Effects of In Vitro Digestion on the Content and Biological Activity of Polyphenols from Acacia mearnsii Bark

Xiao Chen,Jia Xiong,Lingxiao He,Yu Zhang,Xun Li,Liping Zhang,Fei Wang

The stability and bioaccessibility of polyphenol fromAcacia mearnsiibark were measured at various stages during in vitro simulated digestion. Subsequently, the changes in the total polyphenol content (TPC) and biological activity were studied. The results showed that the phenolic compounds fromA. mearnsiiremained stable, and TPC underwent few changes during gastric digestion. Nonetheless, intestinal digestion led to the degradation of proanthocyanidins (PAs) and a significant decrease in TPC (26%). Degradation was determined by normal-phase HPLC and gel permeation chromatography. Only monomers, dimers, and trimers of flavan-3-ols were identified in the serum-accessible fraction for characterization of their bioaccessibility. The results also indicated the obvious antioxidant capacity of PAs fromA. mearnsiibark, and ~53% of the α-glucosidase–inhibitory effect was preserved. All these findings show that PAs fromA. mearnsiibark as a native plant source may be particularly beneficial for human health as a natural nutritional supplement. Keywords:Acacia mearnsii;proanthocyanidins;in vitro digestion;antioxidant;antidiabetic;HPLC/MS

Pickering Emulsions Stabilized by TEMPO-Oxidized Acacia mearnsii Bark Cellulose for Effective Matrine Adsorption

Lingxiao He,Feiyue Zhang,Ning Ma,Mei Hong,Chaoqun You,Fei Wang

TEMPO-oxidized Acacia mearnsii bark cellulose (OABC), pretreated using deep eutectic solvents (DES), was employed to prepare Pickering emulsions, which served as an efficient adsorbent for matrine. The adsorption of matrine from aqueous solutions was systematically characterized and evaluated using an OABC-stabilized Pickering emulsion (OABCPE). At 298 K, the equilibrium adsorption capacity of matrine on OABCPE was determined to be 44 mg g–1. Furthermore, investigations into the Gibbs free energy, entropy change, and enthalpy increment of the adsorption process indicated that the process is exothermic, spontaneous, and predominantly driven by physisorption. During the first 20 min, a noticeable increase in the adsorption rate is observed, with equilibrium being reached after 60 min. Dynamic adsorption studies revealed that equilibrium could be reached within 200 min, with 70% of the adsorbed matrine being desorbed using 70% alcohol. The adsorption isotherms were fitted to both the Langmuir and Freundlich models. The pseudo-first-order and pseudo-second-order models were employed to analyze the adsorption kinetics. Four breakthrough models (Adams–Bohart, Thomas, Clark, and Yoon–Nelson) were applied to analyze fixed-bed adsorption. The rapid adsorption and facile preparation of OABCPE endow it with significant potential for matrine adsorption and exhibit good stability and reusability under varying industrial conditions.

Enzymatic Acylation of Proanthocyanidin Dimers from Acacia Mearnsii Bark: Effect on Lipophilic and Antioxidant Properties

Dunchi Xiao,Xinzhu Jin,Yuanyuan Song,Yu Zhang,Xun Li,Fei Wang

Proanthocyanin (PA) dimers isolated from Acacia mearnsii bark were enzymatically acylated with palmitic acid as the acyl donor by immobilized Candida antarctica lipase on acrylic resin (Novozym 435). The acylation reaction conditions were optimized by comparing the amount of enzyme, the temperature, the reaction solvents, initial water content, substrate molar ratios and reaction time. The highest acylation conversion of 96.53% was achieved under the follow conditions: PA dimers/palmitic acid at a molar ratio of 1꞉10 in tert-amyl alcohol; initial water content of 5% at 60 °C for 12 h with 30 g/L enzyme dosage. Introducing palmitic acid into PA dimers significantly improved both the lipophilicity and antioxidant properties. The 1-octanol/water partition coefficient of the PA dimers and their derivatives showed that the lipophilicity of the derivatives were 2.4 times higher than that of the PA dimers. The derivatives exhibited strong antioxidant scavenging capacities, approximately 1.6 times greater than the original dimers. This work is of great significance to expand the application of natural PA dimers in cosmetic and food industries and also lay a foundation for the high value-added utilization of A. mearnsii .

Analytical Profiling of Proanthocyanidins from Acacia mearnsii Bark and In Vitro Assessment of Antioxidant and Antidiabetic Potential

Xiao Chen,Jia Xiong,Shenlin Huang,Xun Li,Yu Zhang,Liping Zhang,Fei Wang

The proanthocyanidins from ethanol extracts (80%,v/v) ofAcacia mearnsii(A. mearnsii) bark on chemical-based and cellular antioxidant activity assays as well as carbolytic enzyme inhibitory activities were studied. About 77% of oligomeric proanthocyanidins in ethanol extracts ofA. mearnsiibark were found by using normal-phase HPLC. In addition, HPLC-ESI-TOF/MS and MALDI-TOF/TOF MS analyses indicated that proanthocyanidins fromA. mearnsiibark exhibited with a degree of polymerization ranging from 1 to 11. These results of combined antioxidant activity assays, as well as carbolytic enzyme inhibitory activities of proanthocyanidins fromA. mearnsiibark, indicated an encouraging antioxidant capacity for the high polyphenol content and a potential for use as alternative drugs for lowering the glycemic response.Keywords:proanthocyanidins;HPLC/MS;MALDI-TOF/MS;antidiabetic;antioxidant;degree of polymerization
