Electrogenerated Chemiluminescence Resonance Energy Transfer between Ru(bpy)32+ Electrogenerated Chemiluminescence and Gold Nanoparticles/Graphene Oxide Nanocomposites with Graphene Oxide as Coreactant and Its Sensing Application

Yong-Ping Dong,Ying Zhou,Jiao Wang,Jun-Jie Zhu

In the present work, strong anodic electrogenerated chemiluminescence (ECL) of Ru(bpy)32+ was observed at a graphene oxide modified glassy carbon electrode (GO/GCE) in the absence of coreactants. The electrocatalytical effect of GO on the oxidation of Ru(bpy)32+ suggested that GO itself can act as the coreactant of Ru(bpy)32+ ECL, which can be used to fabricate the ECL biosensor. Thiol group terminated adenosine triphosphate (ATP) aptamer was immobilized on the GO film via DNA hybridization. When gold nanoparticles/graphene oxide (AuNPs/GO) nanocomposites were modified on the aptamer through the S–Au bond to form a sandwich-like structure, the ECL resonance energy transfer (ECL-RET) could occur between Ru(bpy)32+ and AuNPs/GO nanocomposites, resulting in an apparent decrease of ECL signal. After the ECL sensor was incubated in ATP solution, the AuNPs/GO nanocomposites were released from the electrode due to the specific interaction between aptamer and ATP, leading to the increased ECL signal. On the basis of these results, an ECL aptasensor was fabricated and could be used in the sensitive and selective detection of ATP in the range of 0.02–200 pM with a detection limit of 6.7 fM (S/N = 3). The proposed ECL aptasensor can be applied in the detection of ATP in real samples with satisfactory results.

Enzyme-responsive aptasensing based on the ATP aptamer and benzothiazole-coumarin G-quadruplex fluorescent probe for label-free detection of adenosine deaminase activity and its inhibitor

Yuxiang Xia,Hongyu Li,Yating Lu,Yanfei Zhang,Kaixin Xue,Jiaheng Zhang,Yujie Zhao,Xingyu Luo

Adenosine deaminase (ADA) is a vital enzyme for regulating the biological process in humans and its abnormal expression level is related to various severe diseases. Nevertheless, conventional approaches for detecting ADA activity are hindered by the drawbacks of complexity and limited detection performances. To address these challenges, we here designed an ADA-responsive aptasensing strategy based on a synthesized benzothiazole-coumarin G-quadruplex (G4) fluorescent probe, which can bind to the G-rich ATP aptamer (G-APA) and form the high-contrast fluorescent complex (S1/G-APA). While the fluorescence of S1/G-APA could be quenched by ATP competition due to the allosteric process, the deamination in ATP catalyzed by ADA would recover the fluorescence of S1/G-APA and allow a facile, turn-on, label-free, and selective detection of ADA activity. This proposed method exhibits favorable detection performances for ADA activity analysis with a wide detection linear range varied from 1 to 100 U/L and a detection limit of 0.82 U/L. Meanwhile, it can be applied for ADA inhibitor (DCF) evaluation and provide a turn-off and sensitive detection performances. Inspired by these detection efficacies, the serum samples spiked with ADA and DCF have been investigated to present the satisfied recovery rates and detection reliability. Collectively, this method will provide a promising way for portable and efficient ADA activity quantification and evaluating the inhibition efficiencies of ADA inhibitors, which hold the potential in promoting ADA-related sensing and pharmaceutical research.

Adenosine triphosphate detection by controlled-release of carboxy fluorescein from mesoporous silica nanoparticles blocked with aptamer-based gold nanoparticles

Ji Xiaoting,Lv Haoyuan,Zhang Wei,Ding Caifeng

In this paper, an adenosine-5′-triphosphate ( ATP ) controlled-release strategy to construct a fluorescence sensing platform has been designed. In the sensing platform, because of ATP aptamer and singlestranded DNA ( ssDNA )-linked mesoporous silica nanoparticles ( Si-MPs ) were hybridized, the pores of Si-MPs were blocked with Au nanoparticles ( AuNPs ) modified with ATP aptamer. Carboxy fluorescein was plugged in channels of Si-MPs. In the presence of target molecule ATP, the ATP aptamer combined with ATP and the AuNPs got away from the pore of the surface of Si-MPs modified by ssDNA. 5-Carboxyfluorescein molecule was released to allow the fluorescent detection. By monitoring the fluorescence at 518 nm, ATP could be quantitatively detected with a detection limit of 6 × 10 –8 M. The linear response range was 6 × 10 –8 to 1 × 10 –6 M. This assay was also able to discriminate ATP from its analogs. The controlled-release aptamer-based biosensor could have an effective application in human breast cancer MCF-7 cells.

A stepwise responsive Au-SS-PEG/Sor/ATPaptamer/LHRH-MPGΔNLS drug delivery vector system for overcoming drug resistance in immunotherapy of hepatocellular carcinoma

Mengting Tong,Guangpeng Chen,Yong Dong,Yubin Pan,Yanan Xue,Da Li

Despite the progress made in novel immunotherapy for hepatocellular carcinoma (HCC), drug resistance remains a challenging problem. In this study, we developed a stepwise nanodrug delivery system, known as Au-SS-PEG/Sor/ATP aptamer /LHRH-MPG ΔNLS , to adapt to the high concentrations of glutathione (GSH) and adenine nucleoside triphosphate/adenosines (ATP/ADO) found in cancer cells and the tumor microenvironment (TME). This system utilizes novel Au nanoclusters conjugated with disulfide-linked PEG as vectors to transport sorafenib (Sor) and an ATP-binding nucleic acid aptamer (ATP apt ). It can enter HCC cells through luteinizing hormone-releasing hormone (LHRH)-MPG ΔNLS (LM). Within the cells, the disulfide bonds of the nanoclusters are cleaved by the high levels of GSH, leading to the release of Sor/ATPapt. This release can be further triggered by ATP/ADO, resulting in a stepwise drug release mechanism. Furthermore, this nanodrug system has exhibited the ability to overcome αPD-1 resistance in HCC tumors. In summary, our novel drug delivery system demonstrates a dramatic anti-HCC effect and holds great potential for treating HCC patients. Synthesis and mechanistic illustration of Au-SS-PEG/Sor/ATP apt /LM nanoclusters for the treatment of hepatocellular carcinoma (HCC).

A Fluorescence Enhancement Sensor Based on Silver Nanoclusters Protected by Rich-G-DNA for ATP Detection

Yuxia Li,Jingxuan Ren,Zeting Meng,Baozhu Zhang

In this study, a turn-on fluorescence sensor for the detection of adenosine 5′-triphosphate (ATP) was developed and tested using ATP-DNA2-Ag NCs. The results showed that the fluorescence of ATP-DNA2-Ag NCs was significantly enhanced with the addition of ATP. The fluorescence enhancement was a result of the specific binding activity of the ATP aptamer and ATP, which caused G-rich sequences to approach the dark DNA-Ag NCs, owing to the alteration in ATP aptamer conformation. The proposed sensor demonstrated a good linear range of 18–42 mM and a limit of detection (LOD) of 2.8 μM. The sensor’s features include sensitivity, selectivity, and simple operation. In addition, the proposed sensor successfully measured ATP in 100-fold diluted fetal bovine serum.

Hemin/G-quadruplex based electrochemical sensor for highly sensitive detection of ATP in fish

Jie Wei,Yumeng Li,Qingyang Si,Qiao Xiao,Qingmin Chen,Tianhui Jiao,Quansheng Chen,Xiaomei Chen

A label-free, sensitive and reliable electrochemical sensor based on hemin/G-quadruplex (G4) DNAzyme was engineered to determine adenosine triphosphate (ATP) in fish. The nucleic acid sequence (DNA1) including the G4 sequence subunit and anti-ATP aptamer segment is anchored on Au electrodes. The nucleic acid sequence (DNA2) is composed of another G4 sequence fragment and anti-ATP aptamer subunit. The probes DNA1 and DNA2 self-assemble into the active hemin/G4 DNAzyme structure on Au electrodes upon exposure to the target ATP. The determination of the target ATP relied on an increase in the catalytic currents occurring on DNAzyme-anchored electrode via the electrochemical reduction of H 2 O 2 . The proposed strategy enables high-performance and specified determination of the target ATP with a detection limit of 0.04 μmol/L. Specifically, the proposed ATP strategy was further implemented to determine ATP in microorganisms in carp samples. Compared with the classical total viable count (TVC) assay, the proposed ATP assay was feasible to assess the freshness of carp, which expands the applications of hemin/G4 DNAzyme and holds promising potential for freshness assessment in food safety.

Cascaded Aptamers-Governed Multistage Drug-Delivery System Based on Biodegradable Envelope-Type Nanovehicle for Targeted Therapy of HER2-Overexpressing Breast Cancer

Tingxizi Liang,Zhigang Yao,Jie Ding,Qianhao Min,Liping Jiang,Jun-Jie Zhu

Tumor-specific therapeutic platforms with improved targeting efficacy and minimized side effect are crucial in cancer therapy. Capitalizing on the recognition capability and biocompatibility of aptamers, we herein designed a multistage targeted drug-delivery system using multiple biodegradable molecules-enveloped nanovehicle that can be employed to efficiently treat human epithelial growth factor receptor (HER2)-overexpressing breast cancer. In this nanovehicle, two aptamers respectively specific to HER2 and ATP were organized in a hierarchical manner. The outmost HER2 aptamer (HB5) governs the recognition to HER2 protein overexpressed in SK-BR-3 cell lines, while the ATP aptamer incorporated with anticancer drug (−)-epigallocatechin gallate (EGCG) and protamine sulfate in the inner core functions as a switch of drug release in response to abundant intracellular ATP. The targeting and drug locker aptamers were cascaded for active targeting effect and stimuli responsiveness, guaranteeing the site-specific drug transportation and endogenous species-triggered drug release inside the tumor cells. Moreover, nanostructured lipid carriers (NLCs) were constructed to wrap and stabilize the loosely bounded ternary complex, minimizing premature drug leakage potentially encountered by the biomolecule assembled nanocarriers. This multiple biomolecules-enveloped nanovehicle demonstrated improved inhibitory actions on tumor growth and minimum side effect to normal organs and tissues both in vitro and in vivo. The presented nanovehicle built from recognition and therapeutic components in a nontoxic framework offered a promising drug-delivery platform with transport precision and biological safety.
