Portable, rapid, accurate and on-site detection of analysis target has always been the goal of continuous efforts in the field of analysis. In this work, a novel intelligent and rapid detection system based on an AIE dye (TPA-DCV) was established for portable and on-site detection of trace water in organic solvents and CN − in water by integrating the smartphone and linear discriminant analysis (LDA). The TPA-DCV displayed remarkable solvatochromism effect and showed different fluorescence colors to different water contents of different solvents, with the minimum detection limit of 54 ppm. In addition, TPA-DCV also exhibited specific response to CN − , and showed significant fluorescence changes to different amounts of CN − . With the assist of this detection system, these different fluorescence color signals could be accurately classified in canonical score plot. What's more, the classification and distribution of fluorescence color in canonical score plot showed regular change with the target concentration, which could be used to quantify the trace water and CN − . The rationality and practicability of this detection system were preliminary verified by a large number of real sample analyses. This detection system greatly simplifies the analysis process, which is in the proof of intelligent and rapid detection just using a simple color analysis without any instrument test, which may be a big step forward.

Molecular organic dyes are classic fluorescent nanoprobes finding tremendous uses in biological and life sciences. Yet, they suffer from low brightness, poor photostability, and lack of functional groups for bioconjugation. Here, we describe a class of biocompatible dye–protein optical nanoprobes, which show long-time photostability, superbrightness, and enriched functional groups. These nanoprobes utilize apoferritin (an intracellular protein for iron stores and release) to encase appropriate molecular organic dyes to produce on-demand fluorescence in aqueous solution. A pH-driven dissociation–reconstitution process of apoferritin subunits allows substantial incorporation of hydrophilic (aggregation caused quenching, ACQ) or hydrophobic (aggregation induced enhancement, AIE) dye molecules into the protein nanocavity (8 nm), producing monodispersed dye–apoferritin nanoparticles (apo-dye-NPs, ∼12 nm). As compared with single dye monomer, single apo-dye-NPs possess hundreds of times larger molar extinction coefficient and 2 orders of magnitude higher absolute luminescence quantum yield (up to 45-fold), multiplying fluorescence brightness up to 2778-fold. We show that varying the type of incorporated dyes entails a precise control over nanoprobe emission profile tunable in a broad spectral range of 370–1300 nm. Mechanical investigations indicate that the diversified microstructures of nanocavity inner surface are able to conform ACQ dyes at reasonable space interval while providing protein-guided-stacking for AIE dyes, thus enhancing fluorescence quantum yield through confining intermolecular quenching and intramolecular rotation. Moreover, apo-dye-NPs are able to emit stable fluorescence (over 13 min) without quenching in confocal imaging of HepG2 cancer cell under ultrahigh laser irradiance (1.3 × 106 W/cm2). These superb properties make them suitable, as demonstrated in this work, for long-term super-resolved structured illumination microscopic cell imaging (spatial resolution, 117 nm) over 48 h, near-infrared (NIR) fluorescence angiography imaging of whole-body blood vessels (spatial resolution, 380 μm), and NIR photoacoustic imaging of liver in vivo.

Preparation and biomedical applications of aggregation induced emission (AIE) dye based fluorescent nanoprobes (FNPs) have been extensively investigated in recent years. And great efforts have been devoted toward fabrication of AIE dye based FNPs through both noncovalent and covalent strategies. However, to the best of our knowledge, the preparation of AIE dye based FNPs via dynamic bonds is rarely reported. In this work, ultrabright FNPs based on AIE dyes have been fabricated using aldehyde-terminated polyethylene glycol (CHO-PEG-CHO) with molecular weight of 2000 Da and 2,2′-diaminotetraphenyl ethylene (TPE-NH 2 ) through formation of Schiff base . The resulting TPE contained copolymers are prone to self-assemble into stabilized core shell nanoparticles (TPE@PEG FNPs). The successful formation of TPE@PEG FNPs was confirmed by 1 H nuclear magnetic resonance spectroscopy, transmission electron microscopy , fluorescence spectroscopy and Fourier transform infrared spectroscopy . Due to their amphipihlic properties, TPE@PEG FNPs displayed high water dispersibility and strong fluorescence intensity . Furthermore, biological evaluation results demonstrated that TPE@PEG FNPs possess excellent biocompatibility and can be facilely internalized by cells. The extraordinary fluorescent properties, excellent water dispersibility as well as the desirable biocompatibility make TPE@PEG FNPs promising candidates for biomedical applications. On the other hand, because of the pH stimuli responsiveness of Schiff base, TPE@PEG FNPs are expected to exhibit some advantages for biomedical applications as compared with the FNPs fabricated by noncovalent and covalent strategies.
