Investigating roasting conditions to explore AGEs formation in chicken breast: A dynamic model approach

Yingshuang Lu,Yu Jiang,Mengyu Sun,Lu Dong,Xiaolong Xing,Yan Zhang,Shuo Wang

This study explores the formation of advanced glycation end products (AGEs) in chicken breast during roasting, focusing on the analytical characterization and simulation of dynamic models for the content of carboxymethyllysine (CML) and carboxyethyllysine (CEL), two key AGEs. To systematically investigate the formation patterns of AGEs, the chicken breasts were subjected to five roasting temperatures ranging from 180 °C to 230 °C. At each temperature, samples were roasted to achieve five distinct core temperatures, from 60 °C to 100 °C. Dynamic models for CML and CEL content were constructed using multiple linear regression and partial least squares methods, with the best-fit models identified based on their R 2 and RMSE values, with R 2 values of 0.886 for CEL, 0.954 for CML, and RMSE values indicating minimal prediction error. Our research provides a robust framework for predicting AGEs formation during roasting with validation, offering valuable insights for optimizing cooking processes to balance product quality and safety.

Effects of baking factors and recipes on the quality of butter cookies and the formation of advanced glycation end products (AGEs) and 5-hydroxymethylfurfural (HMF)

Huiyu Hu,Yuting Wang,Mingyue Shen,Yousheng Huang,Chang Li,Shaoping Nie,Mingyong Xie

Maillard reaction during food processing contributes to the formation of some unpleasant heat-induced toxicants including advanced glycation end products (AGEs) and 5-hydroxymethylfurfural (HMF), which have been linked to various health risks. The effects of baking factors and recipes, such as baking temperature (130°C–180 °C) and time (8 min–15 min), sucrose levels (0 g–20 g), butter levels (0 g–20 g) and egg liquid levels (0 g–12 g) on the formation of free N ε -(carboxymethyl)lysine (CML), free N ε -(carboxyethyl)lysine (CEL), protein-bound CML, protein-bound CEL, HMF, glyoxal (GO), methylglyoxal (MGO), 3-deoxyglucosone (3-DG) and on the sensory qualities were investigated in butter cookies. The results suggested that the levels of AGEs initially increased and then followed by decrease as baking temperature and time increased, HMF is very sensitive to baking temperature and time and grows sharply. The changes of protein-bound AGEs are lagging behind that of free AGEs. The proportions of sucrose, butter and egg liquid in butter cookies were positively correlated with AGEs, with sucrose greatly promoting on the formation of HMF and 3-DG. In addition, the high level of sucrose and butter in cookies is preferred by panelists, especially in terms of appearance, taste and smell.

Mulberry anthocyanins exert anti-AGEs effects by selectively trapping glyoxal and structural-dependently blocking the lysyl residues of β-lactoglobulins

Ibrahim Khalifa,Du Xia,Kunal Dutta,Jinmeng Peng,Yangyang Jia,Chunmei Li

Advanced glycation end-products (AGEs), which instigate many disorders, are mostly mediated by dicarbonyl rearrangements. We studied the corresponding mechanisms of the anti-glycation effects of two anthocyanins purified from mulberry fruits, namely cyanidin 3-glucoside (C3G) and cyanidin 3-rutinoside (C3R), on glycated β-lactoglobulins (β-Lg). Both mulberry anthocyanins (MAs) inhibited the AGEs-formation in a dose-dependent manner, but the effect of C3R was significantly stronger than that of C3G (p < 0.05). MAs inhibited AGEs-formation by selectively trapping dicarbonyls, especially glyoxal. The UPLC-ESI-Q-TOF-MS results characterized that C3R formed mono- and di-glyoxal adducts, where C3G only created di-glyoxal adducts. Additionally, C3R could directly interact with some of the glycation sites of β-Lg. Overall, GO-trapping and β-Lg-MAs covalent/noncovalent binding are disclosed as the key mechanisms of the anti-AGEs activity of MAs on β-Lg, which could be valorised as effectual AGEs inhibitors in proteins-rich matrices.

Characterization of Enzymatic Modified Soluble Dietary Fiber from Rhodomyrtus tomentosa fruits: A Potential Ingredient in Reducing AGEs Accumulation

Deng Yejun,Liu Yong,Zhang Caihong,Xie Pujun,Huang Lixin

The accumulation of advanced glycation end-products (AGEs) from food matrix in human body can induce health crisis. This study firstly investigated the AGEs accumulation reducing potential of soluble dietary fiber (SDF) from Rhodomyrtus tomentosa fruits in antiglycation and trapping AGEs aspects. To obtain superior functionalities, the SDF was prepared by an enzymatic modified extraction method (ESDF). The result showed the extraction yield of SDF increased by 45% with the Viscozyme L modification. The structural characterizations indicated the enzymatic treatment greatly influenced the monosaccharide composition, thermal stability, crystallinity, and apparent viscosity of Rhodomyrtus tomentosa fruits SDF (RTSDF) as compared with SDF obtained from water extraction method (WSDF). The antioxidant assay indicated that RTSDF could scavenge O 2−∙ ,OH ∙ , and H 2 O 2 radicals and chelate Fe 2+ . Additionally, RTSDF, especially ESDF, reduced the more fluorescent AGEs generation in bovine serum albumin (BSA)-fructose model, and protected the BSA from oxidation and cross-linking. Besides, the RTSDF presented the ability to trap those already formed BSA-bound AGEs. ESDF exhibited higher AGEs binding capacity, while WSDF demonstrated stronger ability in restraining the AGEs diffusion. These results suggested the potential utility of RTSDF serving as an essential ingredient in functional foods to reduce AGEs accumulation in human body, controlling AGEs-associated diseases.

Protein-bound AGEs derived from methylglyoxal induce pro-inflammatory response and barrier integrity damage in epithelial cells by disrupting the retinol metabolism

Gang Yu,Jianxin He,Zhongshan Gao,Linglin Fu,Qiaozhi Zhang

Advanced glycation end-products (AGEs) are complex and heterogeneous compounds widely present in processed foods. Previous studies evidenced the adverse effects of AGEs on gut homeostasis, but the precise pathological mechanisms and molecular pathways responsible for the disruption of intestinal barrier integrity by AGEs remain incompletely elucidated. In this study, protein-bound AGEs (BSA-MGO), the most common type of dietary AGEs, were prepared by methylglyoxal-mediated glycation, and an in vitro human epithelial colorectal adenocarcinoma (Caco-2) cell model was employed to evaluate the impact of protein-bound AGEs on gut epithelial function. Results showed that exposure to BSA-MGO significantly increased the permeability of Caco-2 cell monolayers as evidenced by the decreased transepithelial electrical resistance value, increased paracellular transport of FITC-dextran, and down-regulated tight-junction proteins. Parallelly, BSA-MGO induced pro-inflammatory responses and oxidative stress in the monolayers. Transcriptomic profiling further revealed that BSA-MGO disrupted the retinol metabolism, thereby contributing to the barrier integrity damage in epithelial cells. Overall, these results provide valuable insights into the disrupting effects of dietary AGEs on intestinal barrier function, and the perturbed pathways present potential targets for further exploration of the molecular mechanisms underlying the detrimental effect of processed foods on gut health.

Construction and properties of an amyloid fiber ferulic acid chitosan double network hydrogel and its inhibition of AGEs activity

Lili Zheng,Minghan She,Binling Ai,Yang Yang,Xiaoyan Zheng,Shenwan Wang,Dao Xiao,Zhiguo Jiang,Zhanwu Sheng

An amyloid fiber-ferulic acid/chitosan-ferulic acid (AF-FA/CS-FA) double network hydrogel (DN) was synthesized in two steps after prefabricating micronetworks. Ferulic acid (FA) and laccase were used to form protein and polysaccharide micronetworks, respectively. Then, AF-FA and CS-FA were induced to form covalent cross-links using genipin (GP). The phase penetration behavior of the two biopolymers in different proportions could form a homogeneous microstructure. The higher structural complexity and mechanical integrity brought by this structure may be the main reason that it improved the controlled release ability of the gels. The results from a loading capacity experiment and infrared spectra showed that the DN had acquired the AF-FA network to provide a higher load rate (9.50%), possibly because adding the polysaccharide network had increased the proportion of β-sheet structures beneficial to loading FA. The DN hydrogel containing 0.7 wt% CS-FA (DN70) showed the most tightly ordered combination of protein and polysaccharide, which had the highest storage modulus (2552.82 Pa) and the lowest solubility (91.00%). The thermal and photodegradation constants of DN70 were reduced by 75.00% and 58.29%, respectively, compared to the FA monomer . The formation of advanced glycation end products (AGEs) was reduced by 44.75% in the cake with the addition of 0.5% (w/w) DN70. These results suggest that the AF-FA/CS-FA DN can improve the range of FA applications in food products and provide an attractive strategy for polyphenols as AGEs inhibitors in baked goods.

Huang-Lian-Jie-Du Decoction Alleviates Diabetic Encephalopathy by Regulating Inflammation and Pyroptosis via Suppression of AGEs/RAGE/NF-κB Pathways

Guo-Dong Zhuang,Si-Min Deng,Meng-Di Chen,Chao-Fan Deng,Wen-Ting Gu,Shu-Mei Wang,Dan Tang

Ethnopharmacological relevance Cognitive dysfunction associated with diabetes, known as diabetic encephalopathy (DE), is a grave neurodegenerative condition triggered by diabetes, and persistent inflammation plays a vital role in its development. The renowned traditional Chinese medicine Huang-Lian-Jie-Du Decoction (HLJDD) is clinically proven to manage diabetes mellitus and Alzheimer's disease and is famous for its heat-clearing and detoxifying effects. However, the underlying mechanisms through which HLJDD affects DE remain to be elucidated. Aim of the study To explore the beneficial effects of HLJDD on improving cognitive dysfunction in DE mice. Study design and methods A diabetic mouse was established through a high-fat diet and subsequent administration of streptozotocin over five consecutive days. After the animals were confirmed to have diabetes, they were treated with HLJDD. After oral administration of HLJDD or metformin for 14 weeks, behavioral tests were used to assess their cognitive capacity. Biochemical analyses were then performed to detect levels of glucose metabolism, followed by histological analyses to assess pathological damage. Furthermore, AGEs/RAGE/NF-κB axis related proteins were detected by Western blot or immunofluorescence techniques. An advanced UPLC-Q-Orbitrap HRMS/MS analytical technique utilizing a chemical derivatization strategy was employed for comprehensive metabolic profiling of carbonyl compounds in the plasma of DE mice. Results Pharmacological assessment revealed that HLJDD effectively mitigated cognitive dysfunction, normalized glucose metabolic imbalances, and repaired neuronal damage in DE mice. It reduced neuroinflammation by attenuating carbonyl stress, deactivating astrocytes and microglia, and preserving dopaminergic neurons. Additionally, metabolomics analysis revealed 18 carbonyl compounds with marked disparities between DE and control mice, with 12 metabolites approaching normal levels post-HLJDD intervention. Further investigations showed that HLJDD regulated inflammation and pyroptosis through suppressing AGEs/RAGE/NF-κB pathways. Conclusion Our study indicated that HLJDD could ameliorate carbonyl stress via the regulation of carbonyl compound metabolism profiling, and inhibiting the AGEs/RAGE/NF-κB pathway, thereby alleviating inflammation and pyroptosis to exert beneficial effects on DE.

Cinnamomum cassia Presl flavonoids prevent hyperglycemia-induced cognitive impairment via inhibiting of AGEs accumulation and oxidative stress

Hemei Li,Jinling Zhou,Sitong Liu,Xiuting Chen,Tiantian Qin,Guodong Huang,Peizhen Luo,Ying Hu,Xing Xia

Cinnamomum cassia Presl (CCP) has a beneficial impact on diabetes and dementia. In this research, the underlying mechanism of flavonoids of Cinnamomum cassia Presl (CFS) on diabetes-related cognitive impairment (DRCI) were inferred by a network pharmacology approach. The conjectures of network study were verified through the advanced glycation end products (AGEs) systems in vitro and the long-term diabetic rats. The experimental results showed CFS inhibited AGEs generated from nonenzymatic glycosylation reaction in vitro as well as in the brain of long-term diabetic rats in a dose-dependently manner. In morris water maze study, CFS restored the cognitive ability of diabetic rats. Moreover, the hippocampal morphology damage and oxidative stress were ameliorated by CFS, while the hippocampal level of AGEs, RAGE and pJNK/JNK were decreased significantly. These results indicated CFS may suppress AGEs accumulation via inhibiting glycosylation, and alleviating oxidative stress caused by AGEs, thereby prevents the neuronal injury and ameliorates DRCI.

Inhibiting advanced glycation end products (AGEs) of high-performance hemodialysis membranes self-assembled by gallic acid and Ti3C2Tx MXene

Qiqi Wang,Changjun Mu,Ya'nan Yu,Xia Feng,Yang He,Li Chen

Current patients with chronic kidney disease (CKD) suffer from either accumulation of advanced glycation end products (AGEs) or inefficient clearance of the middle molecule uremic toxins, threatening their long-term survival rate. In particular, conventional hemodialysis membranes cannot prevent AGEs formation because of continuous oxidative stress from reactive oxygen species (ROS). Herein, a persistent ROS scavenging and efficient uremic toxin clearance of hemodialysis membrane is proposed by the in situ deposition of a gallic acid-Ti 3 C 2 T x MXene (GA-MXene) n multilayers through a self-assembly method, where n denotes the number of deposition layers. These hemodialysis membranes have a highly interconnecting composite laminate with tunable pore size. In the dialysis experiment, the (GA-MXene) 2 membrane with the vertical nano-slits shows the highest middle molecule uremic toxin clearance of 80.0% compared with state-of-the-art hemodialysis membranes, which has been approximately equal to the clearance of small molecule urea. Furthermore, GA conferring efficient ROS scavenging rate over dialysis treatment time allows anti-oxidative activities to inhibit AGEs’ formation. (GA-MXene) n multilayers enhanced the serum total antioxidant capacity and suppressed lipid peroxidation from oxidative damage. Ultimately, the (GA-MXene) n hemodialysis membranes provide excellent anticoagulant properties and cytocompatibility through the heparin-mimicking sulfonated Ti 3 C 2 T x MXene and biological activity of GA. This work facilitates the blood purification application of MXenes and opens up an avenue for developing functional multilayers for high-performance hemodialysis membranes and beyond.

Study on the inhibitory effects and mechanisms of four polyphenolic compounds from perilla extract on advanced glycation end products (AGEs)

Xue Han,Xue Sun,Zihang Shi,Xiankang Fan,Yangyang Hu,Chen Chen,Qiang Xia,Yangying Sun,Daodong Pan

This study investigates the inhibitory effects and underlying mechanisms of major polyphenolic compounds in perilla extract on the formation of advanced glycation end products using a high-temperature in vitro glycation model. Ultra-performance liquid chromatography coupled with mass spectrometry identified quercetin, luteolin, rosmarinic acid, sinapic acid, and other representative bioactive constituents in PE. These polyphenolic compounds significantly inhibited protein-bound Nε-(carboxymethyl)lysine (41.3 % - 61.1 %), mainly through free radical scavenging, metal ion chelation, and suppression of protein aggregation, thereby mitigating protein oxidation and modulating protein conformation. Further mechanistic analysis revealed that these polyphenols regulated protein surface hydrophobicity, altered protein particle size, and influenced intermolecular interactions, thereby impacting the glycation process. Overall, this study elucidates the molecular mechanisms by which polyphenolic compounds inhibit AGEs formation under high-temperature conditions and provides a theoretical basis for the development of effective AGEs inhibition strategies in processed meat products.

Apigenin and baicalein ameliorate thoracic aortic structural deterioration and cognitive deficit via inhibiting AGEs/RAGE/NF-κB pathway in D-galactose-induced aging rats

Yufang Ma,Zhenming Ma,Yiyuan Zhang,Chunyun Luo,Puxin Huang,Jing Tong,Hong Ding,Honghui Liu

Apigenin and baicalein are structurally related flavonoids that have been reported to have multiple pharmacological activities. The aim of this study was to investigate the protective effects and potential mechanisms of apigenin and baicalein in D-galactose-induced aging rats. First, apigenin and baicalein showed remarkable antioxidant activity and anti-glycation activity in vitro. Secondly, the protective effects of apigenin and baicalein on aging rats were investigated. We found that apigenin and baicalein supplementation significantly ameliorated aging-related changes such as declines in the spatial learning and memory and histopathological damage of the hippocampus and thoracic aorta. In addition, our data showed that apigenin and baicalein alleviated oxidative stress as illustrated by decreasing MDA level, increasing SOD activity and GSH level. Further data showed that they significantly reduced the accumulation of advanced glycation end products (AGEs), inhibited the expression of RAGE, down-regulated phosphorylated nuclear factor (p–NF–κB (p65)). Our results suggested that the protective effects of apigenin and baicalein on aging rats were at least partially related to the inhibition of AGEs/RAGE/NF-κB pathway and the improvement of oxidative damage. Overall, apigenin and baicalein showed almost equal anti-aging efficacy. Our results provided an experimental basis for the application of apigenin and baicalein to delay the aging process.

Dietary myricetin inhibits AGEs formation during thermal processing in different pattern diets and alleviates AGEs-induced intestinal epithelial damage in Caco-2 cells

Luyan Liu,Kaiyue Sun,Xin Xie,Yanghui Chen,Jiahui Wang,Yaxin He,Junjie Yu,Meng Huang,Yan Lin,Juan Chen,Jian Liu,Xian Zhang

Thermal processing promotes advanced glycation end products (AGEs) generation in food. Dietary flavonoids possess anti-oxidative and anti-glycation activities. This study aimed to investigate the effect and underlying mechanism of different pattern diets and flavonoids on AGEs production and AGEs-induced intestinal damage. AGEs production was assessed in chow diet (CD), high-fat diet (HFD), high-fat/high-cholesterol western diet (WD) and flavonoid-supplemented diet during thermal processing and simulated digestion. AGEs-induced intestinal damage and the protective effect of myricetin were evaluated in Caco-2 cells. During thermal processing and simulated digestion, HFD and WD generated more AGEs, N ε -carboxymethyl-lysine and N ε -carboxyethyl-lysine than CD. Flavonoids inhibited AGEs production, with myricetin exhibiting the highest activity. Dietary AGEs impaired intestinal epithelial nutrient transport and barrier function, and promoted intracellular ROS generation through inhibiting Nrf2/Catalase pathway, while myricetin reversed this effect. Altogether, dietary myricetin prevents thermal processing-induced AGEs formation in different pattern diets and mitigates the associated intestinal epithelial damage.
