Glucose-regulated protein 78 ( grp78 ) and activating transcription factor 6α ( atf6α ) are considered vital endoplasmic reticulum (ER) molecular chaperones and ER stress (ERS) sensors, respectively. In the present study, the full cDNA sequences of these two ERS-related genes were first cloned and characterized from black seabream ( Acanthopagrus schlegelii ). The grp78 cDNA sequence is 2606 base pair (bp) encoding a protein of 654 amino acids (aa). The atf6α cDNA sequence is 2168 base pair (bp) encoding a protein of 645 aa. The predicted aa sequences of A. schlegelii grp78 and atf6α indicated that the proteins contain all the structural features, which were characteristic of the two genes in other species. Tissues transcript abundance analysis revealed that the mRNAs of grp78 and atf6α were expressed in all measured tissues, but the highest expression of these two genes was all recorded in the gill followed by liver/ brain. Moreover, in vivo experiment found that fish intake of a high lipid diet (HLD) can trigger ERS by activating grp78 /Grp78 and atf6α/ Atf6α. However, it can be alleviated by dietary betaine supplementation, similar results were also obtained by in vitro experiment using primary hepatocytes of A. schlegelii . These findings will be beneficial for us to evaluate the regulator effects of HLD supplemented with betaine on ERS at the molecular level, and thus provide some novel insights into the functions of betaine in marine fish fed with an HLD.

Alkaline stress is an important environmental factor in saline-alkaline aquaculture, yet its effects on marine euryhaline fish remain poorly understood. In this study, a multi-timepoint acute alkaline stress model was established to investigate hepatic responses in black sea bream ( Acanthopagrus schlegelii ). Fish in both control and treatment groups were maintained at a consistent salinity of 18‰, while the treatment group was exposed to 30 mmol/L NaHCO₃. Liver samples were collected at 0, 6, 12, 24, and 48 h post-exposure for histological, biochemical, and transcriptomic analyses. Alkaline stress induced progressive liver injury, characterized by hepatocellular ballooning, steatosis, and significant increases in ALT and AST activities. Oxidative stress and inflammatory responses were markedly activated, as indicated by altered redox balance, increased lipid peroxidation, and upregulation of pro-inflammatory cytokines. Transcriptomic analysis revealed dynamic changes in gene expression, with significant enrichment of pathways related to metabolism, immune response, and ferroptosis. Notably, ferroptosis-related pathways were consistently enriched, with marked alterations in ACSL4 and GPX4 protein abundance suggesting that ferroptosis-related pathways may be involved inalkaline-induced liver injury. These findings provide new insights into the molecular mechanisms linking oxidative stress, inflammation, and ferroptosis-related responses during alkaline stress adaptation in marine euryhaline fish.

Taurine is a component that tends to be deficient in fish diets that contain higher plant-derivative proteins. Acanthopagrus schlegelii juveniles (IBW − 11.28 g) were fed nine diets containing SPC levels (SPC40, SPC60, SPC80 and SPC100), or supplemented with 5 g/kg of taurine at each replacement level. Our results showed higher WGR and SGR in SPC60 + TAU (WGR; 224.67, SGR; 2.10) followed by SPC40 + TAU (WGR; 221.75, SGR; 2.09). Phosphorus retention efficiency was significantly raised as the SPC increased when compared with the control diet. Taurine addition showed an incline in the nitrogen retention efficiency. An effect was observed within FR between fish and the SPC content. ADCs of dry matter, crude lipid and phosphorous were affected by SPC, while taurine addition affected only crude lipid. A significant effect was seen in ash and phosphorus by the SPC and taurine. TAU supplementation showed an increased T-AOC activity in the SPC80 + TAU (1.90), while malondialdehyde and catalase activity were reduced except in completely replaced fish meal groups. It could be concluded that black sea bream can accept the substitution of FM with 60% of SPC while at the inclusion of dietary TAU (5 g/kg) growth could be improved.

Liver is considered the critical tissue for metabolic processing of dietary nutrients including lipids. High-fat diets (HFD) can induce hepatic lipotoxic injury, but the mechanism is unclear. This study aimed to elucidate the underlying role of endoplasmic reticulum stress (ERS) in mediating HFD-induced hepatic lipotoxic injury in black seabream ( Acanthopagrus schlegelii ) via in vitro and in vivo experiments. For the in vitro experiment, isolated hepatocytes from black seabream were incubated for 24 h in medium (Control), or medium supplemented with 200 μM oleic acid (OA) alone or in combination with 20 μM 4-phenyl butyric acid (OA + 4-PBA). In the in vivo experiment, black seabream juveniles (3.30 ± 0.10 g) were fed diets containing lipid at 12.4% (Control) or 18.6% (HFD) for 8 weeks, after which the HFD-fed fish were divided into three groups and, on alternate days over a period of 8 days, subjected to intraperitoneal injections of 50 mg/kg 4-PBA in hydroxypropyl-β-cyclodextrin (HBC) (treatment 4-PBA) or carrier alone (treatment HFD + HBC) versus no injection (HFD). The contents of triglyceride and non-esterified fatty acid, as well as expression levels of genes and proteins involved in lipid metabolism, ERS, apoptosis and inflammation were determined in liver and isolated hepatocytes, and oil-red O staining, ultrastructural observation, flow cytometry assay of apoptosis rate, and terminal deoxynucleotidyl transferase-mediated dUTP-biotin nick end labeling assay (TUNEL) staining were performed on hepatocytes. The in vitro experiment showed that 4-PBA treatment significantly decreased lipid accumulation, and the OA-induced expression levels of genes/proteins linked to ERS, lipogenesis, inflammation and apoptosis in hepatocytes. Transmission Electron Microscope (TEM) imaging showed that 4-PBA treatment mitigated the ER swelling induced by OA. These results were confirmed in the in vivo experiment, where 4-PBA injection reduced lipid deposition and expression levels of genes/proteins related to ERS, lipogenesis, inflammation and apoptosis in liver compared to the HFD treatment. In conclusion, these results confirmed that ERS plays a contributing regulatory role in HFD- and OA-induced lipotoxic damage including lipid accumulation, apoptosis and inflammation in liver and isolated hepatocytes of black seabream, which supply a new perspective in the cross-talk of ERS and hepatic lipotoxic damage in hepatocytes/liver of marine fish and provide theoretical guidance for the prevention and treatment of hepatic lipotoxic damage.

The present study aimed to reveal the role of inositol-requiring enzyme 1α (Ire1α) in mediating high-fat-diet (HFD) induced inflammation and apoptosis in fish and elucidate underling mechanisms of action. In experiment 1, black seabream juveniles were fed a control diet (Control, 12 % dietary lipid) or a high fat diet (HFD, 19 % dietary lipid) for eight weeks. In experiment 2, primary hepatocytes were isolated from black seabream juveniles and treated with oleic acid (OA, 200 μmol/L), OA + transfection with non-silencing control siRNA (negative control) (OA + NC), and OA + transfection with ire1α -small interfering RNA (OA + si ire1α ) for 48 h versus untreated (Control). Results indicated that fish fed HFD increased lipid deposition in the liver and caused hepatic steatosis. HFD group had significantly higher ire1α /Ire1α mRNA and phosphorylated protein expression and endoplasmic reticulum stress (ERS) related genes expression compared to the Control group, indicating that ERS was triggered. Meanwhile, feeding HFD induced inflammation and apoptosis by evaluated nuclear factor kappa B ( nf-κb ) mRNA and phosphorylated Nf-κb p65 protein expression, and c-Jun N-terminal kinase (jnk ) mRNA and protein expression. However, knock down of ire1α (OA + si ire1α ) in primary hepatocytes alleviated OA-induced increased expression of ire1α /Ire1α mRNA and protein expression, nf-κb /Nf-κb p65 mRNA and phosphorylated protein expression, and jnk /Jnk mRNA and phosphorylated protein expression. These findings revealed the underling mechanism of action of HFD in fish, confirming that HFD increased ESR stress and Ire1α that, in turn, activated Nf-κb and Jnk pathways in hepatocytes and liver mediating HFD-induced inflammation and apoptosis.
