To explore novel terpenoid repellents, 22 candidate terpenoid derivatives were synthesized and tested for their electroantennogram (EAG) responses and repellent activities against Aedes albopictus. The results from the EAG experiments revealed that 5-(2-hydroxypropan-2-yl)-2-methylcyclohex-2-en-1-yl formate (compound 1) induced distinct EAG responses in female Aedes albopictus. At concentrations of 0.1, 1, 10, 100, and 1000 mg/L, the EAG response values for compound 1 were 179.59, 183.99, 190.38, 193.80, and 196.66 mV, demonstrating comparable or superior effectiveness to DEET. Repellent activity analysis indicated significant repellent activity for compound 1, closest to the positive control DEET. The in silico assessment of the ADMET profile of compound 1 indicates that it successfully passed the ADMET evaluation. Molecular docking studies exhibited favourable binding of compound 1 to the active site of the odorant binding protein (OBP) of Aedes albopictus, involving hydrophobic forces and hydrogen bond interactions with residues in the OBP pocket. The QSAR model highlighted the influential role of hydrogen-bonding receptors, positively charged surface area of weighted atoms, polarity parameters of molecules, and maximum nuclear-nuclear repulsion force of carbon-carbon bonds on the relative EAG response values of the tested compounds. This study holds substantial significance for the advancement of new terpenoid repellents.

The Asian tiger mosquito Aedes albopictus transmits several arboviruses, including chikungunya, Zika, and dengue, posing serious public health threats. The excessive use of insecticides has led to increasing resistance to commonly applied insecticides in this species. We previously observed that enhanced expression of a CYP6Z8 homolog (AalbCYP6Z8) was associated with insecticide resistance in two field populations of Ae. albopictus ; however, its capacity in insecticide metabolism had not been biochemically characterized. In this study, to explore the potential role of AalbCYP6Z8 in insecticide metabolism, a fully functional enzyme system was reconstituted in vitro by using Escherichia coli –produced recombinant AalbCYP6Z8 and NADPH-dependent cytochrome P450 reductase along with cytochrome b 5. Our metabolism assays showed that CYP6Z8 was capable of metabolizing organophosphates (temephos and malathion) and carbamates (propoxur and bendiocarb), but not pyrethroids (deltamethrin and permethrin). Mass spectrometry analyses tentatively identified the structures of specific metabolites, revealing that AalbCYP6Z8 transforms organophosphates through oxidative desulfurization and carbamates through dealkylation and hydroxylation. This work demonstrates that AalbCYP6Z8 is a versatile metabolizer of acetylcholinesterase-inhibiting insecticides.

BACKGROUND The use of safe and effective insecticides against mosquito-borne stocks is still one of the most effective ways to rapidly interrupt the spread of diseases such as malaria, dengue etc., but many insects are resistant to most insecticides and most of the traditional pyrethroids (like permethrin, deltamethrin etc.) contain phenoxy groups so it is easy to form dibenzofuran, a pollutant, after photodegradation. It is therefore necessary to synthesize novel pyrethroids which have better mosquito-killing activity, poor resistance to mosquitoes, and no dibenzofuran formation during photodegradation. RESULTS A series of novel dihaloacetylated heterocyclic pyrethroids were synthesized to enhance antimosquito activity. Bioassays based on the guidelines of the Chinese Center for Disease Control and Prevention (CDC) indicate that the synthesized compounds DCA-O (LC 50 = 2.95 ± 0.05 μg L −1 ), DCA-01 (LC 50 = 5.83 ± 0.36 μg L −1 ) and DCA-11 (LC 50 = 5.79 ± 1.51 μg L −1 ) exhibit high insecticidal activities and sensitivity against Aedes albopictus while deltamethrin and permethrin produced moderate levels of resistance. Moreover, the analysis of the photodegradation implies that the novel compounds synthesized by substituting a phenoxy group with a heterocyclic ring do not form dibenzofuran, a pollutant. Lastly, the cytotoxicity data of pyrethroids on human dopaminergic neuroblastoma SH-SY5Y cells confirm that the synthesized compounds have low toxicity for the cells. CONCLUSION Novel dihaloacetylated heterocyclic pyrethroids, made by simple synthesis steps, have better antimosquito activity, lower drug resistance, are less polluting to the environment and show low toxicity to human nerve cells. This also provides an effective idea for the synthesis of pyrethroids. Introduction Aedes albopictus is the primary vector of major arboviral diseases such as dengue fever, chikungunya fever, and Zika virus disease, and its control is highly dependent on chemical insecticides. However, the long-term use of pyrethroid insecticides has led to the development of insecticide resistance in Ae. albopictus , which severely undermines the efficacy of vector control programs. Methods Ae. albopictus populations were collected from five sites in Guangdong and Hainan provinces, China. Beta-cypermethrin resistance levels were determined via bioassays, with resistance ratios at the median lethal concentration (RR 50 ) calculated. Target-site resistance was evaluated via kdr mutation detection in the voltage-gated sodium channel (VGSC) gene. Transcriptome sequencing identified differentially expressed genes (DEGs), and 16S rRNA sequencing characterized gut microbiome alterations. Correlation analysis and Cedecea neteri dietary supplementation assays verified the role of gut microbiota in resistance. Results The results showed that all four populations (except the CP population) exhibited varying degrees of resistance to beta-cypermethrin, with resistance ratios at the median lethal concentration (RR 50 ) ranging from 2.84 to 29.18. Detection of kdr mutations revealed three mutations (F1534C, F1534L, F1534S) at codon 1534 of the voltage-gated sodium channel (VGSC) gene in all field populations, with mutation frequencies ranging from 49.4% to 100.0%, and a low-frequency V1016G mutation at codon 1016. Transcriptome analysis identified a total of 2,566 commonly upregulated genes and 994 commonly downregulated genes across the resistant populations. Gut microbiome analysis revealed a significant alteration in the intestinal microbial community structure of resistant populations; specifically, the relative abundance of the genus Cedecea differed significantly between resistant and susceptible populations and correlated strongly with the expression of most differentially expressed genes. Furthermore, dietary supplementation with Cedecea neteri significantly increased the survival rate of Ae. albopictus exposed to β-cypermethrin (73.86% vs 40.00%; P < 0.0001). Discussion From the perspectives of target-site mutations, gene expression regulation, and gut microbe interactions, this study providing a foundation for further studies on resistance mechanisms in Ae. albopictus , thereby providing a theoretical foundation for further dissection of resistance mechanisms and optimization of vector control strategies.

A large number of studies have shown that matrine (MA) possesses various pharmacological activities and is one of the few natural, plant-derived pesticides with the highest prospects for promotion and application. Fifty-eightMAderivatives were prepared, including 10 intermediates and 48 target compounds in 3 series, to develop novel mosquitocidal agents. Compounds4b,4e,4f,4m,4n,6e,6k,6m, and6oshowed good larvicidal activity againstAedes albopictus, which is both a highly aggressive mosquito and an important viral vector that can transmit a wide range of pathogens. Dipping methods and a bottle bioassay were used for insecticidal activity evaluation. The LC50values of4e,4m, and6mreached 147.65, 140.08, and 205.79 μg/mL, respectively, whereas the LC50value ofMAwas 659.34 μg/mL. Structure–activity relationship analysis demonstrated that larvicidal activity could be improved by the unsaturated heterocyclic groups introduced into the carboxyl group after opening the D ring. TheMAderivatives with oxidized N-1 lost their mosquitocidal activities, indicating that the bareness of N-1 is crucial to maintain their anti-mosquito activity. However, the activity was not greatly influenced by introducing a cyan group at C-6 or a benzene sulfonyl group at N-16. Additionally, compounds4eand4mexhibited good inhibitory activities against acetylcholinesterase with inhibitory rates of 59.12% and 54.30%, respectively, at a concentration of 250 μg/mL, whereas the inhibitory rate ofMAwas 9.88%. Therefore, the structural modification and mosquitocidal activity ofMAand its derivatives obtained here pave the way for those seeking strong mosquitocidal agents of plant origin.

Aedes albopictus is a primary vector of arboviral diseases such as dengue fever, chikungunya fever, and Zika. The rapid development of insecticide resistance is becoming a major challenge to sustainability of insecticide-based interventions for the control of Ae. albopictus borne arboviral diseases. Understanding the molecular basis of resistance is useful for developing effective insecticide-based control action. Our recent study showed that overexpression of the AalbCYP9J26 gene was associated with insecticide resistance in field Ae. albopictus populations in Beijing, but its contribution to insecticide metabolism and resistance remains unknown. In this study, the metabolic capacity of AalbCYP9J26 towards six insecticides was examined using an enzyme system reconstituted with Escherichia coli -produced AalbCYP9J26 protein and its redox partners (AalbCPR and Aalbb5). The results showed that AalbCYP9J26 was able to metabolize temephos, malathion, propoxur, permethrin, and deltamethrin, while no significant activity against bendiocarb was observed. Moreover, the Drosophila GAL4/UAS transgenic system confirmed that enhanced expression of AalbCYP9J26 conferred increased tolerance to malathion, permethrin, deltamethrin, and propoxur. This study demonstrates that AalbCYP9J26 plays a critical role in insecticide metabolism and resistance, suggesting AalbCYP9J26 may serve as a valuable biomarker for monitoring insecticide resistance and a target for new insecticidal agents.

The potential for essential oils (EOs) to act as a natural alternative for controlling mosquito populations has aroused extensive interest in recent years. Four EOs ( Thymus vulgaris (thyme), Foeniculum vulgare (fennel), Citrus medica (lime), and Camellia sinensis (green tea)) were selected in our previous larvicidal screening assays against Aedes albopictus . In this study, the active constituents of each EO were identified. Next, their acetylcholinesterase (AChE) inhibitory activity was evaluated, and the binding mode was studied by molecular docking. To improve EO stability and duration of activity, nanoemulsions were prepared and characterized. Thirteen constituents of the four EOs at the 100 ppm level led to 100% mortality of the larvae, the four most active constituents being α -hexyl cinnamaldehyde (LC 50 = 24.0 ± 0.3 ppm), limonene (LC 50 = 34.1 ± 0.7 ppm), α -terpinene (LC 50 = 50.0 ± 0.5 ppm), and carvacrol (LC 50 = 54.7 ± 0.6 ppm). These compounds, with the exception of α -hexyl cinnamaldehyde, also showed potent AChE inhibitory activity. Nanoemulsions with droplet size of 56.8–224.9 nm were prepared by emulsification at a 1:1 (v/v) ratio of EO and Tween 80. These EO nanoemulsions showed equally potent larvicidal activity as EOs themselves; moreover, they showed better stability and were active for longer time. These results suggest that natural EOs and their constituents have potential as larvicides for mosquito vector control and that nanoemulsification is an effective method for improving their physicochemical properties and efficacy.

Dengue virus, one of the most important mosquito-borne viruses, has shown a sharp upward trend, spreading around the world in recent years. Control of vectorsAedes aegyptiandAe. albopictusremains crucial for blocking dengue transmission. The lethal ovitrap (LO) is one of the cost-effective traps based on the classic “lure and kill” strategy, and finding a proper long-lasting effective toxin is key to achieving the desired effect. The concentration of inorganic salts of habitat environment plays a strong role in affecting oviposition, hatching, and development of mosquitoes, but the potential insecticide activity of Sodium Chloride (NaCl) in habitat water as well as LO still lacks research. In this study, we carried out laboratory experiments to systematically explore the effects of different concentrations of NaCl solutions on oviposition, egg hatching, and larval development ofAe. albopictus. Consequently,Ae. albopictuswas found to prefer freshwater to lay eggs; whereas 48.8 ± 2.6% eggs were laid in freshwater and 20% in ≥1.0% brackish water, few eggs were laid in 3.0% NaCl solution. Compared with egg hatching, larval development ofAe. albopictuspresented a higher sensibility to NaCl concentration. The mortality of the 3rd–4th larvae in 1.0% NaCl solution was 83.8 ± 8.7%, while in 3.0% it reached 100%. Considering the cumulative effect of NaCl, when NaCl concentration was ≥1.0%, no eggs could successfully develop into adults. These data suggested that NaCl solutions with a concentration ≥1.0% can be used as an effective cheap insecticide forAe. albopictusin subtropical inland aquatic habitats, and also as the “kill” toxin in LOs. Meanwhile, the concentration range from 0 to 2.0% of NaCl solution has the potential to be used as the “lure” in LOs. The technological processes of how to use NaCl as insecticide or in LOs still needs further in-depth exploration. Keywords:Aedes albopictus;habitat;Sodium Chloride (NaCl);dengue;50% lethal concentration

Aedes albopictus (Skuse) is a vector of several arboviruses, such as dengue, chikungunya, West Nile, and Zika viruses. At present, the use of synthetic insecticides is the main vector control strategy. However, the widespread and long-term use of insecticides has aroused several problems, including insecticide resistance, environmental pollution, and non-target species effects, thereby encouraging researchers to search for new alternatives derived from natural products. In recent decades, essential oils (EOs) as natural alternatives to control mosquitoes have received increasing attention. In the initial larvicidal activity screen, two Rutaceae plants (Citrus aurantium and Citrus paradisi) EOs were selected and evaluated for killing Ae. albopictus larvae. The LC50 values of C. aurantium and C. paradisi EOs against Ae. albopictus were 91.7 and 100.9 ppm, respectively. The main components of C. aurantium EO include diethyl o-phthalate (37.32%), limonene (10.04%), and methyl dihydrojasmonate (6.48%). The main components of C. paradisi EO include limonene (60.51%), diethyl o-phthalate (11.75%), linalool (7.90%), and styralyl acetate (6.28%). Among these main components of the two EOs, limonene showed potent larvicidal activity, with the LC50 value of 39.7 ppm. The nanoemulsions of limonene were prepared and characterized. The duration of larvicidal activity was greater in the limonene nanoemulsions than when limonene was applied in solvent. This study demonstrates that EOs of plants in family Rutaceae are a potential resource to develop new larvicides, and nanoemulsification is an effective method for improving the physicochemical properties and efficacy of natural products as larvicides.

Simple SummaryDue to the environment and human health concerns of synthetic repellents, essential oils (EOs) as natural alternatives have received increased attention. In this study, the repellent activity of 60 commercial EOs againstAedes albopictuswas evaluated. In the initial screening, 8 active EOs including cinnamon, marjoram, lemongrass, bay, chamomile, jasmine, peppermint2, and thyme were selected. Twenty-one major constituents (>5% relative area) in the 8 active EOs were identified via gas chromatography-mass spectrometry (GC-MS) analysis. Cinnamaldehyde, citral and terpinene-4-ol displayed the highest repellent activity with more than 60% RR, which were more active thanN,N-diethyl-3-methyl benzoyl amide (DEET). Next, their nanoemulsions were prepared and characterized. In the arm-in-cage assay, cinnamaldehyde- and citral-based nanoemulsions have prolonged mosquito protection time compared with their normal solutions.AbstractMosquitoes are one of the most important disease vectors from a medical viewpoint in that they transmit several diseases such as malaria, filariasis, yellow and Dengue fever. Mosquito vector control and personal protection from mosquito bites are currently the most efficient ways to prevent these diseases. Several synthetic repellents such as DEET, ethyl butylacetylaminopropionate (IR3535) and 1-(1-methylpropoxycarbonyl)-2-(2-hydroxyethyl)piperidine) (Picaridin), have been widely used to prevent humans from receiving mosquito bites. However, the use of synthetic repellents has raised several environment and health concerns. Therefore, essential oils (EOs) as natural alternatives receive our attention. In order to discover highly effective mosquito repellents from natural sources, the repellent activity of 60 commercial EOs againstAe. albopictuswas screened in this study. Eight EOs including cinnamon, marjoram, lemongrass, bay, chamomile, jasmine, peppermint2, and thyme, showed a suitable repellent rate (>40%) at the tested dose of 10 μg/cm2. Then, their main constituents were analyzed by GC-MS, and the active constituents were identified. The most active compounds including cinnamaldehyde, citral and terpinen-4-ol, exhibited an 82%, 65% and 60% repellent rate, respectively. Moreover, the nanoemulsions of the three active compounds were prepared and characterized. In the arm-in-cage assay, the protection times of the nanoemulsions of cinnamaldehyde and citral were significantly extended compared with their normal solutions. This study provides several lead compounds to develop new mosquito repellents, and it suggests that nanoemulsification is an effective method for improving the duration of the activity of natural repellents.

Simple SummaryThe essential oil (EO) ofCitrus reticulataBlanco cv. Chachiensis (Chachi) exhibited significant larvicidal activity. The EO from semi-mature fruits and its main components were able to control mosquitoes by effectively inhibiting acetylcholinesterase and three detoxification enzymes and significantly altering the diversity of the internal microbiota in mosquitoes.AbstractThe overuse of synthetic insecticides has led to various negative consequences, including insecticide resistance, environmental pollution, and harm to public health. This may be ameliorated by using insecticides derived from botanical sources. The primary objective of this study was to evaluate the anti-mosquito activity of the essential oil (EO) ofCitrus reticulataBlanco cv. Chachiensis (Chachi) (referred to as CRB) at immature, semi-mature, and mature stages. The chemical compositions of the CRB EO were analyzed using GC-MS. The main components were identified to be D-limonene andγ-terpinene. The contents of D-limonene at the immature, semi-mature, and mature stages were 62.35%, 76.72%, and 73.15%, respectively; the corresponding contents ofγ-terpinene were 14.26%, 11.04%, and 11.27%, respectively. In addition, the corresponding contents of a characteristic component, methyl 2-aminobenzoate, were 4.95%, 1.93%, and 2.15%, respectively. CRB EO exhibited significant larvicidal activity againstAedes albopictus(Ae. albopictus, Diptera: Culicidae), with the 50% lethal doses being 65.32, 61.47, and 65.91 mg/L for immature, semi-mature, and mature CRB EO, respectively. CRB EO was able to inhibit acetylcholinesterase and three detoxification enzymes, significantly reduce the diversity of internal microbiota in mosquitoes, and decrease the relative abundance of core species within the microbiota. The present results may provide novel insights into the utilization of plant-derived essential oils in anti-mosquitoes.
