Throughout the history of traditional Chinese medicine, Angelica pubescens has been extensively employed to treat inflammatory arthritis. However, little is known about the anti-rheumatoid arthritis (RA) effects of A. pubescens polysaccharides. In this study, four crude polysaccharides (APR50, APR70, APR90, and APRB) were isolated from A. pubescens radix, and the rat model of collagen-induced arthritis (CIA) was established to evaluate the anti-RA effects of A. pubescens polysaccharides. A. pubescens polysaccharides could effectively reduce the degree of paw swelling in CIA rats, among which the effect of APR50 was comparable to that of methotrexate. A homogenous polysaccharide APRP50-2-1 was purified from APR50. Structural characterization indicated that APRP50-2-1 was primarily composed of glucose, mannose, galactose, and arabinose. The main skeleton of APRP50-2-1 consisted of →4)- α - d -Glc p -(1→, →6)- α - d -Glc p -(1→, and →6,4)- α - d -Gal p -(1→ residues. The branched chains of →4)- α - d -Glc p -(1→ and →3)- β - d -Man p -(1→ were substituted at O -4 of →4,6)- α - d -Gal p -(1→. APRP50-2-1 inhibited the proliferation and promoted the apoptosis of MH7A cells by down-regulating the expression of the anti-apoptotic protein B-cell lymphoma-2 (BCL-2) and up-regulating the expression of the pro-apoptotic protein BCL2-associated X (BAX). In conclusion, A. pubescens polysaccharides had significant anti-RA effects in rat CIA model, among which APR50 and APRP50-2-1 possessed the promising prospects of ameliorating RA.

Simple SummaryPlutella xylostellais one of the most destructive insect pests affecting cruciferous vegetables. The use of various plant extracts for controlling this pest is increasing in popularity, as they are multi-bioactive, biodegradable, and ecologically safe. In this study, we evaluated the insecticidal oviposition deterrence and repellent activities ofAngelica pubescensextract and its compounds againstP. xylostella. TheA. pubescensextract, caryophyllene oxide, and 3,4-dimethoxycinnamic acid, have been proven to have insecticidal and oviposition deterrent activities againstP. xylostella. TheA. pubescensextract and caryophyllene oxide also mediateP. xylostellarepellence. Overall, our results highlighted the potential ofA. pubescensextract, caryophyllene oxide, and 3,4-dimethoxycinnamic acid in controllingP. xylostellaand in the development of effective eco−friendly “inhibit and kill” insecticide formulations.AbstractPlutella xylostellaL. is a destructive pest affecting cruciferous vegetables, causing massive economic losses worldwide. Plant−based insecticides are considered promising insect control agents. TheAngelica pubescensextract inhibited female oviposition, with an oviposition deterrence index (ODI) of 61.65% at 12.5 mg/mL. We aimed to identify the bioactive compounds inA. pubescensextract. The compounds fromA. pubescensextract were analyzed using LC−MS techniques. The toxicity and behavioral responses of larvae and adults ofP. xylostellato ten compounds were investigated. We found that the caryophyllene oxide and 3,4-dimethoxycinnamic acid inhibited female oviposition; the ODIs were 98.31% and 97.59% at 1.25 mg/mL, respectively. TheA. pubescensextract, caryophyllene oxide, and 3,4-dimethoxycinnamic acid caused larval mortality, with LC50values of 21.31, 4.56, and 5.52 mg/mL, respectively. The EAG response of females was higher than that of males underA. pubescensextract conditions, while the EAG response of males was higher than that of females in caryophyllene oxide and 3,4-dimethoxycinnamic acid conditions. TheA. pubescensextract and caryophyllene oxide showed repellent activity against both female and male adults, while the 3,4-dimethoxycinnamic acid did not elicit any notable behavioral responses fromP. xylostellaadults.A. pubescensextract and caryophyllene oxide are potential insecticides, oviposition deterrents, and behavioral regulators againstP. xylostella, and they could be potential candidates for the development of biological insecticides to controlP. xylostella. Keywords:Plutella xylostella;Angelica pubescens;extract/compounds;toxicity;repellent effect

Immunomodulation plays a key role in maintaining organismal health, and precise molecular mechanisms that control immune activation are key to achieving this balance. Angelica pubescens is in high demand in the Chinese herbal medicine market due to its effective properties in dispelling dampness and relieving pain. Nevertheless, the immunological activity of A. pubescens has not been fully elucidated. This study focuses on exploring the structural characterization and immunomodulatory activities of two homogeneous arabinans (APRP70-1-1 and APRP70-1-2) isolated from A. pubescens . Structural analysis revealed that APRP70-1-1 possesses a higher degree of branching compared to APRP70-1-2. In vitro experiments showed that despite activating macrophages, APRP70-1-2 was less effective than APRP70-1-1 at promoting nitric oxide (NO) release. APRP70-1-1 demonstrated superior immunomodulatory effects, as evidenced by its significantly enhancing macrophage phagocytic capacity and upregulation of tumor necrosis factor -α ( TNF-α ), interleukin -1β ( IL-1β ), and IL-6 mRNA expression. Mechanistic studies indicated that APRP70-1-1 activates the JAK2/STAT3 and NF-κB signaling pathways. In summary, these results indicate that APRP70-1-1 had immunomodulatory activity.

A novel heteropolysaccharide with about twenty sugar residues named DF80-2 was obtained from Angelica pubescens Maxim. f. biserrata Shan et Yuan root, one of the most widely used traditional Chinese medicines for thousands of years in China. The possible structure of DF80-2 was proposed considering the comprehensive results of physicochemical properties, methylation analysis, and 1D/2D NMR spectroscopy, which showed that its main chain was composed of (1→3)-, and (1→4)-linked-α- d -Glc p , (1→4)-linked-β- d -Gal p , (1→6)-linked-α- d -Man p , and (1→3)-linked-α- l -Ara f , and the branch was present as the α- d -Glc p -(1→3)-β- d -Gal p A disaccharides stretched from O -6 position of (1→4)-linked-α- d -Glc moiety in the main chain. Congo red analysis, scanning electron microscopy and atomic force microscopy showed that DF80-2 possessed a triple helical conformation, and its branched monomers were interlaced with one another forming a regular network structure. DF80-2 exhibited antioxidant activity by effectively scavenging DPPH, and hydroxyl radicals, and chelating ferrous ions.
