS01 to ACCase-inhibiting herbicides. A derived cleaved amplified polymorphic sequence (dCAPS) method was developed for the rapid identification of the Cys-to-Arg mutation in the ACCase gene at codon position 2088 in E. crus-galli. Additionally, pretreatment with the cytochrome P450 inhibitor piperonyl butoxide or the glutathione S-transferase inhibitor 4-chloro-7-nitrobenzoxa-diazole had no significant effects (p> 0.05) on the resistance of HS01 to metamifop. To our knowledge, this is the first report of a Cys-2088-Arg mutation in E. crus-galli ACCase that confers cross-resistance to ACCase-inhibiting herbicides.

Acetyl-CoA carboxylase (ACCase)-inhibiting herbicides are among the most commonly used herbicides to control grassy weeds, especially Leptochloa chinensis , in rice fields across China. Herein, we collected a suspected resistant (R) population of L. chinensis (HFLJ16) from Lujiang county in Anhui Province. Whole plant dose response tests showed that, compared with the susceptible (S) population, the R population showed high resistance to cyhalofop-butyl (22-fold) and displayed cross-resistance to metamifop (9.7-fold), fenoxaprop- P -ethyl (18.7-fold), quizalofop- P -ethyl (7.6-fold), clodinafop-propargyl (12-fold) and clethodim (8.4-fold). We detected an amino acid substitution (Cys-2088-Arg) in the ACCase of resistant L. chinensis . However, ACCase gene expression levels were not significantly different ( P > 0.05) between R plants and S plants, without or with cyhalofop-butyl treatment. Furthermore, pretreatment with piperonyl butoxide (PBO, a cytochrome P450 monooxygenase (CYP450) inhibitor) or 4-chloro-7-nitrobenzoxadiazole (NBD-Cl, a glutathione-S-transferase (GST) inhibitor), inhibited the resistance of the R population to cyhalofop-butyl significantly (by approximately 60% and 26%, respectively). Liquid chromatography tandem mass spectrometry analysis showed that R plants metabolized cyhalofop-butyl and cyhalofop acid (its metabolite) significantly faster than S plants. Three CYP450 genes, one GST gene, and two ABC transporter genes were induced by cyhalofop-butyl and were overexpressed in the R population. Overall, GST-associated detoxification, CYP450 enhancement, and target-site gene mutation are responsible for the resistance of L. chinensis to cyhalofop-butyl.

Chinese sprangletop ( Leptochloa chinensis (L.) Nees) is among the most economically damaging and disruptive weeds found in rice fields across China, and it exhibits a high degree of cyhalofop-butyl resistance for the past few years. Here, a suspected cyhalofop-butyl-resistant population HFCH5 (R) was collected from a rice field and analyzed to explore the mechanistic basis for its resistance activity. Compared with the susceptible HFLY1 (S) population, this R population exhibited differing levels of resistance to four ACCase-inhibiting herbicides (cyhalofop-butyl (RI = 11.52), fenoxaprop- P -ethyl (RI = 11.05), metamifop (RI = 6.61), and clethodim (RI = 4.06)), although it did exhibit susceptibility to florpyrauxifen-benzyl, tripyrasulfone, pyraclonil, and anilofos. This cyhalofop-butyl resistance in R plants was not effectively mitigated by pre-treatment with the cytochrome P450 inhibitor piperonyl butoxide or the glutathione S-transferase inhibitor 4-chloro-7-nitrobenzoxadiazole. Further analyses of these R plants revealed the substitution of Trp-1999 (TGG) for a Gly residue (GGG) within the ACCase carboxyltransferase domain. When the wild-type and mutated forms of this ACCase protein were subjected to structural modeling while complexed with a range of herbicides with ACCase-inhibiting activity, this Trp-1999-Gly substitution was found to be associated with a slight decrease in the affinity of this protein for a series of ACCase-inhibiting herbicides. A dCAPS marker was developed to aid in the rapid detection of this Trp (TGG)-1999-Gly (GGG) mutation within L. chinensis . Notably, before or after four ACCase-inhibiting herbicides (cyhalofop-butyl, fenoxaprop- P -ethyl, metamifop and clethodim) treatment, ACCase gene expression level of the R plants was not significantly higher (fold change <2, P > 0.05) than that of the S plants. This study is the first report that in grass species, the newly identified ACCase Trp-1999-Gly mutation is associated with ACCase-inhibiting herbicide resistance.

Wild oat ( Avena fatua L.) is a common and problematic weed in wheat fields in China. In recent years, farmers found it increasingly difficult to control A. fatua using acetyl-CoA carboxylase (ACCase)-inhibiting herbicides. The purpose of this study was to identify the molecular basis of clodinafop-propargyl resistance in A. fatua . In comparison to the S1496 population, whole dose response studies revealed that the R1623 and R1625 populations were 71.71- and 67.76-fold resistant to clodinafop-propargyl, respectively. The two resistant A. fatua populations displayed high resistance to fenoxaprop-p-ethyl (APP) and low resistance to clethodim (CHD) and pinoxaden (PPZ), but they were still sensitive to the ALS inhibitors mesosulfuron-methyl and pyroxsulam. An Ile-2041-Asn mutation was identified in both resistant individual plants. The copy number and relative expression of the ACCase gene in the resistant population were not significantly different from those in the S1496 population. Under the application of 2160 g ai ha −1 of clodinafop-propargyl, the fresh weight of the R1623 population was reduced to 74.9%; however, pretreatment with the application of the cytochrome P450 inhibitor malathion and the GST inhibitor NBD-Cl reduced the fresh weight to 50.91% and 47.16%, respectively, which proved the presence of metabolic resistance. This is the first report of an Ile-2041-Asn mutation and probable metabolic resistance in A. fatua , resulting in resistance to clodinafop-propargyl.

Fenoxaprop- P -ethyl is widely used to control the Italian ryegrass ( Lolium multiflorum Lam.) in China. Recently, Italian ryegrass populations have been reported with resistance to fenoxaprop- P -ethyl in two provinces (Jiangsu and Henan) of China. It was important to clarify the resistance patterns of these populations which had evolved resistance to fenoxaprop- P -ethyl. In this study, we investigated three Italian ryegrass populations (HZGX-2, HZYC-4, and HZYC-5) that were involved with high resistance to fenoxaprop- P -ethyl. The HZGX-2 population showed low cross-resistance to clethodim, haloxyfop- R -methyl, clodinafop-propargyl, and quizalofop- P -ethyl, but was still susceptible to sethoxydim and pinoxaden. The HZYC-4 and HZYC-5 populations showed cross resistance to all tested acetyl-coenzyme A carboxylase (ACCase) inhibiting herbicides. We examined the genetic changes in three Italian ryegrass populations and identified four mutations, named D2078G, C2088R, I1781L, and I2041N and occurring within different frequencies in these populations. These mutations have been reported to underlie resistance to ACCase-inhibiting herbicides. In the HZGX-2 population wild type frequency was 73%, and the mutation frequencies at positions 1781, 2041, and 2078 were 13%, 7%, and 7%, respectively. In the HZYC-4 population, the mutation frequencies were 93% at 1781 and 7% at 2041. In the HZYC-5 population, the mutation frequencies were 72% at 1781, 16% at 2041, and 12% at 2088. There was no wild type found in the HZYC-4 and HZYC-5 populations, either. The cytochrome P450 inhibitors was shown to slightly reduce the ED 50 values of fenoxaprop-P-ethyl-resistant populations, which implied a combination of target-site resistance and non-target-site resistance was involved in resistance. This study can help the weed management of agricultural activities in China and other countries.

BACKGROUND Chinese sprangletop ( Leptochloa chinensis (L.) Nees), a dominant weed species in Chinese rice fields, poses a significant threat to rice yield and quality because of its increasing resistance to herbicides. In particular, resistance to cyhalofop-butyl (Cyb) has become a major concern in weed control. The mechanism by which the Asp-2078-Gly mutation confers resistance to acetyl-CoA carboxylase (ACCase) inhibitors in L . chinensis remains unclear. In this study, we collected Cyb-resistant L . chinensis populations from rice-growing regions and characterized their resistance mechanisms. RESULTS Through molecular analysis, we identified a resistant population harboring the Asp-2078-Gly mutation. Whole-plant dose–response experiments showed that the resistant (R) population exhibited varying levels of resistance compared with the susceptible (S) population: resistance indices values were 15.42 for FOP herbicides Cyb, 12.77 for metamifop, 11.50 for fenoxaprop-P-ethyl; 13.13 for the DIM herbicide clethodim; and 8.61 for the DEN herbicide pinoxaden. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis revealed no significant difference in ACCase gene expression between the Asp-2078-Gly mutant and the susceptible (S) population, indicating that resistance to Cyb was not due to target enzyme overexpression. Furthermore, the resistance of the mutant was not alleviated by P450 or glutathione S -transferase inhibitors, suggesting non-metabolic mechanisms. CONCLUSION Structural analysis confirmed that the Asp-2078-Gly mutation induces significant conformational changes in the herbicide-binding pocket, which reduces herbicide sensitivity and confers resistance. This study elucidates the resistance mechanism conferred by the Asp-2078-Gly mutation in L . chinensis , providing valuable insights for managing herbicide-resistant weeds. © 2026 Society of Chemical Industry.

Digitaria ciliaris var. chrysoblephara is a xerophytic weed severely invading rice fields along with the application of rice mechanical direct seeding technology in China. This study identified one resistant population (M5) with an Ile-1781-Leu substitution in ACCase1 showing broad-spectrum resistance to three chemical classes of ACCase-inhibiting herbicides, including metamifop, cyhalofop-butyl, fenoxaprop-p-ethyl, haloxyfop-p-methyl, clethodim, sethoxydim, and pinoxaden. The other two populations, M2 and M4, without any resistance-responsible mutations, only exhibited resistance to aryloxyphenoxypropionate (APP) herbicides cyhalofop-butyl and fenoxaprop-p-ethyl. Pre-treatment with the cytochrome P450 monooxygenase (P450) inhibitor PBO significantly reduced the cyhalofop-butyl resistance by 43% in the M2 population. Pre-emergence weed control with soil-applied herbicides, such as pretilachlor, pendimethalin, and oxadiazon, can effectively inhibit the germination and growth of D. ciliaris var. chrysoblephara. The present study reported a xerophytic weed species invading rice fields featuring broad-spectrum resistance to ACCase-inhibiting herbicides as a result of Ile-1781-Leu mutation of ACCase. Both target- and P450-involved non-target-site mechanisms may be contributing to resistance in D. ciliaris var. chrysoblephara species.

BACKGROUND Shortawn foxtail ( Alopecurus aequalis Sobol.) is a competitive grass weed infesting winter wheat- and canola-growing fields in China. In May 2016, a suspected A. aequalis resistant population AHTC-06 that survived fenoxaprop- P -ethyl and mesosulfuron-methyl applied at their field-recommended rates was collected from a wheat field in Jinji County, Anhui Province, China. This study aimed to determine the resistance profile of this AHTC-06 population to ACCase- and ALS-inhibitors, and to investigate its mechanisms of resistance to fenoxaprop- P -ethyl and mesosulfuron-methyl. RESULTS Two mutations, a common Ile-2041-Asn ( ACCase gene) and a very rare Pro-197-Tyr ( ALS1 gene), were both identified in resistant individual plants. The homozygous subpopulation AHTC-06F1 for the two mutations was generated, and it showed broad-spectrum resistance to APPs, DENs, and ALS-inhibiting herbicides of all five chemical families tested, with resistance index (RI) values that ranged from 2.2 to 36.5. In vitro ALS activity assays showed the ALS from the resistant population was insensitive to all the tested ALS inhibitors, with RI values ranging from 3.10 to 22.51. Pre-treatment with piperonyl butoxide (PBO) and malathion significantly ( P < 0.05) reversed the weed's resistance to fenoxaprop- P -ethyl and mesosulfuron-methyl, respectively. Two P450 genes, c21190_g1 and c43350_g3 , were constitutively overexpressed and mesosulfuron-methyl-induced upregulated in resistant plants, for which c43350_g3 was also fenoxaprop- P -ethyl-induced upregulated. CONCLUSION This study confirms the first case of a grass weed featuring broad-spectrum resistance to ALS-inhibiting herbicides due to a Pro-197-Tyr mutation in the ALS gene. Fenoxaprop- P -ethyl and mesosulfuron-methyl resistances in AHTC-06 plants were conferred by target site mutations and P450s-based metabolism. © 2018 Society of Chemical Industry

ABSTRACT American slough grass ( Beckmannia syzigachne (Steud.( Fernald) is a worldwide weed and is widely distributed in rice-wheat rotations in China. Fenoxaprop- P -ethyl and mesosulfuron-methyl are two major herbicides used to control B. syzigachne . Resistance has evolved in B. syzigachne under continuous selective pressure from herbicides. This study aimed to establish the cross-resistance pattern of a resistant population and explore the potential non-target-site based resistance mechanisms of B. syzigachne . Sequencing of target enzyme genes (acetyl coenzyme A carboxylase (ACCase( and acetolactate synthase (ALS() revealed that there were no resistance-endowing amino acid substitutions in the resistant B. syzigachne population (R F1 ) compared with the sensitive population (S F1 ), and obtained the purified materials. Furthermore, piperonyl butoxide (PBO) and malathion showed synergistic effects with fenoxaprop- P -ethyl and mesosulfuron-methyl respectively in R F1 . Therefore, we speculate that the resistance observed in B. syzigachne was related with metabolic, mostly involving the cytochrome P450 enzymes. Cross resistance patterns showed that the purified resistant B. syzigachne produce high resistance to fenoxaprop- P -ethyl and pyroxsulam; intermediate resistance to flucarbazone-sodium; low resistance to quizalofop- P -ethyl, clodinafop-propargyl, sethoxydim and mesosulfuron-methyl; sensitive to clethodim pinoxaden and isoproturon. Key words: Herbicide resistance; metabolic resistance; P450 inhibitors; target enzymes sequencing.

Asia minor bluegrass (Polypogon fugax), a widespread Poaceae weed, exhibits broad tolerance to abiotic stresses. Validated reference genes (RGs) for reliable RT-qPCR normalization in this ecologically and agriculturally significant species remain unidentified. This study identified eight candidate RGs using transcriptome data from seedling tissues. We assessed the expression stability of these eight RGs across various abiotic stresses and developmental stages using Delta Ct, BestKeeper, geNorm, and NormFinder algorithms. A comprehensive stability ranking was generated using RefFinder, with validation performed using the target genesCOR413andP5CS. Results identifiedEIF4AandTUBas the optimal RG combination for normalizing gene expression during heat stress, cold stress, and growth stages.EIF4AandACTwere most stable under drought stress,EIF4Aand28Sunder salt stress, andEIF4AandEF-1 under cadmium (Cd) stress. Furthermore,EIF4AandUBQdemonstrated optimal stability under herbicide stress. Additionally, application of validated RGs revealed higher acetyl-CoA carboxylase gene (ACCase) expression in one herbicide-resistant population, suggesting target-site gene overexpression contributes to resistance. This work presents the first systematic evaluation of RGs inP. fugax. The identified stable RGs provide essential tools for future gene expression studies on growth and abiotic stress responses in this species, facilitating deeper insights into the molecular basis of its weediness and adaptability.

Echinochloa crus-galli var. mitis (ECM) is one of the major Echinochloa species in the rice field, with increasing reports of herbicide resistance. This study identified an ECM population (ECM_2312) exhibiting cross-resistance to four acetyl-CoA carboxylase (ACCase)-inhibiting herbicides, including cyhalofop-butyl (8.7-fold), metamifop (15.8-fold), clodinafop-propargyl (16.1-fold) and fenoxaprop-P-ethyl (25.0-fold),compared with the susceptible population ECM_2201. A Cys-2088-Arg mutation was detected in ACCase2C gene copy in ECM_2312, confirming the involvement of that target-site resistance. Liquid chromatography–tandem mass spectrometry (HPLC-MS) analysis revealed that the degradation of the herbicides active forms (cyhalofop-acid and metamifop) were faster in the resistant population than in the susceptible population. Pretreatment with the glutathione S-transferase (GST) inhibitor, 4-chloro-7-nitrobenzoxadiazole (NBD-Cl) reduced the resistance level from 8.7-fold to 4.9 fold for cyhalofop-butyl and from 15.8-fold to 3.0-fold for metamifop. Moreover, GST activity was consistently higher in the resistant population than the susceptible population, both in the absence and presence of herbicide treatment. Furthermore, five GST genes exhibited sustained overexpression for 96 h in the resistant plants following exposure to either cyhalofop-butyl or metamifop. This study demonstrated an ECM population possesses both a target-site mutation (Cys-2088-Arg) and GST-mediated enhanced metabolic resistance to ACCase-inhibiting herbicides.
