Anoectochilus roxburghii (AR) has been used in food, medicine and ornamental industries for a long time. Anion exchange resin was proposed to purify the sub-fraction of water-extracted AR polysaccharide (ARPP-70), and a homogeneous polysaccharide ARPP-70a was obtained. The structural features of ARPP-70a were characterized using gas chromatography–mass spectrometry (GC–MS), nuclear magnetic resonance (NMR) spectroscopy, and high performance size exclusion chromatograph coupled with multi-angle laser light scattering (HPSEC-MALLS). The relative weight average molecular weight for ARPP-70a was determined to be 14.8 kDa, and the molar ratio of glucose to galactose was 1.0:3.2. The structure of ARPP-70a was elucidated to be glucogalactan, with backbone comprising β-1,4-linked Gal p and some α-1,4-linked Glc p . The conformation characteristics of ARPP-70a were supposed to exist as a random coil chain in 0.1 M NaNO 3 solution. Moreover, in vitro antioxidant activity assays revealed ARPP-70a exhibited appreciable antioxidant potential. To the best of our knowledge, this is the first study to obtain this type of glucogalactan, and provide systematic information on its structural and conformational properties. This study improved the understanding of the physicochemical characteristics of AR polysaccharide, which is beneficial for its further application in food and medicinal industry.

Anoectochilus roxburghii is a traditional herb in China that can be potentially used to treat diabetes. A novel polysaccharide ARLP-W was isolated from Anoectochilus roxburghii by chromatography on DEAE-52 cellulose. Chemical analysis indicated that ARLP-W (8.1 × 10 4 Da) was mainly composed of mannose and glucose. The main linkages of glycosidic bonds of ARLP-W were β-1, 4-Manp and α-1, 4-Glcp. The terminal Glcp was connected to Manp-via O-3. RT-qPCR and western blotting analysis showed that ARLP-W caused a significant reduction in the levels of the key gluconeogenesis enzymes phosphoenolpyruvate carboxykinase (PEPCK) and glucose 6-phosphatase (G6Pase) in the liver. The results of the insulin resistance tests indicated that ARLP-W increased glucose absorption. These results indicate that ARLP-W has a good therapeutic effect on type 2 diabetes and can assist with further development and application treatment of diabetes.

Objective Anoectochilus roxburghii is a valuable medicinal and ornamental plant. The aim of this study is to investigate the morphological and biochemical responses during the flower development stages of A. roxburghii , and to assess the effects of exogenous polyamines (PAs) on bud differentiation and metabolism, thereby providing a theoretical basis for understanding the flowering form and physiology of A. roxburghii . Methods In this study, morphological and biochemical responses in flower development stages of A. roxburghii were investigated using paraffin sections and stereomicroscope. A. roxburghii was divided into five periods, including vegetative growth period, flower bud period, flowering period, late flowering period and fruiting period. During the flowering phase, specific biochemical parameters were measured, including soluble sugar content, superoxide dismutase (SOD) activity, soluble protein content, peroxidase (POD) activity, and catalase (CAT) activity. These measurements were conducted to understand the biochemical changes occurring within A. roxburghii during its flowering process. Furthermore, the effects of PAs on bud differentiation were examined. Additionally, the activities of S-adenosylmethionine decarboxylase (SAMDC) and polyamine oxidase (PAO), as well as the content of polyphenols, polysaccharides, and flavonoids in A. roxburghii , were measured after PA treatment to evaluate the metabolic changes induced by exogenous PAs. Results During the flowering phase of A. roxburghii , soluble sugar content and SOD activity were steadily declining. Soluble protein content was initially increasing and then reducing, and POD and CAT activities showed opposite pattern. In addition, the effects of exogenous PAs on bud differentiation were investigated. Results showed that 3 mmol/L putrescine or 0.3 mmol/L spermidine significantly promoted the bud differentiation of A. roxburghii and advanced the flowering. The activities of SAMDC, PAO, and the content of polyphenols, polysaccharides and flavonoids in A. roxburghii significantly increased after PA treatment, demonstrating that exogenous PA can accelerate metabolism and improved the active ingredients content. Conclusion The flower development of A. roxburghii was divided into five stages, with significant changes in soluble sugar, protein, POD, SOD, CAT, MDA, and PRO levels. Exogenous putrescine and spermidine enhanced bud differentiation and accelerated flowering, increasing SAMDC and PAO activities, suggesting accelerated PA metabolism. PAs also improved active component content. These findings provide a theoretical basis for studying flower morphology and PA-induced flowering regulation of A. roxburghii .

Light quality affects plant leaf growth and its metabolism of amino acids and polyphenols. Anoectochilus roxburghii has been used for food , medicine, and ornamental purposes. To obtain the optimal light quality for improved accumulation of metabolites in A. roxburghii in vitro . The seedings were cultured under the white light (W), blue light (B), red light (R), red: blue light = 1:2 (R1B2), red: blue light = 1:1 (R1B1), red: blue light = 2:1 (R2B1). Twenty compounds in raw leaves were in situ rapidly identified within 20 s by neutral desorption-extractive electrospray ionization mass spectrometry (ND-EESI-MS) without any sample pretreatment, including eight amino acids , eleven polyphenols, and kinsenoside. Meanwhile, leaf area and the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were evaluated. Our results showed that leaf area and antioxidant enzyme activities of the R2B1 were significantly higher than B or R group. The red-blue light increased the content of isoleucine , histidine , serine , arginine, and promoted the accumulation of caffeic acid , ferulic acid , quercetin , myricetin , kaempferol , and rutin in phenylpropane metabolism, especially in the R2B1, which could be used as the optimal light quality. ND-EESI-MS could enhance our understanding of the effect of light quality on the accumulation of amino acids and polyphenols in A. roxburghii .

Anoectochilus roxburghii (Wall.) Lindl. is a rare and precious Chinese herbal medicine with medicinal, tonic and ornamental functions. With the wide application of A. roxburghii in the fields of medical care and health care, people's quality requirements are also increasing. Therefore, the aim of this study was to investigate the effects of drying methods on sensory qualities, aroma components, and bioactive compounds of A. roxburghii . Plants underwent one of five drying methods, namely natural drying (ND), hot air drying (HD), microwave drying (MD), vacuum drying (VD), and freeze-drying (FD). We used the spectral data of GC–MS, E-nose, and E-tongue for the first time to evaluate the aromas and flavors of A. roxburghii . The results showed that the FD method is ideal for maintaining the natural appearance, but the HD and MD methods are appropriate for preserving the polysaccharide content and enhancing flavors, respectively. However, the VD (60 ± 2℃) method may be the best option for drying A. roxburghii plants because it can preserve the original color and appearance of the plants and also results in a desirable aroma. Most importantly, it maintains the active ingredient contents of A. roxburghii plants.

CompoundAnoectochilus roxburghii(Wall.) Lindl. (A. roxburghii) oral liquid (CAROL) is a hospital preparation ofA. roxburghiiandGanoderma lucidum(G. lucidum),which have hepatoprotective effects. Eight active components (five nucleosides/nucleobases and three triterpenoid acids) in CAROL,A. roxburghii, andG. lucidumwere simultaneously detected by high-performance liquid chromatography–tandem mass spectrometry (LC–MS/MS). The multiple reaction monitoring (MRM) mode was applied for the detection of analytes. These eight compounds were separated well within 12 min and quantified using the internal standard working curve method. The method showed good linearity (R2> 0.9935) and high sensitivity (limit of detection = 0.29 ng/mL). The analyte recovery ranged from 85.07% to 97.50% (relative standard deviation < 3.31%). The content of the target analytes in four batches of CAROL, and the raw materials ofG. lucidumandA. roxburghiifrom the five regions was determined using this method. The contents of guanosine and ganoderic acid A in four batches of oral liquid were high and stabilized and could be recommended as quality markers (Q-marker) for CAROL. Simultaneous qualitative and quantitative analysis of nucleosides and triterpenoid acids in CAROL,A. roxburghii, andG. lucidumby LC–MS/MS based on the MRM model was reported for the first time. The proposed method provides a sensitive, rapid, and reliable approach for the quality control of Chinese medicinal products.Keywords:LC–MS/MS;Anoectochilus roxburghii(Wall.) Medicinal plants have been widely used in the treatment of various diseases for human health. We developed a novel method for the identification of closely related medicinal plants using a machine learning (ML)-based electrochemical fingerprinting platform. Firstly, the system featured a bare glassy carbon electrode capable of recording the voltammetric response of active components in medicinal plants as electrochemical fingerprints. Subsequently, different algorithms and various datasets were employed to analyze the correlation between the above electrochemical fingerprint data and the medicinal plant species . As a proof-of-concept, 6 species of Anoectochilus roxburghii ( A. roxburghii ) were selected as the verification samples. The electrochemical fingerprints of the samples were measured by differential pulse voltammetry in two buffer solutions. Thereafter, four powerful ML algorithms were utilized for the identification of A. roxburghii with different datasets. The results showed that the accuracy of identifying species reached 94.4 % by the nonlinear support vector machines based on the slope data of electrochemical responses in two buffer solutions, evidencing the successful discrimination of closely related medical plants by this method. Additionally, ML combined with electrochemical fingerprinting approaches had the advantages of being rapid, affordable, and straightforward, which provided potential applications in pharmaceutical research and plant taxonomy.

This study develops a thyme essential oil (TEO)-based bio-fungicide, encapsulated in amino-functionalized mesoporous silica nanoparticles (AMSNs), and evaluates its antifungal efficacy. In vitro , the TEO@AMSNs formulation demonstrated 93.23 % mycelial growth inhibition at 3.84 mg/mL, outperformed free TEO (67.77 % inhibition), with a 13.44 % loading rate. Mechanistic studies indicated that this efficacy results from continuous and strong membrane disruption and high reactive oxygen species (ROS) overaccumulation in fungal cells. Density functional theory (DFT) calculations confirmed that the stable release profiles was driven by hydrogen bonding interactions between the AMSNs carrier and the essential oil. In vivo trials on Anoectochilus roxburghii (A. roxburghii) , a medicinal crop susceptible to F. oxysporum , showed that TEO@AMSNs significantly reduced disease symptoms and promoted root development. Inoculated plants treated with TEO@AMSNs at 2 MIC exhibited nearly normal lateral root counts (∼42) compared to 23 in untreated controls. Microscopic analyses revealed intact chloroplasts and preserved polysaccharide levels in treated plants, suggesting reduced oxidative stress and metabolic disruption. Flavonoid accumulation was also enhanced, further supporting the formulation's role in mitigating pathogen-induced stress. These findings highlight TEO@AMSNs as a promising, sustainable bio-fungicide for managing Fusarium infections and enhancing crop resilience.

As a specialty food that is medicinal and edible, Anoectochilus roxburghii contains many active ingredients. Herein, optimal conditions to preserve A. roxburghii samples and obtain high physiological activity have been studied. The contents of polysaccharides , kinsenoside, and flavonoids of A. roxburghii were monitored during post-harvest storage for a period of eight months at 25 °C with different packaging methods such as stacking, multilayer pocket and vacuum. The results indicated that packaging method and storage time have significant influences on the preservation of active compounds. Polysaccharides, kinsenoside and flavonoids exhibited a general descending trend during storage for a period of eight months in all packaging conditions. However, the vacuum method is more effective in preservation of polysaccharides and flavonoids than stacking and multilayer pockets methods due to the lower activity of microbial community under vacuum condition. Interestingly, kinsenoside, a characteristic compound of A. roxburghii , responsive for the hypoglycemic function, was not affected by packaging methods. This is because kinsenoside was mainly affected by plant endogenous enzymes instead of microbial community. Two-factor variance analysis showed that packaging method and storage time have significant interactions in preservation. It was further disclosed that the changes of active components were closely related to the plant endogenous enzymes, microbial community and their functions. The findings in this study will shed some light on the storage of A. roxburghii and other herb products.

Anoectochilus roxburghii , a famous Chinese herbal medicine, has been commonly used for the treatment of liver disease, diabetes, and rheumatoid arthritis. Our study aimed to investigate the anti-rheumatoid arthritis effects of A . roxburghii polysaccharides (ARP), using the rat's model of type II collagen-induced arthritis (CIA). ARP was prepared by alcohol sedimentation and structurally characterized based on combined chemical, chromatographic and spectroscopic methods. High Performance Size Exclusion Chromatography-Multiangle Laser Light Scattering-Refrative Index (HPSEC–MALLS–RI) analysis revealed that ARP includes two peaks, and the weight-average molecular weight ( Mw ) of the principal one was estimated as 5.90 kDa with a relative content of 98.2%. Pharmacological results exhibited that ARP significantly decreased the arthritis index and ameliorated the inflammatory cell infiltration and the synovial tissue destruction in CIA rats. Additionally, ARP possessed significant NO production inhibitory effects and antioxidant activity. Further anti-inflammatory mechanism investigations indicated that ARP significantly inhibited the activation of nuclear factor κB (NF-κB) pathway by suppressing the phosphorylation of IκB and p65, which subsequently down-regulated the mRNA expressions of IL-1β and IL-6 in LPS-stimulated RAW 264.7 cells. These findings suggested that ARP has great potential in the development of functional foods and dietary supplements for the treatment of rheumatoid arthritis.

The aim of this study was to investigate the effects of light quality on the morphological traits, leaf anatomical characteristics, antioxidant enzyme (superoxide dismutase, catalase, and peroxidase) activities, photosynthetic pigments content, and bioactive compounds (phenols, flavonoids, and polysaccharides) content in Anoectochilus roxburghii. Plants of A. roxburghii were grown under light filtered through four differently colored films for 8 months. The four treatments were red film (RF), blue film (BF), yellow film (YF), and colorless plastic film (control, CK). Compared with the A. roxburghii plants in CK, those in the BF treatment showed significantly greater stem diameter, fresh weight, leaf area, stomatal frequency, chlorophyll content (Chl a, Chl b, Chl a+b), antioxidant enzyme activities, and active compound (polysaccharides, flavones) content. The plants in the RF treatment showed the greatest plant height and phenolics contents. These results show that growing A. roxburghii plants under blue film is a useful technique to improve quality. This technique is conducive to achieving large-scale sustainable production of high-quality plant materials.
