A new adsorbent based on aptamer functionalized magnetic hydrophobic polymer (MHbPA) was developed for specific and efficient adsorption of alternariol (AOH). Through the synergistic effect of aptamer-AOH affinity and hydrophobic interaction of polymer, enhanced adsorption properties had been realized, in which AOH aptamer was the first selected through capture-SELEX with good specificity and affinity, and the targeting polymer was designed based on the hydrophobicity of AOH to increase the interaction. The proposed MHbPA demonstrated a high adsorption capacity of 187.6 ng/mg for AOH. The adsorption behavior was considered as Langmuir adsorption model and pseudo-secondary kinetic adsorption model. Notably, the adsorption of AOH in wheat powder samples could be accomplished within 10 mins with acceptable recoveries. The as designed adsorbent with synergistic effect provides new insights into the development of enhanced pretreatment materials for mycotoxin monitoring in complex food matrices with specific aptamer and targeting polymer.

A simple method based on surface-enhanced Raman scattering (SERS) was developed for the rapid determination of alternariol (AOH) in pear fruits using an easily prepared silver-nanoparticle (AgNP) substrate. The AgNP substrate was modified by pyridine to circumvent the weak affinity of the AOH molecules to the silver surface and to improve the sensitivity of detection. Quantitative analysis was performed in AOH solutions at concentrations ranging from 3.16 to 316.0 μg/L, and the limit of detection was 1.30 μg/L. The novel method was also applied to the detection of AOH residues in pear fruits purchased from the market and in pear fruits that were artificially inoculated with Alternaria alternata. AOH was not found in any of the fresh fruit, whereas it resided in the rotten and inoculated fruits. Finally, the SERS method was cross validated against HPLC. It was revealed that the SERS method has great potential utility in the rapid detection of AOH in pear fruits and other agricultural products.

Alternariol (AOH) is a metabolic product of Alternaria , which has various toxic effects on humans and animals and its pollution is increasingly being discovered in fruits, vegetables, and grains. At present, the detection of AOH is mainly achieved through high-performance liquid chromatography-mass spectrometry and immunological methods. However, there is currently a lack of fast detection methods for on-site use. Considering the unique advantages of immunological testing methods, this study developed an immunolateral chromatographic quantitative method for detecting AOH using quantum dot fluorescent nanospheres (QDFNs) as signal amplification materials. The detection range of AOH is 5.2–4798 μg/kg, 50 % inhibiting concentration (IC 50 ) is 156.4 μg/kg, limit of detection (LOD) value is 2.32 μg/kg, and limit of quantification (LOQ) value is 6.96 μg/kg. The recovery rates of AOH in tomato and strawberry were 95.27%–111.63% and 88.43%–96.22%, respectively. In brief, the developed approach has good accuracy and repeatability and can be used for the detection of AOH.

Alternariol (AOH), a secondary metabolites of the genus Alternaria , is a genotoxic mycotoxin. Because the previous “Mannich antigen” for AOH readily underwent unstable induction, a new AOH hapten was designed for inducing a high-quality antibody against AOH using a conjugate of the AOH carboxymethyl derivative with a carrier protein as the immunogen. A competitive indirect chemiluminescence enzyme immunoassay (ciCLEIA) for AOH was optimized and showed good sensitivity (limit of detection 0.068 ng/mL), a linear range (0.11–1.23 ng/mL), and negligible cross-reactivity with analogues. Average rates of recovery from spiked samples (juice and cereal) were between 72.7% and 115.8%, with coefficients of variation <14.3%. Results from ciCLEIA correlated well with those of high-performance liquid chromatography coupled with tandem mass spectrometry, meaning the proposed method might be an effective screening tool for detection of AOH in food samples.

Alternariol (AOH) is a secondary toxin produced by the metabolism of Alternaria, which poses a significant food safety risk due to its widespread presence in fruits and vegetables. In this study, a dual-mode sensor based on AOH aptamer was developed, which combined with surface-enhanced Raman scattering (SERS) and fluorescence (FL) detection technology to achieve sensitive and specific detection of AOH. The sensor significantly improves the reliability of detection through the synergistic change and mutual verification of SERS and FL signals. The linear detection range of SERS mode was 0.5–100 ng/mL ( R 2 = 0.9927), and the detection limit was as low as 0.064 ng/mL. The linear range of FL mode was 1–100 ng/mL ( R 2 = 0.9792), and the detection limit was 0.14 ng/mL. Experiments showed that the sensor could effectively distinguish AOH from other mycotoxins, and showed good recovery (SERS: 84.09 % -101.40 %, FL: 4.69 % -105.57 %) and precision (RSD < 6.3 % and 8.9 %, respectively) in complex matrices such as kiwifruit, tomato and citrus. The results were consistent with those of the traditional LC-MS method, which verified its reliability. The sensing platform constructed in this study not only provides a reliable technical means for the trace detection of AOH in agricultural products, but also opens up a new way for the rapid screening and quantitative analysis of other mycotoxins in food matrices.

The lack of rapid, user-friendly methods for alternariol (AOH), a prevalent emerging mycotoxin, presents a considerable analytical challenge. This work introduces an integrated sensing strategy advancing from classical homogeneous liquid-phase detection to a portable AI-enhanced paper-based biosensor. The system employed a Cu/Mn-CeO 2 nanozyme as signal generator and amplifier, and exploited the inhibitory effect of AOH on acetylcholinesterase (AChE) to suppress the thiocholine (TCh)-mediated reduction of oxidized 3,3′,5,5′-tetramethylbenzidine (oxTMB), enabling quantitative colorimetric readout. It achieved ultra-sensitive detection in the liquid phase with a limit of detection (LOD) of 0.016 pg/mL. By incorporating an aptamer affinity column for sample cleanup, the assay was transferred to a foldable paper platform. An AI-driven Monte Carlo color analysis method addressed issues of color heterogeneity and subjective interpretation, yielding an LOD of 0.12 μg/kg in wheat. This study provides a point-of-need solution for AOH screening and establishes a generalizable framework for detecting contaminants in complex samples.

Alternariol (AOH) is an emerging mycotoxin widely found in agricultural products. It has drawn attention due to the potential health risks associated with long-term dietary exposure. Therefore, a ratiometric fluorescent aptamer sensor based on PCN-224 was developed. By combining the intrinsic fluorescence properties of PCN-224 with aptamer recognition, sensitive detection of AOH was achieved. PCN-224 served as both a fluorescence platform and a source of internal reference signals, enabling self-calibrated ratio analysis. Under optimal conditions, the sensor exhibited good linear response in the range of 0.001–500 ng/mL. The LOD was 0.449 pg/mL. It also exhibited good selectivity, stability, and reproducibility. This sensor has been successfully applied to the detection of AOH in cereal samples, demonstrating its reliability in the analysis of complex food matrices. This work provided an effective strategy for monitoring mycotoxins by combining MOF fluorescence regulation with aptamer recognition.
