Individual and combined contamination of oxytetracycline and cadmium inhibited nitrification by inhibiting ammonia oxidizers

Cao Xiaoxu,Zhao Wei,Zhang He,Lin Jitong,Hu Jingying,Lou Yanhong,Wang Hui,Yang Quangang,Pan Hong,Zhuge Yuping

Introduction: The large-scale development of animal husbandry and industrialization lead to more and more serious co-contamination from heavy metals and antibiotics in soils. Ecotoxic effects of residues from antibiotics and heavy metals are of increasing concern.Materials and Methods: In this study, oxytetracycline (OTC) and cadmium (Cd) were selected as target pollutants to evaluate the individual and combined effects on nitrification process using four different soil types sampled from North to South China through a 56-day incubation experiment. Results and Discussion: The results demonstrated that the contaminations of OTC and Cd, especially combined pollution had significant inhibitory effects on net nitrification rates (NNRs) as well as on AOA and AOB abundance. The toxic effects of contaminants were greatly enhanced with increasing OTC concentration. AOB was more sensitive than AOA to exogenous contaminants. And the interaction effects of OTC and Cd on ammonia oxidizers were mainly antagonistic. Furthermore, Cd contaminant (with or without OTC) had indirect effects on nitrification activity via inhibiting mineral N and AOA/AOB, while OTC alone indirectly inhibited nitrification activity by inhibiting ammonia oxidizers. The results could provide theoretical foundation for exploring the eco-environmental risks of antibiotics and heavy metals, as well as their toxic effects on nitrification processes.

Cooperation among nitrifying microorganisms promotes the irreversible biotransformation of sulfamonomethoxine

Xueqin Yang,Yijing Shi,Guangguo Ying,Mengyuan Li,Zhili He,Longfei Shu

Ammonia-oxidizing microorganisms, including AOA (ammonia-oxidizing archaea), AOB (ammonia-oxidizing bacteria), and Comammox (complete ammonia oxidization) Nitrospira , have been reported to possess the capability for the biotransformation of sulfonamide antibiotics. However, given that nitrifying microorganisms coexist and operate as communities in the nitrification process, it is surprising that there is a scarcity of studies investigating how their interactions would affect the biotransformation of sulfonamide antibiotics. This study aims to investigate the sulfamonomethoxine (SMM) removal efficiency and mechanisms among pure cultures of phylogenetically distinct nitrifiers and their combinations. Our findings revealed that AOA demonstrated the highest SMM removal efficiency and rate among the pure cultures, followed by Comammox Nitrospira , NOB, and AOB. However, the biotransformation of SMM by AOA N. gargensis is reversible, and the removal efficiency significantly decreased from 63.84 % at 167 h to 26.41 % at 807 h. On the contrary, the co-culture of AOA and NOB demonstrated enhanced and irreversible SMM removal efficiency compared to AOA alone. Furthermore, the presence of NOB altered the SMM biotransformation of AOA by metabolizing TP202 differently, possibly resulting from reduced nitrite accumulation. This study offers novel insights into the potential application of nitrifying communities for the removal of sulfonamide antibiotics (SAs) in engineered ecosystems.

The single and combined effects of sulfamethazine and cadmium on soil nitrification and ammonia-oxidizing microorganisms

Zhou Changrui,Gao Yun,Ma Qiang,Xia Zhuqing,Zhu Mengmeng,Zhang Xinhui,An Siyu,Li Shuailin,Yu Wantai

The coexistence of antibiotics and heavy metals in soil has attracted increasing attention due to their negative effects on microorganisms. However, how antibiotics and heavy metals affect functional microorganisms related to nitrogen cycle remains unclear. The goals of this work were to explore the individual and combined effects of sulfamethazine (SMT) and cadmium (Cd), selected as target pollutants in soil, on potential nitrification rates (PNR) and ammonia oxidizers (ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB)) structure and diversity by 56-day cultivation experiment. Results showed that PNR in Cd- or SMT-treated soil decreased at the beginning of the experiment and then increased over time. PNR was significantly correlated with AOA and AOB- amoA relative abundance ( P  < 0.01). SMT addition (10 and 100 mg kg −1 ) significantly improved AOA activity by 13.93% and 17.93%, respectively, and had no effect on AOB at day 1. Conversely, Cd at 10 mg kg −1 significantly inhibited AOA and AOB by 34.34% and 37.39%, respectively. Moreover, the relative abundance of AOA and AOB in combined SMT and Cd addition clearly higher relative to single Cd at 1 day. The single and combined Cd and SMT increased and reduced the community richness of AOA and AOB, respectively, but reduced the diversity of both after 56 days. Cd and SMT treatments significantly changed the relative abundance of AOA phylum levels and AOB genus levels in the soil. It was mainly manifested in reducing the relative abundance of AOA Thaumarchaeota , and increasing the relative abundance of AOB Nitrosospira. Besides, AOB Nitrosospira was more tolerant to the compound addition of both than single application. Graphical abstract

Biological nitrification inhibition by sorghum root exudates impacts ammonia-oxidizing bacteria but not ammonia-oxidizing archaea

Li Yaying,Zhang Yang,Chapman Stephen James,Yao Huaiying

Sorghum has a great capacity to release biological nitrification inhibitors (BNIs), but the inhibitory effect on nitrification and ammonia oxidizer populations under planted soil conditions is unclear. A pot experiment with three nitrogen (N) application rates (0, 50, and 200 mg N kg −1 ) was set up to detect the influence of sorghum growth on soil nitrification and investigate the function of blocking the activity of ammonia oxidizers. A 15 N-labeled experiment was also conducted to detect the N form absorbed by sorghum. Sorghum root exudates were collected at 30 days after transplanting to hydroponic culture and added into cultured soil to determine the shifts in the populations of nitrifiers. The 15 N labeling experiment showed that the uptake rate by sorghum of ammonium N fertilizer was 24% and that of nitrate N fertilizer was 9%, indicating that sorghum was an ammonium using plant. Compared with unplanted soil, sorghum planting had a significant inhibitory effect on the nitrification process even at the high-N fertilizer rates. Autotrophic nitrification was the prevailing process, and sorghum root exudation inhibited this process as much as dicyandiamide (DCD, 10 mg kg −1 ). Root exudates had a significant inhibitory effect on ammonia-oxidizing bacteria (AOB) but had no effect on ammonia-oxidizing archaea (AOA).

Autotrophic archaeal nitrification is preferentially stimulated by rice callus mineralization in a paddy soil

Liu Haiyang,Pan Hong,Hu Hangwei,Jia Zhongjun,Zhang Qian,Liu Yimeng,Xu Jianming,Di Hongjie,Li Yong

Aims Previous studies suggest that organic carbon (C) and nitrogen (N) can stimulate soil nitrification, but whether autotrophic or heterotrophic nitrification is stimulated and who are active nitrifiers for the nitrification activity is still in debate. We elucidated which nitrification dominated and the active nitrifiers during the decomposition of rice callus. Methods 15 N-labeled callus and acetylene (C 2 H 2 ) inhibition were used to explore the autotrophic or heterotrophic nitrification during the decomposition of callus and DNA-based stable isotope probing (SIP) and high-throughput sequencing were used to investigate the active nitrifiers. Results Autotrophic nitrification dominated the nitrification activity, driven by oxidation of ammonia (NH 3 ) produced from mineralization of the callus-derived organic N. Callus significantly stimulated nitrification activity, which was paralleled by changes in the abundance and community composition of AOA. DNA-SIP further demonstrated that the active AOA outnumbered their bacterial counterparts in the 13 C-DNA from the soil with callus amendment. Phylogenetic analysis revealed the functional importance of soil fosmid 29i4-like and 54d9-like AOA within soil group 1.1b during the active nitrification with callus cells. Conclusions NH 3 released from the mineralization of callus was the main substrate for autotrophic nitrification and preferentially stimulated the growth of AOA within group 1.1b in the paddy soil.

Canonical ammonia oxidizers, rather than comammox Nitrospira, dominated autotrophic nitrification during the mineralization of organic substances in two paddy soils

Haiyang Liu,Hangwei Hu,Xing Huang,Tida Ge,Yongfu Li,Zhenke Zhu,Xingmei Liu,Wenfeng Tan,Zhongjun Jia,Hongjie Di,Jianming Xu,Yong Li

Chemoautotrophic canonical ammonia-oxidizers and complete ammonia oxidizers (comammox) are responsible for ammonia oxidation, which is the rate-limiting step of nitrification in terrestrial ecosystems. However, the relative importance of autotrophic nitrification and the related active nitrifiers during mineralization of organic substances with different C/N ratios have not been fully elucidated to date. In this study, 15 N tracing and 13 CO 2 -DNA-based stable isotope probing (DNA-SIP) techniques were employed to investigate the responses of soil nitrification processes and the active nitrifying phylotypes in acidic and neutral paddy soils with amendment of glutamine (Gln) or rice straw with low C/N (LS) or high C/N (HS). Acetylene (C 2 H 2 ) was utilized to differentiate autotrophic and heterotrophic nitrification. The results indicated that C 2 H 2 completely blocked the production of 15 NO 3 − in Gln and LS treatments, implying the predominance of autotrophic nitrification with the N derived from the mineralization of Gln and LS. Neither 15 NH 4 + nor 15 NO 3 − was detected in the HS-amended soils, suggesting that NH 4 + derived from organic N mineralization was immobilized immediately by microorganisms. In the DNA-SIP microcosms, the abundance of ammonia-oxidizing bacteria (AOB) in the unamended control (CK), Gln and LS treatments of acidic paddy soil increased by 22.1-, 71.9- and 27.6-fold, respectively. A significantly larger proportion of Nitrosospira -like AOB, rather than ammonia-oxidizing archaea (AOA) and comammox Nitrospira , was labelled by 13 CO 2 , indicating that AOB made a stronger contribution to autotrophic nitrification in acidic paddy soil. The active nitrification of neutral paddy soil was predominated by AOA affiliated with the Nitrososphaera viennensis lineage in the CK and LS microcosms but by Nitrosospira -like AOB in the Gln microcosm. These results suggest the significance of autotrophic nitrification catalysed by canonical ammonia oxidizers, rather than comammox, during the mineralization of organic substances with low C/N in paddy soils.

Di-(2-ethylhexyl) phthalate (DEHP) exposure suppressed the community diversity and abundance of ammonia-oxidizers and mitigated N2O emissions in an alkaline soil

Rui Tao,Hanjie Zhang,Xiuping Gu,Baowei Hu,Jun Li,Guixin Chu

Phthalic acid esters (PAEs) pollution has become an increasing issue worldwide, but little is known about its effects on ammonia-oxidizers and nitrous oxide (N 2 O) in the soil environment. Here, a 50-day soil microcosm experiment was conducted using low and high doses bis (2-Ethylhexyl) phthalate (DEHP) to estimate the effect of DEHP exposure on soil N 2 O emissions and the nitrifying community in calcareous soil. The results showed that DEHP exposure at 10 and 100 mg kg -1 doses significantly reduced N 2 O by 67.5% and 73.6%, respectively, relative to the DEHP absent treatment. The microbial biomass carbon and nitrogen (MBC and MBN) were consistently and significantly decreased with DEHP exposure at 5, 22, and 50 days after incubation, especially with high-dose. The bipartite association networks showed that DEHP exposure changed the compositions of both ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) communities. Moreover, the AOA and AOB amoA gene abundances were significantly decreased by DEHP addition to the soil ( P  < 0.05). Random forest modeling showed that the AOB Shannon index and pH were the most important biotic and abiotic factors affecting N 2 O emissions in the soil with DEHP exposure, respectively. Partial least squares path modeling (PLS-PM) demonstrated that the reduction of N 2 O emissions due to DEHP exposure was mainly ascribed to the changes of the AOB community. The results from this study highlight the toxicity of DEHP on the ammonia oxidizers, and its mitigating effect on N 2 O emissions in the soil where ammonia-oxidation is largely driven by AOB.

Treg-enhancing and immunomodulating microgel scaffold promotes cell ingrowth and heart function recovery post-acute myocardial infarction in vivo

Shuqin Wang,Kai Wang,Wangbei Cao,Liang Song,Shifen Li,Zihe Zhai,Liyin Shen,Yang Zhu,Wenxing Liu,Changyou Gao

The ingrowth of endogenous cells into tissue scaffolds and modulation of Treg population take a pivotal function in restoring the structure and function of heart post-acute myocardial infarction (MI). However, the traditional hydrogels are failed to achieve such functions due to their critically small mesh size and inherent bioinertness. In this study, a Treg-regulated microgel scaffold (GTK-TK-drug) was prepared by inter-microgels assembly of microgels having cyclodextrin (CD) or adamantane (Ad) with a particle size of about 220 μm. The raw materials to obtain the microgels included gelatin methacryloyl (GelMA), reactive oxygen species (ROS)-responsive crosslinking agent poly(ethylene glycol)-thioketal-poly(ethylene glycol) (PEG-TK-PEG), ROS-responsive release prodrug aminooxyacetic acid (PEG-TK-AOA), PEG-CD or PEG-Ad, and photoinitiator lithium phenyl-2,4,6-trimethylbenzoylphosphinate (LAP). Quantitative real-time PCR (qPCR), cell counting kit-8, and immunofluorescence staining revealed the effectiveness of AOA for the differentiation of T cells into Treg and the ingrowth of cells in vitro and in vivo . During the short treatment of MI in vivo , the levels of plasma inflammatory factors, cardiomyocyte apoptosis and inflammatory cells were reduced significantly. In the long-term animal experiments, the recovery of cardiac function was significantly enhanced as characterized by echocardiography and the degree of myocardial fibrosis compared to the unassembled microgels control.
