Atg8 and Ire1 in combination regulate the autophagy-related endoplasmic reticulum stress response in Candida albicans

Jiawen Du,He Zhao,Mengsen Zhu,Yixuan Dong,Liping Peng,Jianrong Li,Qiang Zhao,Qilin Yu,Mingchun Li

The unfolded protein response (UPR) is an important pathway to prevent endoplasmic reticulum (ER) stress in eukaryotic cells. In Saccharomyces cerevisiae , Ire1 is a key regulatory factor required for HAC1 gene splicing for further production of functional Hac1 and activation of UPR gene expression. Autophagy is another mechanism involved in the attenuation of ER stress by ER-phagy, and Atg8 is a core protein in autophagy. Both autophagy and UPR are critical for ER stress response, but whether they act individually or in combination in Candida albicans is unknown. In this study, we explored the interaction between Ire1 and the autophagy protein Atg8 for the ER stress response by constructing the atg8 Δ/Δ ire1 Δ/Δ double mutant in the pathogenic fungus C. albicans . Compared to the single mutants atg8 Δ/Δ or ire1 Δ/Δ, atg8 Δ/Δ ire1 Δ/Δ exhibited much higher sensitivity to various ER stress-inducing agents and more severe attenuation of UPR gene expression under ER stress. Further investigations showed that the double mutant had a defect in ER-phagy, which was associated with attenuated vacuolar fusion under ER stress. This study revealed that Ire1 and Atg8 in combination function in the activation of the UPR and ER-phagy to maintain ER homeostasis under ER stress in C. albicans .

Apicoplast biogenesis mediated by ATG8 requires the ATG12–ATG5-ATG16L and SNAP29 complexes in Toxoplasma gondii

Jiawen Fu,Lin Zhao,Yu Pang,Heming Chen,Hayashi Yamamoto,Yuntong Chen,Zhaoran Li,Noboru Mizushima,Honglin Jia

In apicomplexan parasites, the macroautophagy/autophagy machinery is repurposed to maintain the plastid-like organelle apicoplast. Previously, we showed that in Toxoplasma and Plasmodium, ATG12 interacts with ATG5 in a non-covalent manner, in contrast to the covalent interaction in most organisms. However, it remained unknown whether apicomplexan parasites have functional orthologs of ATG16L1, a protein that is essential for the function of the covalent ATG12–ATG5 complex in vivo in other organisms. Furthermore, the mechanism used by the autophagy machinery to maintain the apicoplast is unclear. We report that the ATG12–ATG5-ATG16L complex exists in Toxoplasma gondii (Tg). This complex is localized on isolated structures at the periphery of the apicoplast dependent on TgATG16L. Inducible depletion of TgATG12, TgATG5, or TgATG16L caused loss of the apicoplast and affected parasite growth. We found that a putative soluble N-ethylmaleimide sensitive factor attachment protein receptor (SNARE) protein, synaptosomal-associated protein 29 (TgSNAP29, Qbc SNARE), is required to maintain the apicoplast in T. gondii. TgSNAP29 depletion disrupted TgATG8 localization at the apicoplast. Additionally, we identified a putative ubiquitin-interacting motif-docking site (UDS) of TgATG8. Mutation of the UDS site abolished TgATG8 localization on the apicoplast but not lipidation. These findings suggest that the TgATG12–TgATG5-TgATG16L complex is required for biogenesis of the apicoplast, in which TgATG8 is translocated to the apicoplast via vesicles in a SNARE -dependent manner in T. gondii.Abbreviations: AID: auxin-inducible degron; CCD: coiled-coil domain; HFF: human foreskin fibroblast; IAA: indole-3-acetic acid; LAP: LC3-associated phagocytosis; NAA: 1-naphthaleneacetic acid; PtdIns3P: phosphatidylinositol-3-phosphate; SNARE: soluble N-ethylmaleimide sensitive factor attachment protein receptor; UDS: ubiquitin-interacting motif-docking site; UIM: ubiquitin-interacting motif.

Gypenoside GP5 effectively controls Colletotrichum gloeosporioides, an anthracnose fungus, by activating autophagy

Yujie Liu,Xinyv Li,Chu Gong,Yonghong Cao,Jun Wang,Min Han,Jun-Li Yang

Anthracnose is a plant disease caused by Colletotrichum spp ., known for its widespread infectivity and extreme destructiveness. Colletotrichum gloeosporioides is a representative pathogen of anthracnose in China. Gypenosides GP4 - GP7 , derived from Gynostemma pentaphyllum (Thunb.) Makino, could significantly inhibit the growth of C. gloeosporioides mycelial, with EC 50 values of 96.98, 27.5, 38.48, and 61.59 mg L −1 . The inhibitory effect of these compounds surpassed the commonly used chemical pesticide chlorothalonant and plant-derived pesticide matrine. Among them, the most active compound GP5 also showed a significant inhibitory effect on spore germination and bud tube elongation of C. gloeosporioides . In addition, GP5 could effectively suppress the spread of anthracnose spots in postharvest fruit. Transmission electron microscopy and fluorescence microscopy demonstrated that GP5 primarily exerted its antifungal function by activating cellular autophagy. Additionally, proteomics analysis revealed that GP5 had an antifungal effect against C. gloeosporioides by enhancing cellular autophagy through upregulation of the expression of the autophagy-related protein Atg8. This study presents a novel approach for the control and management of anthracnose in C. gloeosporioides . Consequently, GP5 has the potential to be developed as a plant-derived fungicide for the biological control of anthracnose.

Vrl1 relies on its VPS9-domain to play a role in autophagy in Saccharomyces cerevisiae

Wenjing Li,Zulin Wu,Yongheng Liang

Autophagy is an intracellular degradation process involving many Atg proteins, which are recruited hierarchically to regulate this process. Rab/Ypt GTPases and their activators, guanine nucleotide exchange factors (GEFs), which are critical for regulating vesicle trafficking, are also involved in autophagy. Previously, we reported that yeast Vps21 and its GEF Vps9 are required for autophagy. Later, a third yeast VPS9-domain-containing protein, V A R P- l ike 1 (Vrl1), which was identified as a mutant in major laboratory strains, had partially overlapping functions with Vps9 in trafficking. In this study, we showed that Vrl1 performed roles in autophagy, and its VPS9-domain was crucial for its role in autophagy. We found that localization of Vrl1 differed from the other two VPS9-domain-containing proteins, Vps9 and Muk1, and only Vrl1 changed from multipoint to diffusion after starvation. Like Vps9, Vrl1 suppressed autophagic defects caused by the VPS9 deletion. We further showed that these VPS9-domain-containing proteins, Vps9, Muk1, and Vrl1, all co-localized with Atg8 on autophagosomes in cells blocked in any late step of starvation-induced autophagy, with Vrl1 most often co-localizing with Atg8. A small portion (<25%) of these VPS9-domain-containing proteins were degraded through autophagy. However, a large portion (>60%) of Vrl1 decreased independently of autophagy. We propose that Vrl1 may regulate autophagy in a similar way as Vps9, and the level of Vrl1 partly decreases through both autophagy-dependent and -independent routes.

Plasma membrane CYBDOM proteins catalyse apoplastic AsA regeneration and interact with RbohD to activate autophagy and drought tolerance in plants

Chen Xiuxiu,Jin Songsong,Du Hong,Liu Zebin,Hou Congcong,Tang Ling,Wang Yuanyuan,Wang Yuxi,Yang Zhaolin,Zhao Tong,Ma Jinying,Zhang Lanjun,Li Meijing,Qi Dongdong,Li Wenlong,Wang Xiaohua,Sun Runze,Moore John P.,Liu Yongxiu,Lin Jinxing,Deng Xin

Autophagy and apoplastic ascorbic acid (AsA) play important roles in plant drought tolerance. However, the trans-plasma membrane transport of AsA and its association with autophagy during drought stress are unclear. Here we report an AsA-induced autophagy pathway in plants, wherein plasma-membrane-located cytochrome b561 and DOMON domain (CYBDOM) proteins play positive roles. CYBDOM proteins from the resurrection plant Boea hygrometrica (BhDB) and Arabidopsis (AtDB1) can transport electrons across the plasma membrane using intracellular AsA as an electron donor and apoplastic monodehydroascorbic acid or Fe 3+ as electron acceptors in Xenopus laevis oocytes. Increased apoplastic AsA, autophagy and drought tolerance are observed in BhDB - and AtDB1 -overexpressing Arabidopsis compared with the wild type. CYBDOM proteins interact with respiratory burst oxidase homologue D (RbohD), which serves as the adaptor to bind autophagy related gene 8 protein (ATG8) and cargo protein for autophagic degradation. AsA increases AtDB1 protein level and its interaction with RbohD. Together, the AsA-activated CYBDOM–RbohD synergy to induce autophagy suggests a novel mechanism in plant drought and desiccation tolerance.

Autophagy-regulated ethylene synthesis mediates fruit ripening by affecting the accumulation of lycopene, sugars and organic acids in tomato

Xuelian Zheng,Hongfei Yang,Zhichao Li,Canyu Zhou,Xinlin Chen,Kaixin Wang,Ping Yang,Guanghui Wang,Jie Zhou

Autophagy is a universal cellular process in eukaryotes that plays a critical role in plant growth and stress response. However, the role of autophagy in fruit ripening is largely unknown. Here, we demonstrated that most autophagy-related genes ( ATGs ) were up-regulated during tomato ( Solanum lycopersicum L.) fruit ripening. By using mutants of different autophagy pathway genes ( ATG6 , ATG10 , ATG18a ), we revealed that the deficiency of autophagy delayed the ripening of fruit. Compared with wild-type (WT), the production of ethylene was significantly reduced and the accumulation of lycopene was delayed in atg mutants during fruit ripening. We also observed the contents of glucose and fructose were both significantly decreased in atg mutants compared with WT, while the content of organic acids showed the opposite trend. Additionally, the negative regulator of ethylene production, APETALA2a (AP2a), interacted with ATG8 through a specific ATG8-interacting motif (AIM) and could be degraded through the autophagy pathway. These results demonstrate that autophagy plays a critical role in fruit ripening by regulating ethylene production and the accumulation of pigments, sugars and organic acids in tomato.

Phosphatidylethanolamines link ferroptosis and autophagy during appressorium formation of rice blast fungus

Qiao Liu,Ruhui Long,Chaoxiang Lin,Xinping Bi,Zhibin Liang,Yi Zhen Deng

A cell death pathway, ferroptosis, occurs in conidial cells and is critical for formation and function of the infection structure, the appressorium, in the rice blast fungus Magnaporthe oryzae . In this study, we identified an orthologous lysophosphatidic acid acyltransferase (Lpaat) acting at upstream of phosphatidylethanolamines (PEs) biosynthesis and which is required for such fungal ferroptosis and pathogenicity. Two PE species, DOPE and SLPE, that depend on Lpaat function for production were sufficient for induction of lipid peroxidation and the consequent ferroptosis, thus positively regulating fungal pathogenicity. On the other hand, both DOPE and SLPE positively regulated autophagy. Loss of the LPAAT gene led to a decrease in the lipidated form of the autophagy protein Atg8, which is probably responsible for the autophagy defect of the lpaat Δ mutant. GFP-Lpaat was mostly localized on the membrane of lipid droplets (LDs) that were stained by the fluorescent dye monodansylpentane (MDH), suggesting that LDs serve as a source of lipids for membrane PE biosynthesis and probably as a membrane source of autophagosome. Overall, our results reveal novel intracellular membrane-bound organelle dynamics based on Lpaat-mediated lipid metabolism, providing a temporal and spatial link of ferroptosis and autophagy.
