Arbuscular mycorrhizal fungi (AMF) can form symbioses with most plants in terrestrial environments. In this study, we aimed to investigate root-associated AMF community of white clover in response to AMF inoculation and AgNPs addition in the multi-contaminated soil. Results demonstrated that mycorrhizal colonization was significantly increased by AMF inocultion in the 50 mg/kg AgNPs treatment. Compared with non-AMF inocultion group, AMF inoculation group exhibited higher shoot dry mass. In plants without AMF, root dry mass markedly declined when AgNPs addition level exceed 2000 mg/kg. The relative abundance of Glomus was above 70% both in rhizosphere and endosphere. The endosphere Glomus abundance was higher than rhizosphere in all groups. AMF inoculation and AgNPs addition did not influence AMF diversity in rhizosphere and endosphere. Shannon, simpson, chao and ace index in rhizosphere was higher than in endosphere. AMF community structures in endosphere were significantly affected by AMF inoculation and AgNPs additions. Soil Ca, Mg, Mn, P, Al, Mo, As, NH 4 + -N and pH levels had significant correlations with AMF community in rhizosphere and endosphere. In rhizosphere and endosphere, AgNPs addition (≥100 mg/kg) resulted in more complex networks and AMF inoculation decreased network complexity. We concluded that AgNPs and AMF could influence Trifolium repen root-associated AMF community structure only in endosphere, and co-occurrence networks in rhizosphere and endosphere, but have no effect on alpha diversity.

Based on a sensitive monoclonal antibody against bisphenol A (BPA) and a new tracer named BVA–AMF, a homogeneous fluorescence polarization immunoassay (FPIA) was developed and applied in the determination of bisphenol A in environmental water samples. BVA was selected as the hapten to couple with bovine serum albumin and the conjugate was used as the immunogen for the monoclonal antibody production. Three fluorescein-labeled BVA tracers with different structures (BVA–AMF, BVA–EDF, and BVA–lysFITC) were synthesized. Under the same optimal conditions, BVA–AMF showed the highest sensitivity for FPIA and the detection of BPA exhibited a limit of detection of 5.60 ng mL−1, an IC50 of 140 ng mL−1 and a dynamic range of 11.32–904.21 ng mL−1 approximately. In this assay, several similar compounds were shown of little significantly with the cross-reactivity being less than 0.15%. Four different kinds of water samples were analyzed, with recoveries being 87.91–114.28%. The detection standard curve of BPA exhibited a good linearity (R2 = 0.9913, n = 3). Compared with ELISA and HPLC methods, FPIA showed reliability and a high correlation with ELISA of 0.9964 and HPLC of 0.9971. The proposed immunoassay technique is suitable for detection of BPA in authentic environmental water samples.

Magnetic hyperthermia has been rapidly developed as a potential cancer treatment in recent years. Artificially induced hyperthermia close to a tumor can raise the temperature to 45°C causing tumor cell death. Herein, we introduce a novel method for rapid preparation of anti-cancer magnetocaloric PCL/Fe 3 O 4 mats capable of high-performance hyperthermia using E-jet 3D printing technology. Our 3D printed mats not only maintained the heating efficiency of traditional techniques for magnetic hyperthermia but also prolonged the effective therapy in vivo . When Fe 3 O 4 nanoparticles (NPs) were used in mats at a concentration of 6 mmol/L, 0.07 g PCL/Fe 3 O 4 mats were able to increase the temperature peripherally to 45°C under an alternating magnetic field (AMF) within 45 min. Moreover, the reproducibility experiment indicated that the maximum temperature was achieved following repeated heating and cooling cycles. Cell toxicity tests showed a high cell death rate during one treatment cycle. In vivo experiments indicated clear signs of tumor growth inhibitory and prolonged survival time of tumor-bearing mice after 4 weeks of treatment. The present magnetic mats may be a potential candidate for a novel heat-generating substrate for localized hyperthermia cancer therapy. Furthermore, the main advantage of such implantable magnetic mats is the local and precise delivery of Fe 3 O 4 NPs, ideal for the hyperthermia treatment of easily accessible tumors. © 2017 Wiley Periodicals, Inc. As a crucial reciprocal partner, arbuscular mycorrhizal fungi (AMF) can alleviate the negative effects of a variety of pollutants on their hosts and soil microbes. In our previous studies, such characteristics of AMF on plant growth were documented in response to metal engineered nanoparticle (ENP) treatments. However, the role of AMF in influencing ENP effects on soil microbes remains a matter of debate. To address this issue, we investigated the responses of soil microorganisms to iron oxide nanoparticles (Fe3O4NPs) along a concentration gradient (0.1, 1.0, and 10.0 mg kg−1) in maize plants inoculated with and without AMF. The results showed that a high concentration of Fe3O4NPs significantly decreased the soil bacterial abundance and shifted the community composition, and these negative responses were associated with decreased soil dissolved organic C (DOC) contents in non-AMF-inoculated treatments. However, in AMF-inoculated treatments, no significant changes in soil biota or DOC contents were observed under Fe3O4NPs treatment. These results indicate that AMF alter the effects of Fe3O4NPs on soil microorganisms, possibly by influencing plant growth and organic matter released from plant roots, as DOC contents were impacted by AMF. Our findings suggest that AMF can influence Fe3O4NP-plant-microbe interactions; therefore, more attention should be focused on plant-associated microbes when evaluating the biological effects of nanoparticles.

Repairing infected bone defects remains a severe challenge due to antibiotic abuse and recurrence. Hence, we modified magnetocaloric Fe3O4 nanoparticles and added them to magnesium calcium phosphate bone cement (MCPC) to fabricate multifunctional magnetic composites for sequential bacterial inhibition, angiogenesis and osteogenesis. Nevertheless, high doses of Mg ions and Fe ions were released from MCPC, which adversely affected osteogenesis. Thus, Fe3O4 was modified using gelatin according to the emulsification crosslinking method, which exhibited a controllable magnetocaloric effect and degradation behavior, and favorable anti-bacterial ability under the action of an alternating magnetic field (AMF). In the early stage, the residual MgO created a local strong alkaline microenvironment by hydrolysis, which inhibited the function and activity of S. aureus and E. coli. At the later stage, the MCPC composites were controllably degraded under the function of gelatin and maintained a long-term local slight alkaline microenvironment that promoted the osteogenic differentiation and mineralization of BMSCs. In vivo subcutaneous implantation experiments further indicated that MCPC composites showed good biocompatibility and facilitated angiogenesis, presenting a promising future in magnetic materials design and infectious bone defect repair.

The traditional treatment of atherosclerosis (AS) is mainly focused on lipid-lowering therapy, through oral statins to reduce blood lipid levels and control the progress of AS. However, current oral drug therapy has suffered from poor targeting and single efficacy. Here, a pH-responsive magnetic nanoplatforms (MMNS@AT-CS-DS) was developed. The dextran sulfate (DS) was encapsulated on MMNS@AT-CS to endow the nanoplatforms with the property of targeting atherosclerotic plaques. In vitro and in vivo results showed that MMNS@AT-CS-DS could target lesion areas and release statins to slow the progression of AS due to the magnetocaloric effect under alternating magnetic field (AMF) and dissolution of chitosan (CS) in the AS induced low pH environment. More importantly, its magneto-thermal responsiveness could induce protective autophagy to treat AS synergistically. In addition, the magnetic nanoplatforms could perform magnetic resonance (MR) imaging of atherosclerotic plaques, which shed new light on the integration of diagnosis and treatment of AS.

Nanoparticle-mediated thermotherapeutic research strives innovative, multifunctional, efficient, and safe treatments. Our study introduces a novel nanoplatform: the hollow magnetic vortex nanorings within a polydopamine layer (HMVNp), which exhibit dual functionality as magnetic and photothermal agents. Utilizing a “Dual-mode” approach, combining an alternating magnetic field (AMF) with near-infrared (NIR) laser irradiation, HMVNp demonstrated a significant enhancement in heating efficacy (58 ± 8 %, SAR = 1441 vs 1032 W/g) over traditional solid magnetite nanoparticles coated with polydopamine (SMNp). The unique geometry larger surface area to volume ratio facilitates efficient magnetic vortex dynamics and enhanced heat transfer. Addressing the challenge of heat resistant heat shock protein (Hsp) expression, encapsulated quercetin (Q) within HMVNp leverages tumor acidity and dual-mode thermal therapy to enhance release, showing a 28.8 ± 6.81 % increase in Q loading capacity compared to traditional SMNp. Moreover, HMVNp significantly improves contrast for both magnetic resonance imaging (MRI) and photoacoustic imaging (PAI), with an approximately 62 % transverse relaxation (R2 = 81.5 vs 31.6 mM -1 s −1 [Fe]). In vivo studies showed that while single treatments slowed tumor growth, dual-mode therapy with quercetin significantly reduced tumors and effectively prevented metastases. Our study highlights the potential of HMVNp/Q as a versatile agent in thermotherapeutic interventions, offering improved diagnostic imaging capabilities.

Infectious bone defects pose significant clinical challenges due to persistent infection and impaired bone healing. Icam1+ macrophages were identified as crucial and previously unrecognized regulators in the repair of bone defects, where impaired oxidative phosphorylation within this macrophage subset represents a significant barrier to effective bone regeneration. To address this challenge, dual-responsive iron-doped barium titanate (BFTO) nanoparticles were synthesized with magnetic and ultrasonic properties. These nanoparticles were further loaded with the anti-inflammatory agent curcumin and coated with engineered mesenchymal stem cell membranes (EMM) modified with γ3 peptide, creating BFTO-Cur@EMM nanoparticles specifically designed to target Icam1+ macrophages. These nanoparticles were shown to disrupt bacterial biofilms under alternating magnetic fields (AMF) and to activate oxidative phosphorylation and osteogenic immune responses in Icam1+ macrophages via low-intensity pulsed ultrasound (LIPUS). Transcriptomic sequencing and validation experiments demonstrated that this approach activates oxidative phosphorylation (OXPHOS) by stimulating the JAK2-STAT3 pathway and inhibiting the MAPK-JNK pathway, thereby promoting the polarization of Icam1+ macrophages toward a pro-reparative phenotype and enhancing the secretion of pro-angiogenic and osteogenic cytokines. These nanoparticles were subsequently integrated into quaternized chitosan (QCS) and tricalcium phosphate (TCP) to create a bioink for three-dimensional (3D) printing anti-infection QT/BFTO-Cur@EMM bone repair scaffolds. In vivo studies indicated that these scaffolds significantly improved the healing of infectious bone defects without causing thermal damage to surrounding tissues. This work highlights the potential of this material and the targeting of Icam1+ macrophages as an effective strategy for simultaneously controlling infection and promoting bone regeneration.
