This work investigates the efficiency removal and mechanism of xanthate from flotation tailings wastewater through a coagulation-Fenton oxidation process by introducing Fe 2+ and Mn 2+ to activate H 2 O 2 . Under the conditions of initial 60 mg/L SBX concentration, 50 mg/L H 2 O 2 concentration, 25 mg/L Fe 2+ concentration, 25 mg/L Mn 2+ concentration, 3.5 initial pH, and 60 min reaction time, the degradation rate of xanthate was more than 98 %. The H 2 O 2 + Fe 2+ +Mn 2+ system exhibits markedly superior degradation efficiency for SBX. The results demonstrate that the Fe 2+ +Mn 2+ bimetallic system in simulated AMD facilitates the activation of H 2 O 2 to generate a greater abundance of hydroxyl radicals (•OH). Through characterization of degradation products and analysis of intermediates, two potential degradation pathways of SBX are proposed. In the first pathway, SBX initially undergoes conversion to BPX, which subsequently degrades into small molecular compounds including CO 2 , H 2 O, and SO 4 2− under sustained •OH radical attack. The secondary pathway involves the formation of precipitates composed of xanthates metal (Fe/Mn), iron oxides, and manganese oxides. This work presents a new strategy for efficiently degrading xanthate contaminants in mineral flotation wastewater. It also valorizes AMD by exploiting its intrinsic properties, thereby mitigating AMD's environmental hazards at the source.

Age-related macular degeneration (AMD) is a leading cause of vision loss in the elderly, with neovascular AMD (nAMD) driven by choroidal neovascularization (CNV), persistent inflammation, and oxidative stress. Although combinatorial strategies targeting these pathological factors hold therapeutic promise, their clinical translation is constrained by the lack of effective delivery systems. In this study, we designed a mannose-functionalized poly(aspartic acid)-based nanocarrier bearing quaternary ammonium and boronic acid groups (MDA/QPABA) for the targeted delivery of microRNA-223 (miR-223). The system exploits charge-driven self-assembly to form stable nanoparticles with high loading efficiency, favorable colloidal stability, and tunable surface properties. The mannose moieties enable specific recognition by mannose receptors on target cells, facilitating cellular uptake and subsequent lysosomal escape. The resulting MDA/QPABA/miR-223 nanoparticles demonstrated pronounced anti-inflammatory, antioxidant, and anti-angiogenic activities in vitro . In a laser-induced CNV mouse model, they effectively reduced inflammatory and angiogenic cytokines, suppressed pathological neovascularization, restored retinal and choroidal structure, and preserved photoreceptor function, with an excellent biosafety. These findings highlight the potential of rationally engineered poly(aspartic acid)-based nanoarchitectures with tailored surface chemistry and biointerfacial properties for multifunctional nucleic acid delivery in nAMD therapy.

Catalysts are critical components in advanced oxidation processes (AOPs) for removing recalcitrant antibiotics from water. The preparation of metal catalysts through waste conversion has gradually received growing attention owing to their sustainability and low-cost. Herein, acid mine drainage (AMD) as wastewater offers a tantalizing resource to be harnessed due to its high concentration of metal ions. To reduce the continuous consumption of metallic resources and prevent environmental pollution by heavy metals. This study, utilizing the abundance of transition metal ions in AMD, devises a straightforward methodology for adept transformation into persulfate catalysts—metal sulfides(MS) for antibiotic removal, enabling the MS/PMS system to sustainably degrade tetracycline(TC) over a wide pH range (2.0–10.0). Notably, it recycled more than 97 % of the transition metals in AMD; the removal of TC (30 mg/L) by adsorption and advanced oxidation was as high as 92 %, and MS can achieve 99.5 % removal of TC (10 mg/L) at low concentrations. Transition metal elements such as manganese and iron in the recovered catalytic material dispersed on MS played an indelible role in activating the PMS, and the non-radical pathway dominated by singlet oxygen led to the degradation of the TC. Overall, MS prepared based on the concept of treating pollution with waste had excellent TC removal capability, providing an innovative approach for the reduction of recalcitrant antibiotics (TC) and the resourceization of AMD.

Oxidative stress in the retina and dysregulation of iron homeostasis are established as central pathogenic mechanisms in dry age-related macular degeneration (dAMD). Nonetheless, effective clinical treatments to counteract retinal degeneration in this condition are still absent. Our research began with developing a single-cell transcriptome profile of retinal tissue subjected to sodium iodate (NaIO 3 )-induced oxidative injury. This analysis identified ferroptosis in photoreceptor cells as a key driver of disease advancement. Apoferritin (AFn) is a hollow, spherical protein nanocage formed by 24 self-assembled subunits, enabling supramolecular iron storage through ferroxidase-mediated mineralization while providing dynamic iron buffering and antioxidant protection in biological systems. We then engineered a biomimetic nanoparticle, AFn-MNP, by combining Afn with melanin nanoparticles (MNP). AFn-MNP displayed significant ferrous ion-chelating ability and antioxidant activity, effectively mitigating NaIO 3 -triggered photoreceptor ferroptosis in vitro and in vivo. Significantly, a single intravitreal injection of 10 μg AFn-MNP effectively counteracted visual impairment in NaIO 3 -induced dAMD model mice, reversed damage to photoreceptor synaptic terminals, and significantly alleviated photoreceptor degeneration and retinal oxidative injury. In summary, our results indicate that AFn-MNP holds significant promise as a novel therapy for dAMD and other retinal degenerative diseases linked to oxidative damage and impaired iron metabolism.

Clinically, no effective preventive strategies currently exist to inhibit the progression of early dry age-related macular degeneration (dAMD). Moreover, current retinal therapies rely on intravitreal injections, which carry inherent procedural risks. Inspired by natural melanin and viral structures, we developed virus-biomimetic melanin nanoparticles (VMNPs) surface-modified with ergothioneine (ET), featuring virus-like spike structures and a neutral zwitterionic shell. This study aimed to evaluate whether virus-inspired engineered nanoparticles could enhance retinal delivery and improve the therapeutic potential of melanin-based nanodrugs for dAMD. The VMNPs facilitated cellular uptake through both their virus-mimetic architecture and OCTN1-mediated active transport. They also exhibited potent antioxidant activity, significantly reducing intracellular and mitochondrial reactive oxygen species (ROS) in oxidative stress models of ARPE-19 cells. Transcriptomic analyses revealed that VMNPs activated intrinsic cytoprotective pathways by upregulating HMOX1 and SLC7A11 , while downregulating inflammation-associated genes. Importantly, VMNPs promoted transcytosis and, following topical administration, demonstrated improved retention and deep penetration within the rabbit eye, effectively reaching the retinal pigment epithelium (RPE) and deeper retinal layers. Notably, twice-daily topical administration of VMNPs markedly inhibited retinal degeneration in rabbits, achieving therapeutic efficacy comparable to a single intravitreal injection of pegcetacoplan. Collectively, these findings establish a promising paradigm for the non-invasive clinical treatment of dAMD.

Acid mine drainage (AMD) is a notorious environmental issue, and its economically and environmentally sustainable management remains challenging. A major driving force of AMD formation is the biooxidation of pyrite, largely mediated by acidophilic bacteria like Leptospirillum and Acidithiobacillus . Our previous study demonstrated that lignin can effectively inhibit pyrite oxidation by Acidithiobacillus ferrooxidans, thereby preventing AMD formation at the source. Building upon this, the current study investigated the structure-activity relationship of lignin in inhibiting A. ferrooxidans activity. Our findings indicated that low-molecular-weight lignin exhibits superior antibacterial activity, primarily through the induction of reactive oxygen species both intracellularly and extracellularly. Furthermore, transcriptomic analysis revealed that lignin influences critical bacterial pathways, including membrane biogenesis, carbohydrate metabolism, and signal transduction. This influence leads to an excessive consumption of the bacteria's internal carbon sources and disrupts their internal signal transduction system, ultimately inhibiting biofilm formation. This disruption culminates in cellular damage and bacterial cell death. Overall, this study offers technical guidance for selecting and preparing lignin with high antibacterial activity to enhance AMD control. It also sheds light on the molecular inactivation mechanism of lignin inhibiting A. ferrooxidans , providing a scientific basis for preventing acidification and supporting ecological restoration in new mining areas.

Covalent organic framework (COF) membranes have garnered significant attention as a promising separation technology for dye wastewater treatment, owing to their meticulously ordered pore structure and exceptional chemical stability. The Polyamide COF (Amd-COF) membrane, characterized by amide-linkages, exhibits excellent hydrophilicity and chemical stability, whereas the polyamine COF (Amn-COF) membrane, featuring amine-linkages, demonstrates remarkable flexibility and resistance to chlorine. In this study, we present the synthesis of defect-free Amd/TA-PVA/Amn COF composite membranes with large area and continuous structure through multi-interfacial polymerization. The incorporation of an intermediate layer (TA-PVA) enables stable formation of strong hydrogen bonds between Amd-COF and Amn-COF, thereby enhancing the durability of the composite membranes. Due to the unique structural features and functional groups of the Amd/TA-PVA/Amn COF composite membranes, they exhibit not only high water permeability (119.24 L•m −2 h −1 bar −1 ) but also exceptional rejection efficiency towards Rhodamine B, achieving up to 96.55 % removal. Specifically, the membranes treated with a chlorination intensity of 2000 ppm exhibit stable permeance and dye rejection performance. Moreover, these membranes exhibit exceptional stability in both structure and performance over multiple cycles. This can primarily be attributed to the highest density of amino moieties and phenolic hydroxyl groups on their surface. This work underscores the significant potential of COF membrane incorporating amide and amine functionalities for high-performance separation and anti-chlorine applications.

This study objective is to degrade xanthate from flotation tailings wastewater by using a coagulation-flocculation co-Fenton oxidation process in an acid mine drainage (AMD)-H 2 O 2 system. More than 98% sodium butyl xanthate (SBX) removal rate was achieved by the method under optimal conditions. The acids and Fe 2+ in AMD were sufficient to initiate a Fenton reaction at the aid of H 2 O 2 . Furthermore, iron ions were reduced to an extremely low level (0.19 mg/L) by participating in an oxidation process. Meanwhile, the Cu 2+ ions in AMD facilitated the coagulation-flocculation process. Comparison experiments confirmed that the method was superior to the AMD alone (54.26%) and H 2 O 2 alone (32.23%) in terms of performance in degrading SBX. The kinetic results showed that SBX degradation followed a pseudo first-order kinetic model . Additionally, the electron paramagnetic resonance (EPR) and quenching results suggested that hydroxyl radicals (•OH) were the main active species in AMD-H 2 O 2 system. Degradation products were analyzed, and two possible pathways of SBX degradation were proposed. One pathway displayed that the SBX was first transformed into butyl xanthate peroxide (BPX), CO 3 2− and S 2 O 3 2− , and then further decomposed into CO 2 , H 2 O and SO 4 2− under the ongoing •OH attack. Another pathway showed that precipitates consisting of butyl copper xanthate and iron oxide species were generated during the SBX degradation. This study provides a novel perspective on the innovative application of AMD in Fenton oxidation and provides a strong basis for the green and sustainable treatment of xanthate wastewater in tailings.

Bevacizumab, a vascular endothelial growth factor (VEGF)-targeting drug, is widely used as an off-label therapeutic for age-related macular degeneration (AMD). To reduce the monthly administration frequency, this study investigated microspheres comprising a poly( d , l -lactide-co-glycolide)/poly(cyclohexane-1,4-diyl acetone dimethylene ketal) (PLGA/PCADK) blend that could be loaded with bevacizumab-dextran particles using solid-in-oil-in-water (S/O/W) emulsification. Control microspheres were also prepared through water-in-oil-in-water (W/O/W) emulsification. The structural stability of bevacizumab in the polymer monomers was analyzed using dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), size exclusion chromatography (SEC-HPLC), circular dichroism (CD) and fluorescence spectroscopy. Subsequently, microspheres were prepared and evaluated in terms of their morphology, encapsulation efficiency and release behavior. PLGA/PCADK microspheres prepared by the S/O/W method with <20% PCADK showed a smooth spherical structure with a uniform distribution of bevacizumab. The microspheres exhibited a release behavior comprising a slight initial burst and an increasing total release over 50 days both in vitro and in vivo . Additionally, the microspheres were well tolerated by ocular tissue. Finally, a chorioallantoic membrane (CAM) assay revealed that the bioactivity of bevacizumab was retained by PCADK. In conclusion, these results suggest the potential for bevacizumab-loaded PLGA/PCADK microspheres prepared by the S/O/W method as a means of intravitreal therapy for ocular diseases.

Lycii Fructus (LF) is a medicinal-edible homology material frequently used for treating age-related macular degeneration (AMD) in Chinese medicine. Abundant of literatures verified the retina protection effect of LF on AMD-like animal models. However, previous studies on LF attenuating retinal damage mainly focused on its protection effect on pigment epithelial cells and optic ganglion cells while few reports on Müller cells. In the current study, we found that LF extracts attenuated oxidation damages and inhibited cell apoptosis of retina on a mouse retinal injury model induced by sodium iodate. LF extracts also increased expressions of Müller cell biomarker glutamine synthetase of eyeballs of model mice, which indicated that LF extract protected Müller cell from oxidation injury induced by sodium iodate. In Müller cell MIO-M1 cultures, LF also significantly inhibited cellular apoptosis induced by NaIO 3 by regulating Nrf2/HO-1 signaling pathway and the aqueous extracts exerted better active tendency than ethanol extracts. These data implied that the antioxidant effect of LF on protecting Müller cell might contribute to its retina repair function, thereby providing more scientific evidences for the development of LF as an alternative therapy or preventive measures for AMD.

Thalidomide, a glutamate derivative with teratogenicity, possesses anti-inflammatory, immunomodulatory, and anti-angiogenic properties that enable its use in treating refractory diseases unresponsive to conventional therapies. Dry age-related macular degeneration (AMD), characterized by retinal pigment epithelium (RPE) degeneration and lacking effective therapies, represents a significant unmet medical need. Our findings demonstrated that thalidomide significantly restores mitochondrial function, alleviates G2/M phase cell cycle arrest, and suppresses sustained endoplasmic reticulum (ER) stress in oxidatively injured RPE cells. Mechanistically, these effects are coordinated through E2F2 activation, which subsequently regulates FBXO5 expression. Moreover, thalidomide was able to ameliorate oxidative stress-induced retinal structural disorders and RPE degeneration, and improve visual function in mice. In summary, this study elucidates that thalidomide synergistically regulates cell cycle progression and endoplasmic reticulum homeostasis through the E2F2-FBXO5 signaling pathway, providing a new drug candidate and therapeutic target for the prevention and treatment of dry AMD. Graphical Schematic diagram of the cytoprotective mechanism of thalidomide in RPE cells. Thalidomide activates the transcription factor E2F2, which upregulates FBXO5 expression. This E2F2-FBXO5 pathway coordinately alleviates G2/M phase cell cycle arrest, restores mitochondrial function, and suppresses endoplasmic reticulum stress, thereby protecting RPE cells from oxidative damage.

Background Melatonin is an antioxidant that also has anti-inflammatory effects. It has been reported to delay the progression of age-related macular degeneration (AMD), however, the mechanism has not been fully recognized. Purpose The aim of the present study was to investigate the effects of melatonin on sodium iodate (SI)-induced retinal degeneration and elucidate the specific mechanisms, then, provide novel targets in AMD treatment. Methods Retinal degeneration mouse model and in vitro retinal pigment epithelium (RPE) death model were established by SI treatment. Melatonin was administrated intraperitoneally at a concentration of 20, 40 or 80 mg/kg for in vivo study or treated at 48 h before SI treatment. To confirm the therapeutic effects of melatonin on mouse, the retinal structure and visual function were evaluated. The specific cell death rates were determined by CCK-8 assay, PI staining and protein level of RIPK3. The cytosolic or mitochondrial calcium levels were determined by Fluo-4AM or Rhod-2AM staining. Mitochondrial functions including mitochondrial dynamics, mitochondrial membrane potential, or mitochondrial permeability pore opening were evaluated. The proteins involved in endoplasmic reticulum (ER) stress were measured by western blot assay while the genes expression in calcium signaling pathway were measured by RT-qPCR. Results We show that melatonin protects RPE cells from necroptosis and NLRP3 inflammasome activation induced by SI. Mechanistically, melatonin suppresses ER stress and intracellular calcium overload triggered by SI through restoring the function of SERCA2. Silencing of SERCA2 or blocking of melatonin receptors inhibit the protective effects of melatonin. Melatonin reduces mitochondrial Ca 2+ levels and restores mitochondrial membrane potential. Constant mitochondrial Ca 2+ overload directly promote cell necroptosis through mitochondrial fission. Inhibition of mitochondrial fission by Mdivi-1 prevent necroptosis induced by SI without altering the level of mitochondrial Ca 2+ . Conclusions The results confirmed that melatonin protects RPE cells from SI-induced injury by regulates MT2/SERCA2/Ca 2+ axis. This study highlighted the potential of melatonin in the treatment of AMD and elucidated the mechanism and signaling pathway that mediate the protective effects.
