Co-biodegradation of cyanide and phenol by Alcaligenes faecalis JF101: Investigating interaction effects

Qingyun Li,Shaohui Yan,Zhengfeng Jiang,Liang Xian,Jianjia Du,Haibo Liu,Youyan Liu

Cyanide and phenol are highly toxic chemicals that exert severe inhibitory effects on microorganisms, posing significant challenges for their removal by biological methods in coexistent systems. In this study, we successfully isolated a bacterial strain named JF101, identified as Alcaligenes faecalis , which was capable of degrading both cyanide and phenol. The interactions between cyanide and phenol during biodegradation were investigated. It was found that the presence of 100 mg/L cyanide significantly enhanced the degradation of phenol, leading to the complete degradation of 1000 mg/L phenol within 86 h. In contrast, only 21.9% of phenol was degraded in the absence of cyanide. Additionally, the presence of phenol had minimal impact on cyanide degradation; a concentration of 100 mg/L cyanide could be rapidly and completely degraded within 40 min even in the presence of up to 1000 mg/L phenol. Five empirical kinetic models of Haldane, Aiba, Edward, Yano, and Webb were employed to describe the degradation processes of phenol and cyanide in both single-substrate and dual-substrate systems. The putative degrading enzymes, degradation products, and pathways associated with the co-degradation of cyanide and phenol were systematically analyzed. The formamide and NH 4 + -N produced from the rapid degradation of cyanide served as nitrogen sources, facilitating the resumed growth of strain JF101 and enhancing phenol degradation.

Ali Mohsin,Jingyun Sun,Imran Mahmood Khan,Haifeng Hang,Muhammad Tariq,Xiwei Tian,Waqas Ahmed,Sobia Niazi,Yingping Zhuang,Ju Chu,Muhammad Zubair Mohsin,Salim-ur-Rehman,Meijin Guo

This study presents an engineered approach for sustainable biosynthesis of curdlan by Alcaligenes faecalis using orange peels. To confirm the substrate suitability a four step study was organized. Firstly, drying of substrate was carried within temperature range of 60–120 °C, along with the application of moisture diffusion control model. Secondly, fermentation medium was obtained via saccharification and detoxification, releasing highest sugar at 72.34 g/L with phenolics removal of 95–98%. Thirdly, curdlan fermentation was conducted in detoxified orange peel hydrolysate followed by optimization of batch culture fermentation via kinetic modeling using Logistic and Luedeking-Piret equations. In 5 L bioreactor, highest specific growth rate ( μ m  = 0.233/h), highest curdlan production ( P m = 23.24 g/L) and growth associated rate constant ( α  = 3.403) were achieved. Moreover, the total sugar consumption and conversion rates were 83.27% and 53.20%. Lastly, characterization techniques such as FTIR, NMR, XRD, TGA, HPGPC and EDS were applied to biosynthesized curdlan for qualitative validation.

Microbial Degradation of Nicotinamide by a Strain Alcaligenes sp. P156

Hu Chunhui,Zhao Shuxue,Li Kuiran,Yu Hao

A novel Alcaligenes sp. strain P156, which can utilize nicotinamide as its sole source of carbon, nitrogen and energy, was enriched and isolated from soil in a solid waste treatment plant. Aerobic growth and degradation with nicotinamide were characterized. Seven nicotinamide degradation-related genes were obtained by sequence alignment from the genome sequence of strain P156. Four genes, designated naaA , naaD , naaE and naaF , were cloned and heterologously expressed. Nicotinamide degradation is initiated by deamination to form nicotinic acid catalyzed by the nicotinamidase NaaA, which shares highest amino acid sequence identity (27.2%) with nicotinamidase from Arabidopsis thaliana . Nicotinic acid is converted to 6-hydroxynicotinic acid, which is further oxidized to 2,5-dihydroxypyridine (2,5-DHP). 2,5-DHP is then transformed to a ring-cleavage product, N -formylmaleamic acid, by an Fe 2+ dependent dioxygenase NaaD. N -formylmaleamic acid is transformed to fumaric acid through maleamic acid and maleic acid by NaaE and NaaF, respectively. To our knowledge, this is the first report of the complete microbial degradation of nicotinamide in bacteria. Nicotinamide is considered as a model compound for the study of microbial degradation of pyridinic compounds, and the nicotinamide degrading related genes in strain P156 were distributed differently from the reported similar gene clusters. Therefore, this study contribute to the knowledge on the degradation of pyridinic compounds.

Effects of carbon nanotube on denitrification performance of Alcaligenes sp. TB: Promotion of electron generation, transportation and consumption

Zeyu Wang,Cong Chen,Huan Liu,Dzmitry Hrynshpan,Tatsiana Savitskaya,Jianmeng Chen,Jun Chen

Multi-walled carbon nanotubes (MWCNTs) promote biodegradation in water treatment , but the effect of MWCNT on denitrification under aerobic conditions is still unclear. This investigation focused on the denitrification performance of MWCNT and its toxic effects on Alcaligenes sp. TB which showed that 30 mg/L MWCNTs increased NO 3 - removal efficiency from 84% to 100% and decreased the NO 2 - and N 2 O accumulation rates by 36% and 17.5%, respectively. Nitrite reductase and nitrous oxide reductase activities were further increased by 19.5% and 7.5%, respectively. The mechanism demonstrated that electron generation (NADH yield) and electron transportation system activity increased by 14.5% and 104%, respectively. Cell membrane analysis found that MWCNT caused an increase in polyunsaturated fatty acids, which had positive effects on electron transportation and membrane fluidity at a low concentration of 96 mg/kg but caused membrane lipid peroxidation and impaired membrane integrity at a high concentration of 115 mg/L. These findings confirmed that MWCNT affects the activity of Alcaligenes sp. TB and consequently enhances denitrification performance.

Improvement of Alcaligenes sp.TB performance by Fe-Pd/multi-walled carbon nanotubes: Enriched denitrification pathways and accelerated electron transport

Zeyu Wang,Luyao Dai,Jiachao Yao,Tianjiao Guo,Dzmitry Hrynsphan,Savitskaya Tatsiana,Jun Chen

Aiming at the accumulation of NO 2 − -N and N 2 O during denitrification process, this work focused to improve the denitrification performance by Pd-Fe embedded multi-walled carbon nanotubes (MWCNTs). The main conclusions were as follows: 30 mg/L Pd-Fe/MWCNTs have shown an excellent promotion on denitrification (completely TN removal at 36 h). Meanwhile, enzyme activity results indicated that the generation of NO 2 − -N, NH 4 + -N by Pd-Fe/MWCNTs led the occur of short-cut denitrification by increasing 203.9% the nitrite reductase activity. Furthermore, electrochemical results and index correlation analysis confirmed that the electron exchange capacity (1.401 mmol eg −1 ) of Pd-Fe/MWCNTs was positively related to nitrite reductase activity, indicating its crucial role in electron transport activity (0.46 μg O 2 /(protein·min) at 24 h) during denitrification process by Pd-Fe/MWCNTs played a role of chemical reductant and redox mediator.

Co-Treatment with Single and Ternary Mixture Gas of Dimethyl Sulfide, Propanethiol, and Toluene by a Macrokinetic Analysis in a Biotrickling Filter Seeded with Alcaligenes sp. SY1 and Pseudomonas Putida S1

Yiming Sun,Xiaowei Lin,Shaodong Zhu,Jianmeng Chen,Yi He,Yao Shi,Hua Liu,Lei Qin

The biotrickling filter (BTF) treatment is an effective way of dealing with air pollution caused by volatile organic compounds (VOCs). However, this approach is typically used for single VOCs treatment but not for the mixtures of VOC and volatile organic sulfur compounds (VOSCs), even if they are often encountered in industrial applications. Therefore, we investigated the performance of BTF for single and ternary mixture gas of dimethyl sulfide (DMS), propanethiol, and toluene, respectively. Results showed that the co-treatment enhanced the removal efficiency of toluene, but not of dimethyl sulfide or propanethiol. Maximum removal rates (rmax) of DMS, propanethiol and toluene were calculated to be 256.41 g·m−3·h−1, 204.08 g·m−3·h−1and 90.91 g·m−3·h−1, respectively. For a gas mixture of these three constituents, rmaxwas measured to be 114.94 g·m−3·h−1, 104.17 g·m−3·h−1and 99.01 g·m−3·h−1, separately. Illumina MiSeq sequencing analysis further indicated that Proteobacteria and Bacteroidetes were the major bacterial groups in BTF packing materials. A shift of bacterial community structure was observed during the biodegradation process. Keywords:biotrickling filter;decomposition;dimethyl sulfide;pollution;propanethiol;toluene

Enhanced denitrification performance of Alcaligenes sp. TB by Pd stimulating to produce membrane adaptation mechanism coupled with nanoscale zero-valent iron

Zeyu Wang,Cong Chen,Huan Liu,Dzmitry Hrynshpan,Tatsiana Savitskaya,Jianmeng Chen,Jun Chen

The microbial reduction of nitrate in the presence of nanoscale zero-valent iron (nZVI) was evaluated to assess the feasibility of employing nZVI for biological denitrification treatment. The effect of modified nZVI on the growth, metabolism, and denitrification performance of Alcaligenes sp. TB under aerobic conditions was studied. Results showed that Alcaligenes sp. TB with nZVI/Pd had 31.5% increase in nitrate removal and 18.1% decrease in nitrite accumulation within 28 h. nZVI/Pd exhibited less inhibition on the cell growth (OD 600  = 0.725), NADH/NAD + ratio (86% of control), and electron transfer system activity (68.5% of control). In addition, nZVI/Pd decreased the membrane fluidity by increasing the trans/cis isomerization ratio (317.7% of control) to enhance the resistance of nZVI. This study underlines the effects of nZVI/Pd on membrane susceptibility via membrane fatty acid transformation during denitrification and suggests the influence of engineered nanomaterials on denitrification.

Remediation of arsenic and polycyclic aromatic hydrocarbon contaminated soils using electro-microbial combined remediation: effects of soil physicochemical properties

Jiang Chengchen,Zhou Shuai,Shu Xiaohua,Yu Guo,Zeng Honghu,Zhu Zongqiang,Yang Chen,Dang Zhi

Composite pollution poses a significant challenge for soil remediation. This study simulated arsenic and PAH-contaminated soil in karst regions to evaluate the effectiveness of electro-bioremediation under varying soil conditions. Results showed that Alcaligenes faecalis subsp. faecalis exhibited strong PAH degradation and moderate As(V) reduction capabilities. PAH removal efficiency was maximized (55.93%), while arsenic removal efficiency declined as pH increased. Higher soil organic carbon and available nitrogen reduced arsenic removal, whereas increased available phosphorus enhanced it. Under optimized conditions (35 g/kg organic carbon, 150 mg/kg nitrogen, 50 mg/kg phosphorus), residual arsenic was minimized (118.80 mg/kg), and PAH removal peaked at 45.47%. These findings underscore the pivotal role of soil parameters in remediation efficiency, offering practical insights for optimizing electro-bioremediation strategies for soil contaminated by arsenic and PAH.

Catabolic characterization of dipicolinic acid in Alcaligenes faecalis strain JQ135

Yang Mu,Fuyin Zhang,Ning Li,Shanshan Pi,Ang Li,Qing Hong,Jian He,Jiguo Qiu,Jiandong Jiang

Dipicolinic acid (DPA) is a widely distributed pyridine derivative containing two carboxylate groups. However, the catabolism of DPA mediated by microorganisms are rarely reported. Alcaligenes faecalis strain JQ135, previously isolated in our lab, was able to degrade various pyridine derivatives, including nicotinic acid (NA). In this study, it was found that strain JQ135 could utilize DPA as the sole carbon and nitrogen source for growth and degrade 1.8 mM (300 mg/L) DPA to a non-detectable level within 12 h. The catabolism of DPA by strain JQ135 extracted two intermediates, i.e., 3-hydroxy-2,6-dipicolinic acid and 4-imino-pent-2-enedioic acid. Moreover, deletion of the key genes essential for NA catabolism (such as moaCDEBmoeA and maiA ) did not affect DPA catabolism, indicating that the maleamate pathway and molybdenum cofactor are not involved in the DPA catabolism by strain JQ135. This study will provide insights into the bacterial catabolism of DPA.

Enhanced degradation of arsanilic acid and in situ recovery of inorganic arsenic in a two-stage bioelectrochemical process

Shi Lin,Zhan Cetao,Bai Wenjing,Wang Wei,Yuan Shoujun,Hu Zhen-Hu

Arsanilic acid ( p -ASA), an organoarsenic additive found in livestock wastewater, can release toxic inorganic arsenic into the environment. While bioelectrochemical systems have proven effective in decomposing organoarsenics, managing the resulting inorganic arsenic remains a challenge. This study demonstrated the feasibility of a two-stage bioelectrochemical process designed to facilitate p -ASA degradation and in situ recover inorganic arsenic from contaminated livestock wastewater. It consisted of two sequential stages: (I) anodic stimulation for p -ASA degradation and (II) reversing electrode polarities for the cathodic reduction of inorganic arsenic. In Stage I, the anode significantly enhanced the degradation of p -ASA, resulting in 18 μg/L of As(III) and 700 μg/L of As(V) released into the bulk solution. In Stage II, the cathode further reduced the As(III) and As(V) to 8.9 and 35.5 μg/L, respectively, through the synergistic action of the cathode and suspended microbes. The inorganic arsenic was recovered as a layer of As(V)-O on the cathode. Microbial analysis indicated that Alcaligenes was responsible for the degradation of p -ASA, while Anaerobacillus and Desulfitibacter played key roles in reducing As(V) and As(III) on the cathode, respectively. This study provided a promising alternative approach for the removal of organoarsenics and in situ recovery of inorganic arsenic from organoarsenic-bearing wastewater.

Physiological response and molecular mechanism of glyphosate degradation by Pseudomonas alcaligenes Z1–1

Zhimei Zhang,Chao Zhao,Tianming Wu,Yanfeng Xu,Lu Wang,Yusheng Niu

The accumulation of glyphosate during its application in agriculture and its toxicity seriously threaten ecosystems and human health. Currently, glyphosate residual contamination is mainly accomplished through bioremediation techniques based on enhanced microbial degradation activity. However, there are drawbacks, such as poor environmental adaptability of strains and low degradation efficiency. Therefore, in this study, an efficient glyphosate-degrading strain, Pseudomonas alcaligenes Z1–1, was isolated from herbicide-contaminated environments and was capable of completely degrading glyphosate at a concentration of 200 mg/L within 7 days. Kinetics analysis showed that glyphosate degradation was concentration-dependent, with a maximum tolerant concentration of 800 mg/L. Mass spectrometric analysis indicated that AminoMethylPhosphonic acid (AMPA) was the predominant intermediate produced in the degradation pathway of glyphosate, revealing that glyphosate destruction began with breaking the C-N bond. Whole genome sequencing identified the key genes potentially involved in glyphosate degradation, including the thiO , glpA , aroA , soxB , and argA genes. Furthermore, in contrast to the majority of the metabolic pathways previously reported for glyphosate degradation via glyphosate oxidoreductase , the breaking of the C-N bond was primarily catalyzed by glycine oxidase . Overall, this research provides novel insights into the mechanisms of glyphosate degradation, offering valuable degradation enzyme resources for future applications.

Beyond Enzymatic Pathways: Contribution of Chemical Reactions to Hydroxylamine Oxidation during Heterotrophic Nitrification by Alcaligenes faecalis

Su Qingxian,Zhi Mei,Zhong Yijie,Wu Jiapeng,Smets Barth F.,Ng How Yong

The heterotrophic nitrifier Alcaligenes faecalis oxidizes ammonium (NH 4 + ) to dinitrogen gas (N 2 ) via the key intermediate hydroxylamine (NH 2 OH), accompanied by the formation of nitrite (NO 2 – ), nitric oxide (NO), and nitrous oxide (N 2 O). Although NH 4 + and NH 2 OH oxidation by A. faecalis were previously attributed to the direct nitrogen formation ( dnf ) gene cluster, recent evidence suggests that NH 2 OH oxidation to N 2 proceeds through chemical reactions. However, the exact mechanism and relative importance of chemical reactions remain unknown. Here, we characterized the formation of N intermediates in batch incubations of A. faecalis and determined the kinetics of NH 2 OH-mediated chemical reactions in abiotic tests under varying C/N ratios, oxygen, pH, and matrix conditions. Combined with transcriptomic analyses, we resolved the respective enzymatic and chemical N turnover pathways across cultivation conditions. Distinct formation and removal pathways of N intermediates were observed: NH 2 OH was oxidized via both biotic and abiotic processes, with abiotic contribution accounting for 23–64% of NH 2 OH removal under environmentally relevant conditions (C/N = 8–16, 21% O 2 , pH 7) and increasing to 35–86% at elevated O 2 (50%) and alkaline pH (9). NO 2 – formation was exclusively enzymatic, whereas N 2 O was produced predominantly through chemical reactions of NH 2 OH with NO. These findings advance our understanding of inorganic N metabolism of A. faecalis and highlight the previously underestimated role of abiotic reactions in N loss.
