A photoelectrochemical (PEC) assay strategy for the detection of matrix metalloproteinase-9 (MMP-9) was fabricated. RuSe 2 /CuO nanohybrids were first prepared by hydrothermal method. As a transition metal element semiconductor, RuSe 2 has excellent optical and electrical properties. By combining the excellent conductivity and adjustable bandgap of CuO, a binary heterojunction nanoflower with high photoelectric response was obtained. The combination of RuSe 2 and CuO reduces the electron hole recombination rate; the RuSe 2 /CuO nanohybrids exhibits a higher photocurrent response. In the MMP-9 PEC strategy determination, the magnetic beads (MB)-peptide-alkaline phosphatase (ALP) recognition probe was formed by modifying C-terminal of the peptide chain with biotin and binding to ALP, and the N-terminal was connected to carboxylated MB. When the target MMP-9 exists, the recognition probe, MB-Peptide-ALP, was recognized by MMP-9 to cut the substrate area and coding area and release ALP, catalyzing the hydrolysis of O-phosphonoxyphenol (OPP) to produce benzene-1,2-diol in situ, the photocurrent response was generated. This strategy provides lower background PEC noise for detecting MMP-9, with the advantage of simplicity and the sensitivity than conventional ELISA. The results showed that the recognition peptide probe PEC method achieved ultra-high sensitivity detection of target MMP-9, with a linear detection range of 1 pg/mL–100 ng/mL and a detection limit of 0.3 pg/mL (S/N = 3). The results of PEC assay strategy in human serum sample give the recoveries rate of 94.3 % to 108.1 % and relative standard deviation of 0.45–5.87 %.

Clustered regularly interspaced short palindromic repeats (CRISPR)-based diagnostics have been considered as the next generation molecular diagnosis tool. Single-readout mode has been extensively employed in massive CRISPR/Cas12a-based biosensors. In this work, we propose a one-tube dual-readout biosensor for ultrasensitive nucleic acids and non-nucleic acids detection by harnessing CRISPR-ALP tandem assay for the first time (termed CRISAT). In the presence of target, the Cas12a is activated to randomly cut the single-stranded hyDNA sequence of MB@hyDNA-cALP, thus releasing abundant alkaline phosphatase (ALP) in the supernatant solution. By using 4-aminophenol phosphate as the substrate of ALP, p-aminophenol is produced and then reacts with N-[3-(trimethoxysilyl)propyl]ethylenediamine or diethylenetriamine to generate silicon-containing polymer carbon dots (Si PCDs) or polymer carbon dots (PCDs) in situ, which can be observed by naked eye or detected by a fluorescent device in the same solution. Using this strategy, a fluoroscence and colorimetry dual-readout nanoplatform for CRISPR-based biosensor can be rationally developed. We ascertain the applicability of CRISAT through detecting SARS-CoV-2 pseudovirus, achieving superior sensitivity and specificity. With simple modification of crRNAs, the CRISAT platform can be also employed to detect monkeypox virus (MPXV) and non-nucleic acids of adenosine triphosphate (ATP). This work provides a great potential for nucleic acids and non-nucleic acids detection.

Poor cellular permeability greatly hampers the utilization of anionic Ir(III) complexes, though efficiently emissive and remarkably stable, in cell-based diagnosis. To overcome this barrier, we present the development of an alkaline phosphatase (ALP)-responsive, anionic, and aggregation-induced emission (AIE)-active Ir(III) complex (Ir1) for specific recognition of osteosarcoma cells. Containing phosphate moieties, Ir1 exhibits a net −1 charge, enabling charge repulsion from the cell membrane and resulting in low cellular uptake and good biocompatibility in normal osteoblast cells. Upon ALP-mediated hydrolysis of phosphate groups, the resulting dephosphorylated product, Ir2, demonstrates a positive charge and increased lipophilicity, promoting cellular uptake and activating its AIE properties for specific recognition of osteosarcoma cells that express elevated levels of ALP. This study elucidates the role of ALP as an ideal trigger for enhancing the cellular permeability of phosphate ester-containing Ir(III) complexes, thus expanding the potential of anionic Ir(III) complexes for biomedical applications.

Ovarian cancer (OC) remains a prevalent gynecological malignancy, posing significant threats to women's health globally. Emerging evidence underscores the critical involvement of alkaline phosphatase (ALP) in OC pathogenesis, prompting the development of targeted diagnostic tools. Herein, we report the successful application of a novel fluorescent probe, HD-ALP, for fluorescent imaging of ALP in ovarian cancer cells and preclinical models. After being triggered by ALP, HD-ALP responded in the fluorescent signal reporting manner. More importantly, the fluorescence signal showed a reliable linear correlation with ALP levels in the range of 0-10000 U/L. The HD-ALP probe demonstrated high sensitivity, selectivity, and stability under the necessary biological conditions. The intracellular imaging results indicated that HD-ALP enabled the visualization of ALP levels within ovarian cancer cells. The in vivo experiments demonstrated that HD-ALP could accurately differentiate between healthy and cancerous models (subcutaneous transplantation tumor model of ovarian cancer, orthotopic ovarian cancer model, primary ovarian cancer model). Notably, In cellular and in vivo imaging, HD-ALP could effectively reflect the decrease in fluorescence signal after the addition of ALP inhibitors. By integrating ALP detection, which utilizes the HD-ALP fluorescence mode, with traditional methods like immunohistochemistry staining and serum biomarker detection, potential strategies for more accurate diagnosis of OC may be developed for preclinical trials in the future. According to the research, this is also one of the few developments of fluorescent detection tools for measuring ALP levels in ovarian cancer models.

Alkaline phosphatase (ALP) is a reliable biomarker for various diseases, making the development of new detection methods highly significant. Herein, we report a simple and accurate dual-signal optical detection strategy for measuring ALP activity across the entire clinical range in adults. Based on this, we designed a smartphone app to eliminate the limitations of large instruments, enabling real-time field detection. In this work, ALP catalyzes the conversion of the phosphate monoester of 2-phospho-L-ascorbic acid (PAA) into ascorbic acid (AA), a compound with strong reducing properties. AA facilitates the in-situ reduction of Ag + to form Au@Ag core-shell nanoparticles, accompanied by significant changes in the ultraviolet-visible absorption spectra and color. These changes allow for the qualitative and quantitative detection of ALP activity. In UV absorption spectroscopy and colorimetric detection, the signal shows good linearity with ALP activity concentrations, with detection ranges of 0 U/L to 200 U/L (R 2 =0.9961) and 25 U/L to 250 U/L (R 2 =0.9771), respectively. Subsequent selective analysis and serum sample analysis demonstrated the high selectivity and accuracy of the proposed method, highlighting its potential for field application.

Sensitive and accurate detection of interleukin 6 (IL-6) is crucial for the early diagnosis of cerebral infarction to improve patient survival rates. However, the low-abundance of IL-6 in cerebral infarction presents a significant challenge in developing effective diagnosis method. Herein, we studied and analyzed the strong fluorescence property of 4-aminophenol phosphate (APP) and developed an enzyme-linked immunosorbent assay (ELISA) for IL-6 detection. The detection was based on the integration of optical signal change induced by alkaline phosphatase (ALP)-catalyzed APP hydrolysis and ALP-mediated ELISA. The generated colorimetric signal of 4-aminophenol, APP hydrolysis product, was used for ELISA of IL-6 with a detection limit of 0.1 ng/mL, and the visual detection of IL-6 was achieved. The changes in APP fluorescence have a good linear relationship with the logarithm of IL-6 concentration in the range of 0.005 ng/mL to 5.0 ng/mL, with a detection limit of 0.001 ng/mL, which was 100 times lower than that of conventional pNPP-based ELISA. Furthermore, the constructed ELISA effectively distinguished between samples from patients with cerebral infarction and volunteers with non-cerebral infarction, and the severity of symptoms was well distinguished based on IL-6 measurement. The dual-mode ELISA demonstrated high feasibility of low-abundance biomarker detection and displayed good potential for accurate in vitro diagnosis.

A simple homogeneous photoelectrochemical (PEC) sensing platform based on an alkaline phosphatase (ALP)–mediated pesticide assay was established for the sensitive detection of omethoate (OM). The Bi 2 S 3 @Bi 2 Sn 2 O 7 heterojunction was used as a photoactive material to provide stable background photocurrent signals. The inhibition of OM on ALP and PEC determination was carried out in the homogeneous system. In the absence of OM, dephosphorylation of L-ascorbic acid 2-phosphate trisodium salt (AAP) was catalyzed by ALP to produce the enzyme-catalyzed product (L-ascorbic acid, AA). AA, as an electron donor, could capture photogenerated holes on the Bi 2 S 3 @Bi 2 Sn 2 O 7 heterojunction, thus inhibiting the recombination of electron holes to achieve an increase of the photocurrent signal. When the OM was introduced, the enzyme activity of ALP was reduced due to the organophosphorus pesticides (OPs)–based enzyme inhibition, and the AA produced by catalytic hydrolysis was also reduced, thus reducing the photocurrent signal. Compared with the traditional PEC sensor for OPs, this homogeneous PEC sensor avoided immobilization procedures, covalent labeling, separation, and the steric hindrance effect caused by immobilized biomolecules, which achieved high recognition efficiency and caused a reduction in analysis time. Additionally, an ALP-mediated pesticide assay for the determination of OPs with a simplified experimental process further improved the stability and reproducibility of the PEC sensor. The PEC sensor showed high sensitivity to the target OM within a dynamic range of 0.05 ~ 500 ng mL −1 , and the detection limit was 0.0146 ng mL −1 . Additionally, the PEC biosensing system showed good selectivity and anti-interference ability, and exhibited a satisfactory result in spinach and mustard samples. Graphical abstract A homogeneous PEC biosensor based on ALP inhibition strategy was constructed for OM detection in vegetable samples via Bi 2 S 3 @Bi 2 Sn 2 O 7 heterojunction as the photoactive substrate material

Cholestatic hepatitis is a progressive inflammatory liver disease characterized by disrupted bile acid homeostasis, excessive reactive oxygen species (ROS) generation, and chronic inflammation, remains clinically challenging due to limited diagnostic precision and lack of effective, targeted therapies. Here, we developed a multifunctional theranostic nanoplatform, CyP-CuGA-UDCA nanosheets (NSs), that integrates therapeutic intervention with enzyme-responsive disease monitoring. The platform features copper-gallic acid (CuGA) nanozymes with superoxide dismutase-, peroxidase-, and catalase-like activities for effective ROS scavenging and NLRP3 inflammasome inhibition. Co-loaded with ursodeoxycholic acid (UDCA) to alleviate bile acid toxicity, and with an alkaline phosphatase (ALP)-responsive near-infrared fluorescent probe (CyP), the nanoplatform enables targeted treatment and real-time imaging of cholestatic lesions. In a DDC-induced mice model, CyP-CuGA-UDCA NSs significantly suppressed pro-inflammatory cytokine production, reduced hepatic macrophage infiltration, attenuated oxidative stress and hepatocyte apoptosis, and alleviated fibrosis, outperforming monotherapies. Concurrently, the ALP-activated fluorescence allowed precise visualization of cholestatic progression in vivo. This study presents a first-in-class nanostructured system that couples NLRP3 inflammasome modulation and bile acid regulation with enzyme-specific diagnostics, offering a robust strategy for precision therapy and monitoring of cholestatic hepatitis.

Quantitative detection of alkaline phosphatase (ALP) activity is crucial in clinical diagnosis and bioanalysis. Herein, we have developed a highly sensitive electrochemiluminescence (ECL) biosensor that employs a biomimetic zirconia interface as its core sensing platform. The interface was constructed by immobilizing o-phosphorylethanolamine (PEA) onto zirconium oxide nanofilms (ZrO2NFs), forming a surface rich in Zr-O-P bonds. This design mimics phosphate recognition and enzyme-triggered dephosphorylation processes, where ALP catalyzes the hydrolysis of these bonds, triggering a direct switch in the ECL signal from Ru(bpy)32+-loaded gold nanocage (Ru-AuNCs) emitters. This sensor achieves a wide linear range of 0.100–100 U/L and a low detection limit down to 0.0899 U/L. Its practical utility was validated through the accurate detection of ALP in fetal bovine serum samples, confirming high recovery and reliability. This strategy highlights the potential of biomimetic zirconia interfaces in developing robust biosensors for early disease diagnosis.

Hierarchical porous bioglass films on the tantalum were designed to enhance osteointegration of metallic implants. The films were prepared by a sol-gel method using P123 as the mesopore template and polystyrene microsphere as the nanopore template. The films with 5.4 nm mesopores and 100 nm nanopores (MBG-100) elicited an obviously elongated morphology of the cultured MC3T3-E1 cells, as a result, a higher alkaline phosphatase level was expressed. It is suggested that the nanopores play an important role in regulating cellular behavior by initial protein adsorption through nanopore curvatures. The mesopores were proven very effective for loading rhBMP-2, and the rhBMP-2 loaded on MBG-100 films showed a better function of enhancing osteogenic differentiation, which is attributed to that the nanopore structure could expedite rhBMP-2 release and provide a microenvironment for intensifying the interaction of rhBMP-2 with the cells. Hence, the cell osteogenic differentiation can be enhanced by hierarchical porous bioglass films through both the porous structure and rhBMP-2 induction.

The detection of enzyme activity can provide valuable insights into clinical diagnosis. Herein, we synthesize gold nanobipyramids@MnO2 nanoparticles (AMNS) as the surface-enhanced Raman spectroscopy (SERS) substrate for the first time and design a “turn-on” SERS strategy for the detection of enzyme activity without the need for a complicated SERS nanotag preparation process. In the presence of alkaline phosphatase (ALP), 2-phosphate-L-ascorbic acid trisodium salt (AAP) can be hydrolyzed to ascorbic acid (AA), which can etch the shell of AMNS by reducing MnO2 to Mn2+. The cracked MnO2 shell-caused electromagnetic field enhancement from AMNS can give rise to a significant increase in the Raman intensity of the adsorbed molecules (i.e., crystal violet, CV) on the surfaces of nanobipyramids. Thus, the ALP activity can be accurately quantified based on the MnO2 shell thickness dependent Raman signal output from CV. A linear dynamic range from 0.4 to 20 mU mL−1 with a detection limit of 0.04 mU mL−1 is achieved, which is more sensitive than other spectroscopic methods for ALP detection. Because of its advantages of sensitivity, convenience and versatility, this approach provides a new perspective to disease-related biomolecular detection in the future.

Alkaline phosphatase (ALP) is a crucial biomarker for clinical diagnosis, which is closely related to the physiological homeostasis regulation process of human body. And the abnormal level of ALP is associated with numerous diseases, such as liver dysfunction, bone diseases, diabetes, and so on. In order to meet the demand of personalized healthcare, it is particularly important to develop a miniaturized point-of-care testing (POCT) device for ALP detection. Herein, a portable solid-phase colorimetric sensor based on enzyme-induced metallization signal amplification strategy was constructed for ALP detection. The AuNPs modified on the glass slides acted as crystal seeds, allowing Ag + in the solution to be reduced and deposited on the surface of AuNPs, which further formed the gold core and silver shell (Au@Ag) complex and generated visual signals. The visual signals were recorded by a smartphone and quantified using open-source ImageJ software. Under the optimal conditions, the proposed method exhibited a good linear relationship from 2.0 to 16.0 pM, and the detection limit was as low as 0.9 pM. In addition, it was further successfully applied for ALP detection in non-transparent and complex samples (milk, different types of cells). A sensitive, low cost, rapid and convenient solid-phase sensor was developed for ALP detection, which was expected to provide a promising strategy for POCT devices.
