Aeromonas is a ubiquitous aquatic bacteria, and it is a significant factor contributing to meat spoilage during processing and consumption. The abilities of Aeromonas salmonicida 29 and 57, which exhibit spoilage heterogeneity, to secrete protease, lipase, hemolysin, gelatinase, amylase, and lecithinase were confirmed by plate method. A total of 3948 proteins were identified by ITRAQ in extracellular secretions of A. salmonicida , and 16 proteases were found to be potentially related to spoilage ability. The complete genome sequence of A. salmonicida 57 consists of one circular chromosome and three plasmids, while A. salmonicida 29 consists of one circular chromosome, without a plasmid. Transcriptomic analysis revealed a significant number of DEGs were up-regulated in A. salmonicida 29, which were mainly enriched in metabolic pathways (e.g., amino acid metabolism, carbohydrate metabolism), indicating that A. salmonicida 29 had better potential to decompose and utilize nutrients in meat. Six protease genes (2 pepB , hap , pepA , ftsI , and pepD ) were excavated by combined ITRAQ with transcriptome analysis, which potentially contribute to bacterial spoilage ability and exhibit universality among other dominant spoilage bacteria. This investigation provides new insights and evidence for elucidating metabolic and spoilage phenotypic differences and provides candidate genes and strategies for future prevention and control technology development.

Lactic acid bacteriaLigilactobacillus salivariushas been shown to be a substitute for antibiotics in the treatment of bacterial disease in high animals. However, its beneficial mechanism in fish farming is still unclear. This study evaluated the antagonistic effects of theLigilactobacillus salivariusGX118 strain onAeromonas salmonicidaand its regulation of rainbow trout growth in vivo and in vitro. The results found that GX118 produces an antibacterial substance that can directly destroy the cell wall ofA. salmonicida. Whole-genome sequencing of GX118 revealed that Enterolysin A is a type III bacteriocin with antibacterial properties. An in vivo experiment showed that the supplementation of GX118 in diet competitively inhibited the colonization ofA. salmonicidain the intestine. In addition, it was able to improve the growth performance of rainbow trout within a 21-day feeding experiment. The supplementation of GX118 increased the diversity of gut microbiota, in which the abundance ofBacteroidota,Blautia, andRhodobacteraceaeincreased. In addition, the use of GX118 activated the expression ofIFN-γandNF-κBgenes and reduced the expression level ofIL-6andIL-8, thus exhibiting a certain effect on activating the immunity of rainbow trout. This study provides a scientific basis for the development of antibacterial probiotics in the healthy farming of rainbow trout.

TheAeromonas salmonicidais responsible for causing furunculosis in various fish species. Furunculosis is a ubiquitous disease that affects the aquaculture industry and causes the mass mortality of turbot.Vibrio vulnificusis a pathogen that causes skin ulcers and hemorrhagic septicemia in fish, resulting in significant mortality in aquaculture. In this study, we have established a bivalent inactivated vaccine againstA. salmonicidaandV. vulnificuswith Montanide™ ISA 763 AVG as an adjuvant. This bivalent inactivated vaccine was used to immunize turbot by intraperitoneal injection, and the relevant immune indexes were detected. The results demonstrate that the bivalent inactivated vaccine exhibited a relative percent survival (RPS) of 77% followingA. salmonicidaandV. vulnificusintraperitoneal challenge. The vaccinated group exhibited higher levels of acid phosphatase activity and lysozyme activity compared to the control group. ELISA results showed a significant increase in serum antibody levels in immunized turbot, which was positively correlated with immunity. In the kidney tissue, related immune genes (TLR5,CD4,MHCIandMHCII) were up-regulated significantly, showing that the vaccine can induce cellular and humoral immune responses in turbot. In conclusion, the bivalent inactivated vaccine againstA. salmonicidaandV. vulnificuswas immunogenic, efficiently preventing turbot from infection, which has the potential to be applied in aquaculture.

Microbial spoilage of meat products is a significant problem in the food industry. Aeromonas salmonicida is a significant microorganism responsible for spoilage in chilled meat. Its effector protein, hemagglutinin protease (Hap), has been identified as an effective substance for degrading meat proteins. The ability of Hap to hydrolyze myofibrillar proteins (MPs) in vitro demonstrated that Hap has obvious proteolytic activity, which could alter MPs’ tertiary structure, secondary structure, and sulfhydryl groups. Moreover, Hap could significantly degrade MPs, focusing primarily on myosin heavy chain (MHC) and actin. Active site analysis and molecular docking revealed that the active center of Hap was bound to MPs via hydrophobic interaction and hydrogen bonding. It may preferentially cleave peptide bonds between Gly44-Val45 in actin, and Ala825-Phe826 in MHC. These findings suggest that Hap may be involved in the spoilage mechanism of microorganisms and provide crucial insights into the mechanisms of meat spoilage induced by bacteria.

An inulin-aluminum composite adjuvant achieved 61.54% RPS against A. salmonicida. • It enhanced lysozyme, total protein, and specific IgM vs. single adjuvants. • It promoted antigen uptake and upregulated Th1-type cytokines and T cell markers. • This adjuvant offers a safe, effective strategy for fish vaccine development. Inulin is a type of plant polysaccharide that is widely distributed in nature. Research has demonstrated that inulin possesses adjuvant activity, stimulating balanced cellular and humoral immune responses. However, the role of inulin as an adjuvant has been relatively underexplored within aquatic vaccine research. In this study, a composite aluminum adjuvant containing inulin was developed, and an inactivated vaccine against Aeromonas salmonicida was formulated based on this adjuvant. Subsequently, the immunoprotective effects of the vaccine in turbot were evaluated to assess the potential application of inulin as an adjuvant in aquaculture vaccines. As a result, the composite of aluminum and inulin significantly promoted immune cell recruitment and antigen uptake at the injection site. Vaccination with the composite adjuvant significantly reduced mortality following A. salmonicida challenge, achieving a relative percent survival (RPS) of 61.54% compared to the control group. Moreover, the vaccine exhibited increased levels of specific IgM along with elevated serum protein levels and lysozyme activity. Furthermore, gene expression analysis revealed significant upregulation of MHC-I , MHC-IIβ , CD4 , CD8 as well as some cytokines including IL-4 , IL-10 , IL-1β , IFN-γ , IL-12 and TNF-α , indicating a robust humoral and cellular immune response. In brief, these findings suggest that inulin acts as a natural immune stimulant and, when combined with aluminum, confers enhanced protective efficacy compared with either adjuvant alone, while balancing both humoral and cellular immune responses. Further studies are needed to clarify the nature of this complementary effect. This novel composite of adjuvants presents a leading candidate strategy for improving fish vaccination practices within aquaculture aimed at bolstering disease prevention efforts. Further studies are warranted to optimize this formulation and investigate the mechanisms underlying immune enhancement provided by the composite adjuvant system for teleost fish.

Aeromonasspecies are among the main pathogens causing rainbow trout infections. Silver nanoparticles (AgNPs) have a broad spectrum of antimicrobial properties and are usually produced by various green-synthesis methods. However, the application of commercialized AgNPs has not fully been clarified. Thus, the objective of this study was to evaluate the antibacterial activities of commercialized AgNPs (range of sizes 10–12 nm) on two contrastingA.salmonicidastrains (I-1 and I-4), isolated from rainbow trout; the antibacterial mechanism, histopathological alterations and the expression of immune-related genes were investigated. In vitro, the minimal inhibitory concentration (MIC) was 10 µg/mL for I-1, and lowered to 9.5 µg/mL for I-4, respectively. AgNPs were shown to disrupt both the cell wall and membrane of I-1 and I-4, resulting in cell lysis and degradation. In vivo, rainbow trout challenged by immersed or intraperitoneally injected infection, the 10 µg/mL AgNP-treated groups, both showed delayed deaths and lower mortalities compared to the control groups, without any clinical signs and pathological changes. Especially for the virulent I-4, the enhanced expressions of immune-related genes TNF-α, IL-1β, IL-10 and IL-11 were significantly reduced in the AgNP-treated group, indicating a lesser inflammation due to the application of AgNPs. This study would lay theoretical foundation for the wide application of silver nanoparticles in fish diseases.
