CompoundAnoectochilus roxburghii(Wall.) Lindl. (A. roxburghii) oral liquid (CAROL) is a hospital preparation ofA. roxburghiiandGanoderma lucidum(G. lucidum),which have hepatoprotective effects. Eight active components (five nucleosides/nucleobases and three triterpenoid acids) in CAROL,A. roxburghii, andG. lucidumwere simultaneously detected by high-performance liquid chromatography–tandem mass spectrometry (LC–MS/MS). The multiple reaction monitoring (MRM) mode was applied for the detection of analytes. These eight compounds were separated well within 12 min and quantified using the internal standard working curve method. The method showed good linearity (R2> 0.9935) and high sensitivity (limit of detection = 0.29 ng/mL). The analyte recovery ranged from 85.07% to 97.50% (relative standard deviation < 3.31%). The content of the target analytes in four batches of CAROL, and the raw materials ofG. lucidumandA. roxburghiifrom the five regions was determined using this method. The contents of guanosine and ganoderic acid A in four batches of oral liquid were high and stabilized and could be recommended as quality markers (Q-marker) for CAROL. Simultaneous qualitative and quantitative analysis of nucleosides and triterpenoid acids in CAROL,A. roxburghii, andG. lucidumby LC–MS/MS based on the MRM model was reported for the first time. The proposed method provides a sensitive, rapid, and reliable approach for the quality control of Chinese medicinal products.Keywords:LC–MS/MS;Anoectochilus roxburghii(Wall.) Anoectochilus roxburghii (Wall.) Lindl. is a rare and precious Chinese herbal medicine with medicinal, tonic and ornamental functions. With the wide application of A. roxburghii in the fields of medical care and health care, people's quality requirements are also increasing. Therefore, the aim of this study was to investigate the effects of drying methods on sensory qualities, aroma components, and bioactive compounds of A. roxburghii . Plants underwent one of five drying methods, namely natural drying (ND), hot air drying (HD), microwave drying (MD), vacuum drying (VD), and freeze-drying (FD). We used the spectral data of GC–MS, E-nose, and E-tongue for the first time to evaluate the aromas and flavors of A. roxburghii . The results showed that the FD method is ideal for maintaining the natural appearance, but the HD and MD methods are appropriate for preserving the polysaccharide content and enhancing flavors, respectively. However, the VD (60 ± 2℃) method may be the best option for drying A. roxburghii plants because it can preserve the original color and appearance of the plants and also results in a desirable aroma. Most importantly, it maintains the active ingredient contents of A. roxburghii plants.

A novel, simple, and efficacious analytical method for determining of nucleotides in Anoectochilus roxburghii (Wall.) Lindl ( A. roxburghii ) was developed. Magnetic dispersive solid-phase micro-extraction (MDSPME) combined with high-performance liquid chromatography was applied for extraction and determination of three nucleotides, such as adenosine-5ʹ-monophosphate (AMP), uridine-5ʹ-monophosphate (UMP) and guanosine-5ʹ-monophosphate (GMP) in A. roxburghii from different sources. The structure and morphology of magnetic nanoparticles, Fe 3 O 4 @GO, were illustrated by Fourier-transform infrared (FTIR), X-ray diffraction (XRD), transmission electron microscopy (TEM), and Thermagravimetric analysis (TGA) techniques. The effects of different extraction conditions on extraction efficiency were investigated and optimized. The optimum extraction conditions were performed as follows: 40.0 mg Fe 3 O 4 @GO were dispersed in 30 mL adsorption solution (pH 3.50, 2 μg/mL), 50 mM NaOH was employed for elution with 12 min of ultra-sonication at 40 °C. Under the aforementioned extraction conditions, the Fe 3 O 4 @GO nano-adsorbent obtained an excellent adsorption property. The corresponding linearity range for all three analytes exhibited a good linearity (r 2 ≥ 0.9982) and notable added recoveries ranging from 88.4% to 109.8%, whereas the limit of quantitation was between 0.8–8 ng/mL. The enrichment factors (EF) were between 174 and 255. The proposed method showed the advantages of full purification, high EF, simplicity, and good recovery. The method was also successfully applied to nucleotides extraction and determination in A. roxburghii, showing superior reproducibility and high sensitivity. Based on this, the method could be expected to provide a novel experimental means and developmental direction for improving pretreatment and purification of nucleotides, reducing matrix effects as much as possible, in traditional Chinese medicinal materials or biological specimens.

Luteolin is one of the bioactive components from the compound Anoectochilus roxburghii (Wall.) Lindl. oral liquid (CAROL), which was reported to have excellent hepatoprotective and anti-inflammatory activities. However, the enrichment and quantitation of luteolin from CAROL is challenging due to the low content and complex aqueous matrix. In this study, a bifunctional monomer surface molecularly imprinted polymer (MIP) with metal–organic frameworks (MOFs) as cores was prepared for the selective adsorption of luteolin from the aqueous system CAROL. Compared with conventional MIPs, this unique nanocomposite adsorbent (MOF@MIPs) has the advantages of short kinetic equilibrium time, good selectivity, and high adsorption capacity in aqueous solution. The theoretical maximum adsorption capacity of MOF@MIPs for luteolin was 36.99 mg/g. After adsorption enrichment of luteolin from CAROL using MOF@MIPs, liquid chromatography–tandem mass spectrometry was applied to analyze the target. The corresponding linearity range for analyte was 10–6000 ng/mL with good linearity (R 2 =0.9992), and the added recoveries varied from 85.70% to 99.25%. The present method has been successfully employed for the analysis of luteolin in five different batches of CAROL. Notably, we found no significant difference in the content of luteolin between these batches, which proved that the composition was stable between batches. The novel structure MIPs are suitable for the specific recognition of template molecules in aqueous solution. Therefore, this study provides a technical reference for the special identification and determination of trace components in complex samples, while the novel MOF@MIP nanocomposite can also provide valuable references for the extraction and purification methods of specific substances in traditional Chinese medicine and expand the application environment of MIPs material.

The analysis of plant growth regulators presents a challenge due to their trace quantities and complex matrices. A novel, simple, and effective analytical method for the determination of three trace acidic plant growth regulators in Anoectochilus roxburghii (Wall.) Lindl was developed to address this issue. Three-phase hollow fiber liquid-phase microextraction combined with high-performance liquid chromatography was applied for the enrichment, purification, and determination of three acidic plant growth regulators, namely, indole-3-acetic-acid, indole-3-butyric-acid, and (+)-abscisic acid. The factors affecting extraction performance, including extractant species, pH of donor and acceptor phases, salt addition dosage, extraction time, temperature, and stirring rate, were investigated and optimized. Under optimum conditions, the proposed method provided good linearity ( R 2 , 0.9994–0.9999), low limit of detection (0.038–0.12 ng/mL), and acceptable relative recoveries (56.7–117.6%). The enrichment factors were between 153 and 328. The developed method was successfully applied to the enrichment and determination of plant growth regulators in Anoectochilus roxburghii (Wall.) Lindl and exhibited increased purification capacity, higher sensitivity, and decreased organic solvent consumption compared with conventional sample preparation methods. This method may provide a testing platform for the monitoring of plant growth regulator residues, ensuring the safe and effective use of traditional Chinese medicine.
