Microbiota regulation by different Akebia trifoliata fruit juices upon human fecal fermentation in vitro

Sun Yuhao,Hu Yaru,Hu Danqi,Xiao Zhuqian,Wang Hongpeng,Huang Jun,Mao Jianwei

Three different Akebia trifoliata fruit juices were prepared, and their effects on modulation of human fecal microbiota were elucidated through an anaerobic fermentation in vitro. Results indicated that the introduction of inoculatedly-fermented Akebia trifoliata fruit juice promoted short-chain fatty acids productivity. Fecal microbiota analysis demonstrated up-regulations for abundances of Limosilactobacillus , Megamonas , Bifidobacterium , and Escherichia_Shigella , and down-regulations for numbers of Bacteroides , Prevotella_9 , Parasutterella , and Sutterella . Correlation analysis confirmed relationships among sample components, short-chain fatty acids productivity, and microbial abundances, suggesting that sugars and organic acids stimulated growth of Actinobacteriota and suppressed proliferation of Proteobacteria , thus uncovering the underlying mechanism for the better ability of inoculatedly-fermented Akebia trifoliata fruit juice to regulate microbiota structure. Besides, clusters of orthologous groups of proteins analysis indicated that metabolite biosynthesis, energy metabolism, homeostasis maintenance and other physiological functions were ameliorated.

Extraction, moisturizing activity and potential application in skin cream of Akebia trifoliata (Thunb.) Koidz polysaccharide

Xin Li,Jing Wei,Lezhen Lin,Guodong Zheng

Akebia trifoliata (Thunb.) Koidz is gaining popularity, because of its high nutritional, economic and medicinal values. The purpose of this study was to optimize the extraction conditions of A. trifoliata (Thunb.) Koidz polysaccharide (ATKP) and investigate moisturizing activity to prepare a novel moisturizing cosmetic material. It has been observed that using polysaccharide yield as the evaluation criterion showed optimal extraction at a material-liquid ratio of 1:20, an ultrasonic temperature of 80 °C, an ultrasonic time 50 min and ultrasonic power of 175 W. Furthermore, three deproteinization methods of ATKP process, including Sevag, enzymatic hydrolysis and their combination method, were evaluated. The deproteinization efficiency of the Sevag and enzymatic hydrolysis combination method was the highest, with a deproteinization efficiency of 87.52 %. The moisturizing capacity of ATKP was significantly higher than that of chitosan and sodium alginate . ATKP-containing skin cream, particularly the combination of 4.2 % glycerol and 0.8 % ATKP, had an excellent moisturizing effect, and was added to cosmetic formulations to form skin creams. The findings revealed that ATKP has the potential synergistic effect with glycerol in the development of cosmetic formulations, which could be used as a new material for natural moisturizers.

Phytochemical composition and anticancer effect of Akebia trifoliata seed in non-small cell lung cancer A549 cells

Yuanquan Ran,Lanlan Yang,Xiaoyan Jia,Huan Zhao,Qiong Hu,Bing Yang,Dongxin Tang,Minyi Tian

Background Akebia trifoliata is a widely distributed medicine and food homology plant. Its fruit is used to treat tumors and the seed’s weight proportion exceeds 50%. However, the anti-tumor effect of A. trifoliata seed remains poorly studied. Methods UHPLC-Q-Orbitrap-MS was used to identify the chemical components of A. trifoliata seed water extract (WE) and ethanolic extract (EE), and their cytotoxicity was evaluated using MTT assay. Further, colony formation and cell cycle assays examined the anti-proliferative effect of A. trifoliata seed EE. Next, morphology observation, AO/EB staining, Hoechst 33,258 staining, Annexin V-PE/7-AAD staining, and JC-1 assays were employed to detect apoptosis. The influence of A. trifoliata seed EE on A549 cell metastasis was assessed by wound healing and transwell invasion tests. In addition, western blotting and network pharmacology were applied to further analyze its anticancer mechanism. Results Eighty-two components were characterized from the A. trifoliata seed WE and EE. For anticancer activity, A. trifoliata seed EE exhibited high cytotoxicity against cancer cells A549 (IC 50 : 52.38 ± 1.04 μg/mL) and NCI-H1299 (IC 50 : 70.29 ± 0.58 μg/mL) and revealed low cytotoxicity against non-cancer cells MRC-5 (IC 50 : 101.28 ± 2.84 μg/mL) and L929 (IC 50 : 113.44 ± 0.55 μg/mL). A. trifoliata seed EE suppressed A549 cell proliferation by inducing S-phase arrest via down-regulation of CDK2 and p21 as well as up-regulation of cyclin E1 and E2. It increased the Bax/Bcl-2 ratio, reduced mitochondrial membrane potential (ΔΨm), activated caspase-9 and caspase-3, cleaved PARP, and triggered apoptosis through the mitochondrial pathway. Besides, it repressed migratory and invasive ability by reducing MMP-2, MMP-9, and N-cadherin levels. Conclusion A. trifoliata seed EE exhibits outstanding anticancer properties and has the exploitation potential as an anticancer drug in the pharmaceutical industry.

Differential lipidomics sheds light on the great improvement prospect of Akebia trifoliata as a new edible oil crop

Rui Han,Xueying Li,Chen Chen,Jie Li,Xiaoxiao Yi,Huai Yang,Feiquan Tan,Tianheng Ren,Wei Chen,Peigao Luo

Akebia trifoliata has demonstrated large potential as a novel edible oil crop. However, its lipidomic profile remains poorly understood. To explore the dynamics of lipid metabolism and support the agricultural development of this crop, we conducted untargeted liquid chromatography-mass spectrometry analysis on two A. trifoliata genotypes: high-oil content “H543” and low-oil content “L2127.” This approach simultaneously identified 477 lipids in seed oil, which were categorized into 6 classes and 46 subclasses, along with a well-balanced fatty acid composition. Differential lipid analysis revealed 160 and 82 lipid compounds specific to different developmental stages in “L2127” and “H543,” respectively. Furthermore, 139 lipid compounds exhibited significant differential expression between the two genotypes, primarily associated with glycerophospholipid (Ko00564) and glycerolipid (Ko00561) metabolic pathways. Efficient phosphatidic acid/phosphatidylcholine-diacylglycerols-triacylglycerols pathways were identified as key contributors to the high oil content in “H543.” These results provide valuable insights into the metabolic mechanisms underpinning lipid biosynthesis, laying a robust foundation for the utilization of A. trifoliata as an oil crop.

Improved Antioxidant Capacity of Akebia trifoliata Fruit Inoculated Fermentation by Plantilactobacillus plantarum, Mechanism of Anti-Oxidative Stress through Network Pharmacology, Molecular Docking and Experiment Validation by HepG2 Cells

Yuhao Sun,Zhenzhen Wang,Jing Dai,Ruyi Sha,Jianwei Mao,Yangchen Mao,Yanli Cui

In this work, spontaneously fermented and inoculation-fermentedAkebia trifoliatafruit Jiaosu (SFAJ/IFAJ) were compared. The key metabolites and antioxidant activities of SFAJ and IFAJ were tracked and tested during fermentation. The antioxidant effect of fermentedAkebia trifoliatafruit and the underlying mechanisms were explored using network pharmacology for the prediction and verification of the molecular targets and pathways of theAkebia trifoliatafruit’s action against oxidative stress. Furthermore, the results were verified by molecular docking and then investigated, based on a HepG2 cell model. The results of correlation analysis and principal component analysis (PCA) showed that there were significant positive correlations between the phenols, flavonoids, and terpenoids in SFAJ and IFAJ and their antioxidant activities. Network pharmacology and molecular docking analysis disclosed the antioxidation mechanism at the molecular level. In addition, both SFAJ and IFAJ were effective at alleviating oxidative stress in HepG2 cells. In particular, IFAJ performed better than SFAJ in protecting cells with an intracellular reactive oxygen species (ROS) level of 99.96 ± 4.07 U/mg prot, superoxide dismutase (SOD) activity of 41.56 ± 0.06 U/mg prot, catalase (CAT) activity of 91.78 ± 3.85 U/mg prot, and glutathione peroxidase (GSH-Px) activity of 39.32 ± 2.75 mU/mg prot in the IFAJ group. Collectively, this study revealed the changes in bioactive metabolite contents and the in vitro antioxidant activity during fermentation and investigated the protectiveness of SFAJ and IFAJ against oxidative stress within HepG2 cells, promoting the study of the antioxidant efficacy of IFAJ, thereby providing valuable reference data for the optimization of its preparation and the development of relevant products with health benefits. Keywords:Akebia trifoliatafruit;bioactive metabolites;antioxidant activity;network pharmacology;molecular docking;oxidative stress
