Avian leukosis virus subgroup J (ALV-J) is an immunosuppressive virus which has caused heavy losses to the poultry breeding industry. Currently, there is no effective medicine to treat this virus. In our previous experiments, the low-molecular-weightSargassum fusiformepolysaccharide (SFP) was proven to possess antiviral activity against ALV-J, but its function was limited to the virus adsorption stage. In order to improve the antiviral activity of the SFP, in this study, three new SFP long-chain alkyl group nanomicelles (SFP-C12M, SFP-C14M and SFP-C16M) were prepared. The nanomicelles were characterized according to their physical and chemical properties. The nanomicelles were characterized by particle size, zeta potential, polydispersity index, critical micelle concentration and morphology. The results showed the particle sizes of the three nanomicelles were all approximately 200 nm and SFP-C14M and SFP-C16M were more stable than SFP-C12M. The newly prepared nanomicelles exhibited a better anti-ALV-J activity than the SFP, with SFP-C16M exhibiting the best antiviral effects in both the virus adsorption stage and the replication stage. The results of the giant unilamellar vesicle exposure experiment demonstrated that the new virucidal effect of the nanomicelles might be caused by damage to the phospholipid membrane of ALV-J. This study provides a potential idea for ALV-J prevention and development of other antiviral drugs. Keywords:Sargassum fusiforme;polysaccharide;long-chain alkyl group;antiviral activity;ALV-J

A novel pH-responsive sandwich-type electrochemiluminescence immunosensor was successfully developed for the detection of avian leukosis virus subgroup J (ALV-J). The immunosensor consisted of graphene oxide functionalized with tannic acid (rGO-TA) and primary antibodies (Ab 1 ) as the main ALV-J sensing platform. For signal amplification , pH-responsive hollow MnO 2 (hMnO 2 ) nanospheres encapsulating Ru(bpy) 3 Cl 2 and carrying secondary antibodies (Ab 2 ) were employed. Under optimal conditions, the immunosensor exhibited a wide ALV-J detection range of 10 1.80 to 10 4.30 TCID 50 /mL (TCID 50 : 50% tissue culture infective dose) and a low detection limit of 10 1.71 TCID 50 /mL (S/N = 3). Importantly, the immunosensor showed high sensitivity, good reproducibility and excellent operational stability. Finally, the immunosensor was evaluated using a real avian serum sample containing ALV-J, with excellent results being achieved.

DNA hydroxymethylation is an important epigenetic modification, which plays crucial roles in many biological processes, such as gene expression and cell differentiation. Herein, a novel photoelectrochemical immunosensor was built for detecting 5-hydroxymethylcytosine (5hmC), where the perovskite Bi 4 TaO 8 Cl sensitized by 3,4,9,10-perylenetetracarboxylicacid (PTCA) was implemented as photoactive material, anti-5hmC antibody was served as target recognition reagent, ZrO 2 was used for connecting more modifications, and ferrocene was adopted as signal amplification unit. The prepared nanomaterials of Bi 4 TaO 8 Cl, PTCA and ZrO 2 were well characterized. The possible mechanism for the improved PEC response of Bi 4 TaO 8 Cl sensitized by PTCA was investigated carefully, indicated that the matched energy band facilitated the migration of photogenerated electron. In addition, the possible mechanism for signal amplification caused by ferrocene was investigated, showing that ferrocene acted as electron donor to accelerate electron transfer. Under the optimal condition, the biosensor manifested wide detection range from 0.02 to 100 nM with the low detection limit of 5.2 pM (S/N = 3). Moreover, the applicability of the biosensor was demonstrated by investigating the effect of heavy metal Cd 2+ and perfluorooctanoic acid on 5hmC content in the leaves and roots of wheat seedlings, as well as the effect of ALV-J on 5hmC content in chicken serum.
