Aleuroglyphus ovatus, a common mite harming stored grain, poses a threat to human health. Paeonol is the main component isolated from peony roots and exhibits various medicinal values. In our previous study, we found that paeonol shows strong acaricidal activity. In the present study, the acaricidal mechanism of paeonol against A. ovatus was uncovered through RNA-seq transcriptomics and TMT proteomics. According to the transcriptomics results, paeonol treatment noticeably up-/downregulated genes associated with “oxidoreductase activity” (peptidase activity, ATPase activity, and hydrolase activity). As shown by proteomic analysis, concordant changes were identified in proteins linked with hydrolase activity, serine-type peptidase activity, antioxidant activity, and oxidoreductase activity. Paeonol target proteins were FMO, ACAD8, GPX, CTSL, and PRSS1, as determined by transcriptomics-proteomics correlation analysis. We propose that paeonol may play an acaricidal role by interfering with enzyme activities in A. ovatus, especially serine-type peptidase, thereby causing an imbalance in amino acid ratios and eventually leading to the death of A. ovatus. However, the specific mechanism of action needs to be further studied. In addition, we verified the key enzymes at the transcriptional level.

This study was designed to evaluate the acaricidal effect and the mode of action of lemongrass essential oil (LEO) against Aleuroglyphus ovatus. Contact and fumigant mortality indicated that LEO had higher acaricidal activities at a low concentration, and the acaricidal action of LEO was significantly more effective by contact effects than by fumigant bioassay. Repellent rate against A. ovatus at different concentrations of LEO increased with application time. Observation on the toxic symptoms revealed that almost all mites were in immobilized type (IM type) in 30 min, but the symptom of knock-down type (KD type) appeared later. Cuticles of mites killed by LEO exhibited obvious colour change and deformation. Detoxification enzyme activity assay showed that glutathione S-transferase (GST) activity increased in mites treated by LEO compared to the controls. LEO increased catalase (CAT) activity of A. ovatus but inhibited superoxide dismutase (SOD) after 20 h. LEO also increased nitric oxide synthetase (NOS) and inhibited acetylcholine esterase (AChE) activities. Moreover, LEO acted as a fast-acting neurotoxin on A. ovatus. Our results suggest that LEO has an acaricidal potential and is a promising agent in control of A. ovatus.

Rosemary (Rosmarinus officinalis L) essential oil (REO) has an inhibitory effect against pests and is regarded as a safe alternative to synthetic acaricides. This study investigated the acaricidal and repellent activity of REO and underlying mechanisms against Aleuroglyphus ovatus, an important mite pest of stored products. Gas Chromatography-Mass Spectrometry analysis of REO indicated that the compounds of 1,8-cineole (25.20%) was the highest. REO at 13% concentration provided the highest repellent effect (92.67%) on A. ovatus, and the lowest repellent activity (48.67%) was at 0.8% concentration. Microscopic observations revealed that the mite epidermis started to shrink after treatment with REO at 6 h post-treatment. The enzyme level of catalase (CAT), acetylcholinesterase (AChE) and superoxide dismutase (SOD) decreased significantly (P < 0.05), whereas there was a significant increase (P < 0.05) in nitric oxide synthase (NOS) and glutathione-s-transferases (GST) activities in A. ovatus treated with REO. In the molecular docking study, camphor and 1,8-cineole, the main monoterpenes of REO, bind more closely with CAT and AChE, and PI-conjugated interaction, hydrogen bonding, and hydrophobic were found by molecular docking 3D model. Overall, our results indicate the potential of REO as an effective botanical acaricide to manage A. ovatus.

Aleuroglyphus ovatus (Acari: Acaridae) is a major pest mite of stored grains that is distributed worldwide. Paeonol, a phenolic component of the essential oil extracted from the Chinese herb Paeonia moutan , possesses a range of biological activities, including antiviral, antifungal and acaricidal activity. This study investigated the bioactivity of paeonol against A. ovatus and its effect on the activity of detoxification enzymes. The bioactivity of paeonol against A. ovatus was determined by contact, fumigation and repellency bioassays, and the mechanism was preliminarily explored via morphological observation of the color changes of mite epidermis and determination of the changing trend of some important enzymes associated with acaricidal efficacy in the mites. The results showed that the median lethal concentration (LC 50 ) in the contact and fumigation bioassays was 9.832 μg/cm 2 and 14.827 μg/cm 3 , respectively, and the acaricidal activity of paeonol was higher under direct contact than under fumigation. Dynamic symptomatology studies registered typical neurotoxicity symptoms including excitation, convulsion and paralysis in A. ovatus treated with paeonol. The enzyme activity of catalase (CAT), nitric oxide synthase (NOS) and glutathione-S-transferase (GST) was higher, whereas the activity of superoxide dismutase (SOD) and acetylcholinesterase (AChE) was lower, compared to the control group. CAT, NOS and GST were activated, whereas SOD and AChE activities were inhibited after paeonol intervention. Our findings suggest paeonol has potent acaricidal activity against A. ovatus and thus may be used as an agent to control the stored-product mite A. ovatus .
