Aeromonas hydrophila and A. veronii are widespread and important critical pathogenic bacteria in the aquaculture industry and cause severe economic damage. At present, magnolol has been proved to be a broad-spectrum antibacterial activity, such as A. hydrophila , Staphylococcus aureus and Streptococcus mutans . In order to explore the cause of in vivo disease resistance of magnolol and promote its safe application in aquaculture, the pathological detection and changes in immune indicators of fish after feeding with magnolol were conducted in this paper. Results showed that the diets supplemented with magnolol (3 g magnolol/kg commercial feed) significantly increase the expression level of anti-inflammatory cytokines ( IL-10 , TGF-β and IL-4 ) in the liver of goldfish ( p < .05). Additionally, the expression levels of proinflammatory cytokines ( IL-1β , IL-8 and IFN-γ ) did not increase significantly. Subsequently, this study investigated the resistance of goldfish to A. hydrophila and A. veronii infection after feeding with magnolol. The results showed that the survival rates of treatment groups fed 3 g magnolol/kg commercial feed daily increased by 23.1% and 38.5% after 10 days post A. hydrophila and A. veronii ( p = .0351) infection, respectively. Meanwhile, growth performance (body weight and length), major internal organs (liver, spleen, kidney and intestine) and the serum biochemistry indicators (ATL and AST) all exhibited no significant adverse effects after the goldfish fed with magnolol for 30 days. TP showed an increasing concentration in the treatment group ( p < .05). Results of the mRNA expression of stress response indicated that the expression level of cyp1a and hsp70 was significantly down-regulated after a 30-day treatment ( p < .05), and the two genes recovered to the similar level as the control group after a commercial feed diet. In brief, the diets supplemented with magnolol protected the host from the excessive immune response caused by A. hydrophila and A. veronii via enhancing its anti-inflammatory capacity and had no adverse effects with feeding.

848 proteins were commonly identified in five tissues of both DR-GC and GC. • The intestine was identified as the most divergent tissue between DR-GC and GC. • GCHV-induced protein and hemoglobin were critical in defense mechanism of DR-GC. Sustainable aquaculture of grass carp ( Ctenopharyngodon idella , GC) is consistently threatened by bacterial diseases, particularly those caused by Aeromonas veronii . A disease-resistant grass carp (DR-GC) has been developed by backcrossing female gynogenetic GC with normal male GC, exhibiting improved resistance. However, the systemic molecular mechanisms of DR-GC defending against Aeromonas veronii infection remain largely unexplored. Here, a label-free quantitative proteomics approach was employed to systematically compare proteomic profiles across five tissues (intestine, liver, muscle, skin, and kidney) in DR-GC and GC under healthy and infected conditions. The intestine was identified as the central defense tissue, exhibiting the highest number of differentially abundant proteins (DAPs). In DR-GC, A0A3N0YEK7 (small ribosomal subunit protein eS28), A0A3N0YGT8 (ATP synthase-coupling factor 6) and A0A3N0YNS7 (apolipoprotein A-I) were significantly upregulated in intestine, while D5KZW6 (GCHV-induced protein), A0A3N0Z0A1 and Q8JH84 (hemoglobin subunit alpha) were significantly dysregulated across multiple tissues, which playing the critical roles in defense mechanisms at the protein level. Furthermore, cytochrome P450-associated pathways, cytosolic DNA-sensing and RIG-I-like receptor signaling pathways were identified as crucial coordinators mediating immune and metabolic responses. This study provides the first comprehensive proteomic view of multi-tissue defense mechanisms in DR-GC, and identifies key DAPs and pathways for subsequent functional validation. Micropterus salmoides , a freshwater fish of significant economic value, faces substantial challenges due to frequent infection by Aeromonas veronii and excessive antibiotic use. Bacillus tequilensis emerges as a promising antibiotic alternative, enhancing fish immunity and gut health. However, its specific effects on M. salmoides have not been fully clarified. This study examined the impact of different concentrations of B. tequilensis Bt-CO (0, 6.0 × 10 9 , 6.0 × 10 10 , and 6.0 × 10 11 CFU/kg) on juvenile M. salmoides , focusing on growth, survival, body indexes, immune and antioxidant responses, digestive enzymes, disease resistance, intestinal microbiota, and metabolism. Following an 8-week feeding trial, Bt-CO had no significant impact on the growth performance, hepatosomatic index, and condition factor of M. salmoides . However, at a concentration of 6.0 × 10 11 CFU/kg, it significantly enhanced immune indicators, glutathione peroxidase activity, digestive enzyme activities, and post- A. veronii infection survival rates of M. salmoides . Gut microbiota analysis revealed that Bt-CO enhanced beneficial bacteria while reducing harmful ones. Metabolomic analyses indicated that Bt-CO modulated intestinal metabolite levels, including SM(d16:1/24:1(15Z)), d -Threitol, and PC(18:1(9Z)/P-16:0), involving 25 metabolic pathways like d -Glutamine and D-glutamate metabolism. Correlation analysis highlighted significant links between specific gut bacteria and differential metabolites. Overall, this study suggests that incorporating Bt-CO at a concentration of 6.0 × 10 11 CFU/kg in the diet of juvenile M. salmoides could: (i) improve intestinal health by modulating microbial composition and metabolite profile in the intestine; and (ii) enhance fish digestive capacity, immunity, and disease resistance.

Aeromonas veronii is associated with food spoilage and some human diseases, such as diarrhea, gastroenteritis, hemorrhagic septicemia or asymptomatic and even death. This research investigated the mechanism of the growth, biofilm formation, virulence, stress resistance, and spoilage potential of Bacillus subtilis lipopeptide against Aeromonas veronii . Lipopeptides suppressed the transmembrane transport of Aeromonas veronii by changing the cell membrane's permeability, the structure of membrane proteins, and Na + /K + -ATPase. Lipopeptide significantly reduced the activities of succinate dehydrogenase (SDH) and malate dehydrogenase (MDH) by 86.03% and 56.12%, respectively, ultimately slowing Aeromonas veronii growth. Lipopeptides also restrained biofilm formation by inhibiting Aeromonas veronii motivation and extracellular polysaccharide secretion. Lipopeptides downregulated gene transcriptional levels related to the virulence and stress tolerance of Aeromonas veronii . Furthermore, lipopeptides treatment resulted in a considerable decrease in the extracellular protease activity of Aeromonas veronii , which restrained the decomposing of channel catfish flesh. This research provides new insights into lipopeptides for controlling Aeromonas veronii and improving food safety.
