Insight into Fructose-to-Sucrose Ratio as the Potential Target of Urinalysis in Bladder Cancer

Dewang Zhou,Jianxu Huang,Haoxiang Zheng,Yujun Liu,Shimao Zhu,Yang Du

Bladder cancer usually has been diagnosed in elderly patients as it stays asymptomatic until it presents. Current detection methods for bladder cancer cannot be considered as an adequate screening strategy due to their high invasiveness and low sensitivity. However, there remains uncertainty about targets with high sensitivity and specificity for non-invasive bladder cancer examination. Our study aims to investigate the actionable non-invasive screening biomarkers in bladder cancer. Here, we employed scRNA-seq to explore the crucial biological processes for bladder cancer development. We then utilized bidirectional Mendelian randomization (MR) analysis to explore the bidirectional causal relationship between ATP-associated metabolites in urine and bladder cancer. Lastly, we used a BBN-induced mouse model of bladder cancer to validate the crucial gene identified by scRNA-seq and MR analysis. We found that (1) the ATP metabolism process plays a critical role in bladder cancer development; (2) there is a bidirectional and negative causal relationship between fructose-to-sucrose ratio in urine and the risk of bladder cancer; and (3) the higher expression of TPI1, a critical gene in the fructose metabolism pathway, was validated in BBN-induced bladder tumors. Our results reveal that fructose-to-sucrose ratio can serve as a potential target of urinalysis in bladder cancer.

ROCK-Mediated Actin Remodeling Contributes to the Reduced Proliferation and Migration of bEnd.3 Endothelial Cells Treated With Florfenicol

Dongfang Hu,Zihui Shen,Xueke Hou,Zhishuai Wan,Xiaoyu Guo,Lingli Chen,Zhihong Yin,Hongmei Ning,Yaming Ge

Florfenicol (FLO), a widely used antibiotic, can cause early embryonic death and impaired angiogenesis. However, the mechanism by which FLO inhibits angiogenesis and its role in FLO-induced embryonic toxicity are not yet fully understood. This study aimed to investigate the effect of FLO on the proliferation and migration of mouse bEnd.3 endothelial cells (bECs) and the underlying mechanism, given the crucial role of endothelial cells in angiogenesis. The results showed that FLO treatment at 7.5–15 μg/mL significantly inhibited proliferation and migration in a time-dependent manner, with stronger effects observed after 48 h compared to 24 h. RNA sequencing analysis identified 1351 genes differentially expressed in response to FLO. Functional analysis indicated that FLO disrupted ATP metabolism, angiogenesis, vasculature development, and actin filament organization. The F-actin cytoskeleton remodeling was morphologically confirmed, and activation of the ROCK/Cofilin signaling pathway was detected. Subsequent experiments demonstrated that pharmacological inhibition of the pathway normalized cytoskeletal rearrangement and promoted angiogenesis, as evidenced by enhanced wound closure and increased cell viability. Our results demonstrated that ROCK-mediated F-actin remodeling plays a crucial role in the FLO-induced inhibition of proliferation and migration in bECs. These findings may explain why FLO inhibited angiogenesis. However, further investigations are necessary to validate our findings in in vivo experiments.

A general strategy to prepare transition metal sulfide functionalized hierarchically porous carbons with multiple enzyme-like activities for simultaneous electrochemical sensing of ATP metabolites

Decai Zeng,Kai Zhang,Haoran Huang,Xiaohua Zhang,Zongde Wang,Shangxing Chen,Guangbin Liu,Yangping Wen,Peng Wang

A general strategy for the in-situ hydrothermal synthesis of transition metal sulfides functionalized hierarchically porous carbons (FeS 2 -HPC, Co 9 S 8 -HPC, Ni 3 S 2 -HPC, Cu 2 S-HPC, and ZnS-HPC) is proposed using different transition metal chlorides as metal sources and activators, sustainable agroforestry waste biomass ( Citrus sinensis peel sponge layer) as a carbon source, sulfuric acid as a sulfur source, and silica colloid as a hard template. The introduction of both transition metal salts and silica colloids facilitate the adjustment of the pore structure. The added sulfuric acid promotes the degradation of the agroforestry waste biomass, which are conducive to the formation of porous carbons. The in-situ synthesis of transition metal sulfide particles and the sulfur doping improve the conductivity, electrocatalytic and enzyme-like activities of obtained composite materials. The FeS 2 -HPC with multiple enzyme-like activities and the best electrocatalytic activity is applied as a nanozyme electrochemical sensing platform for the simultaneous detection of three ATP metabolites with a limit of detection (LOD) of 0.069 μM for uric acid in a linear range of 0.3–200 μM, a LOD of 0.047 μM for xanthine in 0.3–100 μM, and a LOD of 0.21 μM for hypoxanthine in 0.7–300 μM. This work provides a new idea for the development of transition metal sulfides functionalized porous carbons from agroforestry waste biomass, and a theoretical basis for the fabrication of nanozyme electrochemical sensing platform for health assessment and disease prevention.

Metabolomics, volatolomics, and bioinformatics analyses of the effects of ultra-high pressure pretreatment on taste and flavour parameters of cured Culter alburnus

Dongyin Liu,Mingzhu Zhou,Hongyuan Tan,Guangquan Xiong,Lan Wang,Liu Shi,Chuan Li,Wenjin Wu,Yu Qiao

The effects of ultra-high pressure (UHP) pretreatment (50–250 MPa) on the fish curing were studied. UHP increased the overall volatile compound concentration of cured fish. Among 50–250 MPa five treatment groups, 150 MPa UHP group exhibited the highest total free amino acid content (294.34 mg/100 g) with that of the control group being 92.39 mg/100 g. The activity of cathepsin L was increased under 50–200 MPa UHP treatment (62.28–58.15 U/L), compared with that in the control group (53.80 U/L). UHP treatment resulted in a significant increase in small molecule compounds, especially the amino acid dipeptides and ATP metabolic products. Under UHP treatments, the bacterial phyla Actinobacteriota (1.04–5.25 %), Bacteroidota (0.20–4.47 %), and Deinococcota (0.00–0.05 %) exhibited an increased abundance, and they promoted taste and flavor formation . Our results indicated that UHP is a promising pretreatment method to improve taste and flavour in cured fish by affecting the microorganisms, cathepsin, and proteins.

Bioactive calcium silicate nanowires-containing hydrogel for organoid formation and functionalization

Wenping Ma,Yi Zheng,Guangzhen Yang,Hongjian Zhang,Mingxia Lu,Hongshi Ma,Chengtie Wu,Hongxu Lu

Organoids, which are 3D multicellular constructs, have garnered significant attention in recent years. Existing organoid culture methods predominantly utilize natural and synthetic polymeric hydrogels. This study explored the potential of a composite hydrogel mainly consisting of calcium silicate (CS) nanowires and methacrylated gelatin (GelMA) as a substrate for organoid formation and functionalization, specifically for intestinal and liver organoids. Furthermore, the research delved into the mechanisms by which CS nanowires promote the structure formation and development of organoid. It was discovered that CS nanowires can influence the stiffness of the hydrogel, thereby regulating the expression of the mechanosensory factor Yes-associated protein (YAP). Additionally, the bioactive ions released by CS nanowires in the culture medium could accelerate Wnt/β-catenin signaling, further stimulating organoid development. Moreover, bioactive ions were found to enhance the nutrient absorption and ATP metabolic activity of intestinal organoids. Overall, the CS/GelMA composite hydrogel proves to be a promising substrate for organoid formation and development. This research suggested that inorganic biomaterials hold significant potential in organoid research, offering bioactivities, biosafety, and cost-effectiveness.
