Agastache rugosa (AR), a traditional edible and medicinal herb, is often used for treating gastrointestinal (GI) motility disorder. But little effort has been done on its gastrointestinal motility modulation (GMM) efficacy-related components and quality control of AR. In this study, a novel strategy was proposed to find GMM efficacy-related chemical markers for the quality control of AR. Firstly, network pharmacology and serum pharmacochemistry were applied to predict potential GMM efficacy-related marker components. Secondly, the GMM efficacy-related marker components were verified through literature matching, target isolation/identification and activity evaluation. Lastly, a quantitative analysis of multiple components by a single marker (QAMS)-based method for simultaneous quantification of marker components was established and validated by HPLC–DAD. The results showed that nine components in AR were screened as potential GMM related components, five of which (rosmarinic acid, tilianin, apigenin, acacetin, and cirsimaritin) were matched by literatures, and four (acacetin-7- O -(6''- O -malonyl)- β -D-glucopyranoside, agastachoside, acacetin-7- O -(2''- O -acetyl-6''- O -malonyl)- β -D-glucopyranoside, and isoagastachoside) were chemically identified and newly evaluated on zebrafish model. The nine components were used as marker compounds to develop an effective QAMS-based method for the quantitative evaluation of 26 batches of commercial AR samples.

In present study, a comprehensive strategy integrating multiple chromatographic and chemometric methods to simultaneously characterize the volatile and non-volatile components was developed for the holistic quality evaluation of commercial Agastache rugosa (AR), a common edible and medicinal herb, collected in China. The volatile components and the non-volatile components were characterized by GC-MS and UPLC-QTOF-MS/MS, respectively. And the data were analyzed either independently or integratively by multivariate statistical analysis (MVS) for the quality assessment of commercial samples. The results revealed that the commercial AR samples were different in both the composition and the content of volatile components. However, the compositions of non-volatile components in commercial AR were generally similar, whereas the contents of some components were different. All the results indicated that the holistic quality of commercial AR was inconsistent, and the commercial samples collected could be classified into two main groups, the volatile components were majorly responsible for the classification. Whether or not the holistic quality variations affect the efficacy of AR deserves further investigation.

The abundance of monoterpenoids and phenolic compounds determines the medicinal quality and anti-insect properties of Agastache rugosa , which can be compromised by biotic stress such as herbivore attacks. The traditional use of chemical pesticides to mitigate herbivore interference is increasingly incompatible with sustainable agriculture. In response, nanotechnology-based biostimulants, which can activate metabolic processes to enhance plant growth and stress resistance, offer a more cost-effective and environmentally-friendly alternative. However, to date, it remains unknown how nano-biostimulants improve the therapeutic value and insect resistance of medicinal plants simultaneously. This study investigates the effect of 0–1000 mg/L of a nano-biostimulant salicylic acid functionalized chitosan nanocomposite (SCN) on the pharmacological and anti-herbivore properties of medicinal plant A. rugosa . Results showed that 100 mg/L SCN significantly inhibited Spodoptera litura growth by 62.9 %, and increased plant shoot and root biomass by 107.2 % and 77.6 %, respectively. Moreover, 100 mg/L SCN significantly upregulated the expression of the key genes ( e.g. , LS , L3OH , and CHS ) involved in monoterpene and phenolic compounds biosynthesis by 1.4–10.1 folds, thus boosting the production of active compounds such as pulegone, β-myrcene, and chlorogenic acid by 1.5–24.4 folds. These enhancements were superior to salicylic acid or chitosan alone. Altogether, our findings promote the sustainable and eco-friendly application of nano-biostimulant in improving the quality of medicinal plants and green pest control in agroecosystems.

Agastache rugosa (AR) is a wildly cultivated medicinal and aromatic crop, which usually recommended to be harvested when the plant is exuberant with not flowering or in full flower stage. But there is no scientific basis for the support of the cognition. Therefore, the accumulation patterns of major active components during flower development (inflorescences with buds, full bloom and after full bloom stages) in two chemotypes (menthone type and estragole type) of AR were investigated by newly developed GC–QQQ–MS/MS and UPLC–QTOF–MS/MS methods. Five volatile (pulegone, menthone , estragole, methyleugenol and β -caryophyllene) and nine non-volatile components (rosmarinic acid, tilianin, acacetin 7- O -(6"- O -malonyl)- β - D -glucopyranoside, apigenin , agastachoside, isoagastachoside, acacetin 7- O -(2"- O -acetyl-6"- O -malonyl)- β - D -glucopyranoside, cirsimaritin and acacetin) in two chemotypes of AR samples at three stages were simultaneously quantified. It was found that the relatively higher contents of main volatile components were present in menthone type of AR collected at the full flower stage and in estragole type of AR at the after full bloom stage, whereas the relatively higher contents of main non-volatile components were present in menthone type of AR collected at the inflorescences with buds stage and in estragole type of AR at the after full bloom stage. Considering major active volatile and non-volatile components with higher levels, full bloom stage and after full bloom stage should be optimal harvesting time for menthone type and estragole type of AR, respectively.
