At present, the 100% case fatality and the cross-infection of virus strains make the ASFV 's harm to society continue to expand. The absence of an effective commercial vaccine poses early detection remains the most effective means of curbing ASFV infection. Here, we report a cascaded detection platform based on the CRISPR-Cas12a system combined with graphene field-effect transistor sensors. The cascade platform could detect ASFV as low as 0.5 aM within 30 min and achieve typing of wild and vaccine strains of ASFV in a single detection system. The evaluation of 16 clinical samples proved that, compared with the gold standard Real-time PCR method, this platform has outstanding advantages in sensitivity, specificity and typing. Combining CRISPR-Cas12a's high specificity with the bipolar electric field effect of graphene field-effect transistor, the cascade platform is expected to achieve clinical application in the field of DNA disease detection, and provides a new direction for multi-strain disease typing.

African swine fever virus (ASFV) is a highly infectious and pathogenic DNA virus with a mortality rate of nearly 100%. Here, we developed a flexible biosensor based on a AgNWs/hydrogel/PS(polystyrene)/Fe 3 O 4 composite film for the detection of ASFV P72 protein gene fragments. A large number of carboxyl groups in the hydrogel provide modification sites for capture probes (cp) simplifying the preparation process of the biosensor. Combined with the excellent magnetic properties of Fe 3 O 4 , the excellent electrical conductivity of silver nanowires (AgNWs) and the unique optical properties of two-dimensional photonic crystals (2DPC), the flexible biosensor can convert biological signals into optical signals and electrical signals, and the micro-deformation of the film can be verified by simple optical methods. In order to improve the sensitivity, we introduce Fe 3 O 4 and apply an external magnetic field to amplify the thin film micro-deformation. The concentration of P72 protein gene fragment and the relative change rate of resistance showed a good linear relationship, and the linear equation is y = − 0.00126x − 0.31729; the detection limit (LOD) is as low as 0.208 μM. The composite film was used to detect real serum samples, and the recovery of the composite film fluctuated in the range 91.89 to 103.19%, indicating that the composite film has practical application potential in clinical detection of ASFV. In addition, the biosensor also shows good biocompatibility, stability, and specificity. Graphical

African swine fever (ASF) is a highly contagious and hemorrhagic disease caused by infection with the African swine fever virus (ASFV), resulting in a mortality rate of up to 100%. Currently, there are no effective treatments and commercially available vaccines for ASF. Therefore, it is crucial to identify biochemicals derived from host cells that can impede ASFV replication, with the aim of preventing and controlling ASF. The ASFV is an acellular organism that promotes self-replication by hijacking the metabolic machinery and biochemical resources of host cells. ASFV specifically alters the utilization of glucose and glutamine, which are the primary metabolic sources in mammalian cells. This study aimed to investigate the impact of glucose and glutamine metabolic dynamics on the rate of ASFV replication. Our findings demonstrate that ASFV infection favors using glutamine as a metabolic fuel to facilitate self-replication. ASFV replication can be substantially inhibited by blocking glutamine metabolism. The metabolomics analysis of the host cell after late-stage ASFV infection revealed a significant disruption of normal glutamine metabolic pathways due to the abundant expression of PLA (phenyllactic acid). Pretreatment with PLA also inhibited ASFV proliferation and glutamine consumption following infection. The metabolomic analysis also showed that PLA pretreatment greatly slowed down the metabolism of amino acids and nucleotides that depend on glutamine. The depletion of these building blocks directly hindered the replication of ASFV by decreasing the biosynthetic precursors produced during the replication of ASFV’s progeny virus. These findings provide valuable insight into the possibility of pursuing the development of antiviral drugs against ASFV that selectively target metabolic pathways.

ASFV is the only known double-stranded insect-borne DNA virus, which can rapidly infect domestic pigs and wild boars with ticks as transmission medium. Since it was first discovered in 1921, it quickly spread to all parts of the world and brought huge economic losses to the pig industry all over the world. At present, there is still no safe and effective vaccine for ASFV. Here, we developed a quantum-dot labeled antibody test strip for the detection of antibodies against ASFV pp62. The pp62 protein was labeled with quantum dots, and the antibody test strip was developed uses it in a detection mode of labeled antigen-SPA interceptor-monoclonal antibody quality control. The test strip showed high sensitivity, the positive detection limit of the strip was 1: 10 6 by continuous multiple dilution using the positive standard serum of ASFV antibody as reference. The test strip showed good specificity, and there was no cross reaction with other swine diseases virus (PCV2, PRRSV, CSFV, PPV). Using the detection results of commercialized kit for African swine fever virus as reference, 80 ASFV antibody negative serum and 4 different ASFV antibody positive serum were detected using the ASFV pp62 quantum-dot labeled antibody test strip. The results were consistent with the commercial kit. This study provides a new detection method for the prevention and control of African swine fever.

Abstract African swine fever virus (ASFV) causes a highly contagious and often lethal swine viral disease, and leads to tremendous economic losses to the swine industry. Unfortunately, there are no vaccines and effective antiviral agents available to prevent and control ASFV outbreaks. Therefore, it is necessary to develop simple and rapid strategies to monitor ASFV-infected pigs to restrain its spread. In the current study, ASFV capsid protein p72 was expressed along with its chaperone pB602L to form trimers in human embryonic kidney 293 (HEK293) cells. The p72 trimers were subsequently labeled with colloidal gold to develop a immunochromatographic strip. The strip showed high specificity to ASFV-positive serum and no cross-reactivity to other swine virus positive sera. Importantly, the strip showed a higher sensitivity of detecting ASFV antibodies in both positive standard serum and clinical serum samples than a commercial enzyme-linked immunosorbent assay (ELISA) kit. Taken together, these results demonstrate the strip as a reliable diagnostic tool against ASFV infection, which will be appropriate for application in prevention and control of ASFV.
Key points
• ASFV p72 trimers were successfully generated . • A colloidal gold strip was developed based on ASFV p72 trimers . • The strip is appropriate for detecting ASFV antibodies in the field .

The lack of simple, rapid, accurate, and low‐cost on‐site diagnostic technology has seriously hindered the effective prevention and control of major infectious diseases such as African swine fever (ASF). This study presents an integrated, portable diagnostic system comprising a low‐cost microfluidic chip and a fully automatic, portable device for simultaneously detecting nine samples. The aim is to achieve rapid on‐site detection of African swine fever virus (ASFV) nucleic acid. The system integrates magnetic bead nucleic acid extraction, transfer, loop‐mediated isothermal amplification (LAMP), and optical detection functions, and can perform fully automatic, closed‐tube analysis of 9 samples in 25 min, effectively avoiding aerosol pollution. The detection limit for the ASFV P10 gene is 10 2 copies/μL. In a blind test involving 87 clinical samples, the system demonstrated excellent performance with a clinical sensitivity and specificity of 97.6% (42/44) and 98.0% (42/43), respectively, and an accuracy of 96.6% (84/87). When the sample concentration exceeds the detection limit, sensitivity can reach 100 %. The core advantages of this method are that it is fast, accurate, and low cost, and does not require professional laboratories or operators. Its modular design makes it easy to expand to a 100‐channel, high‐throughput detection system at low cost. This system provides a powerful technical tool for large‐scale epidemic screening and prevention in areas with limited resources, offering significant public health and economic value.

African swine fever (ASF) is a swine viral disease that could cause highly contagious and extremely high mortality, causing huge economic losses to the pig industry. As there is currently no vaccine and effective treatment methods. Therefore, early monitoring is one of the most important solutions to prevent and control ASF. In this study, the dual QDM recombinant virus protein 30 and 54 (P30 and P54) probes and pre-incubation in vitro were proposed for the first time as QDM based-ASFV immunosensor (QAIS) for the ultra-sensitive quantitative detection of ASFV antibodies in serum. In the range from serum dilution of 1:1000 to 1:64000, it showed a good linear relationship (R 2 = 0.9947), and its detection sensitivity was 1:64000 dilution. Compared with commercial enzyme-linked immunosorbent assay (ELISA) and colloidal gold immunochromatographic strip (CGICS), its detection sensitivity was improved by at least one order of magnitude and four orders of magnitude respectively. In addition, the whole ASFV antibody screening test can be completed in 25 min with simple operation. The performance and practicability of the established QAIS sensor have been verified by ASF-ELISA kit, and its coincidence rate was as high as 98.7% in 151 clinical samples. We firmly believe that the proposed QAIS sensor could potentially be applied to point-of-care testing (POCT) for quantitative ASFV antibody in pig farms.
