The emerging understanding highlights indole as a disruptive factor to quorum sensing (QS) mechanisms, prompting further investigation into its role in anaerobic digestion (AD) system inhibition. However, relevant studies are still scarce and the potential mechanism linking indole and AD inhibition remains unclear. This study showed that indole (1, 2, and 3 mM) significantly reduced cumulative methane production by 8.47–51.89% and extended the lag phase by 1.34–6.68 days. Time-series AHLs quantification, metagenomics, and circular clustering heatmaps analysis revealed that indole might disrupt microbial communication between hydrolysis-acidification bacteria and acetoclastic methanogens by reducing the AHLs level (C6-HSL, 3-oxo-C8-HSL, C10-HSL, C12-HSL, 3-oxo-C10-HSL, and C18-HSL). Notably, indole degradation alleviated the inhibition of C10-HSL, C18-HSL, and 3-oxo-C10-HSL, which might restore hydrolysis and acidification and mitigate AD inhibition. Exogenous AHLs (1 and 5 µM) restored methane production by 48.44–55.59% in 3 mM indole-inhibited reactors ( p < 0.05), while the quorum quenching agent vanillin further reduced methane production by 72.55%, suggesting that AHLs play an important role in helping microorganisms resist indole stress. These findings highlight the importance of AHLs-mediated inter-microbial communication in counteracting indole-inhibited methanogenesis inhibition, suggesting potential practical strategies to enhance AD stability and efficiency in challenging conditions.

Bisphenol A (BPA) poses multiple risks to human health, and it has become a critical environmental pollution issue. The advantage of bacterial degradation of BPA has become more prominent, and enhancement methods have become increasingly diverse. Using quorum sensing to regulate functional bacteria and genes to enhance the bacterial degradation efficiency of BPA serves as a highly promising new approach. This study investigated the effect of adding exogenous N-hexanoyl-L-homoserine lactone (C6-HSL) on the degradation of BPA by Pseudomonas asiatica P1 (strain P1) and analyzed the regulatory mechanism of the signaling molecules. The results showed that exogenous homoserine lactone signaling molecules (AHLs) could enhance the BPA degradation efficiency of strain P1, with 10 μM C6-HSL demonstrating the most significant enhancement, increasing the degradation rate by 12.4 % compared with the control group and producing products such as benzoic acid and succinic acid. In terms of the regulatory mechanism, C6-HSL preferentially induced a 5.18-fold upregulation in the expression of the AHL receptor-encoding gene LasR. Subsequently, LasR regulates the upregulation of hub genes such as bvgS and aruS , ultimately achieving an upregulation of BPA degradation genes through hierarchical regulation. This led to an enhancement in the initial degradation step of BPA and the activation of the benzoic acid degradation pathway. This study analyzed the potential of C6-HSL to enhance microbial BPA degradation and, for the first time, clarified the mechanism by which C6-HSL enhanced bacterial BPA degradation efficiency through hierarchical regulation.

Prolonged idle conditions pose a major challenge to aerobic granular sludge (AGS) systems by compromising granule integrity and pollutant removal performance. This study investigates the in situ reactivation of AGS after three months of static storage using two quorum sensing molecules, N-hexanoyl-L-homoserine lactone (C6-HSL) and N-octanoyl-homoserine lactone (C8-HSL). All reactors rapidly restored COD and NH 4 + -N removal efficiencies to >92% and >98%, respectively. C6-HSL significantly accelerated phosphorus recovery, reaching removal efficiencies above 90% by day 26, compared to day 34 in the control and C8-HSL groups. C8-HSL enhanced EPS secretion and granule growth, yielding the largest granule size (1210 μm), which was 1.26-fold and 1.71-fold larger than those in the control and C6-HSL groups, respectively. Metagenomic analysis revealed comparable microbial structures at the phylum level, but distinct functional responses. C6-HSL increased the abundances of phosphorus metabolism genes ( ppk, ppx, ppa ), while C8-HSL notably upregulated genes related to the biosynthesis of tyrosine, tryptophan, and structural polysaccharides (e.g., alginate and Psl), supporting enhanced EPS production and granule stability. These results demonstrate molecule-specific regulatory roles of individual N-acyl-homoserine lactones during AGS reactivation, linking functional recovery and structural regeneration to distinct quorum sensing pathways. This study provides mechanistic and engineering insights into an energy-efficient strategy for restoring AGS performance after prolonged ambient idle conditions, with direct relevance to the stable operation and management of full-scale wastewater treatment systems.

As the secondary metabolites of cyanobacterial harmful algal blooms (Cyano-HABs), microcystins (MCs) were generated under various environmental and cellular conditions. The understanding of the causes of MCs generation is of great interest in the field of water treatment and environmental science. In this work, we studied how Microcystis aeruginosa (FACHB-905) cell densities affect the MCs synthetase genes ( mcy ) expression, microcystin-LR (MC-LR) and quorum sensing molecules (Acyl-homoserine lactones (AHLs)) production. An electrochemical sensor was developed here for sensitive and quantitative detection of MC-LR that cultured at different cell densities. The results showed that mcy expression and MC-LR concentration started to increase when the cell density reached ca. 22 × 10 6 cells/mL, and was significantly increased with increasing cell densities. Moreover, the up-regulation of AHLs with increasing cell densities revealed that MC-LR is quorum sensing-mediated. Our results undoubtedly confirmed that MC-LR was produced in a cell density-dependent way that mimics quorum sensing, and the minimum cell density ( ca. 22 × 10 6 cells/mL) that was required to produce MC-LR was provided and offered a reference standard for the prevention and control of MCs pollution in the actual water environment.

Proteobacteria primarily utilize acyl-homoserine lactones (AHLs) as quorum-sensing signals for intra-/interspecies communication to control pathogen infections. Enzymatic degradation of AHL represents the major quorum-quenching mechanism that has been developed as a promising approach to prevent bacterial infections. Here we identified a novel quorum-quenching mechanism revealed by an effector of the type IVA secretion system (T4ASS) in bacterial interspecies competition. We found that the soil antifungal bacterium Lysobacter enzymogenes OH11 (OH11) could use T4ASS to deliver the effector protein Le1288 into the cytoplasm of another soil microbiome bacterium Pseudomonas fluorescens 2P24 (2P24). Le1288 did not degrade AHL, whereas its delivery to strain 2P24 significantly impaired AHL production through binding to the AHL synthase PcoI. Therefore, we defined Le1288 as LqqE1 ( L ysobacter q uorum- q uenching e ffector 1 ). Formation of the LqqE1-PcoI complex enabled LqqE1 to block the ability of PcoI to recognize/bind S -adenosy-L-methionine, a substrate required for AHL synthesis. This LqqE1-triggered interspecies quorum-quenching in bacteria seemed to be of key ecological significance, as it conferred strain OH11 a better competitive advantage in killing strain 2P24 via cell-to-cell contact. This novel quorum-quenching also appeared to be adopted by other T4ASS-production bacteria. Our findings suggest a novel quorum-quenching that occurred naturally in bacterial interspecies interactions within the soil microbiome by effector translocation. Finally, we presented two case studies showing the application potential of LqqE1 to block AHL signaling in the human pathogen Pseudomonas aeruginosa and the plant pathogen Ralstonia solanacearum .

A magnetic fluorescence probe was fabricated by coating carbon quantum dots-doped molecularly imprinted polymers (MIPs) layers on the surface of Fe 3 O 4 particles (MFMP) for detection of N -acyl homoserine lactones (AHLs) signaling molecules. N-Z- L -homoserine lactone molecular was used as the template to prepare AHLs MIP layers, employing MAA and HEMA as functional monomers. The developed MFMP owned superparamagnetism, fluorescence, fast response and class-selectivity. If AHLs (C 4 -HSL, C 6 -HSL, C 8 -HSL, C 10 -HSL, C 12 -HSL and C 14 -HSL) were captured by the MFMP, they quenched the fluorescence of the probe. Fluorescence dropped linearly in the concentration ranges of 3.65 × 10 −3 μmol/L–0.96 × 10 −1 μmol/L for AHLs. The MFMP was applied to the analysis of fish juice and milk samples, and recoveries ranged from 83.10% to 90.74% with relative standard deviation less than 5.1%. This study offered a novel strategy to fabricated AHLs fluorescence probe with great potential for wide-ranging application in agri-food products.

Serratia marcescens NJ01 and Erwinia carotovora , two pathogenic bacteria involved in vegetable spoilage, can persist in leafy vegetables and subsequently form biofilms. The aim of this study was to evaluate the effect of propyl gallate (PG) on the susceptibility of antibiotics against the biofilms of S. marcescens NJ01 and E. carotovora and uncover its anti-virulence mechanism. PG notably inhibited the production of AHLs and enhanced the susceptibility of antibiotics against the biofilm formation of S. marcescens NJ01 and E. carotovora and significantly improved the disrupted effect on the preformed biofilms of S. marcescens NJ01. PG exposure decreased the minimum inhibitory concentrations of these five antibiotics and reduced the production of virulence factors. Further investigation indicated that PG exposure resulted in the downregulation of genes involved in virulence, biofilm formation, and antioxidant enzymes of S. marcescens NJ01. The suppression of antioxidant enzymes caused the enhancement of oxidative stress, increased membrane permeability, and ultimately promoted the susceptibility of bacterial/biofilm cells to antibiotics. The tomato and lettuce infection assay indicated that PG could notably attenuate the pathogenicity of S. marcescens NJ01 and E. carotovora . These findings suggest that PG has the potential to function as an antibiotic accelerant to defend against spoilage pathogens.

Large quantities of low-concentration uranium-contaminated water from the nuclear industry poses a critical environmental challenge due to the limitations of current methods on removal efficiency and adsorption selectivity. This study developed a novel biofilm cathode for enhanced uranium adsorption by leveraging the quorum sensing (QS) system of Shewanella oneidensis MR-1. Biofilm formation was induced using exogenous acyl-homoserine lactones (AHLs), with C4-HSL (10 μmol/L) identified as the most efficient signal molecule. It significantly improved biofilm properties, increasing thickness by 107.7 % versus the control without AHLs, boosting extracellular protein content, and increasing the proportion of living cells. Microstructure analysis (CLSM) revealed a “base-polysaccharide-gel” three-dimensional structure, where proteins, lipids, and β-polysaccharides form a functional gel layer that provides the primary functional matrix for uranium binding. The C4-HSL-induced biofilm exhibited a 43.4 % higher reduction peak current and a 33.3 % decrease in electron transfer resistance, confirming improved electron transfer efficiency. Furthermore, coupling CLSM, SEM-EDS, FT-IR and XPS analysis indicated uranium capture was primarily dominated by complexation/coordination with functional groups on extracellular polymeric substances (EPS), supplemented by electrochemical reduction of 25.91 % soluble U(VI) to insoluble U(IV). Accordingly, a possible mechanism model of QS-driven biofilm cathode is proposed for optimizing biofilm structure and enhancing uranium capture. Finally, verification tests in actual uranium-contaminated groundwater demonstrated the biofilm cathode exhibited exceptional performance with a 99.4 % uranium recovery rate and high selectivity (K d,U at 34.57 L/g). These findings highlight the significant potential of AHLs-triggered QS as a powerful strategy to optimize bioelectrochemical properties for highly-efficient uranium remediation and resource recovery.

The widespread emergence of antibiotic resistance genes (ARGs) in drinking water systems endangers human health, and may be exacerbated by their horizontal gene transfer (HGT) among microbiota. In our previous study, Quorum sensing (QS) molecules produced by bacteria from biological activated carbon (BAC) biofilms were demonstrated to influence the transfer efficiency of a model conjugative plasmid, here RP4. In this study, we further explored the effect and mechanism of QS on conjugation transfer. The results revealed that Acyl-homoserine lactones producing (AHL-producing) bacteria isolated from BAC biofilm play a role in the propagation of ARGs. We selected several quorum sensing inhibitors (QSIs) to study their effects on AHL-producing bacteria, including the formation of biofilm and the regulating effect on conjugation transfer. In addition, the possible molecular mechanisms for AHLs that promote conjugative transfer were attributable to enhancing the mRNA expression, which involved altered expressions of conjugation-related genes. We also found that QSIs could inhibit conjugative transfer by downregulating the conjugation-relevant genes. We believe that this is the first insightful exploration of the mechanism by which AHLs will facilitate and QSIs will inhibit the conjugative transfer of ARGs. These results provide creative insight into ARG pollution control that involves blocking QS during BAC treatment in drinking water systems.
