BACKGROUND Chlorantraniliprole (CAP) is a diamide insecticide with high efficacy against many pest insects, including the black cutworm, Agrotis ipsilon . Agrotis ipsilon is a serious pest causing significant yield losses in crops. Glutathione- S -transferases (GSTs) belong to a family of metabolic enzymes that can detoxify a wide range of pesticides. However, little is known about the functions of GSTs in CAP tolerance in A. ipsilon . RESULTS A cDNA sequence (designated AiGSTu1 ) encoding an unclassified GST was identified from A. ipsilon . AiGSTu1 is highly expressed during the 3 rd -instar larval and the pupal stages. Most of the mRNA transcripts were found in larval Malpighian tubules. Exposure to CAP strongly enhanced AiGSTu1 expression, GST activity, hydrogen peroxide (H 2 O 2 ) and malondialdehyde levels in larvae. H 2 O 2 treatment upregulated the transcription level of AiGSTu1 , suggesting that CAP-induced oxidative stress may activate AiGSTu1 expression. The activity of recombinant AiGSTu1 was inhibited by CAP in a dose-dependent manner. Metabolism assay results demonstrated that AiGSTu1 is capable of depleting CAP. Overexpression of AiGSTu1 enhanced the tolerance of Escherichia coli cells to H 2 O 2 and the oxidative stress inducer, cumene hydroperoxide. Silencing of AiGSTu1 by RNA interference increased the susceptibility of A. ipsilon larvae to CAP. CONCLUSION The findings of this study provide valuable insights into the potential role of AiGSTu1 in CAP detoxification and will improve our understanding of CAP tolerance in A. ipsilon . © 2023 Society of Chemical Industry.

The attractiveness of a series of mixtures of (Z)-7-dodecenyl acetate (Z7-12:Ac), (Z)-9-tetradecenyl acetate (Z9-14:Ac), and (Z)-11-hexadecenyl acetate (Z11-16:Ac), the Agrotis ipsilon (Hufnagel) (Lepidoptera: Noctuidae) pheromone, were evaluated in four locations in China. The ternary blend of Z7-12:Ac, Z9-14:Ac, and Z11-16:Ac was the complete pheromone blend for A. ipsilon and the ratio of Z7-12:Ac and Z9-14:Ac was optimal at 3:1. The most attractive ratio of Z11-16:Ac to the other components depended on geographic location. The optimal ratio was 3:1:6 in Yunnan and Shanxi, 3:1:2 in Sichuan and ranged from 3:1:2 to 3:1:12 in Shanghai, which differs significantly from the ratio of 3:1:16 in Japan. The dose of the blend in the pheromone lure influenced attractiveness to male moths and was related to the temperature in the test locations. Attractiveness of sugar–acetic acid-baited and pheromone-baited traps to male and female moths was different before and after the start of flowering of the oilseed rape crop; large numbers of female moths were attracted to sugar–acetic acid traps before flowering but none after flowering had started. This was similar for male moths and there was no synergistic effect when sugar–acetic acid solutions and pheromone were used together. These studies suggest that pheromone trapping based on the blends of three components can be an effective tool to improve the efficiency of monitoring of this pest in China.

In insects, the transcription factor cap ‘n’ collar isoform C (CncC) plays a critical role in the regulation of multiple genes involved in insecticide detoxification. Knockdown of CncC genes leads to increased susceptibility to different types of insecticides in several insect species. However, the CncC gene has not yet been fully characterized in the black cutworm Agrotis ipsilon , a notorious insect pest that causes severe damage to field crops. In this study, the CncC gene (designated AiCncC ) was identified from A. ipsilon . Exposure to a median lethal concentration (LC 50 ) of chlorantraniliprole (CAP) or phoxim (PHO) strongly increased the expression of AiCncC . Silencing of AiCncC by RNA interference significantly increased the susceptibility of A. ipsilon larvae to both insecticides. Moreover, CncC/Maf binding sites were predicted in the putative promoters of two glutathione S -transferase (GST) genes ( AiGSTe1 and AiGSTu1 ) involved in the detoxification of CAP and PHO. The transcription levels of AiGSTe1 and AiGSTu1 were dramatically decreased by silencing AiCncC . These findings indicate that AiCncC is associated with CAP and PHO susceptibility through the regulation of GST genes.

Exposure to insecticides can cause abnormally high accumulation of reactive oxygen species (ROS) and oxidative stress in insects. Thioredoxin peroxidases (TPXs) play critical roles in protecting insects from insecticide-induced oxidative injury. The black cutworm, Agrotis ipsilon (Hufnagel) (Lepidoptera: Noctuidae), is an important agricultural pest. Lambda-cyhalothrin (LCT) is a pyrethroid insecticide used to control A. ipsilon . However, whether LCT causes oxidative stress and whether TPXs are involved in the antioxidant defense in A. ipsilon remain unknown. In this study, a TPX gene, AiTPX6 , was identified from A. ipsilon . AiTPX6 consists of five exons and four introns and encodes a protein of 227 amino acid residues. The AiTPX6 protein has characteristic features of TPXs, including the peroxidase active site motif PVCTTE with a conserved catalytic cysteine residue. Phylogenetic analysis revealed that AiTPX6 belongs to the “1-Cys” TPX group. The mRNA transcripts of AiTPX6 were most abundant in the larval midgut and at the fifth- and sixth-instar larval stages. Exposure to a sublethal concentration (LC 50 ) of LCT significantly increased the H 2 O 2 and malondialdehyde (MDA) concentrations and elevated the mRNA level of AiTPX6 , suggesting that LCT-induced oxidative stress activates AiTPX6 transcription. Recombinant AiTPX6 expressed in Escherichia coli displayed potent antioxidant activity, protecting the cells against oxidative stress. Knockdown of AiTPX6 via RNA interference (RNAi) significantly increased the mortality rate of A. ipsilon following LCT exposure. These findings revealed that AiTPX6 contributes to LCT tolerance via antioxidant defense and could therefore be used as a potential target for RNAi-based control of A. ipsilon .

In insects, heat shock proteins (HSPs) are crucial for protecting against environmental stressors, particularly insecticides. However, in the important agricultural pest Agrotis ipsilon , the specific protective function provided by HSPs against different insecticides remains unclear. Herein, we identified a small HSP (sHSP) gene in A. ipsilon : AisHSP19.6 . Peak AisHSP19.6 expression occurred during the sixth-instar larval stage in the larval midgut. Treatment with phoxim (PHO), lambda-cyhalothrin (LCT), and chlorantraniliprole (CAP) causes oxidative stress in A. ipsilon larvae. Furthermore, PHO, LCT, and CAP exposure, as well as H 2 O 2 injection, significantly elevated AisHSP19.6 transcription. Although AisHSP19.6 did not metabolize PHO, LCT, and CAP, it protected the cells from oxidative stress. Moreover, A. ipsilon larvae in which AisHSP19.6 was silenced using RNA interference (RNAi), exhibited a significantly higher mortality rate in response to PHO, LCT, and CAP exposure relative to AisHSP19.6 -expressing larvae. This study provides new insights into sHSP-mediated insecticide susceptibility in A. ipsilon . These findings also provide a potential RNAi-based strategy for the management of this agricultural pest. Summary Exposure to PHO, LCT, and CAP causes oxidative stress, which activates AisHSP19.6 transcription. AisHSP19. 6 can protect against oxidative stress. AisHSP19.6 knockdown increases susceptibility to PHO, LCT, and CAP.

To better understand the sublethal effects of cyantraniliprole on the black cutworm Agrotis ipsilon (Lepidoptera: Noctuidae), several studies were carried out to investigate sublethal effects on development stages, population parameters, feeding indices and nutrient content of A . ipsilon . The result of a bioassay showed that cyantraniliprole had high toxicity against A . ipsilon fourth-instar larvae with an LC 50 of 0.354 μg.g −1 using an artificial diet. Compared with controls, sublethal doses of cyantraniliprole at LC 5 , LC 20 and LC 40 levels prolonged larval and pupal duration and extended mean generation time and total preovipositional period. In addition, survival rate, reproductive value, intrinsic and finite rates of increase and net reproduction rate declined significantly. Meanwhile, cyantraniliprole had markedly antifeedant effects; decreased the relative growth rate (RGR), the relative consumption rate (RCR), the efficiency of conversion of ingested food (ECI), the efficiency of conversion of digested food (ECD); and increased the approximate digestibility (AD) significantly. This phenomenon contributed to the decrease of nutrient contents, including lipids, protein and carbohydrates, to the point that insufficient energy was available for normal growth. Therefore, sublethal concentrations of cyantraniliprole decreased growth speed and reduced population reproduction of A . ipsilon . This result provides information useful in integrated pest management (IPM) programs for A . ipsilon .
