Tumor Microenvironment-Responsive Hydrogel for Direct Extracellular ATP Imaging-Guided Surgical Resection with Clear Boundaries

Jingyi Yang,Jiahao Qu,Xuanming Teng,Weiping Zhu,Yufang Xu,Yangyang Yang,Xuhong Qian

Most solid tumors are clinically treated using surgical resection, and the presence of residual tumor tissues at the surgical margins often determines tumor survival and recurrence. Herein, a hydrogel (Apt-HEX/Cp-BHQ1 Gel, termed AHB Gel) is developed for fluorescence-guided surgical resection. AHB Gel is constructed by tethering a polyacrylamide hydrogel and ATP-responsive aptamers together. It exhibits strong fluorescence under high ATP concentrations corresponding to the TME (100–500 µ m ) but shows little fluorescence at low ATP concentrations (10–100 n m ) such as those in normal tissues. AHB Gel can rapidly (within 3 min) emit fluorescence after exposure to ATP, and the fluorescence signal only occurs at sites exposed to high ATP, resulting in a clear boundary between the ATP-high and ATP-low regions. In vivo, AHB Gel exhibits specific tumor-targeting capacity with no fluorescence response in normal tissue, providing clear tumor boundaries. In addition, AHB Gel has good storage stability, which is conducive to its future clinical application. In summary, AHB Gel is a novel tumor microenvironment-targeted DNA-hybrid hydrogel for ATP-based fluorescence imaging. It can enable the precise imaging of tumor tissues, showing promising application in fluorescence-guided surgeries in the future.

Ratiometric Fluorescent DNA Nanostructure for Mitochondrial ATP Imaging in Living Cells Based on Hybridization Chain Reaction

Lei Luo,Min Wang,Yuan Zhou,Dongliu Xiang,Qing Wang,Jin Huang,Jianbo Liu,Xiaohai Yang,Kemin Wang

For intracellular molecular detection, the appropriate probes should include the abilities to enter target cells noninvasively, target specific sites, and then respond to the analytes reliably. Herein, a ratiometric fluorescent DNA nanostructure (RFDN) was designed for mitochondrial adenosine triphosphate (ATP) imaging in living cells. The DNA nanostructure was constructed by continuous hybridization of two hairpin DNA strands (HS1-Cy3 and HS2-Cy5) under the initiation of the trigger. HS1-Cy3 and HS2-Cy5 contained split aptamer fragments of ATP and are labeled with a fluorescent donor (Cy3) and acceptor (Cy5), respectively. The RFDN integrated multiple split aptamer fragments and increased the local concentration of sensing probes. The binding of ATP to aptamer fragments on the RFDN shortened the distance between Cy3 and Cy5, resulting in obvious ratiometric signals (fluorescence resonance energy transfer). The RFDN showed good biocompatibility and can be internalized into cells in a caveolin-dependent endocytosis pathway. The co-localization imaging results indicated that the DNA nanostructure could target the mitochondria via Cy3 and Cy5. Moreover, the confocal imaging results showed that the intracellular ATP changes stimulated by drugs in living cells could be indicated by the RFDN. In this way, the RFDN is expected to be a simple, flexible, and general platform for chemo/biosensing in living cells.

An Integrated Afterglow and MRI Probe for In Vivo Imaging of ATP in Tumor Metabolism

Huan Du,Fengrong Lv,Renye Yue,Yuzhen Yu,Xinyu Xu,Zhe Li,Baode Chen,Cheng Zhang,Yu Peng,Wei Cheng,Sulai Liu,Xiaobing Zhang,Guosheng Song

Adenosine triphosphate (ATP) is a central mediator of cancer cell metabolism and signaling, driving processes such as proliferation, invasion, metastasis, and therapeutic resistance. Current ATP imaging modalities-including fluorescence, chemiluminescence/bioluminescence, photoacoustic imaging, and magnetic resonance imaging (MRI)-face inherent trade-offs among sensitivity, spatial resolution, and tissue penetration capability. Here, a dual-mode probe is reported that synergistically couples the anatomical resolution of MRI with the high-sensitive, long-lasting signals of afterglow imaging for visualization of ATP in the tumor microenvironment. The probe is constructed via DNA hybridization between an afterglow unit and a T 1 -weighted MRI contrast agent, with an ATP-specific aptamer mediating probe disassembly upon ATP binding. This design generates simultaneous turn-on afterglow and MRI signals, enabling the imaging of intracellular and extracellular ATP fluctuations. Metabolic modulators-coenzyme Q10 (an ATP activator) and 2-deoxy-D-glucose (2-DG, a glycolysis inhibitor)-are applied to demonstrate dynamic, bidirectional tracking of ATP levels in vivo. Furthermore, by monitoring ATP depletion during synergistic metabolic therapy (cisplatin + 2-DG), a correlation between imaging readouts and therapeutic efficacy is established. The work provides anintegrated MRI/afterglow imaging platform for high-contrast, real-time ATP sensing in cancer, offering a powerful tool for elucidating tumor metabolism and evaluating metabolic interventions.
